Screening Analysis of 10 Adrenal Steroids by Matrix-Assisted Laser Desorption Ionization-Tandem Mass Spectrometry

  • Defective synthesis of the steroid hormones by the adrenal cortex has profound effects on human development and homeostasis. Due to the time-consuming chromatography procedure combined with mass spectrometry, the matrix-assisted laser desorption ionization method coupled to the linear ion-trap tandem mass spectrometry (MALDI-LTQ-MS/MS) was developed for quantitative analysis of 10 adrenal steroids in human serum. Although MALDI-MS can be introduced for its applicability as a high-throughput screening method, it has a limitation on reproducibility within and between samples, which renders poor reproducibility for quantification. For quantitative MALDI-MS/MS analysis, the stable-isotope labeled internal standards were used and the conditions of crystallization were tested. The precision and accuracy were 3.1~35.5% and 83.8~138.5%, respectively, when a mixture of 10 mg/mL ${\alpha}$-cyano-4-hydroxycinnamic acid in 0.2% TFA of 70% acetonitrile was used as the MALDI matrix. The limit of quantification ranged from 5 to 340 ng/mL, and the linearity as a correlation coefficient was higher than 0.988 for all analytes in the calibration range. Clinical applications include quantitative analyses of patients with congenital adrenal hyperplasia. The devised MALDI-MS/MS technique could be successfully applied to diagnosis of clinical samples.

Near-Field Detection of Aβ Proteins Using Micro Beads

  • In this paper, we present the possibility of quantification analysis for $A{\beta}$ captured by micro beads using Near-filed detection. In order to evaluate detection efficiency, detected signals were compared with different sizes of micro beads and a varied number of micro beads. Also, $A{\beta}$ deposits and $A{\beta}$ binding to micro beads were measured, therefore, we observed the $A{\beta}$ deposit and light scattering around the surface of micro beads induced by attached $A{\beta}$. This method can be used for quantitative analysis for not only the number of $A{\beta}$, but also the binding ratio of $A{\beta}$ to micro beads.

  • As the convergence among technologies is reorganizing industry sectors, it is quite important to evaluate the probability of technological convergence, and to analyze how the technologies in a certain industry sector affect the same or other industry sectors. As a result, the large number of studies have been focused on the industry convergence. However, most of them have dealt mainly with case studies or strategy and policies and few efforts have been made to study it using quantitative data. The investigation of industry convergence using quantitative data will help understand the characteristics of industry and forecast the future of the industry from an objective point of view. Therefore, this research proposes a methodology to evaluate the possibilities of industry convergence using patent data. In particular, we emphasize the possibilities of technology convergence and suggest a technology relationship matrix to evaluate the technology convergence, as an antecedent of industry convergence. The feasibility and utility of the suggested methodology was verified with a case study on the convergence of IT and BT. The research results are expected to provide a useful guideline for developing a measure of convergence.

  • 현대 사회는 인터넷과 SNS를 통해 발생하는 복합적이며 대량의 데이터를 수집 집계 분석하는 빅데이터 처리 기술이 여러 분야에서 요구되고 있다. 그 중 대표적인 활용분야가 기업이나 대학에 대한 평판을 평가하는 평판시스템이다. 대학평판을 측정하고 수치화하기 위해서는 공정하고 객관적인 자료와 효율적인 데이터 처리가 무엇보다도 중요하다. 이를 위하여 공공데이터 지표를 활용하여 정량지수를 구하였고, 뉴스 기사를 활용한 감성분석을 통해서 정성지수를 구한 후 혼합 대학평판 지수를 산출하였다. 본 논문에서는 대학평판을 측정하기 위하여 정량지수로 객관성을 확보하면서 감성적 평판을 반영한 혼합 대학평판 지수를 산출하였고 이를 바탕으로 혼합 대학평판 시스템을 제안하였다.

