Antioxidant Activity and Protective Effect of Leaf Extract from Diospyros lotus on Oxidative Stress of Red Blood Cells (고욤 잎 추출물의 항산화 활성 및 적혈구 산화적 손상에 대한 보호 효과)

  • This study was to evaluate the antioxidant properties of the leaf extracts of Diospyros lotus (DLE) on the chemical-induced free radical and rat red blood cell (RBC) oxidative damage in vitro. DLE were prepared by extracting with water. DLE showed the high antioxidant activities on the scavenging of 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid)-induced radicals. An antioxidant activities of DLE was similar to the reference antioxidant butylated hydroxytoluene (BHT) and (${\pm}$)6-hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid (Trolox). Reducing power of $1,000{\mu}g/mL$ DLE also was similar to the vitamin C. In RBC, oxidative hemolysis induced by the aqueous peroxyl radical generator (2,2'-Azobis (2-methylpropionamidine) dihydrochloride (AAPH)) were significantly suppressed by DLE in a dose-dependent manner. Furthermore, DLE prevented the depletion of cytosolic antioxidant glutathione in RBC damaged with AAPH. These results suggest that DLE may has value as natural product with its high quality antioxidant properties against oxidative stress.

Identification and Functional Analysis of RelA/SpoT Homolog (RSH) Genes in Deinococcus radiodurans

  • To identify the global effects of (p)ppGpp in the gram-positive bacterium Deinococcus radiodurans, which exhibits remarkable resistance to radiation and other stresses, RelA/SpoT homolog (RSHs) mutants were constructed by direct deletion mutagenesis. The results showed that RelA has both synthesis and hydrolysis domains of (p)ppGpp, whereas RelQ only synthesizes (p)ppGpp in D. radiodurans. The growth assay for mutants and complementation analysis revealed that deletion of relA and relQ sensitized the cells to $H_2O_2$, heat shock, and amino acid limitation. Comparative proteomic analysis revealed that the bifunctional RelA is involved in DNA repair, molecular chaperone functions, transcription, the tricarboxylic acid cycle, and metabolism, suggesting that relA maintains the cellular (p)ppGpp levels and plays a crucial role in oxidative resistance in D. radiodurans. The D. radiodurans relA and relQ genes are responsible for (p)ppGpp synthesis/hydrolysis and (p)ppGpp hydrolysis, respectively. (p)ppGpp integrates a general stress response with a targeted re-programming of gene regulation to allow bacteria to respond appropriately towards heat shock, oxidative stress, and starvation. This is the first identification of RelA and RelQ involvement in response to oxidative, heat shock, and starvation stresses in D. radiodurans, which further elucidates the remarkable resistance of this bacterium to stresses.

Type of Coping with Stress of Nursing Students (간호학과 학생의 스트레스 대처 양상)

  • This study designed to know subjective structure about type of coping with stress of nursing students using Q-methodology. The statement positions on the sorting continuum were scored from +4(representing the highest level of agreement) through 0(at the neutral position on the continuum) through -4(representing the highest level of disagreement). Factor analysis of the data was undertaken using PC QUANL software to determine if any patterns were discernible. The four types extracted eigenvalues greater than 1.0 and explained approximately 52.14% of the variation in responses(31.73, 10.13, 5.73 and 4.65% respectively).: (1) Positive resolve type, (2) Ignore problem type, (3) Self resolution type and (4) Conversation type. The study results can be utilized in both an effective nursing education and clinical practice.

  • The flow characteristics in a bifurcated duct are investigated experimentally. Physical properties such as mean velocity vectors, mean x-y stress distributions, mean vorticity and total pressure distributions are Obtained for three different Reynolds numbers(578, 620, 688) Using PIV measurements and CFD analysis. Also, three different rates of discharge Q=26.11 l/min, Q=28.11 $\ell$/min, Q=31.17 $\ell$/min) were selected foy experimental conditions. The results of this study would be useful to the engineer in designing the flow systems for heating, ventilation, air conditioning and wastewater purification plants.

Optimal response of conical tool semi angle in ductile metal sheets indentation and its governing mechanics

  • The nonlinear dependence aspect of various conical tool indentation parameters leading to an optimum tool semi angle value for easiest perforation is plotted and discussed explicitly in this work with the conclusion that tool angle has an optimum response towards most of the indentation parameters. Around this optimum angle, the aluminium sheets showed minimum fracture toughness as well as minimum work input to overcome the offered resistance. At the end, the mechanism leading to this phenomenon is presented with the conclusion that plastic flow dominates as the dimple semi cone angle reaches 35 and both pre and post plastic flow perforations lead the tool semi cone angle value towards this dimple cone semi angle of plastic flow initiation for its optimum performance. It is also concluded that specimen material failure is solely under tensile hoop stress and hence results into radial cracks initiation and propagation.

