Effects of oral administration with fermented product from sewage in land-based seawater fish farm on haematological factors of olive flounder, Paralichthys olivaceus (양식장 배출물 발효물의 어류 사료 첨가에 따른 넙치, Paralichthys olivaceus의 혈액학적 변동에 미치는 영향)

  • Effects of oral administration with fermented product from sewage in land-based seawater fish farm on haematological disturbance in the olive flounder, Paralichthys olivaceus was investigated. After 4 weeks of conditioning with a basal diet, fish were divided into 4 groups and provided experimental diet (0.1, 0.5, 1.0 and 2.0%) supplement of fermented sewage for 80 days. Proximal analysis was performed for the product of sewage which was fermented by lactic acid and yeast. RBC count, hemoglobin concentration and hematocrit value were increased according to the treated periods, however, no statistical difference was observed between control and treatment groups. There were no significant difference in serum organic, inorganic compounds and enzyme activities between control and treatment groups. This study hypothesized that the supplement of fermented product from sewage in land-based seawater fish farm might be an additive supplement for source of fish diet in view of haematological examination. Recycling of the sewage may be an economic artificial sources of diet for fish aquaculture practices.

  • In late summer of 2001, a new disease "overturn disease" occurred among mud loach (Misgurnus mizolepis) cultured in a farm Chonbuk Province. The fish exhibited a lethargic, erratic and spiral swimming at the water surface. Affected fish showed swollen abdomen with accumulation of air bubbles in the intestinal canal. The bubbles were much larger in the anterior part than in the posterior part. Hematological and histopathological examinations were done. No significant differences were found in RBC count, Ht value and Hb concentration between the diseased and the healthy fish. Gill filaments showed proximal hyperplasia of the respiratory epithelia and fusing of the gill lamella. Hemorrhage and necrosis occurred in the intestinal epithelia and within the lamina propria, and the mucosal epithelia were separated from the muscularis. Hepatocytes underwent atrophy. In the fish experimentally tied between the annal fin and the anus by a thread to confirm accumulation of air bubbles within intestine, accumulation of air bubbles was confirmed. This disease in mud loach appeared to be induced by circulatory disturbances in the intestinal wall owing to accumulation of air bubbles in the alimentary canal after fed high protein diet throughout all culturing period.

Changes of Growth and Hematological Constituents in the Rock Bream Oplegnathus fasciatus Exposed to TBT (TBT 노출에 따른 돌돔, Oplegnathus fasciatus의 성장 및 혈액성분의 변화)

  • Experiments were carried out to investigate the growth and hematological parameters of rock bream Oplegnathus fasciatus exposed to several different concentrations (0, 1, 2, 4, $8{\mu}g/L$) of tributyltin (TBT) for 4 weeks. Growth rate of the fish exposed to $8{\mu}g/L$ of TBT concentration was significantly lower than that of control group fish. The major hematological findings were significant decreases in the red blood cell count, hematocrit value and hemogobin concentration in the fish exposed to $8{\mu}g/L$. Although serum glucose concentration was significantly reduced compared to the control group, total protein concentration was found to be significantly increased over the control group. Exposure to $8{\mu}g/L$ of TBT concentration resulted in significant increase in the enzyme activities, such as glutamic oxalate transminase and glutamic pyruvate transminase in the fish. The present findings suggest that exposure to $8{\mu}g/L$ concentration of TBT can cause significant changes in growth and hematological parameters of rock bream Oplegnathus fasciatuso.

Effects on lethal concentration 50%, hematological parameters and plasma components of mirror carp, Cyprinus carpio nudus exposed to waterborne ammonia (수인성 암모니아 급성노출에 의한 향어, Cyprinus carpio nudus의 반수치사농도, 혈액학적 성상 및 혈장성분의 영향)

  • Mirror carp, Cyprinus carpio nudus (weight 26.2±3.1 g, length 11.8±0.5 cm) were exposed to waterborne total ammonia nitrogen (TAN) at 0, 2, 4, 8, 16, 32, 64 and 128 mg TAN/L. The lethal concentration 50 (LC50) of mirror carp, C. carpio nudus exposed to waterborne ammonia was 60.38 mg TAN/L. The red blood cell (RBC) count was significantly reduced by ammonia exposure, whereas there was no significant changes in the hemoglobin concentration and hematocrit value. In the plasma components, glucose, total protein, aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by ammonia exposure. The results of this study suggest that the ammonia exposure to C. carpio nudus affects the survival rates, hematological parameters and plasma components as toxicity.

