The phenetic relationships among six natural populations of Equisetum pratense in Korea were investigated at the population level by constructing a tree based on Random Amplified Polymorphic DNA (RAPD) markers. RAPD analysis was also conducted to estimate genetic diversity and the population structure of E. pratense. A mean of 26.7% at the six population levels indicated polymorphism. E. pratense was found to have fewer alleles per locus (1.267) and fewer effective alleles per locus (1.176). Genetic diversity (0.102) in E. pratense is lower than the average for species with similar life history traits. Total genetic diversity values (HT) varied between 0.112 (OPD-07) and 0.445 (OPD-16), for an average overall polymorphic locus of 0.141. Inter-locus variation in the within-population genetic diversity ($H_S$) was low (0.102). Asexual reproduction, small population size, and the colonization process are proposed as possible factors contributing to the observed low genetic diversity in E. pratense. On a per-locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) ranged from 0.129 for OPD-07 to 0.455 for OPD-09, with a mean of 0.277. This indicated that about 27.7% of the total variation was among populations. Thus, genetic variation (72.3%) resided within populations. This study contributes new information for research on the taxonomy and population genetics of E. pratense.

Pigs have been extensively used as mediators of xenotransplantation research. Specifically, the Massachusetts General Hospital (MGH) miniature pig was developed to fix major histocompatibility antigens for use in xenotransplantation studies. We generated transgenic pigs for xenotransplantation using MGH pigs. However, it has not been studied yet whether these pigs show similarity of reproductive physiological characteristics to wild types of MGH miniature pig. In this study we analyzed the estrous cycles and pregnancy characteristics of wild type (WT) and transgenic MGH miniature pigs, which were ${\alpha}1,3$-galactosyltransferase (GalT) heterozygous and homozygous knock-out, and membrane cofactor protein (MCP) inserted in its locus, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs. Estrous cycles of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $20.9{\pm}0.74$, $20.1{\pm}1.26$, and $17.3{\pm}0.87days$, respectively, and periods of estrous were $3.2{\pm}0.10$, $3.1{\pm}0.12$, and $3.1{\pm}0.11days$. The periods of gestation of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $114.2{\pm}0.37$, $113.3{\pm}0.67$, and $115.4{\pm}0.51days$, respectively. Litter sizes of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $4.8{\pm}0.35$, $4.8{\pm}1.11$ and $3.0{\pm}0.32$ respectively. There were no significant differences on estrous cycle, periods of estrous and gestation, and litter size among WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs, meaning that GalT knock-out and additional expression MCP of the MGH miniature pig did not effect on reproduction traits. These results provide relevant information to establish breeding system for MGH transgenic pig, and for propagation of $GalT^{-MCP/-MCP}$ pig to supply for xenotransplantation research.
This study was conducted to provide primary data through analysis of zoysia grass genetic resources to develop grass cultivars with beneficial novel properties. Zoysia grass (Zoysia. spp) is native to Korea, and is mainly propagated through stolons. However, since seed coat treatment technology was developed, the breeding of sexually reproductive grass variants has become possible, necessitating characterization of the floret appearance rate in the secured zoysia grass genetic resource before developing sexually reproductive cultivars. In this experiment, 549 grass lines were examined, revealing that florets appear in only 43 lines (7.81%). Survival rates after transplantation, and stolon generation rates displayed a significant positive correlation (Rho = 0.44). Survival rates after transfer, and rates of stolon production displayed very low correlations with floret appearance (Rho = -0.11 and Rho = -0.06). No significant results were obtained in 43 lines that displayed >20% floret appearance. To breed sexually reproductive grass variants, it is thus necessary to secure more genetic resources, considering the low rate of floret appearance. Finding traits that predict floret appearance at an early stage is also required.
