β-Carotene, Cucumisin Content and Fruit Morphology of Melon (Cucumis melo L.) Germplasm Collections

  • Melon fruits are a year-old plant in the family Cucurbitae and are the most cultivated fruit in tropical countries. Melon flesh is an important source of anti-cancer, antioxidant effects, attracting attention as a functional food. We investigated the morphological properties, β-carotene content, and cucumisin activity of seeds of 58 melon genetic resources. Melon resources have shown various morphological properties. Melons grouped by morphological properties were the five groups. β-carotene content varied between 82.34 mg/kg, 86.75 mg/kg, 25.56 mg/kg, 86.25 mg/kg, and 54.65 mg/kg. Between β-carotene, cucumisin activity and other quantitative fruit morphological properties, the color of the pulp and the firmness of the pulp had a significant amount of correlation between the β-carotene content. However, cucumisin activity and β-carotene content had significant negative correlation, and the color of the fruit and shape of the fruit had significant negative correlation with β-carotene content and activity of cucumisin. Our study vindicated that high diversity in melon morphological characters of genetic sources that provide beneficial baseline data for the future and in the activity of β-carotene and cucumisin, and these results could predict indicators of β-carotene content by the length of leaf, the width of fruit, the length of joint, and the length of seed, and collectively the morphological properties of melons could help predict indicators of β-carotene content and help develop functional sarcoma and farmhouse cultivation.

A Study on the Characteristics of Re-Organized Shortform Contents (재가공형 숏폼 콘텐츠의 특성 연구)

  • The purpose of this study is to clarify the meaning and characteristics of re-organized shortform contents, which is centered on media companies to edit and service existing broadcast content. For this, KBS, MBC, SBS, JTBC, and tvN's entire drama videos and representative entertainment programs opened on the Naver TV platform from 2014 to 2021 were selected for analysis and a synchronic and diachronic approach was conducted at the same time. As a result of the analysis, quantitative and qualitative expansion was made, with the number and form of videos provided by both dramas and entertainment programs diversifying from a synchronic approach. In particular, in the case of special videos, the meaning as independent content was also strengthened, such as sequencing centered on characters, themes, and materials. It was confirmed that thumbnails and titles were also formalized as tags as paratexts that act as curation for searches. From a diachronic point of view, it was found that re-organized shortform contents is considered to be character-oriented contents and independent viewing context through comparison with real-time views and original videos. This study is significant as an attempt to capture the meaning and phase change of shortform contents, which was considered incidental.

Genetic Insights into Domestication Loci Associated with Awn Development in Rice

  • Rice (Oryza sativa L.) is a widely studied domesticated model plant. Seed awning is an unfavorable trait during rice harvesting and processing. Hence, awn was one of the target characters selected during domestication. However, the genetic mechanisms underlying awn development in rice are not well understood. In this study, we analyzed the genes for awn development using a mapping population derived from a cross between the Korean indica cultivar 'Milyang23' and NIL4/9 (derived from a cross between 'Hwaseong' and O. minuta). Two quantitative trait loci (QTLs), qAwn4 and qAwn9 were mapped on chromosome 4 and 9, respectively, increased awn length in an additive manner. Through comparative sequencing analyses parental lines, LABA1 was determined as the causal gene underlying qAwn4. qAwn9 was mapped to a 199-kb physical region between markers RM24663 and RM24679. Within this interval, 27 annotated genes were identified, and five genes, including a basic leucine zipper transcription factor 76 (OsbZIP76), were considered candidate genes for qAwn9 based on their functional annotations and sequence variations. Haplotype analysis using the candidate genes revealed tropical japonica specific sequence variants in the qAwn9 region, which partly explains the non-detection of qAwn9 in previous studies that used progenies from interspecific crosses. This provides further evidence that OsbZIP76 is possibly a causal gene for qAwn9. The O. minuta qAwn9 allele was identified as a major QTL associated with awn development in rice, providing an important molecular target for basic genetic research and domestication studies. Our results lay the foundation for further cloning of the awn gene underlying qAwn9.

