  • 본 연구는 육성돈에 있어 인분해효소인 phytase의 첨가가 환경친화성사료개발을 위한 기초자료로 사용하기 위해 영양소 이용률을 측정하였다. 대조구는 옥수수-대두박의 육성돈 사료이고, 처리구는 대조구 사료에 Phytase의 첨가 수준을 500 unit 및 1,000 unit을 첨가한 구로 3×3 Latin Square 방법으로 실시하였다. 건물과 질소에 대한 소화율에서는 각 처리구간에 유의적인 차이가 없었다(P>0.05). 회분, 칼슘 및 인의 소화율의 경우 phytase 첨가구가 대조구에 비해 높은 수준을 보였다(P<0.05). 돼지가 섭취한 인의 양은 phytase 첨가구가 대조구에 비해 증가하였다. 또한 인의 흡수량에서 phytase 첨가구가 대조구에 비해 높았다(P<0.05) 분과 뇨의 총 인 배설량은 유의적인 차이를 나타내지 않았다. 칼슘의 섭취량은 처리구가 대조구에 비해 높았다. 칼슘의 분 배설량은 각 처리구간에 별 다른 차이를 보이지 않았으나(P>0.05), 뇨의 배설량은 처리구가 대조구보다 적은 양이 배설되었다. 소화된 아미노산 중 cystine, isoleucine, tyrosine, phenylalanine은 처리구가 대조구에 비해 소화율이 현저히 증가된 것을 알 수 있었다(P<0.05). Asparagine, threonine, serine, methionine, leucine의 경우 phytase를 500 unit 첨가한 구가 높은 소화율을 나타내었다(P<0.05). 한편 lysine, histidine, arginine 등의 다른 아미노산은 처리구가 대조구보다 약간의 소화율 증가를 보였으나 통계적 유의차는 나타나지 않았다(P>0.05). 결론적으로 인분해효소 phytase를 사료내 첨가하므로서 분중 영양소를 줄일 수 있는 환경친화성 사료를 개발 가능하다고 사료된다. 체결홈이 형성된 체결부를 갖는 안경집과 이것에 결합되는 체결구가 마련되어 개폐되는 커버가 형성된 케이스로 구성된다.로 금형설계 및 제작기간을 단축하고자 한다.ere demonstrated in rats after i.c.v., intraperitoneally and orally administration, respectively.   • Disposal repository for high-level radioactive waste secures its safety by means of engineered and natural barriers. The performance of these barriers should be tested and verified through various aspects in terms of short and/or long-term. KAERI has been conducting various in-situ demonstrations in KURT (KAERI Underground Research Tunnel). After completing previous experiment, a conceptual design of an improved in-situ experiment, i.e. K-COIN (KURT experiment of THMC COupled and INteraction), was established and detailed planning for the experiment is underway. Preliminary characterizations were conducted in KURT for siting a K-COIN test site. 15 boreholes with a depth of about 20 m were drilled in three research galleries in KURT and intact rock specimens were prepared for laboratory tests. Using the specimens, physical measurements, uniaxial compression, indirect tension, and triaxial compression tests were conducted. As a result, specific gravity, porosity, elastic wave velocities, uniaxial compressive strength, Young's modulus, Poisson's ratio, Brazilian tensile strength, cohesion, and internal friction angle were estimated. Statistical analyses revealed that there did not exist meaningful differences in intact rock properties according to the drilled sites and the depth. Judging from the uniaxial compressive strength, which is one of the most important properties, all the specimens were classified as very strong rock so that mechanical safety was secured in all the regions.