Background: Necdin (NDN), a member of the melanoma antigen family showing imprinted pattern of expression, has been implicated as causing Prader-Willi symptoms, and known to participate in cellular growth, cellular migration and differentiation. The region where NDN is located has been associated to QTLs affecting reproduction and early growth in cattle, but location and functional analysis of the molecular mechanisms have not been established. Methods: Here we report the sequence variation of the entire coding sequence from 72 samples of cattle, yak, buffalo, goat and sheep, and discuss its variation in Bovidae. Median-joining network analysis was used to analyze the variation found in the species. Synonymous and non-synonymous substitution rates were determined for the analysis of all the polymorphic sites. Phylogenetic analysis were carried out among the species of Bovidae to reconstruct their relationships. Results: From the phylogenetic analysis with the consensus sequences of the studied Bovidae species, we found that only 11 of the 26 nucleotide changes that differentiate them produced amino acid changes. All the SNPs found in the cattle breeds were novel and showed similar percentages of nucleotides with non-synonymous substitutions at the N-terminal, MHD and C-terminal (12.3, 12.8 and 12.5%, respectively), and were much higher than the percentage of synonymous substitutions (2.5, 2.6 and 4.9%, respectively). Three mutations in cattle and one in sheep, detected in heterozygous individuals were predicted to be deleterious. Additionally, the analysis of the biochemical characteristics in the most common form of the proteins in each species show very little difference in molecular weight, pI, net charge, instability index, aliphatic index and GRAVY (Table 4) in the Bovidae species, except for sheep, which had a higher molecular weight, instability index and GRAVY. Conclusions: There is sufficient variation in this gene within and among the studied species, and because NDN carry key functions in the organism, it can have effects in economically important traits in the production of these species. NDN sequence is phylogenetically informative in this group, thus we propose this gene as a phylogenetic marker to study the evolution and conservation in Bovidae.
Environmental changes can affect life-history traits, such as growth rate and reproduction, and organisms adapt on a given environmental condition to maximize ecological fitness. This study shows the effects of water temperature and dissolved oxygen level on early growth and accumulated damage in fish using a dynamic-state-dependent model. I have hypothesized that the level of foraging activity is related to growth and stress and so the optimal level can maximize reproductive success - ultimately, fitness. The critical temperature and dissolved oxygen (DO) is also defined as inducing the maximum growth rate at the level. So, the model predicts the highest growth rate at oxygen saturation and lower growth rate at lower or higher level of DO in water. Lower DO (i.e., hypoxia) causes slower growth rate through higher amount of accumulated stress whereas higher DO (i.e., hyperoxia) induces faster growth rate, but smaller body size. In addition, I show that there is lower impact when considering simple or independent environmental factors on environmental assessment. My findings suggest that multiple environmental factors as physiological ecology approach should be considered to improve impact assessment in environmental changes and a further study is needed to develop advanced assessment tools considering multiple environmental factors.
Ostrinia scapulalis is one of important pests in leguminous crops, especially red bean. In order to understand the biological characteristics of the insect, we investigated the effects of temperature on development of each life stage, adult longevity and fecundity of O. scapulalis at eleven constant temperatures of 7, 10, 13, 16, 19, 22, 25, 28, 31, 34, and 36℃. Eggs and larvae successfully developed next life stage at most temperature subjected except 7, 10 and 13℃. The developmental period of egg, larva and pupa decreased as temperature increased. Lower and higher threshold temperature (TL and TH) were calculated by the Lobry-Rosso-Flandrois (LRF) and Sharpe-Schoolfield-Ikemoto (SSI) models. The lower developmental threshold (LDT) and thermal constant (K) from egg hatching to adult emergence of O. scapulalis were estimated by linear regression as 13.5℃ and 384.5DD, respectively. TL and TH from egg hatching to adult emergence using SSI model were 19.4℃ and 39.8℃. Thermal windows, i.e., the range in temperature between the minimum and maximum rate of development, of O. scapulalis was 20.4℃. Adults produced viable eggs at the temperature range between 16℃ and 34℃, and showed a maximum number, ca. 416 offsprings, at 25℃. Adult models including aging rate, age-specific survival rate, age-specific cumulative oviposition, and temperature-dependent fecundity were constructed, using the temperature-dependent adult traits. Temperature-dependent development models and adult oviposition models will be useful components to understand the population dynamics of O. scapulalis and will be expected using a basic data for establishing the strategy of integrated pest management in leguminous crops.

