The Use of Fungal Inoculants in the Ensiling of Potato Pulp: Effect of Temperature and Duration of Storage on Silage Fermentation Characteristics

  • A $3{\times}3$ factorial design experiment was conducted to investigate the effect of temperature and duration of storage on the fermentation quality of potato pulp ensiled with two fungal inoculants under laboratory conditions. The inoculants, Rhizopus oryzae (R) and Amylomyces rouxii (A) were each added to potato pulp material to contain at least $1{\times}10^6$ CFU/g fresh matter, and silages without additives served as controls. The silages were stored under three temperature regimes; 4, 12 and $25^{\circ}C$. Three silos per treatment from every temperature regime were opened on days 7, 24 and 40 days after ensiling to investigate treatment effects on fermentation quality, starch and sugar concentrations. Increase in temperature and duration of storage had a positive significant effect (p<0.01) on the fermentation quality of potato pulp silage (PPS). The inoculants had little effect (p>0.05) on the fermentation quality of the silages. Sugar concentration in the silages decreased with increase in temperature (p<0.01) but increased (p<0.05) with progression of duration of storage. The fungal inoculants had no effect on starch degradation in PPS. The results suggest that storage temperature and duration of storage are more important in determining the rate of fermentation than addition of the fungal inoculants in PPS.

Microcomputer-controlled Koji Incubation System and Its Application to Barley Koji Manufacture (마이크로컴퓨터 제어(制御) 종국배양장치(種麴培養裝置)와 보리코오지 제조(製造)의 자동화(自動化))

  • For the automation of Koji incubation process, microcomputer based Koji incubation system was built and applied to acquisition of the process variables, and to control of the Koji incubation process. The incubation variables included the relative humidity and Koji weight. And data measured were sent to the microcomputer by the interface device built with MC 6821 PIA. Incubation environment conditions -temperature and humidity- were controlled by the actuation of heater and mist sprayer with on/off signal generated by ASIC program. Aspergillus oryzae as a starter of the Koji and steamed barley as media were used and Koji was successfully manufactured both at $25^{\circ}C,\;70%$ RH and at $27^{\circ}C,\;80%$ RH. During the Koji preperation, the temperature was linearly increased and substrate was consumed stepwise showing 3 steps in the weight loss curve.

Antioxidant and Neuroprotective Effects of Doenjang Prepared with Rhizopus, Pichia, and Bacillus

  • A new type of doenjang was manufactured by mixing soaked soybean, koji (Rhizopus oryzae), cheonggukjang (Bacillus amyloliquefaciens MJ1-4 and B. amyloliquefaciens EMD17), and Pichia farinosa SY80 as a yeast, salt, and water, followed by fermentation with koji that was made by fermenting whole wheat with R. oryzae. The mixed culture doenjang was designed to have a more palatable flavor and stronger biological activities than the conventional product. The extract of mixed culture doenjang showed higher antioxidant activity than the commercial doenjang as evaluated by the ferric reducing antioxidant power assay although it was not significantly different from the commercial product in 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activities. Further, the mixed culture doenjang reduced intracellular reactive oxygen species levels and protected cells from glutamate-induced cytotoxicity more efficiently in human hippocampal HT22 neuroblastoma cells than the commercial doenjang. In conclusion, a newly-developed mixed culture doenjang had a strong antioxidant activity in vitro and cultured cell model systems, exhibited a potential to prevent oxidative stress-associated disorders although animal and clinical studies are needed to confirm its in vivo efficacy.

Use of Restriction Fragment Length Polymorphism Analysis to Differentiate Fungal Strains in Sunchang Meju

  • Twenty-three fungal strains were isolated from meju that had originated from the Sunchang province, the famous location for making fermented soybean foods in Korea. The restriction fragment length polymorphism (RFLP) of the internal transcribed spacer (ITS) region of the rDNA (ITS-RFLP) was applied to differentiate the isolated fungal strains. First, the ITS region by polymerase chain reaction (PCR) with specific primers was amplified and then cleaved the products with different restriction enzymes. Cleavage of the amplified fragments with the restriction enzymes AluI, HaeIII, HhaI, and TaqI revealed extensive polymorphisms. The ITS-RFLP results highly correlated with ITS sequence analysis. All of the 23 fungal strains were classified into 5 groups by ITS-RFLP analysis. Aspergillus oryzae was the major fungal strain isolated from Sunchang meju (12 out of 23), while Aspergillus fumigatus was the next most frequently isolated strain (7 out of 23). In contrast, it was found that Fusarium asiaticum, Aspergillus sydowii, and Arthrinium sp. were the minor fungal strains in meju.

