Reproductive cycle with the gonadal development of Ruditapes philippinarum can be classified into five successive stages by histological observations: early active stage (January to March), late active stage (February to May), ripe stage (April to August), partially spawned stage (May to October), and spent/inactive stage (August to February). Changes in total protein contents in the adductor muscle tissues reached the maximum in the early and late active stages (February) and appeared the minimum in the ripe and partially spawned stages (May), while changes in their contents in the visceral mass tissues reached the maximum in the ripe and partially spawned stages (June) and gradually decreased in the partially spawned stage (June to October). On the whole, changes in total protein contents showed a negative correlationship between the adductor muscle and visceral mass tissues (r = -0.292, p < 0.05). Changes in total lipid contents in the adductor muscle tissues reached the maximum in the inactive and early active stages (January) and sharply decreased in the early and late active stages (February), while their contents in the visceral mass tissues reached the maximum in the ripe and partially spawned stages (April) and gradually decreased in the partially spawned stage (to October). On the whole, changes in total lipid contents showed a negative correlationship between the adductor muscle and visceral mass tissues (r = -0.699, p<0.05). Changes in glycogen contents in the adductor muscle tissues reached the maximum in the late active and ripe stages (April) and rapidly decreased in the partially spawned stage (May to October), while their contents in the visceral mass tissues reached the maximum in the early and late active stages (February) and rapidly decreased in the late active stage (March). Thereafter, their levels gradually increased in the ripe and partially spawned stages (April to July). On the whole, changes in glycogen contents appeared no correlationship between the adductor muscle and visceral mass tissues (r = 0.062, p > 0.05). These results indicate that the adductor muscle and visceral mass tissues are an important energy storage and nutrient supply organ in the Manila clams, and the nutrient contents of the adductor muscle and visceral muscle tissues change in response to gonadal energy needs.

R. Philippinarum is dioecious and oviparous. In the early vitellogenic oocyte, the Golgi apparatus and mitochondria present in the perinuclear region are involved in the formation of lipid droplets and in lipid granule formation. In the late vitellogenic oocyte, the endoplasmic reticulum, mitochondria in the cytoplasm are involved in the formation of proteid yolk granules. At this time, exogenous lipid granular substance and glycogen particles in the germinal epithelium are passed into the ooplasm of oocyte through the microvilli of the vitelline envelope. Ripe oocytes are about 55-60 $\mu$ m in diameter. The spawning period was once a year between early June and early October, and the main spawning occurred between July and August when seawater temperature was approximately 20 C. The reproductive cycle of this species can be categorized into five successive stages: early active stage (February to March), late active stage (April to May), ripe stage (April to August), partially spawned stage (June to October), and spent/inactive stage (August to March). Gonad developmental phases by histological qualitative analysis showed similar results with those of quantitative image analysis.

