Effects of Parathyroid Hormone on the Fluidity of the Plasma Membrane Vesicles of Cultured Osteoblasts

  • Intramolecular excimer formation of 1,3-di(1-pyrenyl)propane (Py-3-Py) and fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH) were used to investigate the effects of parathyroid hormone (PTH) on the bulk bilayer fluidity of the plasma membrane vesicles isolated from cultured osteoblasts (OB-PMV). In a dose-dependent manner, rat PTH-(1-34) [rPTH-(1-34)] increased the excimer to monomer fluorescence intensity ratio (I'/I) of Py-3-Py and decreased the anisotropy (r) of DPH in OB-PMV. This indicates that PTH increased both the lateral and rotational diffusion of the probes in OB-PMY. Selective quenching of DPH fluorescence by trinitrophenyl groups was utilized to examine the transbilayer fluidity asymmetry of OB-PMV. The anisotropy, limiting anisotropy, and order parameter of DPH in the inner monolayer were 0.024, 0.032, and 0.062 greater than calculated for the outer monolayer of OB-PMY. Selective quenching of DPH fluorescence by trinitrophenyl groups was also utilized to examine the transbilayer effects of PTH on the fluidity of OB-PMV. rPTH-(1-34) had a greater fluidizing effect on the outer monolayer as compared to the inner monolayer of OB-PMV. Thus, it has been proven that PTH exhibits a selective rather than nonselective fluidizing effect within transbilayer domains of OB-PMV.

  • The effect of tempering temperature on the ultrasonic propagation velocity at SCM 440 steel quenched from $870^{\circ}C$ and $1000^{\circ}C$ has been studied by metallurgical and crystallographical observation. The measurements of ultrasonic velocity were made on the specimen by appling an immersion ultrasonic pulse-echo technique with a constant frequency of 10 MHz. The quenched microstructure of this steel was a lath martensite. As the tempering temperature was increased, the martensite was transformed into the tempered martensite composed of cementite and carbide. The ultrasonic velocity increased with increasing the tempering temperature. It was thought that these were resulted from the microstructural transformation. The change of ultrasonic propagation velocity with quenching and tempering heat treatment was resulted from microstrain due to the change of internal stress. Considering these results concerning to the change of ultrasonic propagation velocity. the phenomena of microstructural transformation were estimated. Consequently, it was thought that the degree of quenching and tempered heat treatment of steel could be nondestructively evaluated with the change of ultrasonic propagation velocity.

Transbilayer Effects of n-Alkanols on the Fluidity of Phospholipid Model Membranes

  • Selective quenching of 1, 6-diphenyl-1, 3, 5-hexatriene (DPH) by trinitrophenyl groups was utilized to examine the transbilayer fluidity asymmetry of model membranes of phospholipids (SPMVPL) extracted from synaptosomal plasma membrane vesicles (SPMV). The polarization (P), anisotropy (r), limiting anisotropy $(r_\infty$), and order parameter (S) of DPH in the inner monolayer were 0.019, 0.014, 0.018, and 0.047, respectively, greater than calculated for the outer monolayer of SPMVPL. Selective quenching of DPH by trinitrophenyl groups was also utilized to examine the effects of n-alkanols on the individual monolayer structure of SPMVPL. n-Alkanols fluidized the hydrocarbon region of bulk SPMVPL and the potencies of n-alkanols up to 1-nonanon increased with carbon chain length. It appears that the potencies in bilayer fluidization increase by 1 order of magnitude as the carbon chain length increases by two carbon atoms. The cut-off phenomenon was reached at 1-decanol, where further increase in hydrocarbon length resulted in a decrease in pharmacological activity. The n-alkanols had greater fluidizing effects on the outer monolayer as compared to the inner monolayer of SPMVPL, even though these selective effects tended to become weaker as the carbon chain length increased. Thus, it has been proven that n-alkanols exhibit selective rather than nonselective fludizing effects within transbilayer domains of SPMVPL.

An estimation and radioactivity measurement for radiocarbon(14C) in the Korean nuclear power plants

  • Radiocarbon (14C), with a radioactive half-life of approximately 5730 years, poses a long-term environmental contamination risk when released into the atmosphere. The quantification analysis of its release estimates plant-specific generation rates based on factors such as plant power, core neutron flux distribution, and the volume of water exposed to this flux. Utilizing the improved estimation method, the 14C production rate for several Korean Pressurized Water Reactors (PWRs) was calculated. Also, improvements in measurement methods through sampling have also been made. These enhancements include the verification of the absorption method versus the mixing method. The results of this study indicate that plant-specific 14C production rates range from 0.213 to 0.317 TBq/yr, which are comparable to the global range observed in PWRs. Furthermore, the study evaluated a quenching correction curve for a liquid scintillation counter using two quenching correction methods: the external standard method and the internal standard method. The accuracy of these methods with 72 samples was validated with an average relative error within ±2.5%. The relative error of the mixing method, when compared to the direct absorption method, was found to be within ±20%. This finding underscores the validity of the improved measurement technique.