  • Non-aflatoxigenic Aspergillus oryzae and aflatoxigenic A. flavus cannot be clearly identified by partial sequencing of the internal transcribed spacer (ITS) and 18S ribosomal ribonucleic acid (18S rRNA) regions. This study aimed to compare the accuracy among three aflatoxin detection methods using ultra-performance liquid chromatography (UPLC), high-performance liquid chromatography (HPLC), and an enzyme-linked immunosorbent assay (ELISA) kit and to select the non-aflatoxigenic Aspergillus sp. isolated from soybean paste. All analytical methods were suitable according to the international standards of Codex Alimentarius FAO-WHO (CODEX) or the Ministry of Food and Drug Safety (MFDS). UPLC exhibited the best of limit of detection (LOD) and limit of quantification (LOQ). Based on UPLC, HPLC, and the ELISA kit assay, the P5 and P7 strains isolated from soybean paste had 1,663.49, 1,468.12, and >20 ㎍/kg and 1,470.08, 1,056.73, and >20 ㎍/kg, respectively, detected and re-identified as A. flavus. In contrast, the P3 and P4 strains (A. oryzae), which were detected below the MFDS standards in all assays, were confirmed as non-aflatoxigenic fungi. Among the methods evaluated for quantitative analysis of aflatoxin, UPLC and HPLC are superior in terms of accuracy, and the ELISA kit rapidly detects low concentrations of aflatoxin. Furthermore, this study demonstrates that any Aspergillus sp. isolated for use as a fermentation starter should be analyzed for potential aflatoxin production using UPLC and HPLC for accurate quantitative analysis or ELISA for the rapid detection of low-level concentrations of aflatoxin.

Studies on the Histones of the Genus Rhizopus (Rhizopus속의 histones에 관한 연구)

  • The chromatin of all higher eukaryotic cells contains a group of very basic low-mole-cular weight proteins, the histones. But much less is known about histones in lower eukaryotes. Our purpose was to study the histones of the genus Rhizopus. After isolation and purification of nucleoprotein the basic nucleoproteins were analyzed by gel electrophoresis, in sodium dodecyl sulfate as well as acid/urea gels and compared with calf thymus histones. Their electrophoretic mobility in polyacrylamide gel indicate that they are histone homologous, although not identical, to the H2A, H2B, H3 and H4 histones of mammals with the exception of H1. The result suggests that Rhizopus thus appears to contain histone proteins which are homologous to the histones from in higher eukaryotes. The similarity between the calf thymus histone H1 and the Rhizopus high band group remains to be discussed.

Establishment of hot water extraction conditions for optimization of fermented Smilax china L. using response surface methodology (반응표면분석에 의한 발효 청미래덩굴(Smilax china L.) 잎 열수 추출조건의 최적화)

  • In this study, we investigated the contents of total polyphenol (TP), total flavonoid, and absorbance at 475 nm ($OD_{475}$) which may produced in solid-fermented leaf of Smilax china L. by Aspergillus oryzae as a new functional components with reddish brown color, contents of water soluble substance (WSS), electron donating ability (EDA), Hunter $L^*$, $a^*$, $b^*$ values, sensory overall acceptability (OA) and also, the inhibitory activities (XOI and AOI) against partial purified xanthine oxidase (XO) and aldehyde oxidase (AO) from rabbit liver which were well known to relate the gout, and alcoholic liver disease, respectively in order to optimize water extraction using response surface methodology (RSM). All the $R^2$ values of the second-order polymonials ranged from 0.85 to 0.98, except for the EDA (0.69) and the XOI (0.78). However, the activities of the EDA and XOI were relatively high in the lower concentration of the fermented Smilax china L. leaf. The effects on the water extraction were highest in the concentration, among the dependent variables, and showed significant differences at the 1% level in the TP, TF and WSS contents and the $a^*$, $b^*$ and $OD_{475}$ values, but the OA showed significant differences at the 5% level. The optimal values of AOI, which was the most important functionality in the Smilax china L. that was predicted via RSM, were 59.48% at the 2.19% concentration, a $90.02^{\circ}C$ extraction temperature and a 4.03 minute extraction time ($R^2$: 0.93, p<0.007). The ranges of all the dependent variables of the optimal water extraction were 1.6~1.8% for the concentration, $83{\sim}93^{\circ}C$ for the temperature and 3.4~4.4 minutes for the extraction time; and the optimal water extraction conditions were a 1.7% concentration, an $88^{\circ}C$ extraction temperature and a 3.9-min extraction time.