  • The study was carried out to investigate the changes of haematological constituents induced by dietary exposure of Hexachlorobenzene (HCB) and Polychlorinated biphenyls (PCBs) in nile tilapia, Oreochhromis niloticus. The nile tilapia were fed pellet with 0.05, 0.25 and 0.50 mg/kg body weight/day of HCB and PCBs for 30 days, respectively. HCB ($\ge$ 0.25 mg/kg) induced a significantly decreased the red blood cell count at 30 days, and the hemoglobin concentration and hematocrit value were significantly decreased after 20 days to 0.50 mg/kg. The diminution of red blood cell count, hemoglobin concentration and hamatocrit value by PCBs was found after 20 days at $\ge$0.25 mg/kg. The concentrations of calcium decreased significantly at 30 days at $\ge$0,25 mg/kg of PCBs-diet. However, magnesium and inorganic phosphorus did not show any noticeable changes in the range of 0.05$\~$o.50 mg/kg of HCB and PCBs compared to control group during the experimental period, The low concentration of total protein was observed at the last day of experiment to 0.50 mg/kg of HCB-diet, while total protein initiated to increase from 20 days at the 0.25 mg/kg of PCBs-diet. The glucose was significantly increased after 20 days at $\ge$0,25 mg/kg of HCB and PCBs, A significant increment GOT and GPT activities was noticed after 20 days at 0.50 mg/kg of HCB and PCBs, These results indicate that hematological and serum biochemical changes in the nile tilapia by dietary exposure to HCB and PCBs are affected at more than 0,25 mg/kg after 20 days.

  • The effect of captafol on the hematological value, erythrocyte membrane and plasma biochemical value was investigated using blood of SPF Sprague-Dawley male rats in vitro. For the anticoagulations, we used 0.5mg of heparin per $10m{\ell}$ of blood from vena cava. Three con-centrations($0.1{\times}10^{-4}M$, $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$) captafol in ethanol were added to the each $2.5m{\ell}$ blood so that the linal concentration of ethanol was 1%. The blood contained with each concentration of captafol was incubated at $37^{\circ}C$ C for 2 hours under 5% $CO_2$ gas The whole blood and plasma were examined for hematological vaJues, erythrocyte membrane damage and biochemical values, respectively. The results obtained were summarized as follows ; 1. The number of RBC in $1{\times}10^{-2}M$ group and the concentration of MCHC in $1{\times}10^{-3}M$ group were significantly (p<0.05) decreased and increased from that of control values, respectively. The percentage of Hct was significantly (p<0.05) decreased with dose-response. 2. Erythrocyte fragility rate of $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$ group were significantly (p<0.01) increased from that control with dose response. 3. Potassium ion level of $1{\times}10^{-2}M$ was significantly (p<0.05) increased from that of control. 4. The concentration of total bilirubin in the $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$ groups were significantly increased from that of control. The enzyme level of creatine kinase in $1{\times}10^{-2}M$ group was significanlty (p<0.05) decreased from control value.

Studies on the Reproductive Toxicant and Blood Metabolite in Pups Born After Bisphenol A Administration in Pregnant Mice (임신중인 생쥐에 Bisphenol A 투여 후 태어난 차산자의 생식독성과 혈액성분에 관한 연구)

  • Bisphenol A (BPA) is a monomer used in the manufacture of a multitude of chemical products, including epoxy resins and polycarbonate. The objective of this study was to evaluate the effects of BPA administration on reproductive characteristics and blood hematological and chemical values in offspring of pregnant dams treated with BPA. BPA was administrated to pregnant mice by intraperitoneally injection with 0, 0.05, 0.5 and 5.0 mg/kg B.W. for 5 times at 3 days interval on gestation days 1-16. There were no treatment-related effects of BPA on reproductive organ weight in male offsprings at 45 days-of-age, but body weight was the lowest in 5.0mg BPA group when compared to other groups (P<0.05). No differences in semen characteristics (sperm concentration, viability, motility and abnormality) were observed between the control and BPA treatment groups. The WBC, HB, HT, MCV, MCH, MCHC, albumin, BUN and total protein of blood hematological and chemical values in male offsprings were not difference for any treatment groups, but RBC value in BPA groups was significantly increased comparing to the control group (P<0.05). The PLT value was slightly higher in 5.0mg BPA groups than in any other group, but not significantly difference among the experimental groups. In female offsprings, the effects of BPA didn't affect to the body and ovary weight, but the uterus weight in 5.0mg BPA group was slightly heavier than that of control group (P>0.05). No statically significant difference in blood hematological values in female offsprings were observed between the control group and BPA groups, but the concentration of albumin and BUN were significantly higher in 0.5mg BPA group when compared to control and other BPA treatment groups (P<0.05). The histological evaluation of testis and ovary in growing offspring at 45 days-of-age was not difference between the control group and BPA groups, but endometriosis of the uterus in female offspring was dramatically increased in 0.5 and 5.0mg BPA groups. These founding suggest that low concentration of BPA might not have a important role on reproductive ability or blood metabolite in offspring of pregnant dams treated with BPA.

  • This study was performed to evaluate the effect of hemoglobin (Hb) on bone mineral density (BMD) in university students by performing a quantitative analysis. The subjects included healthy university students aged 20 to 30 years. Although osteoporosis has traditionally been considered as a disease of aging women, it is becoming an increasingly concerning male health problem. Diagnosis of osteoporosis is calculated with a quantitative assessment of BMD. Laboratory blood and urine tests are mainly used with low BMD or fragility fractures to identify any possible causes of bone metabolism disorders. In this study, there was no difference in BMD according to gender. The average red blood cell (RBC), Hb, and Hematocrit (HCT) were significantly higher in males (p<0.01). The correlation between lumbar spine, skeletal muscle mass (SMM), and basal metabolic rate (BMR) was statistically significant (p<0.01). Hb showed a 51.7% statistical influence on BMD by multiple regression analysis. These findings are useful to understand the relationship between BMD and Hb; lower Hb level is associated with lower BMD. The Hb level was the strongest predictor of abnormal BMD. In conclusion, this study showed that a low Hb value was significantly correlated with low bone mass, suggesting that a low Hb value is a risk factor for changes in bone turnover that leads to a decrease bone density.