A Review of Multivariate Analysis Studies Applied for Plant Morphology in Korea (국내 식물 형태 연구에 사용된 다변량분석 논문에 대한 재고)

  • A review was given of the role of traditional morphometrics in plant morphological studies using 54 published studies in three major journals and others in Korea, such as Journal of Korean Forestry Society, Korean Journal of Plant Taxonomy, Korean Journal of Breeding, Korean Journal of Apiculture, Journal of Life Science, and Korean Journal of Plant Resources from 1997 to 2008. The two most commonly used techniques of data analysis, cluster analysis (CA) and principal components analysis (PCA) with other statistical tests were discussed. The common problem of PCA is the underlying assumptions of methods, like random sampling and multivariate normal distribution of data. The procedure was intended mainly for continuous data and was not efficient for data which were not well summarized by variances or covariances. Likewise CA was most appropriate for categorical rather than continuous data. Also, the CA produced clusters whether or not natural groupings existed, and the results depended on both the similarity measure chosen and the algorithm used for clustering. An additional problems of the PCA and the CA arised with both qualitative and quantitative data with a limited number of variables and/or too few numbers of samples. Some of these problems may be avoided if a certain number of variables (more than 20 at least) and sufficient samples (40-50 at least) are considered for morphometric analyses, but we do not think that the methods are all mighty tools for data analysts. Instead, we do believe that reasonable applications combined with focus on objectives and limitations of each procedure would be a step forward.

The Test of Combining Ability and Heterosis on the Silkworm(Bombyx mori) Breeding (누에 견.사질에 관한 잡종강세 및 조합능력검정)

  • The study was conducted to obtain the genetic information on heterosis and combining ability of the quantitative characters for F1 hybrid breeding in silkworms. Six parental varieties and each set of 30 diallel crosses in F1's were used as materials, and bred on the randomized complete block design with three replications. Fourteen characters were observed with the twenty samples in each tray. The data were analyzed for (1) heterosis and combining ability in F1 hybrid. The heterosis in the weight and the length of cocoon showed positively high at 24.51%, and 23.4%, respectively and the weight of the whole cocoon as well as the weight of the whole cocoon layer showed a siginificant heterosis ranging from 15.56% to 15.71% and from 17.14% to 19.01%, but the fifth and the total instar period showed negative heterosis. It was found that the combination between, C70XRomogua and N9 X Romogua showed highly a negative heterosis on the fifth instar period and for the cocoon weight. The female of N9+Sansuian and the male of Romogua X Sansurian have a high heterosis effect, on the cocoon shell weight, and Sansurian X Romogua(reciprocal) on the length and the weight of cocoon filament with no regard to sexuality. The significant maternal and cytoplasmic effect on heterosis of the cocoon weight and the cocoon shell weight were observed with the combinations, N9 X C5, N63 X C70 and on the length of the cocoon filament with the combinations, Sansurian X N63, Sansurian X C5, Sansurian X C70 and N9 X C70, N63 X C70 on the weight of cocoon filament. As mean squared of GCA, SCA and RCA were significant with these combining ability for all characters resulted from additive and non-additive altogether and there is a significant difference between reciprocals. Sansurian showed a negative GCA effect on the fifth and total larval duration, but the higher positive GCA effects took places with varieties N9 and C5 on the length, width, weight of cocoon, cocoon shell weight, percentage of cocoon shell weight, length and weight of cocoon filament, percentage of raw-silk with no regard to both generations and silkworm sexuality. The values of SCA between the cross combinations varied generation-wise and sex-wise. It was shown that SCA value for the fifth instar period was highly negative for Sansurian X C70, Romogua X C70, Sansurian X C5, Romogua X C5, but it was positive effect on the cocoon weight, cocoon shell weight with N9 X C5, and C70 X Sansurian, on the length of cocoon filament with N9 X C5, Romogua X Sansurian on the weight of cocoon filament between Romogua and N63 and on the percentage of raw-silk between the combination of Sansurian X Romoga.