A Study on the Occurrence of Benzo(a)pyrene in Fats and oils by Heat Treatment(I) (유지가열시 Benzo(a)pyrene 생성에 관한 연구(I) -대두유가열시-)

  • Benzo(a)pyrene [B(a)P], one of the polycyclic aromatic hydrocarbons (PAHs) is known as a potent carcinogen. As lipid consumption increases recently, the toxic effect of overheated lipid foods and fats & oils were reported increasingly. In this study, the contents of B(a)p, other PAHs and rancidities of soybean oil were determined, and then the proper heating temperature, time and frequency were recommended, The work was carried out using soybean oil heated at $180\pm$5^{\circ}C$$, $200\pm$5^{\circ}C$$, and $300\pm$5^{\circ}C$$,for 50 hours. Acid Value(AV) and Conjugated Diene Value of samples were determined. The contents of B(a)P and other PAHs contents of all samples were masured by HPLC/UV method. The results obtained were as follows; Each content of PAHs in the fresh soybean oil was: Pyr 1.093, B(a)A 0.986, Ch 1.147, DMBA 1.082, B(e)P 0.664, Per 1.135, B(a)P 0.146, DBA 1.053, 3-MC 0.05 rg/kg. When the soybean oil was heated at $180\pm$5^{\circ}C$$, for 10, 20, 30, and 50 hours, B(a)P conterlts in heated soybean oils were 0.391, 0.692, 0.451, and 0.372 $\mu\textrm{g}$/kg respectively. Acid value of them were 0.26, 0.26, 0.29, and 0.33, and conjugated diene value was 0.67, 0.76, 0.99, and 1.04, respectively. When the soybean oil was heated at $200\pm$5^{\circ}C$$,and $300\pm$5^{\circ}C$$,for 10, 20, 30, and 50 hours, B(a)P coiltents in soybean oil heated at $200\pm$5^{\circ}C$$,were 0.844, 0.512, 0.479 and 0.247 Ig/kg respectively, Acid value 0.22, 0.21, 0.23 and 0.51 and CDNV 0.39, 0.49, 3.27, and 3.89. B(a)P contents in soybean oil heated at $300\pm$5^{\circ}C$$,were 0.466, 0.706, 0.607 and 0.247$\mu\textrm{g}$/kg respectively, Acid value 0.47, 1.57, 3.90, and 6.42 and CDNV 0.65, 2.15, 3.00, and 3.88.

Cultivation of Lactic Acid Bacteria for the Development of Probiotic Products using Red Ginseng Starch (프로바이오틱스 개발을 위한 홍삼 전분을 활용한 유산균 배양)

  • To reduce the production cost of Lactobacillus, discarded red ginseng starch was collected from a factory of red ginseng extract in order to develop the Lactobacillus culture medium. According to the analysis of the gensenoside composition of red ginseng starch, the total gensenoside content of starch was 2.73 mg/g, and the gensenoside $Rb_1$, $Rb_2$ and $Rg_3$ contents were 0.1, 0.29 and 0.52 mg/g, respectively. For the preparation of the liquid media, red ginseng starch was added at rates of 0, 5, 10 and 20%. Further, Lactobacillus plantarum 15357 and Leuconostoc mesenteroides sub sp. strains were then inoculated to these prepared broths. With the red ginseng starch medium, the growth rates ($1.42{\times}10^7$ and $2.96{\times}10^{10}$ CFU/mL) and the final cell concentrations were higher than the MM medium ($1.0{\times}10^7$ CFU/mL). The optimal concentration of red ginseng starch and yeast extract as a medium were 20% and 10 g/L, respectively. Under these conditions, the cell mass of L. plantarum 15357 and L. mesenteroides sub sp. reached $5.11{\times}10^{10}$ and $8.17{\times}10^{10}$ CFU/mL. These results show a great possibility for the utilization of red ginseng starch as economic medium sources in the production of cell mass of lactic acid bacteria. This is the first trial of development of economic LAB growth medium using discarded red ginseng starch.

