Effects of Glycerol and Shikimic Acid on Rapamycin Production in Streptomyces rapamycinicus

  • Rapamycin, derived from Streptomyces rapamycinicus, is an important bioactive compound having a therapeutic value in managing Parkinson's disease, rheumatoid arthritis, cancer, and AIDS. Because of its pharmaceutical activity, studies over the past decade have focused on the biosynthesis of rapamycin to enhance its yield. In this study, the effect of rapG on rapamycin production was investigated. The rapG expression vector was constructed by utilizing the integration vector pSET152 under the control of the erythromycin resistance gene (ermE), a strong constitutive promoter. The rapamycin yield of wild type (WT) and WT/rapG overexpression mutant strains, under fermentation conditions, was analyzed by high-performance liquid chromatography (HPLC). Our results revealed that overexpression of rapG increased rapamycin production by approximately 4.9-fold (211.4 mg/l) in MD1 containing 15 g/l of glycerol, compared to that of the WT strain. It was also found that Illicium verum powder (10 g/l), containing shikimic acid, enhanced rapamycin production in both WT and WT/rapG strains. Moreover, the amount of rapamycin produced by the WT/rapG strain was statistically higher than that produced by the WT strain. In conclusion, the addition 15 g/l glycerol and 15 g/l I. verum powder produced the optimal conditions for rapamycin production by WT and WT/rapG strains.

대형직접전단시험에 의한 RAP 복합지반의 전단강도 특성 연구 (A Study on the Shear Strength Characteristics of Composited Ground applying RAP Method by Large Direct Shear Test)

  • To secure stability and availability of Rammed Aggregate Pier method as the foundation of a structure, the shear strength characteristics according to the area replacement ratio of RAP and the relative density of in-situ ground was studied through soil laboratory tests and large direct shear tests in a model ground. As a result, the internal friction angle tends to increase in proportion to in-situ relative density(Very Loose, Loose, Medium) in composite ground formed by the same area replacement ratio of RAP and also increase in proportion to increasing the area replacement ratio(30, 40, 50%) of RAP in the same ground condition. Furthermore, the comparative analysis between the experimental value and theoretical value of the shear strength is carried out.

  • Shaking table tests were performed to investigate the response of liquefaction mitigation of rammed aggregate piers(RAP) on soft ground. The displacements of the soft ground reinforced by RAP under area replacement ratio 7, 14, 28% during seismic loading were measured. The result of tests showed that effects of liquefaction mitigation were affected various area replacement ratios and ground acceleration on RAP systems.

Effects of a Phosphomimetic Mutant of RAP80 on Linear Polyubiquitin Binding Probed by Calorimetric Analysis

  • RAP80 plays a key role in DNA damage responses by recognizing K63-linked polyubiquitin moieties through its two ubiquitin-interacting motif (UIM) domains. The linker between the two UIMs possesses a phosphorylation site, but the relationship between phosphorylation and polyubiquitin recognition remains elusive. We investigated the interaction between a phosphorylation-mimic RAP80 mutant S101E and linear polyubiquitins, structurally equivalent to the K63-linked ones, using isothermal titration calorimetry (ITC). ITC analysis revealed differential binding affinities for linear tetraubiquitin by otherwise equivalent UIMs in S101E. Mutational analysis supported such differential polyubiquitin recognition by S101E. Our results suggest a potential crosstalk between polyubiquitin recognition and phosphorylation in RAP80.

애성환자에서 음향지표인 RAP, PPQ 및 APQ의 유용성 (Significance of Acoustic Parameter - RAP, PPQ, APQ- in Hoarseness)

  • Change of voice, espicially hoarseness show irregular vibration of vocal cord. So, computerized acoustic analysis has presented many acoustic parameters for objective evaluation of voice. We objectively investigated the vocal vibration of normal persons and hoarseness patients in Korea. The RAP(relative average perturbation), PPQ(pitch period perturbation quotient) and APQ(amplitude perturbation quotient) of normal persons were compared with that of hoarseness patients with multidimensional voice program for the possibility of distinguishing the pathologic vocal vibration from normal. Authors agree that RAP, PPQ and APQ showed interesting differences between the normal and the hoarseness patients by the multivariate statistical analysis. In conculusion, relative average perturbation, pitch period perturbation and amplitude perturbation quotient might be meangingful screening parameters distinguishing hoarseness patients from normal.

Self-Consolidating Concrete Incorporating High Volume of Fly Ash, Slag, and Recycled Asphalt Pavement

  • The use of sustainable technologies such as supplementary cementitious materials (SCMs), and/or recycled materials is expected to positively affect the performance of concrete mixtures. However, it is imperative to qualify and implement such mixtures in practice, if the required specifications of their intended application are met. This paper presents the results of a laboratory investigation of self-consolidating concrete (SCC) containing sustainable technologies. Twelve mixes were prepared with different combinations of fly ash, slag, and recycled asphalt pavement (RAP). Fresh and hardened concrete properties were measured, as expected the inclusion of the sustainable technologies affected both fresh and hardened concrete properties. Analysis of the experimental data indicated that inclusion of RAP not only reduces the ultimate strength, but it also affected the compressive strength development rate. The addition of RAP to mixes showed a consistent effect, with a drop in strength after 3, 14, and 28 days as the RAP content increased from 0 to 50 %. However, most of the mixes satisfied SCC fresh properties requirements, including mixes with up to 50 % RAP. Moreover, several mixes satisfied compressive strength requirement for pavements and bridges, those mixes included relatively high percentages of SCMs and RAP.