Immunohistochemical and ultrastructural experiments were conducted to find out heavy metal accumulation in some selected organs such as the kidney, the digestive gland, and the gill of the Antarctic clam Laternula elliptica and R. philippinarum. According to the immunohistochemical study the subject organs of the clam showed reactions indicating the presence of MT (metallothionein), a metal-binding protein involved in metal detoxifying process. Examination under the transmission electron microscope also revealed that other ligands may play a role in metal accumulating and detoxifying process in L. elliptica and R. philippinarum. In the artificial exposure of the clam to Cd, the clams showed immediate subcellular responses. The level of the anti-MT reactions became higher in the proportion to the degree of pollution of their habitat and to the period of Cd exposure. These suggest that the two species can be used as efficient biomarkers for Cd exposure in the natural environment.
Life cycle stages, including daughter sporocysts, cercariae, and metacercariae, of Parvatrema duboisi (Dollfus, 1923) Bartoli, 1974 (Digenea: Gymnophallidae) have been found in the Manila clam Ruditapes philippinarum from Aphae-do (Island), Shinan-gun, Jeollanam-do, Korea. The daughter sporocysts were elongated sac-like and 307-570 (av. 395) ㎛ long and 101-213 (av. 157) ㎛ wide. Most of the daughter sporocysts contained 15-20 furcocercous cercariae each. The cercariae measured 112-146 (av. 134) ㎛ in total length and 35-46 (av. 40) ㎛ in width, with 69-92 (av. 85) ㎛ long body and 39-54 (av. 49) ㎛ long tail. The metacercariae were 210-250 (av. 231) ㎛ in length and 170-195 (av. 185) ㎛ in width, and characterized by having a large oral sucker, genital pore some distance anterior to the ventral sucker, no ventral pit, and 1 compact or slightly lobed vitellarium, strongly suggesting P. duboisi. The metacercariae were experimentally infected to ICR mice, and adults were recovered at day 7 post-infection. The adult flukes were morphologically similar to the metacercariae except in the presence of up to 20 eggs in the uterus. The daughter sporocysts and metacercariae were molecularly (ITS1-5.8S rDNA-ITS2) analyzed to confirm the species, and the results showed 99.8-99.9% identity with P. duboisi reported from Kyushu, Japan and Gochang, Korea. These results confirmed the presence of various life cycle stages of P. duboisi in the Manila clam, R. philippinarum, playing the role of the first as well as the second intermediate host, on Aphae-do (Island), Shinan-gun, Korea.
The present study was conducted to confirm that a bivalve Ruditapes philippinarum can be used as a biomarker for the monitoring of the heavy metal pollution in the silt of the marine environment. The clams were collected from the silt of Cheonsu-bay, Buheung-ri, and Tan-island of the West Sea, Korea. To observe the normal structures of the target organs (hepatopancreas and gill), they were dissected out for the immunohistochemical study and the electron microscopy with TEM, SEM, and SEM-EDS. The immunohistochemical study showed that the interdiverticular connective tissues of the hepatopancreas, and the outer epithelium of the gill lamellae was strongly reacted to anti-metallothionein (MT), indicating the presence of MT, a metal-binding protein, involved in metal detoxifying process. According to the examinations under the TEM, the epithelial cells of the hepatopancreas of the clams collected from polluted area (Tan-island) showed certain changes such as swollen rER, swollen nuclear envelope and inclusion bodies in the nulcei. In the SEM-EDS analysis, tissue of the hepatopancreas showed relatively higher concentration of S, Zn, and Cd. These elements are supposed to be concerning with the MT-reaction in the hepatopancreas. Considering that the coastal bivalve R. philippinarum showed immediate subcellular responses to heavy metal pollution in the overall experiments conducted, this species might act as one of efficient biomarkers for the heavy metal contamination in the marine environment.
Mass mortality of the Manila clam, Ruditapes philippinarum has been reported all along the west and south coast of Korea for the past several years. As a pathogenic agent, Perkinsus sp., an endoparasitic protozoan has been identified in this study and believed to be responsible for the mass mortalities. Prevalence and infection intensity of Perkinsus sp. was investigated from a Manila clam population inhabiting at Komsoe Bay in the west coast where mass mortality of the clam has been reported. A total of 142 Manila clam, 50 oyster, Crassostrea gigas, 10 ark shell, Scapharca broughtonii, and 5 predatory gastropada, Rapana venosa were examined for the presence and the quantity of Perkinsus sp. Ray's fluid thioglycollate medium method (FTM method) with modified Mackin's infection intensity scale and Choi's quantitative method were used in detecting and quantifying the parasite. All individuals of R. philippinarum examined in this study were infected with Perkinsus sp., indicating $100\%$ prevalence while none of the oysters and the gastropods exhibited the parasite. Six to ten individual hypnospores of Perkinsus sp. were counted from the ark shells. The number of hypnospores in the clam tissues varied from 16,667 to 4,091,667, with a mean number of 1,077,628. Average infection intensity according to Mackin's was 2.87, indicating a moderate infection. A negative correlation was observed between the number of Perkinsus sp. in the tissue and the condition index, a ratio tissue wet weight to shell cavity volume. The clam size and the infection intensity in terms of total number of parasites were positively correlated; the bigger clam, the heavier infection. Such high number of Perkinsus sp. counted in the clams could be enough to cause physiological disturbance of clams, such as retarded growth and reproduction. It is also believed that such a high infection leads mortality of the clam via continuous draining of the energy by metabolic activities and reproduction of the parasites. Correlation between the condition index and the infection intensity observed in this study supports this hypothesis.

The characteristics of the populations of shortnecked clam (Ruditapes philipinarum) originated from three different seed-production sites, Hadong, Kochang and Ulsan along the coast of Korea, were analysed by the morphological differences and the random amplified polymorphic DNA (RAPD) profiles. The morphology of the shell and survival rate for each population were also investigated 13-months after transplantation to a farming site in Shinsung. The morphology of the populations from Hadong and Kochang showed significant differences (P<0.05), but one year after transplantation to Shinsung, the morphology of these three populations was no longer significantly different (P>0.05). The template DNA for RAPD was efficiently extracted from the digestive diverticula of the clams. Up to 13 of amplified fragments were detected using arbitrary primers. Within the species of R. philipinarum, the genetic similarities ranged from 0.196 to 0.259. The populations from Hadong and Ulsan showed the highest similarity. The survival rates of the populations from Hadong $(69.4\%)$ and Ulsan $(63.8\%)$ were higher than that from Kochang $(41.7\%)$ 13-months after transplantation. From the RAPD analysis, it could be used as one of the primary criterion in determining which shellfish populations among various seed-production sites tend to be genetically similar and more adaptable and transplantable to a farming site.