Plants interact with various biotic and abiotic environmental factors. It requires much information to understand the traits of a plant species. A shortage of information would restrict the assessment, especially in the evaluation of what kind of factors influence a plant species to face extinction. Polygonatum stenophyllum Maxim. is one of the northern plants of which Korea is the southern distribution edge. The Korean Ministry of Environment had designated it to be the endangered species until December 2015. Although it is comparatively widespread, and a large population has recently been reported, it is assessed to be vulnerable due to the low population genetic diversity. This study evaluated the current distribution of Polygonatum stenophyllum Maxim. We investigated the vegetational environment, population structures, phenology, soil environment, and self-incompatibility based on the results. Lastly, we evaluated the current threats observed in the habitats. The habitats tended to be located in the areas where the masses at the edge of the stream accumulated except for those that were located on slopes of some mountainous areas. Most of them showed a stable population structure and had re-established or recruited seedlings. Polygonatum stenophyllum Maxim. had the difference in time when the shoots appeared above the ground depending on the depth of the rhizome located in the underground. In particular, the seedlings and juveniles had their rhizome located shallow in the soil. Visits by pollinator insects and success in pollination were crucial factors for bearing of fruits by Polygonatum stenophyllum Maxim. The threats observed in the habitat of Polygonatum stenophyllum Maxim. included the expansion of cultivated land, construction of new buildings, and construction of river banks and roads. Despite such observed risk factors, it is not likely that there would be rapid population reduction or extinction because of its widespread distribution with the total population of more than 2.7 million individuals and the new populations established by the re-colonization.
Data from five Hanwoo breeding farms were collected to analyze the effects of parity, season, nutrition and reproduction on body conditions of heifers and cows and to analyze the relationships of body conditions with reproductive performances and body growth. Analytic models included farms and parity which were run through SAS to estimate least squares means and correlation coeflicients between traits body condition scores(BCS) of I(very emaciated) through 9(very obese) scale at service and at calving. calving interval, days to rebreeding, gestation length. number of services per conception. Overall averages were 4.55 for BCS at service, 5.42 for BCS at calving. 406.7days of calving interval, 287.7days of gestation length, 66.2days from calving 10 first service and 1.78 times of services per conception. Number of services per conception was 1.50${\sim}$1.74 times for the cows of BCS 5 or under and 2.00${\sim}$3.00 for those of BCS 6 or higher. Body weight of calves from cows observed averaged 23.3kg at birth and 70.7kg at weaning. 57 percent of cows showed BCS 4${\sim}$5 at service while 46 percent of those showed 5${\sim}$6 at calving. Estimated phenotypic correlation coefficients of BCS at service with BCS at calving, calving interval, gestation length, number of services per conception were low but positive: 0.16, 0.26. 0.08 and 0.06. respectively. Phenotypic correlations of BCS at calving with calving interval, gestation length and number of services per conception were estimated to be also low and positive: 0.10, 0.13 and 0.10. respectively. However, phenotypic correlations between calving interval and gestation length, and between gestation length and number of services per conception were negative but low as - 0.11 and- 0.13 each.
One of the domesticated species; the dog has been selectively bred for various aims by human. The dog has many breeds, which are artificially selected for specific behaviors and morphologies. Dogs contribute their life to human as working dogs for guide, rescue, detection or etc. Working dogs requires good personality, such as gentleness, robustness and patience for performing their special duty. Many studies have concentrated on finding genetic marker for selecting the high-quality working dog. In this study, we confirmed quantitative expression patterns of eight genes (ABAT; 4-Aminobutyrate Aminotransferase, PLCB1; Phospholipase C, Beta 1, SLC10A4; Solute Carrier Family 10, Member 4, WNT1; Wingless-Type MMTV Integration Site Family, Member 1, BARX2; BarH-Like Homeobox 2, NEUROD6; Neuronal Differentiation 6, SEPT9; Septin 9 and TBR1; T-Box, Brain, 1) among brains tissues from four dog breeds (Beagle, Sapsaree, Shepherd and Jindo), because these genes were expressed and have functions in brain mostly. Specially, BARX2, SEPT9, SLC10A4, TBR1 and WNT1 genes were highly expressed in Beagle and Jindo, and Sapsaree and German Shepherd were vice versa. The biological significance of total genes was estimated by database for annotation, visualization and integrated discovery (DAVID) to determine a different gene ontology (GO) class. In these analyses, we suppose to these eight genes could provide influential information for brain development, and intelligence of organisms. Taken together, these results could provide clues to discover biomarker related to functional traits in brain, and beneficial for selecting superior working dogs.