공통특허분류 분석을 활용한 안전기술융합분야 탐색 : Association Rule Mining(ARM) 접근법 (Exploring Convergence Fields of Safety Technology Using ARM-Based Patent Co-Classification Analysis)

  • As the safety fields are expanding to a variety of industrial fields, safety technology has been developed by convergence between industrial safety fields such as mechanics, ergonomics, electronics, chemistry, construction, and information science. As the technology convergence is facilitating recently advanced safety technology, it is important to explore the trends of safety technology for understanding which industrial technologies have been integrated thus far. For studying the trends of technology, the patent is considered one of the useful sources that has provided the ample information of new technology. The patent has been also used to identify the patterns of technology convergence through various quantitative methods. In this respect, this study aims to identify the convergence patterns and fields of safety technology using association rule mining(ARM)-based patent co-classification(co-class) analysis. The patent co-class data is especially useful for constructing convergence network between technological fields. Through linkages between technological fields, the core and hub classes of convergence network are explored to provide insight into the fields of safety technology. As the representative method for analyzing patent co-class network, the ARM is used to find the likelihood of co-occurrence of patent classes and the ARM network is presented to visualize the convergence network of safety technology. As a result, we find three major convergence fields of safety technology: working safety, medical safety, and vehicle safety.

기능적 자기공명영상 사용유무에 따른 확산텐서영상 분석의 유효성 평가 (Evaluation of Quantitative Effectiveness of MR-DTI Analysis with and without Functional MRI)

  • Purpose: This study was conducted in order to evaluate the quantitative effectiveness of region of interest (ROI) setting in MR-DTI analysis with and without fMRI activation results. Methods: Ten right-handed normal volunteers participated in this study. DTI and fMRI datasets for each subject were obtained using a 1.5T MRI system. For neural fiber tracking, ROIs were drawn using two methods: The drawing points were located in the fMRI activation areas or areas randomly selected by users. In this study, the neural fiber tract targeted the corticospinal tract (CST) Quantitative analyses were performed and compared. The pixel numbers passing through the fiber tract in the individual brain volume were counted. The ratios between the ROI pixel numbers and the extracted fiber pixel numbers, and the ratios between the fiber pixel numbers and the whole-brain pixel numbers were also calculated. Results: According to our results, extracted CST fiber tract in which the ROI was drawn with fMRI activation areas showed higher distribution than drawing the ROI by users' hands. In addition, the quantitatively measured values represented higher pixel distribution: The counted average pixel numbers were 4553.8 and 1943.3. The average ratios of the ROI areas were 33.87 and 22.52. The average percentages of the individual whole-brain volume numbers were 2.06 and 0.87. Conclusion: Results of this study appear to indicate that use of this method can allow for more objectives and significant for study of the recovery of neural fiber mechanisms and brain rehabilitation.

나눔교육을 통한 아동의 변화 연구: Multiple Convergence Model의 적용 (Effects of Philanthropy Education on Elementary School Students in Korea : Analysis Using a Multiple Convergence Model)

  • 본 연구는 다중수렴모델(Multiple Convergence Model)을 적용하여 나눔교육을 통한 아동의 변화를 보다 다각적이고 심층적으로 이해해 보고자 한다. 양적 연구를 통해서는 초등학교 고학년 아동을 대상으로 친사회성 행동 향상에 초점을 두고 변화를 살펴보았으며, 질적 연구를 통해서는 나눔교육 후의 아동 변화에 대해 아동, 학부모, 교사의 시각에서 다차원적으로 변화의 내용과 의미를 분석하였다. 분석결과, 나눔교육을 받은 고학년 아동들은 친사회적 행동에서 통계적으로 유의미한 긍정적 변화가 나타났으며, 아동, 학부모, 교사의 시각에서도 그러한 변화는 확인되었다. 또한 양적 결과를 질적 결과를 통해 확증하는 수렴적 방법을 활용해 종합해 본 결과, 나눔교육이 아동에게 매우 긍정적인 의미있는 변화를 유발하는 것으로 나타났다. 이러한 결과를 바탕으로 본 연구는 나눔교육의 주요 장(주체)인 가정 학교 지역사회에서 나눔교육의 효과를 극대화시키기 위한 방안들을 논의한다. 본 연구는 '나눔교육을 통한 아동의 변화'라는 현상에 대해 포괄적 심층적 이해 제고를 위해 국내 최로로 다중수렴모델을 적용하는데, 이는 사회복지연구방법의 지평을 확대하는 효과, 즉 새로운 연구방법론 활용의 기초를 마련할 것으로 기대한다.