Helicobacter pylori 감염과 소아 복통 (Helicobacter pylori infection and abdominal pain in children)

  • The relationship between H. pylori(Hp) infection and recurrent abdominal pain(RAP) in children is not clear. Current data in the literature regarding a causal relationship between Hp infection and childhood RAP are conflicting. However, meta-analysis and most of the recently published studies have not supported an association between Hp infection and an increased prevalence of abdominal pain. Most published studies have some problems and have been criticized on methodological grounds. Most of the existing studies have not been designed to include double-blind placebo controls. And, because of the low incidence of Hp infection in children, they have a problem of obtaining sufficient numbers of the Hp infected children. There is also a concern about the heterogeneity of testing methodology in the studies and diagnostic accuracy problems of the non-invasive tests such as low sensitivity of the serology tests and false positive results of the UBT in children. And conflicting results may be explained, in part, by the poor definition of RAP or dyspepsia for the study subjects. To date, no consensus exists for the treatment of Hp-infected children with RAP. Some authors suggest the clinical efficacy of Hp. But others suggest that eradication of Hp is not associated with improvement of RAP. The problem of the most treatment studies is that they had usually no control groups and the number of the subjects was small. In order to make reasonable decisions about treatment of Hp infection in children with RAP, long-term, randomized, double blind, well controlled studies in large numbers of children will be required.

  • To secure stability and availability of Rammed Aggregate Pier method as the foundation of a structure, the bearing capacity and failure behavior characteristics was studied through soil laboratory tests in a model ground. In this study, soil laboratory tests use carried out to find the applicability of RAP method as the foundation of a structure. And bearing capacity and the failure mechanism of RAP method was studied according to relative density($60\%,\;70\%,\;90\%$), diameter(45mm, 60mm, 70mm) of each pier ana depth(5cm, l0cm, 15cm, 20cm, 25cm, 30cm). Earth pressure cell is set up approach RAP and 1.0D space at RAP center. Bearing acpacity and the failure mechanism of RAP is investigated by load test As a result, bulging failure was happened in $5\~10cm\;(1.0D\~2.00)$ depth which the maximum lateral earth pressure is acting. Especially, diameter changing of RAP are in inverse proportion to the relative density and the lateral stress is very much influenced by the lateral earth pressure in every layer and tends to decrease according to depth.

호안용 돌망태 매트리스의 수리학적 특성 연구 (A Study on the Hydraulic Characteristics for River Bank Protection using Gabion Mattress)

  • The condition of initial movement far the river bank protection using rip rap is a limit condition beyond which the lining is progressively destroyed as the separate elements are removed by the flow. In the case of the river bank protection using gabion mattress, however, after the initial movement the containment offered by the mesh remains. A new situation of equilibrium with a deformed river bank protection using gabion mattress is obtained, allowing it to withstand more severe conditions without compromising the resistance and without further deformation. Shield's coefficient for the river bank protection using gabion mattress is twice the value of that fur river bank protection using rip rap. This means that with the same hydraulic behavior conditions, the average dimension of the rocks to be used in the river bank protection using gabion mattress is half that of river bank protection using rip rap rock. When the same size rocks are used the allowable velocity for the river bank protection using gabion mattress is more than twice, even as much as 3 or 4 times that for river bank protection using rip rap.

그룹 랩만들기의 치료적 적용: 경도지적장애 청소년의 의사소통기술 향상 사례 (Case Study on Enhancing Communication Skills of Adolescents With Mild Intellectual Disabilities Through a Group Rap Making)

  • 본 연구는 경도지적장애 청소년 대상 그룹 랩만들기가 해당 청소년의 의사소통기술을 향상시키는지 알아보고자 실시된 사례연구이다. 연구 대상은 경도지적장애로 진단받은 만 14-15세의 청소년 3명이다. 본 연구에서 그룹 랩만들기는 랩의 구성요소인 비트, 벌스, 훅, 추임새를 치료적으로 활용하여 소통 맥락 구성, 소통 내용 완성 및 창작, 소통 내용의 적용이라는 3단계로 구성되었고, 회기별 50분씩, 주 2회, 총 6회기 진행되었다. 랩만들기가 적용된 후 경도지적장애 청소년의 의사소통기술 변화를 알아보기 위해 대상자들이 구성하고 부른 랩 가사를 분석하였고, 음악 활동 안에서 나타나는 요구·질문하기 및 정보제공하기 행동을 사건 기록법으로 측정하였다. 또한 프로그램 전후 대상자의 보호자들이 사회적 의사소통 체크리스트를 평정하도록 하였다. 연구 결과, 랩 가사에서 대상자들이 자신의 의견을 자발적으로 표현하는 시도가 증가하는 것을 확인할 수 있었고, 랩 구성 과정에서 자신의 의견을 요구하거나 질문하는 행동 역시 증가하는 것으로 나타났다. 사회적 의사소통 체크리스트에서도 대상자의 기능수준에 따라 개별 차이가 있긴 했지만 전반적으로는 평정 점수가 향상된 것으로 나타났다. 이러한 연구결과를 통해 일부 임상 대상군에게 제한적으로 사용되었던 랩만들기가 경도지적장애 청소년에게도 적용할 수 있음을 제시할 수 있었고, 자신의 의견을 표현하고 타인과 의견을 공유하고 조율하는 능력을 촉진할 수 있는 의미 있는 중재 방안이 될 수 있음 역시 확인할 수 있었다.