Manila clam, Ruditapes philippinarum, is commercially and ecologically important marine bivalve in Korea and japan. However, clam landings in the two countries have dramatically declined since the 1980-1990's. In the present study, the protozoan parasite, Perkinsus sp., lectin (host's defense-related glycoprotein) and histopathological features were investigated in Manila clams collected from Gomso Bay in Korea and Ariake Bay in japan (one of the largest clam beds in each country) during summer and fall, 2002-2003. DNA sequences of non-transcribe spacer (NTS), internal transcribed space. (ITS) and 5.85 rRNA of Perkinsus sp. were identical to those of P. atlanticus that was reported in Europe and Korea. For diagnosis of Perkinsus, the fluid thioglycollate medium (FTM) and the 2 M NaOH lysis methods were used. Prevalence of the parasite varied from 92.5-98.7% in Gomso Bay and 35.5-37.9% in Ariake Bay. Infection intensity, in terms of the number of Perkinsuscells per gram tissue wet weight, in the clams of Gomso Bay in fall 2002 averaged 1,010,077-470,937 recording approximately100 times higher than that of Ariake Bay, and these were twice higher than those of summer samples in each location. Mean hemagglutination titer of the clams from Gomso Bay was approximately 60-folds higher than that of clams from Ariake Bay in 2002. In histological preparation of the clams from Gomso Bay in 2002, trophozoites of P. atlanticus were in groups and resulted in severe inflammatory response of host clam. Prevalence of the trematod, Cercaria tapes-like in the clams of Gomso Bay and Ariake Bay were 8.8 % and 10.5% respectively. In conclusion, the clams from Gomso Bay showed more severe pathologic symptoms and higher immune response than those of the clams from Ariake Bay.

This research reports the intersexuality of bivalves, such as Crassostrea gigas, Mytilus galloprovincialis, Rupiditapes philippinarum, Gomphina veneriformis and Barnea davidi discovered during the process of investigating the ecological health status of coastal waters of Korea. In intersex ovaries, the opposite germ cells were observed either individually or in groups in the interfollicular space and inside the oogenic follicle. Oocytes in the intersex testis were at the previtellogenic or initial vitellogenic stage. They were either scattered individually or in groups in the interfollicular space and inside the spermatogenic follicle. The intersexuality in C. gigas was 10.4% (n = 19/183), while female (12.2%, n = 6/49) exhibited a higher proportion than male (9.7%, n = 13/134). The intersexuality in M. galloprovincialis was 31.7% (n = 19/60), while female (36.4%, n = 12/33) exhibited a higher proportion than male (25.9%, n = 7/27). The intersexuality in R. philippinarum was 11.2% (n = 11/98), while male (16.7%, n = 7/42) exhibited a higher proportion than female (7.1%, n = 4/56). The intersexuality in G. veneriformis was 28% (n=30/107), while male (31.5%, n=17/54) exhibited a higher proportion than female (24.5%, n=13/53). The intersexuality in B. davidi was 18.4% (n = 7/38), while female (35.7%, n =5 /14) exhibited a higher proportion than male (8.3%, n = 2/24).
Macrobenthic fauna were studied on a tidal flat in Garolim Bay in July 2002, in order to understand their spatial distribution with the surface sediment types. Forty-nine stations were set on the tidal flat. A total of 147 species was sampled with the mean density of $1,140ind./m^2$ and the mean biomass of $157.2g/m^2$. The polychaetes were speices- and density-dominant faunal group with 64 species and the mean density of $791ind./m^2$. Mollusks were biomass-dominant faunal group with $132.0g/m^2$. Major dominant species were two polychaetes, Mediomastus californiensis with the mean density of $374ind./m^2$ and Heteromastus filiformis with $224ind./m^2$. Also mollusks such as Umbonium thomasi, Batillaria cumingi and Ruditapes philippinarum was relatively high in the mean density, showing the density of 45, 42, $32ind./m^2$ respectively. Especially, U. thomas was distributed in the mouth of the bay, where sediment grain size is rather coarse, and R. philippinarum at near tidal channel. Based on the cluster analysis, the macrobenthic community on the tidal flat was classified into seven station groups depending on the benthic environmental parameters(sediment types). Compared with other Korean tidal flats, species richness of Garolim tidal flats is more and less low.
