  • The characteristics of $CO_2$ exchange between plant and human modules, absorption rate of NO$_2$, and growth of lettuce were examined in an urban-type plant factory(UPF). With 150 lettuce plants, $CO_2$ concentrations of plant module were 600 ~ 700$\mu$mol mol$^{-1}$ at average leaf weight of 130g.plant$^{-1}$ and 900~1100$\mu$mol.mol$^{-1}$ at 75g.plant$^{-1}$ for one and two persons' stay in the human module, respectively. When the air of 0.13, 0.30 and 0.45 $\mu$molㆍmol$^{-1}$ NO$_2$ in a human module was circulated ON/OFF 10/20min between the human and plant modules, $NO_2$ decrement in the chamber during 10 min was 0.040, 0.109, and 0.149 $\mu$mol, respectively. The lettuces grown at 0.45 $\mu$molㆍmol$^{-1}$ $NO_2$ during experimental period showed no significant differences in growth factors such as leaf width, leaf length, leaf area and fresh weigh, and in the quality between treated and control.

In vitro Regeneration of Phragmites australis through Embryogenic Cultures

  • Phragmites australis (reed) has received much attention as being one of the principle emergent aquatic plants for treating industrial and civil wastewater. Plant regeneration via plant tissue culture in p. australis was investigated. Three types of callus were identified from seeds on N6 medium plus 4.5 UM 2,4-dichlorophenoxyacetic acid (2,4-D). Yellow compact type showed the best redifferentiation, whereas white compact type and yellow friable were not competent to differentiate into plane. Solid medium culture was better than liquid suspension culture for enhancing callus growth when N6 medium supplemented with 4.5 ${\mu}M$ 2,4-D was used. Phytagel, as a gelling agent, was superior to agar in plant regeneration on N6 medium, supplemented with 9.4 ${\mu}M$ kinetin and 0.54 ${\mu}M$ $\alpha$-naphthaleneacetic acid (NAA). Transfer of the plantlets regenerated from kinetin and NAA-supplemented N6 medium to growth regulator-free MS medium enhanced the further development of the plantlets. Plantlets on subsequently grown to maturity when tansferred to potting soil. The regenerated plants exhibited morphologically normal. The system for plant regeneration of P. australis enables to propagate elite lines on a large scale for water purification in the ecosystem

Selection of Optimum Water Plant in Constructed Wetland by Natural Purification Method for Municipal Sewage Treatment (자연정화공법에 의한 인공습지 하수처리장에서 최적 수생식물의 선정)

  • Objectives of this experiment were to select of the optimum water plant and to investigate the removal efficiency of the major nutrients in the constructed wetland by natural purification method for municipal sewage treatment in the farming and fishing village. For the experiment we used "constructed wetland" which was consisted of aerobic (vertical filter system) and anaerobic systems (horizontal filter system). Both systems were filled with gravel and filter media, and grew water plants on top of them. And then, we investigated several items such as sewage treatment efficiency, growth status of water plants and the absorbed amount of inorganic element in water plants with periodical periods. In aerobic area, removal efficiencies of BOD, COD, T-N and T-P were over 92%, 74%, 25% and 57%, respectively, and then when the water is passed through anaerobic area, the efficiency was over 96%, 84%, 44% and 71%, respectively, which was increased more treatment efficiency than that of aerobic area. Absorption amount of nitrogen and phosphorous in Miscanthus sacchariflorus BENTH were the highest in the water plants as 17.7 and 2.41 g/plant in the aerobic area, respectively. Absorption amounts of nitrogen and phosphorous in Scirpus tabernaemontani GMEL were the highest in the water plants as 8.7 and 1.13 g/plant in the anaerobic area, respectively. For the selection of optimum water plants in the constructed wetland by natural purification method for municipal sewage treatment, it were observed that there were Miscanthus sacchariflorus BENTH, Phragmites japonica STEUD and Phragmites communis TRINIUS in the aerobic area and were Zizania latifolia TURCZ, Scirpus tabernaemontani GMEL, Typha orientalis PRESL, Iris pseudoacorus L and Cares dispalata BOOTT in the anaerobic area.

Prepurification of paclitaxel by micelle and precipitation

  • A novel prepurification method was developed aiming at increasing yield and purity, also reducing solvent usage for purification of paclitaxel. This method was a simple and efficient procedure, for the isolation and prepurification of paclitaxel from the biomass of Taxus chinensis, consisting of micelle formation, followed by two steps of precipitation. The use of a micelle and precipitation in the prepurification process allows for rapid separation of paclitaxel from interfering compounds and dramatically reduces solvent usage compared to alternative methodologies. This prepurification process serves to minimize the size and complexity of the HPLC operations for paclitaxel purification. This process is readily scalable to a pilot plant and eventually to a production environment where multikilogram quantities of material are expected to be produced. As much as possible, the process has been optimized to minimize solvent usage, complexity, and operating costs.

  • The measures for water quality improvement have been planned to introduce for several reservoirs which were badly polluted among the sites included in the Network of Agricultural Water Quality Survey (NAWQS). Considering conditions of circumstances around reservoirs, self-purification systems such as natural ecosystem, oxidation ponds with plants, grassed waterways, weirs, and manmade plant-islands are taking into account enhancing to trap nutrients in waters running off from agricultural lands. The Pollutant Run off Ratios were analysed to predict the effects of water quality improvement for self-purification systems. The cost of water quality improvement was evaluated. The correlation equation between cost and irrigation area showing high correlation coefficient was derived.