This study was carried out for the objectives to collect the basic informations on the health behaviors of the elementary school children in an urban area in Korea. Seven hundred students were drawn to fill in the designed questionnaire which carries variety of Questions on health re-lated behaviors in general, eating habits, disease history, mental health, and sex education. Questionnaire were filled in by their parents. Major findings are as follows: ① 55.7% had habits of washing the hands before eating whereas 59.8% trashing their hands after toilet. The others had no idea of washing hands before eating and after toilet. ② 26,5% had habits of brushing the teeth twice a day 54.7% only once in the morning, and 2.6% once only in the evening. Thus, the idea of prevention from decayed teeth seems to be lacking among the school children. ③ Bathing habits were also inquired to get 40.3% of bathing more than once a week, 43.1% once every two weeks, and the rest of 16.6% once every one to three months. ④ 41.7% keep the regular bedding time whereas 58.3% irregular. Physical exercises were con-ducted by 76.6% on the ground while 23.5% did not practice any physical exercises at all. Of those physical exercises, rope skipping occupied 37.5%, and the other 66.9% consisted of 14 different kinds of individual type physical exercises such as gymnastic exercise. The main reasons for not enjoying exercises were different by sex; boys largely complained the inadequacy and lack of gymnastic facilities and girls felt in short of friends who could join the exercises. ⑤ 31.9% of the school children had been taking not much of food while 28.3% had unbalanced diets. Of these unbalanced diets, meat occupied 33.2% to be the priority to have an order of the following items such as vegetables, bread or noodle, and fishes as next to each. For eating habits, 88.5% take simple snack such as bread (38.4%, cookies, fruits, and candies in order. 25.8% of the children were provided such snacks or their parents regularly. Breakfast was sufficiently taken by 45.0% whereas 8.4% had never sufficiently. As to the lunch, 63.6% had sufficiently while 16.8% insufficiently. 70.6% take breakfast with all family members together and 30.4% separately. Correlation of sufficient taking of breakfast and eating together of tile family member's seems to be significant when we compare 72.5% of sufficient takers who enjoy breakfast together with the family members with 55.6% of insufficient takers who enjoy it with the family. This finding allows the investigator to point out the importance of table circumstances for children's eating. ⑥ The most common disease was catching a cold (38.8%), and the second was stomach trouble to be followed by the frequency of car sickness, headache, and skin infection. Doctors are consulted only by 23.9% when they are sick whereas 59.7% resorted to the drug stores. The lower the educational attainment of the parents, the lower the rate of visiting clinics. ⑦ 36.7% of their parents pointed out the problems of personality guidance as the most difficult thing at home 71.3% of their parents worried about and unsatisfied with their children's personality traits. Of these complains of the parents, impatience stood at the top to be tabulated at 24.1%, and 21.1% indicated narrow-mindedness. In line with this primary socialization at home, the most crucial problem seems to be related with the lack or recognition of the parents'own role when we find only 43.1% of the parents understood the importance of their own role for the home education of children; the latter group attributed tile responsibility of personality formation to the children themselves. ⑧ As to the sex educational aspects, 30.9% of children have ever asked about the physiology of reproduction or sexual matters to their parents, of those parents only 17.0% could give the constructive responses to the inquiries of the children. In companies on with these data, 25.6% recognized their own role in sex education for their own children while the large segments of the parents (51.1%) attributed the responsibility of sex education to tile low level of 38.3% who recognized the importance of sex education in the school curriculum and 25.1% of the parents insisted to wait until they get to know naturally about sex. 38.1% of the parents said they had some knowledge on sex from books while 16.9% through mass media. The next groups had common senses of sex from their own parents, school friends and other sources.