Development of New Reverse Micellar Microencapsulation Technique to Load Water-Soluble Drug into PLGA Microspheres

  • The objective of this study was to develop a new reverse micelle-based microencapsulation technique to load tetracycline hydrochloride into PLGA microspheres. To do so, a reverse micellar system was formulated to dissolve tetracycline hydrochloride and water in ethyl formate with the aid of cetyltrimethylammonium bromide. The resultant micellar solution was used to dissolve 0.3 to 0.75 g of PLGA, and microspheres were prepared following a modified solvent quenching technique. As a control experiment, the drug was encapsulated into PLGA microspheres via a conventional methylene chloride-based emulsion procedure. The micro­spheres were then characterized with regard to drug loading efficiency, their size distribution and morphology. The reverse micellar procedure led to the formation of free-flowing, spherical microspheres with the size mode of 88 ~m. When PLGA microspheres were prepared follow­ing the conventional methylene chloride-based procedure, most of tetracycline hydrochloride leached to the aqueous external phase: A maximal loading efficiency observed our experimental conditions was below $5\%$. Their surfaces had numerous pores, while their internal architecture was honey-combed. In sharp contrast, the new reverse micellar encapsulation technique permitted the attainment of a maximal loading efficiency of 63.19 $\pm$$0.64\%$. Also, the microspheres had smooth and pore-free surfaces, and hollow cavities were absent from their internal matrices. The results of this study demonstrated that PLGA microspheres could be successfully prepared following the new reverse micellar encapsulation technique.

Characterization of glasses composed of PbO, ZnO, MgO, and B2O3 in terms of their structural, optical, and gamma ray shielding properties

  • The amorphous glasses containing PbO, ZnO, MgO, and B2O3 have been fabricated using the melt quenching technique. The structural properties have been analysed using the Fourier-transform infrared (FTIR) and Raman spectroscopy. Derivative of Absorption Spectra Fitting (DASF) method have been used to estimate the band gap energy from the UV-Vis absorption data which decreases from 3.02 eV to 2.66 eV with increasing the concentration of the PbO.The four glass samples 0.284 and 0.826 MeV showed unique variations in terms of gamma attenuation ability. LZMB4 glass sample proved to be the mist effective in terms of shielding of gamma radiation as it requires little distance compared to LZMB3, LZMB2 and LZMB1 to attenuate. RPE revealed a raise with increase in the thickness of the material and reduces as the energy raises. TF is superior in LZMB1 compared to LZMB2, LZMB3 and LZMB4, confirming that, LZMB4 will attenuate better. The ZEff of the materials was seen falling as the energy increases, confirming that the linear attenuation coefficient of the glass materials decreases when the energy is increased. The results confirmed that, glass material LZMB4 is the best option especially for gamma radiation shielding applications compared to LZMB3, followed by LZMB2, then LZMB1.

Stydy of Pool Boiling under Steady State using Ultrasonic Measurement (초음파 측정법을 이용한 정상상태의 푸울비등 연구)

  • A recently developed new technique for measuring the fraction of wetted area has applied to pool boiling of water. The basis of the new applied technique of ultrasonic makes use of the reflection of ultrasonic from the vapour surface to measure the fraction of wetted area values. The results are the measured fraction of wetted area values in nucleate and transition boiling and the pool boiling curve for water under steady state conditions. The measurement of this paper shows a fraction of wetted areaf around 0.98 at the critical heat flux for water.

  • It is well recently recognized that quench is one of the serious problems for the integrity of superconducting magnets, which is mainly attribute to the rapid temperature rising in the magnet due to some extrinsic factors such as conductor motion, crack initiation etc. In order to apply acoustic emission(AE) technique effectively to monitor and diagnose superconducting magnets, it is essential to identify the sources of acoustic emission. In this paper, an acoustic emission technique has been used to monitor and diagnose quenching phenomenon in racetrack shaped superconducting magnets at cryogenic environment of 4.2K. For these purposes special attention was paid to detect AE signals associated with the quench of superconducting magnets. The characteristics of AE parameters have been analyzed by correlating with quench number, winding tension of superconducting coil and charge rate by transport current.

  • In this study, acoustic emission(AE) technique has been applied to detecting quench which is one of the serious peoblems to assure the integrity of superconducting coil at cryogenic temperature. The characteristics of AE parameters have been analyzed by correlating with the number of quenches, whinding tension of superconducting coil and charge rate of transport current. The quench localization was also performed using AE signals and there was also good correlation between quench current and AE parameters such as AE energy and AE events. In this study, it was confirmed that AE signals were mainly due to the conductor motion which caused by premature quenching. It was also found that optimized winding tension at superconducting coil was needed to prevent quench caused by conductor motion.

Quantitative Vapor Phase Exciplex Fluorescence Measurements at High Ambient Temperature and Pressure

  • The exciplex fluorescence technique with the TMPD (tetamethyl-Ρ-phenylene-diamine) / naphthalene dopant system was applied in a combustion-type constant-volume spray chamber. A detailed set of calibration experiments has been performed in order to quantify the TMPD fluorescence signal. It has been demonstrated that the TMPD fluorescence intensity was directly proportional to concentration, was independent of the chamber pressure, and was not sensitive to quenching by either water vapor or carbon dioxide. Using a dual heated-jet experiment, the temperature dependence of TMPD fluorescence up to 1000 K was measured. The temperature field in the spray images was determined using a simple mixing model, and an iterative solution method was used to determine the concentration and temperature field including the additional effects of the laser sheet extinction. The integrated fuel vapor concentration compared favorably with the measured amount of injected fuel when all of the liquid fuel had evaporated.