  • Parallel testing means ordering a number of tests at the same time so abnormalities in any of the tests can be found quickly and used in making the diagnosis. This is a good medical strategy to eliminate diseases and it is relatively inexpensive if all the tests are potential sources of information and performed on the same analyzer. In regression, the equation for the straight line is recast as y = bx + a. This change in terminology leads to confusion. Here a is the y-intercept or constant and b is the coefficient or slope of the line. A few more words of caution about regression - as in all of statistics there are certain assumptions: the x value is a true measure, both X and Y distributions are normal, and homoscedasticity, i.e., the variance of y is the same for each value of x. In this study the linearity classification made by different scientists were always in agreement. Typical examples of curves that were considered linear are presented in Fig. 1-5. Because these automated procedures values were usually within five percent of each other the curvature could be easily detected. The plot of the WBC, RBC, hemoglobin, hematocrit and platelet concentrations from approximately 74.4 to $0{\times}10^3/{\mu}L$ and $80.4-0{\times}10^3/{\mu}L$, $5.6-0{\times}10^6/{\mu}L$ and $6.1-0{\times}1106/{\mu}L$, 18.3-0 g/dL and 19.0-0 g/dL, 54.1-0% and 56.8-0% and 642.0 to $0.03{\times}10^3{\mu}L$ and $754.0-0{\times}10^3/{\mu}L$ on the ADVIA 2120 C Versus and A and B typical of an acceptable linear study as shown in Fig. 1-5. The grand mean of R2, intercept and slope is 0.99898, 0.99459 and 1.54626.

Effects of Dioxin on the Body Weight, No. of Sperm, Motility, Testis and Organ Weight in Mice (Dioxin의 투여가 마우스의 체중, 정자수, 정자활력, 정소 및 장기중량에 미치는 영향에 관한 연구)

  • In this study, we observed the effects of dioxin on weights of body, testes and other organs, the number and motility of sperm in the a various dose after two days' of administration in mice. Animals were treated with oral dose of dioxin 10, 20, 30, 40 mg/kg body weight, respectively. 1, After administration dioxin at doses of 10, 20, 30 and 40 $\mu\textrm{g}$/kg to the mice, the changes in body weights were 30.6 $\pm$ 2.g~40.7 $\pm$ 3.9g and 30.8 $\pm$4.1g~39.5g $\pm$3.1 for 10 and 20 $\mu\textrm{g}$/kg dosed group, 31.0 $\pm$ 3.5g ~ 39.0 $\pm$ 3.5g, 30.6 $\pm$ 3.4g~38.3 $\pm$ 4.0g for 30 and 40 $\mu\textrm{g}$/kg dosed group. The body weight of dioxin-administered group showed lower value when compared to 30.6 $\pm$ 2.8g ~ 44.5 $\pm$ 3.1g of which is control group's. 2. After administration of dioxin at doses of 10, 20, 30 and 40 $\mu\textrm{g}$/kg to the mice, the increase in the number of WBC was prominent, but the increase in the number of RBC wasn't significant, though the values of Hb, PCV, and PLT were higher than those of control group's. 3. After administration of dioxin at doses of 10. 20, 30 and 40 $\mu\textrm{g}$/kg to the mice, the changes in sperm number were 112.5 $\pm$ 3.7~119.4 $\pm$4.2 $\times$ 10$^{6}$ $m\ell$, 103.9 $\pm$3.8 ~ 110.2 $\pm$ 3.6 $\times$ 10$^{6}$ $m\ell$, 97.5 $\pm$ 3.4 ~105.7 $\pm$ 4.4 $\times$ 10$^{6}$ $m\ell$, 87.2 $\pm$ 3.7~98.5 $\pm$ 3.8 $\times$ 10$^{6}$ $m\ell$, respectively. The sperm number of dioxin-administered group showed lower value than that of control group's, which was 119.0 $\pm$ 4.3 ~ 120.7 $\pm$ 4.8 $\times$ 10$^{6}$ $m\ell$. After administration of dioxin at doses of 10~40 $\mu\textrm{g}$/kg to the mice, the sperm motility were 69.4$\pm$ 3.0 ~ 86.6 $\pm$4.7%. The sperm motility of dioxin-administered group showed lower value than that of control group's. 4. After administration of dioxin at doses of 10, 20, 30 and 40$\mu\textrm{g}$/kg to the mice, the organ weight of each dioxin-administered group's was decreased a little compared to that of control group's. 5. After administration of dioxin at doses of 10, 20, 30 and 40 $\mu\textrm{g}$/kg to the mice, the weights of spleen, kidneys, and liver showed increase a little.