Thioltransferase (Glutaredoxin) from Chinese Cabbage: Purification and Properties

  • Thioltransferase, also known as glutaredoxin, was purified from Chinese cabbage (Brassica campestris ssp. napus var. pekinensis) by a combination of ion-exchange chromatography and gel filtration. Its purity was confirmed by SDS-polyacrylamide gel electrophoresis and its molecular weight was estimated to be about 12,000 which is comparable with those of most known thioltransferases. The enzyme utilizes 2-hydroxyethyl disulfide, S-sulfocysteine, ${\alpha}-chymotrypsin$, insulin, and trypsin as substrates in the presence of reduced glutathione. The enzyme has Km values of 0.03-0.97 mM for these substrates. It appeared to contain dehydroascorbate reductase activity. The pH optimum of the enzyme was 8.5, when 2-hydroxyethyl disulfide was used as a substrate. It was greatly activated by reduced glutathione. Its activity was not significantly lost when stored at high temperature, indicating its thermostable character. It may play an important role in thiol-disulfide exchange in plant cells.

  • The mixing of water and chemicals is an important process in the water purification plant. In this paper, we compare PDM(pump diffusion mixer) and hydraulic mixer at the basin in the mixing process. The proper flow rates are predicted and compared in both mixers using CFD technique. As a result, the flow rate of purifier chemical liquid should be 5% of that of water for the optimal performance of mixing process. The characteristics of the two mixing methods are compared with each other for strong and weak points on the operation of the purification system, discussed in the view point of CFD simulations.

A Study on Water Quality Purification Function by Using Planting Concrete and Porous Concrete (다공성 콘크리트와 식생 콘크리트에 의한 수질정화기능에 관한 연구)

  • The purpose of this research provides the basic solution about waterfront and promotes the inherent capability, that recoveries both the river and gives the river for water quality to improve plan. In this study, we compare and analysis the slope contact between filter media of the porous concrete and planting concrete. As a result, when appling the porous concrete, it can show the removal efficiency is SS 53%, BOD 39%, COD 20%, T-N 36% T-P 42% and appling planting concrete is SS 58%, BOD42%, COD 26%, T-N 45%, T-P 53%. Therefore, planting concrete is higher removal efficiency (SS 5%, BOD 3%, COD 6%, T-N 9%, T-P 10%) than porous concrete. The experimental results show that using purification filter media on planting concrete is better than on porous concrete, because it have the higher purification filter efficiency. The quality of water improves vegetarian concrete, that can expect the increase of the self-purification capacity and improve the spectacle for providing the waterside and planting of planting concrete. In addition, it can complete and apply the research if having enough time for experimentation and accurate study for mechanism by plant, we can use both planting concrete filter media and the existing dike. As a result, we can gain the better quality of the water of the city's rivers and good economic value, that is spread by all cities applicable technologies. So it can be expected using well for future.

Analysis of Water Purification Capability of the Spent Fuel Storage Pool Using Consolidated Fuel Storage in Uljin 1&2 (조밀화 핵연료 집합체 저장에 의한 울진 1&2호기의 사용후 핵연료 저장조 정화능력 해석)

  • The radioactivity in the spent fuel storage pool is calculated to ensure to maintain its concentration below the permissible limit, when the storage capacity of Uljin nuclear power plant unit 1&2 is extended from 9/3 to 32/3 core using consolidated fuels in maximum density rack (MDR). For this evalulation, two models to calculate the spent fuel pool activities on the continuous and intermittent operating its purification system are developed and these results compared, The results of above two cases show that the current water purification system can not guarantee the radioactivity concentration below the design limit, 5$\times$10$^{-4}$ $\mu$Ci/ml, for the extention to 32/3 core. Therefore, it has been concluded that a modification of the current purification system is necessary to extend the spent fuel storage capacity with the above method. The alternative way suggested in this study is to increase the number of cation bed demineralizers.

  • Recently, the advancement of information and communication technology(ICT) is expanding the connectivity through Internet of Things(IoT), and the media of connection is also expanding from wire/cable transmission to broadband wireless communication, which has significantly improved mobility. This hyperconnectivity has become a key element of the fourth industrial revolution, whereas the supervisory control network of purification plants in korea is operated as a communication network separated from the outside, thereby lagging in terms of connectivity. This is considered the best way to ensure security, and thus there is hardly any consideration of establishing alternatives to operate an efficient and stable communication network. Moreover, security for management of a commercialized communication network and network management solution may be accompanied by immense costs, making it more difficult to make new attempts. Therefore, to improve the conditions for the current supervisory control network of purification plants, this study developed a industrial security L2 switch that supports modbus TCP(Transmission Control Protocol) communication and encryption function of the transmission section. As a result, the communication security performance improved significantly, and the cost for implementing the network management system using Historical Trend and information of HMI(Human Machine Interface) could be reduced by approximately KRW 200 million. The results of this study may be applied to systems for gas, electricity and social safety nets that are infrastructure communication networks that are similar to purification plants.