Fermentable and Nutritional Characteristics of Brewery Meal-Based Fermented Feedstuffs Supplemented with Aspergillus Oryzae and Saccharomyces Cerevisiae (Aspergillus Oryzae 및 Saccharomyces Cerevisiae를 첨가하여 제조한 맥주박 위주 발효사료의 발효 특성 및 영양학적 특성)

  • This study was conducted to compare and evaluate fermentable and nutritional characteristics of brewery meal-based fermented feedstuffs supplemented with Aspergillus oryzae (AO) and Saccharomyces cerevisiae (SC). Experiments were divided into three treatment groups; fermented feedstuff supplemented with $1\%$ of AO(FFAO), fermented feedstuff supplemented with $1\%$ of SC(FFSC), and fermented feedstuff supplemented with $0.5\%$ of AO and $0.5\%$ of SC(FFAS). For changes of crude protein contents by 48 h fermentation, there were no significant differences among treatments. Ether extract(EE) contents were significantly increased by 48 h fermentation (p<0.05). Neutral detergent fiber(NDF) contents of FFAO, FFSC and FFAS were significantly decreased by 48 h fermentation(p<0.05), but acid detergent fiber(ADF) and acid detergent lignin (ADL) contents were not different. The pH of FFAO and FFAS was decreased more rapidly than that of FFSC(p<0.05), reaching a plateau after 24 h. Alcohol content was increased rapidly until 18 h in FFAO and was increased rapidly until 12 h in FFSC and FFAS, and alcohol content of FFAO, FFSC and FFAS was maintained constantly after 24 h. The ammonia N content of FFAO, FFSC and FFAS was 0.022, 0.073 and $0.040\%$ at 48 h, respectively, and then ammonia N was over twice higher in FFSC than in FFAO and FFAS(p<0.05). Dextrose content was increased until 6 h in FFAO but was rapidly decreased in FFSC and FFAS until 6 h(p<0.05). Lactate content was higher in FFAO and FFAS than in FFSC(p<0.05). Consequently, when we added AO in formulation of fermented feedstuff with brewery meal which moisture content was high, EE, alcohol, and lactate contents were increased, but NDF and ammonia N contents were reduced. Therefore, it is expected that AO will be effective to increase the feed value and the preservation of fermented feedstuff with a high moisture content.

Quality Properties of Capsule Type Meju Prepared with Aspergillus oryzae (Aspergillus oryzae를 이용한 캡슐형 메주의 품질특성)

  • In order to improve some problems such as contamination of undesirable mold, mycotoxin production and excessive drying on the surface of traditional meju, we developed a capsule type-meju(CM) coated with steamed black bean and Aspergillus oryzae - rice koji(0.3%, w/w) mixture to surface of traditional meju and fermented at 25$^{\circ}C$ for 14 days under 80% relative humidity. Contamination of undesirable mold on the surface of CM was not detected within 2 weeks and some genus Penicillium molds on control meju without koji were found naturally after 12 days of fermentation. The moisture content of meju was showed to be in the range of 34.7 - 29.4% being 32.7%(w/w) of mean value. Titratable acidities in CMs prepared with black bean(BCM) and soybean(SCM) were much higher than that in control meju, and BCM was similar to SCM. Free sugar content in BCM(123.98 mg%) was 10 times and 2.1 times higher than that in control meju(15.02 mg%) and SCM(59.85 mg%), respectively. Amino type nitrogen content in control meju was 147.00 mg% and its content in BCM(255.50 mg%) was 1.37 times higher than that in SCM(187.25 mg%). Total organic acid content in BCM(95.98 mg%) and SCM(1l9.98 mg%) were much higher than that in control meju(26.44 mg%), and then lactic and malic acid contents were markedly changed according to capsulation of meju. Lightness value(L) of Hunter color index was much higher in BCM than in SCM. Fatty acid composition of CM was not different as compared to control meju. Total free amino acid content in BCM(1039.70 mg%) was 4.4 times and 2.4 times higher than that in control meju(236.45 mg%) and SCM(556.07 mg%), respectively.

Damage Analysis of Rice Panicle Blast on Disease Occurrence Time and Severity

  • The structural differences between healthy and diseasedpanicle necks caused by Pyricularia oryzae were observed using electron-microscope. In the diseased panicle neck, the infection hyphae of the rice blast pathogen grew through the sclerenchymatous fiber tissue and reached to the central internal lacuna. Since the pathogen grew through the sclerenchymatous fiber tissues, the vascular bundle composed with xylem and phloem had been destroyed and finally the nutrients from the leaf and stem were not able to be transported into the grains. Infection of panicle base by the blast pathogen until 20 days after heading caused more than 50% of yield loss in both Jinmibyeo and Chucheongbyeo. There was a positive correlation between incidence of the panicle blast and rice yield losses. The regression equations between incidence of the panicle blast and yield losses were y= -3.61+496.7 ($R^2$=0.70) in Jinmibyeoand y=-3.93+520.2 ($R^2$=0.82) in Juanbyeo. The panicleblast caused deterioration of grain quality. Healthy grain rate was reduced by increase of panicle blast infection.