Taxonomic reconsideration of the Philadelphus schrenkii complex (고광나무분류군(Philadelphus schrenkii complex)의 실체에 대한 형태 고찰)

  • The Philadelphus schrenkii complex including P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, P. tenuifolius, P. pekinensis, P. seoulensis, P. lasiogynus, and P. scaber, is distributed in China, Korea, japan, and far eastern Russia. Their taxonomic delimitations are obscure because quantitative characters, such as presence/absence and density of hair on leaf, petiole, style, disc, calyx-tube, and pedicel, are highly variable with no apparent characteristics that consistently distinguish from each other within the group. Thus, these morphological characters are reexamined using univariate analyses. P. pekinenesis is distinct from other taxa in having glabrous disk, calyx, pedicel, and leaf blade. In addition, the number of flowers in an inflorescence [(5)7-9(11) vs 5-7 in other taxa], smaller calyx tube [2.5-3 mm vs. (2.5)3-4(5.9) in other taxa], and slightly divided styles help distinguish P. pekinenesis from other related taxa in Korea. However, P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, and P. lasiogynus show continuous variation in the pubescence of disk, calyx, pedicel, and leaf blade, and this may reflect a lack of divergence among them. Particular emphases are placed on P. tenuifolius and P. seoulensis, because of the high degree of morphological intergradation with respect to the degree of pubescence in disk, calyx, pedicel, and leaf blade and their inconsistent circumscription across various systematic treatments. P. scaber, distributed in the southern Korea, has often been treated as an independent species having exfoliated branchlets, distinct serrated leaf margin, and recurved styles. However, this current study suggests that P. scaber should be recognized as a form of P. tenuifolius because these distinguishing characters are continuous between P. tenuifolius and P. scaber. In geographical distribution, P. pekinensis is mainly distributed in the southern part of Korea, while P. tenuifoilus and P. schrenkii are commonly found all over the country.

  • Silkworm is known to be damaged under the unfavorable environmental conditions. Recently, it has been often argued that nicotine is a main factor to affect the silkworm nutritional balance and to result in decrease of quantitative characters, and unbalance of larval nutrition. Especially in spring season, there is tendency to produce nicotine damage to silkworm fed with the mulberry leaf near to tobacco field early cultivated in mulching system. In these connections, the trial was made in the different pots placing by the different distance 10rn, 20m, 40m, and 60m in the direction of east-north from the tobacco field. Location of pot near to the tobacco fold is 40m below the tobacco Held and gradually higher than the tobacco field by 10 degree of slope. In addition, when the mulberry field is divided into two areas from the east-south to west-north, southern east zone is lowering down below 10 degree of slope and has 36% of east-south wind, 21% of south-east wind and 16% of south wind, etc. Pots are left on the mulberry field from the completion of lower part leaf harvest to just before upper part leaf harvest is completed for 32 days. Then silkworm at 4th-5th stage was fed with the leaf grown during the period and larval characters are statistically analyzed. The results are summarized as follows: 1. 10-40m distant pot statistically at the same level has the longest larval duration and 60m distant pot is. not significant at 5% level. Whole larval duration has same tendency as in the 5th larval duration. 2. Percentage of delayed molting larvae at the 5th stage is increased in 10m distant pot and there is no significance statistically in other pots, as compared with that of control. 3. It is not showed that there is no damage to mortality of the old larvae in 60m distant pot. 4. 10-20m distant pot produces a low pupation and others are at the same level statistically at 5% significance, 5. It is showed that nicotine damage is occurred in single cocoon freight and cocoon shell weight up to 60m distant pot, rather higher damage in cocoon shell weight than in single cocoon weight. 6. It is showed that cocoon yield from 10, 000 molted larvae including double cocoon is decreased up to 40m distant pot far from the tobacco field in which 40% of cocoon yield in 10m distant pot by 7kg, 26% of cocoon yield in 20m distant pot by 3.9kg, 24% of cocoon yield in 40m distant pot by 3.6kg, and 12% of cocoon yield in 60m distant pot by 1.8kg, as compared with control. However, there is no significance statistically at 5% level in 60m distant pot. With these above results, it is concluded that nicotine da mage could be occurred in cocoon yield and cocoon quality up to 60m distant far from the tobacco field.

  • The seventeen barnyardgrass [Echinochloa crus-galli (L.) Beauv.] accessions, which were collected national-widely in 1986 and selected two times through 1987, were experimented at 1988. To study the degree of ecological variations of the collected accessions of native barngardgrass species as a weed, the experiment was conducted with Wagner pots in size of 1/5000a and under P.E. film house. Among 57 characters, 100 grains weight, rate of tiller increase from transplanting to 30 days, the number of the maximum tillers and rate of dry weight increase from 15 DAS to 22 DAS were become intra-specific variarions as higher coefficient of varation than 30%, the size of the first leaf, fresh weight and dry 22 seedlings, rate of productive tiller, panicle number and tillering divergence made intra-specific variations as more and less C. V. than 25%, while the required days upto heading since seeding at June 6. rate of fresh/dry weight and plant height of 22 days old seedling made intra-specific variations as the coefficient of variation of about 50%. Among the attributes the color in leaf, leaf sheath, mid-ribs, node, spike, grain, and plant type, length of leaf marginal tooth, grain shape, purple bend on leaf were seemed useful to indentify the accessible specific.