  • This paper deals with a novel application of the Generalized Differential Quadrature (G.D.Q.) method to the linear elastic static analysis of isotropic rotational shells. The governing equations of equilibrium, in terms of stress resultants and couples, are those from Reissner-Mindlin shear deformation shell theory. These equations, written in terms of internal-resultants circular harmonic amplitudes, are first put into generalized displacements form, by use of the strain-displacements relationships and the constitutive equations. The resulting systems are solved by means of the G.D.Q. technique with favourable precision, leading to accurate stress patterns.

Effect of Stress History on CPT-DMT Correlations in Granular Soil (응력이력이 사질토의 CPT-DMT 상관관계에 미치는 영향)

  • Stress history increases the residual horizontal stress of granular soil and, consequently, the penetration resistance. This study analyzes the effect of stress history on the cone resistance ($q_c$), horizontal stress index ($K_D$) and dilatometer modulus ($E_D$) of CPT and DMT from calibration chamber specimen in OC as well as NC state. Test results show that the normalized cone resistance by mean effective stress correlates well with the relative density and the state parameter, whereas the normalized cone resistance by vertical effective stress is a little affected by stress history. The influence of stress history is more reflected on $K_D$ than $E_D$ and $q_c$. The $K_D/K_0$, in which the effect of stress history on $K_D$ is compensated by the at-rest coefficient of earth pressure, $K_0$, is related to relative density, state parameter and the normalized cone resistance by mean effective stress. It is also observed that the normalized dilatometer modulus by mean effective stress ($E_D/{\sigma}_m'$) shows a unique correlation with the state parameter, regardless of stress history.

  • Small punch-creep(SP-Creep) test technique has been applied for evaluating the creep characteristics for high temperature materials. However, in order to evaluate the damage and predict the remaining life, it is necessary to establish a quantitative correlation between SP-Creep and uniaxial-creep test results. This paper presents analytical and experimental results of useful correlation between SP-Creep and uniaxial-creep properties for 9Cr1MoVNb steel at $600{\sim}650^{\circ}C$ in terms of stress(load) and activation energy during creep deformation. Especially, the activation energy obtained from SP-Creep test is linearly related to that from uniaxial-creep test at $650^{\circ}C$ as follows: $Q_{sp-p}{\fallingdotseq}1.37\;Q_{TEN},\;Q_{sp-{\sigma}}{\fallingdotseq}1.53\;Q_{TEN}$.

Propagation of plane waves in an orthotropic magneto-thermodiffusive rotating half-space

  • The present article is aimed at studying the reflection phenomena of plane waves in a homogeneous, orthotropic, initially stressed magneto-thermoelastic rotating medium with diffusion. The enuciation is applied to generalized thermoelasticity based on Lord-Shulman theory. There exist four coupled waves, namely, quasi-longitudinal P-wave (qP), quasi-longitudinal thermal wave (qT), quasi-longitudinal mass diffusive wave (qMD) and quasi-transverse wave (qSV) in the medium. The amplitude and energy ratios for these reflected waves are derived and the numerical computations have been carried out with the help of MATLAB programming. The effects of rotation, initial stress, magnetic and diffusion parameters on the amplitude ratios are depicted graphically. The expressions of energy ratios have also been obtained in explicit form and are shown graphically as functions of angle of incidence. It has been verified that during reflection phenomena, the sum of energy ratios is equal to unity at each angle of incidence. Effect of anisotropy is also depicted on velocities of various reflected waves.

Identification of Candidate Transcripts Related to Drought Stress using Secondary Traits and qRT-PCR in Tropical Maize (Zea mays L.)

  • Global climate change exerts adverse effects on maize production. Among abiotic stresses, drought stress during the tasseling stage (VT) can increase anthesis-silking intervals (ASI) and decrease yield. We performed an evaluation of ASI and yield using a drought-sensitive line (Ki3) and a drought-tolerant line (Ki11) to analyze the correlation with ASI and yield. Moreover, the de novo data of Ki11 were analyzed to find putative novel transcripts related todrought stress in tropical maize. A total of 182 transcripts, with a log2 ratio >1.5, were found by comparing drought conditions to a control. The top 40 transcripts of high expression levels in the de novo analysis were selected and analyzed with PCR. Of the 40 transcripts, six novel transcripts were detected by quantitative real-time PCR (qRT-PCR) using seedling and VT stage samples. Five transcripts (transcripts_1, 12, 34, 35, and 40) were up-regulated in the Ki11 shoot at seedling stage, and transcripts_1, 12, and 40 were up-regulated at the re-watering stage after 12 h of drought stress. The transcripts_32 and 34 were up-regulated at the VT stage. Hence, transcript_34 possibly plays a significant role in drought tolerance during the seedling and VT stages. The transcript_32 was identified as chloramphenicol acetyltransferase (CAT) by Pfam domain analysis. The function of the other transcripts remained unknown. Further characterization of these novel transcripts in genetic regulation will be of great value for the improvement of maize production.