Comparison of digital PCR platforms using the molecular marker

  • Assays of clinical diagnosis and species identification using molecular markers are performed according to a quantitative method in consideration of sensitivity, cost, speed, convenience, and specificity. However, typical polymerase chain reaction (PCR) assay is difficult to quantify and have various limitations. In addition, to perform quantitative analysis with the quantitative real-time PCR (qRT-PCR) equipment, a standard curve or normalization using reference genes is essential. Within the last a decade, previous studies have reported that the digital PCR (dPCR) assay, a third-generation PCR, can be applied in various fields by overcoming the shortcomings of typical PCR and qRT-PCR assays. We selected Stilla Naica System (Stilla Technologies), Droplet Digital PCR Technology (Bio-Rad), and Lab on an Array Digital Real-Time PCR analyzer system (OPTOLANE) for comparative analysis among the various droplet digital PCR platforms currently in use commercially. Our previous study discovered a molecular marker that can distinguish Hanwoo species (Korean native cattle) using Hanwoo-specific genomic structural variation. Here, we report the pros and cons of the operation of each dPCR platform from various perspectives using this species identification marker. In conclusion, we hope that this study will help researchers to select suitable dPCR platforms according to their purpose and resources.

냉동제조 시설의 암모니아 누출사고 위험 분석 (Risk Analysis of Ammonia Leak in the Refrigeration Manufacturing Facilities)

  • 국내 냉동제조시설에서 암모니아 누출사고가 여전히 발생하고 있음을 볼 수가 있다. 암모니아는 가연성가스 및 독성가스이므로 사고 발생할 때 인체와 생태계에 큰 피해를 일으킬 수 있다. 국내 냉동제조시설의 암모니아 사고유형을 파악한 후 사고시나리오를 선정하여 피해범위를 산정하고 사고 빈도와 위험도를 분석하여 사고 피해를 최소화하는 대책 수립이 필요하다. 본 연구에서는 정량적 위험성 평가(QRA: quantitative risk assessment)의 분석 방법에 따라 암모니아 냉동시스템의 리시버 탱크에 대한 위험도를 분석하였다. 시나리오 분석 조건은 화학물질관리법에서 정하는 '사고시나리오 선정에 관한 기술지침' 및 미국 화학공정안전센터(CCPS: center for chemical process safety)의 가이드라인에 따라 정하였다. DVN사의 SAFETI 프로그램을 활용하여 시나리오에서 선정된 모든 사고 영향범위를 산정하고 빈도 분석을 통하여 리시버 탱크에 대한 위험도를 도출하였다. 빈도 값은 사건수 분석(ETA: event tree analysis)기법과 Part count 기법을 활용하였다. 연구 결과 암모니아 냉동시스템의 개인적 위험도는 7.71E-04/yr으로 도출되었으며, 사회적 위험도 1.17E-03/yr으로 도출되었다. 도출된 위험도는 국제 화재방지협회 (NFPA: national fire protection association)의 ALARP (as low as reasonably practicable) 범위를 적용하여 위험도의 적합성을 확인하였으며, 본 연구에서 제안한 위험도 산정 방법이 사고 피해 최소화 방안을 모색하는데 활용된다면 보다 좋은 결과가 도출될 것으로 판단된다.