  • 한국천문연구원은 차세대소형위성 1호의 근적외선 영상분광기 NISS (Near-infrared Imaging Spectrometer for Star formation history) 탑재체를 개발하여 2017년 6월 30일에 최종 비행모델을 납품하였고, 이 발표는 탑재체 NISS 구조체의 비행모델 개발 결과를 보고한다. NISS는 0.9 - 2.5um (R~20) 근적외선 파장에서 관측을 해야 하기 때문에, 구조체의 배경잡음을 없애기 위해서 200K까지 passive cooling으로 냉각되며, H2RG 검출기는 소형 냉동기에 의해 약 88K에서 운영된다. NISS 구조체의 passive cooling을 효율적으로 수행하기 위해서 방열판, Kevlar 지지대, MLI, 표면제어용 필름 등을 조립하였고, 실제 지상 시험을 통해서 그 성능을 확인하였다. NISS 구조체는 최종 시스템 조립 과정에서 전자부 하네스 조립을 함께 수행했으며, 온도 모니터링 센서를 부착하고 소형 냉동기 피드백 온도를 반복 시험을 통해서 결정하였다. NISS 구조체는 미러 및 렌즈를 지지하는 광기계부를 함께 포함하기 때문에 발사 및 우주환경에서 광학 성능을 유지하기 위한 설계를 거쳐서 제작 되었으며, 최종 시스템 검교정 시험, 진동 및 열진공 시험을 통해서 그 성능을 확인하였다. NISS를 탑재한 차세대소형위성 1호는 2018년 상반기에 미국의 Falcon 9 발사체에 실려서 발사될 예정이다.

Composition of Saponin and Free Sugar of Some White Ginsengs with Processing Conditions (가공방법에 따른 몇가지 백삼 제품의 사포닌 및 유리당 조성 변화)

  • To analyse components of fresh ginseng and some white ginsengs with different processing conditions, approximate composition, extraction yield, total saponin content and composition and free sugar composition of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were examined. Yield of hot water extraction was two times higher than that of 80% methanol extraction. Hot water extraction yields of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were 56.4, 39.9, 42.9 and 46.6%, respectively, while the 80% methanol extraction yields ranged from 15.8% to 21.9%. Total saponin contents of the above were 2.40, 1.73, 1.45 and 1.79%, respectively, with hot water extraction and were 2.15, 2.99, 2.81 and 2.35%, respectively, with 80% methanol extraction. Ginsenoside compositions of the above varied with processing conditions and extraction solvents. Hot water and 80% methanol extracts of fresh and white ginseng composed of fructose, glucose, sucrose and maltose. Rhamnose was detected only in the extract of Taeguksam A and B.

  • Objective: Quantitative polymerase chain reaction (qPCR) has been extensively used in the field of mesenchymal stem cell (MSC) research to elucidate their characteristics and clinical potential by normalization of target genes against reference genes (RGs), which are believed to be stably expressed irrespective of various experimental conditions. However, the expression of RGs is also variable depending on the experimental conditions, which may lead to false or contradictory conclusions upon normalization. Due to the current lack of information for a clear list of stable RGs in bovine MSCs, we conducted this study to identify suitable RGs in bovine MSCs. Methods: The cycle threshold values of ten traditionally used RGs (18S ribosomal RNA [18S], beta-2-microglobulin [B2M], H2A histone family, member Z [H2A], peptidylprolyl isomerase A [PPIA], ribosomal protein 4 [RPL4], succinate dehydrogenase complex, subunit A [SDHA], beta actin [ACTB], glyceraldehyde-3-phosphate dehydrogenase [GAPDH], TATA box binding protein [TBP], and hypoxanthine phosphoribosyltrasnfrase1 [HPRT1]) in bovine bone marrow-derived MSCs (bBMMSCs) were validated for their stabilities using three types of RG evaluation algorithms (geNorm, Normfinder, and Bestkeeper). The effect of validated RGs was then verified by normalization of lineage-specific genes (fatty acid binding protein 4 [FABP4] and osteonectin [ON]) expressions during differentiations of bBMMSCs or POU class 5 homeobox 1 (OCT4) expression between bBMMSCs and dermal skins. Results: Based on the results obtained for the three most stable RGs from geNorm (TBP, RPL4, and H2A), Normfinder (TBP, RPL4, and SDHA), and Bestkeeper (TBP, RPL4, and SDHA), it was comprehensively determined that TBP and RPL4 were the most stable RGs in bBMMSCs. However, traditional RGs were suggested to be the least stable (18S) or moderately stable (GAPDH and ACTB) in bBMMSCs. Normalization of FABP4 or ON against TBP, RPL4, and 18S presented significant differences during differentiation of bBMMSCs. However, although significantly low expression of OCT4 was detected in dermal skins compared to that in bBMMSCs when TBP and RPL4 were used in normalization, normalization against 18S exhibited no significance. Conclusion: This study proposes that TBP and RPL4 were suitable as stable RGs for qPCR study in bovine MSCs.