The Variation of Leaf Characterics in 6 Natural Populations of Stewartia koreana Nakai (노각나무 6개 천연집단(天然集團)의 엽형질(葉形質) 변이(變異))

  • In order to examine the natural distribution variations between groups of the Stewartia koreana, the leaf form characteristics of the investigation sites were analyzed by each group. As a result, the Mt. Kumsan group showed a smaller value in leaf length, width, area, and the number of veins, but not in the petiole length and serration number. Among each character, the coefficient of variation(CV) of the characters excluding petiole length and leaf area was in a comparatively narrow range, from 11.6~17.4%. On the other hand, the CV of petiole length and leaf area between the groups was 34.9% and 28.4% respectively. The CV of these characters within the group was also extraordinary- petiole length showed 29.5~42% and leaf area showed 27.7~40.7%. Also, the simple correlation analysis between 12 leaf characteristics showed that the correlation between leaf width and leaf area was high (r=0.975). The correlations between leaf length and leaf area, between leaf length and leaf width were 0.971 and 0.969, respectively. A negative correlation between angle of leaf base and ratio of leaf length to leaf width was discovered (r= -0.843), meaning that the ratio of leaf length to leaf width decreases as angle of leaf base increases. A cluster analysis was enforced among leaf characteristics of the selected group as a standard on the similarity of quantitative, qualitative measurements. The results showed that at a 0.4 distance level, the subjects could be classified into 4 groups. Group 1 was the Mt. Jogyesan and Mt. Kayasan group, group 2 was Mt. Paegunsan, group 3 was Mt. Unmunsan and Mt. Mudungsan, and group 4 was Mt. Kumsan. At a distance level of 0.6, the subjects were classified into two groups. Group 1 was the Mt. Ktimsan group and group 2 was Mt. Mudungsan, Unmunsan, Paegunsan, Kayasan, and Cogyesan. Especially the Mt. Kumsan group had the smallest value in the leaf characteristics of leaf length, width, area, and the number of veins, showing an obvious difference from the other five groups. There were five principal components that had a meaningful eigenvalue over 1.0 among the 12 extracted components. The explanatory power of the top two main components (leaf length and width) on the total variation was 52.7%. The explanatory power was 91.3% when all 5 main components were included.

Microsatellite Markers Linked to Quantitative Trait Loci Affecting Fatness in Divergently Selected Chicken Lines for Abdominal Fat

  • Abdominal fat characters are complex and economically important in the poultry industry. Their selection may benefit from the implementation of marker-assisted selection (MAS). The objective of this study was to identify the markers linked to QTL responsible for fatness traits. The Northeast Agricultural University broiler lines divergently selected for abdominal fat content (NEAUHLF) were used in the study. A total of 596 individuals from the divergent tails from the 6th to the 10th generations were genotyped at 23 microsatellite markers on chromosome 1. The differences of allele frequencies of all marker alleles between the divergent tails across the five generations were recorded. The allele frequencies of five markers, including LEI0209, LEI0146, MCW0036, ADL328 and MCW0115, had significant differences between the two tails in all five generations. The resulting p-values using Fisher's exact test on eleven markers, containing MCW248, MCW0010, MCW0106, LEI0252, LEI0068, MCW0018, MCW0061, LEI0088, MCW200, MCW283 and ROS0025, had a decreasing tendency from the 6th to the 10th generation. Statistical analysis showed that polymorphisms of the eight markers, including LEI0209, LEI0146, ROS0025, MCW0115, MCW0010, MCW0036, MCW283, ADL328, were significantly (p<0.0011) or suggestively (p<0.05) associated with abdominal fat content (AFW and AFP) across generations. It is concluded that the eight markers could be associated with the QTL affecting the deposition of abdominal fat in broiler chickens.