13 weeks repeated oral dose toxicity studies with LMK02-Jangwonhwan in SD rats (LMK02의 Sprague-Dawley 랫드를 이용한 13 주간 반복 경구투여 독성시험)

  • Objectives : The oriental medicine Jangwonhwan, a boiled extract of 12 medicinal herbs/mushrooms, has been prescribed to patients with cognitive dysfunction, as originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan (LMK02-Jangwonhwan) consisting of seven medicinal plants/mushrooms, was shown to reduce the ${\beta}$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer's disease. The toxicity of LMK02-Jangwonhwan was investigated in SD rats, by a daily oral administration for 13 weeks and NOAEL(No observed adverse effect dose), a definite toxic dose and target organ, as well. Methods : Quality control of the tablet form of LMK02-Jangwonhwan was established by estimating the indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02-Jangwonhwan was investigated in 6 week old, specific pathogen free (SPF), Sprageu-Dawley rats by oral administration. Each test group consisted of 10 male and 10 female rats. The groups received doses of 500, 1,000 or 2,000 mg/kg/day of test substance for 13 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of the control group. Results : The 13-week repeated oral treatment doses didn't result in any specific symptoms or death. There were no significant changes in the rat's weight and food consumption. Further, ophthalmic examination, urinalysis, hematological, serum biochemistry test and organ weight revealed no significant differences. Conclusions : The no-observed-adverse-effect level(NOAEL) of LMK02 for male and female Sprague-Dawley rats was determined as 2,000mg/kg/day and the target organ wasn't confirmed. Because no significant adverse effects were observed, the target organ could not be determined.

Each test group were consist of 5 male and 5 female and they received doses of 500, 1,000 and 2,000 mg/kg/day of test substance for 4 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of control group. Urinalysis : We observed increase of PRO(p<0.01), SG(p<0.01) in female rats of 1,000 mg/kg/day and 2,000 mg/kg/day(p<0.01). Also, we observed increase of pH and KET in female rats of 1,000 mg/kg/day(p<0.05) and of 2,000 mg/kg/day(p<0.01). WBC in female rats in 1,000 mg/kg/day and 2,000 mg/kg/day were on increase. Hematological test : We observed increase of MCV in male rats of 250 mg/kg/day. (p<0.05) Serum biochemistry test : We found increase of CHO in female rats of 2,000 mg/kg/day(p<0.05). During the experimental period, there were no animals dead or moribund. There were no treatment related changes of general symptom, food and water consumption, organ weight and autopsy According to the results of 4-week repeated dose range finding study, the highest dose was established as 1000 mg/kg for 13-week repeated dose toxicity study and we determined to put 2 more groups by common ratio two.

IGRINS Mirror Mount Design for Three Off-Axis Collimators and One Slit-Viewer Fold Mirror

  • The Korea Astronomy and Space Science Institute and the Department of Astronomy at the University of Texas at Austin are developing a near infrared wide-band high resolution spectrograph, immersion grating infrared spectrometer (IGRINS). The compact white-pupil design of the instrument optics uses seven cryogenic mirrors, including three aspherical off-axis collimators and four flat fold mirrors. In this study, we introduce the optomechanical mount designs of three off-axis collimating mirrors and one flat slit-viewer fold mirror. Two of the off-axis collimators are serving as H and K-band pupil transfer mirrors, and are designed as system alignment compensators in combination with the H2RG focal plane array detectors in each channel. For this reason, the mount designs include tip-tilt and parallel translation adjustment mechanisms to properly perform the precision alignment function. This means that the off-axis mirrors' optomechanical mount designs are among the most sensitive tasks in all IGRINS system hardware. The other flat fold mirror is designed within its very limitedly allowed work space. This slit-viewer fold mirror is mounted with its own version of the six-point kinematic optics mount. The design work consists of a computer-aided 3D modeling and finite element analysis (FEA) technique to optimize the structural stability and the thermal behavior of the mount models. From the structural and thermal FEA studies, we conclude that the four IGRINS mirror mounts are well designed to meet all optical stability tolerances and system thermal requirements.
