PVA degrading bacteria were isolated from water system, and identified as Pseudomonas cepacia and Pseudmonas pseudomallei, which were named as Pseudomonas sp. G5Y and Pseudomonas sp. PW. It was found out that those two kinds of bacteria have a symbiotic relationship to degrade PVA. For the mixed culture of these bacteria, the optimal conditions of pH, temperature, nitrogen source, and polymerization degree of PVA were found to be 7.5, $35^{\circ}C$, ammonium sulfate, and 500, respectively. Also, the growth of these bacteria was promoted by trace elements such as vitamin B1, B12, pyridoxine, and p-aminobenzoate, respectively. The specific growth rate of mixed bacteria was inhibited when the concentration of PVA was more than 20g/l. The substrate inhibition kinetics of the mixed culture was $${\mu}=\frac{0.065S}{2.56+S+(S^2/156}hr^{-1}$
In this study nitroreductase from Pseudomonas sp. HK-6 capable of degrading 2,4,6-trinitrotoluene (TNT) was characterized. Through a series of purification process including ammonium sulfate precipitation, DEAE-sepharose, and Q-sepharose, three different fractions I, II, and III having the enzyme activity of NTRs whose molecular weights were approximately 27 kDa were detected in fractions from HK-6 cells. Specific activity of the three fractions were approximately 4.85 unit/mg, 5.47 unit/mg, and 5.01 unit/mg, and concentrated to 9.0-, 10.1-, and 9.3-fold compared to crude extract, respectively. The optimal pH and temperature for the three NTR fractions were approximately 7.5 and $30^{\circ}C$, respectively. Metal ions, $Ag^{+}$ , $Cu^{ 2+}$, $Hg^{2+}$ inhibited approximately 70% of enzymes activities of all NTR, while $Fe^{2+}$ did not stimulate or inhibit the activities. Monitoring the effect of chemicals on the enzyme activity revealed that those NTR fractions lost enzyme activity in presence of $\beta$-mercaptoethanol, but were a little influenced by dithiothreitol, EDTA and NaCl. The three NTR fractions demonstrated enzyme activities for nitrobenzene and RDX as well as TNT.
Two forms of extracellular inulinase, designated as PI and PII were detected in the crude enzyme preparation from n species of Pseudomonas isolated from soil. PI and PII were purified to homogeneity by ammonium sulfate fractionation, DEAE Sephadex A-50 chromatography, Sephadex G-100 and Sephadex G-200 gel filteration. Both isoenzymes catalyzed specifically and endowise the cleavage of the $\beta$-2,1-fructofranoside linkage of inulin, and displayed no action upon sucrose, raffinose and levan. The optimal pH values for the PI and PII enzyme were pH 5.5 and 6.0, respectively and the highest activity of the two enzymes was observed at 55$^{\circ}C$. The Km values of PI and PII were calculated to be 2$\times$10$^{-3}$M and 5$\times$10$^{-3}$M, respectively.
A bacterial strain which utilizes phenol under denitrifying condition was isolated from the industrial waste water collected from the Chong-ju Industrial Complex. The strain was identified as Pseudomonas species from the morphological, physiological, and biochemical characteristics and designated as HL100. The strain can utilize phenol as the sole source of carbon and energy when nitrate is provided as the terminal electron acceptor. The isolated strain completely degraded 3 mM of phenol within 110 hour with concomitant reduction of nitrate to nitrite. The observed maximum doubling time was 20 hours. Under appropriate condition, complete reduction of nitrate to atmospheric N$_2$ was observed indicating that the isolated strain could perform complete steps of denitrification. The strain showed optimal growth at pH 7.0 and temperature of 37$^{\circ}C$ under denitrifying phenol-degrading condition. The strain can also utilize toluene as the sole carbon and energy source under the same growth condition. However, no growth was detected on xylene and benzene.
Chromosomal gene transferable hybrid plasmids, RP4::Mu cts and RP4::mini-Mu, were transferred by conjugation from E. coli to Pseudomonas strains. In order to use for recipient cells of RP4::Mu cts and RP4:: mini-Mu, plasmid-free Pseudomonas strains were characterized for their antobiotic resistance, aromatic hydrocarbon utility and degradation patterns of chlorinated herbicide. Transfer frequencies of RP4::mini-Mu exhibited about $10^{-2}$ to $10^{-4}$, while those of RP4::Mu cts exhibited very low value of $10^{-7}$ in recipients tested except Pseudomonas aeruginosa KU557. Existance of hybrid plasmids in Pseudomonas transconjugants were identified by their antibiotic resistance and agarose gel electrophoresis. In case of RP4::Mu cts transconjugants it was also confirmed by demonstrating that they were capable of releasing phage and forming plaques at $43^{\circ}C$. Plaque forming unit of the transconjugants was about $10^{5}$. It was shown by the stability test that RP4::Mu cts and RP4::mini-Mu in Pseudomonas were relatively stable.

In this study, we isolated bacteria from petroleum contaminated soil which were near to underground storage tanks(UST). Through the screen test, we selected high efficiency bacterium, KDi19, for biodegradation of diesel. KDi19 was identified as Pseudomonas sp. by 16S rDNA, fatty acid, and morphological physiological characteristics. KDi19 degraded 956.3 mg/L(95.6%) of 1,000 mg/L diesel for 48 hours(incubation condition : temperature; $30^{\circ}C$, cell concentration; 1.0 g/L, pH 7). At low temperature, $20^{\circ}C$, $15^{\circ}C$, $10^{\circ}C$, KDi19 respectively removed 63.9%, 18.5% and 17.0% of 1,000 mg/L diesel for 48 hours(cell concentration 1.0 g/L, pH 7). At low concentration of diesel, 50 mg/L and 100 mg/L, KDi19 degraded 97.9% and 96.2% of diesel for 24 hours(temperature; $30^{\circ}C$, cell concentration: 1.0 g/L, pH 7), respectively.
Methanol assimilating bacterium, Pseudomonas sp. ILS-003 was used to investigate the optimum conditions for the production of $poly-{\beta}-hydroxybutyric$ acid from methanol. For PHB production, the optimum initial pH was 6.4 and the optimum temperature was $30^{\circ}C$. Also the optimum methanol concentration was found to be 1.0%(v/v). In the PHB production, $(NH_4)_2SO_4$ was the most effective nitrogen source and the optimum concentration was 0.8 g/l, which was eqivalent to 17.4 in C/N ratio. Also, deficiency of the 2 valence metal ions in the medium had stimulating effect on PHB accumulation. Under the optimum substrate concentration, successive feeding of 0.25%(v/v) methanol was the most effective on PHB production. Under the optimum conditions, 1.94 g/l of PHB and 2.78 g/l of dry biomass were produced in 96 hours, and the yield was 69.8%(w/w).

Accumulation of Pseudomonas sp., P. fluorescens and Erwinia carotovora in 60 min treatment was greater in extracts from soil, exudate from ginseng root and solutions than distilled water. In bacterial movement toward rubber tube soil from chamber, accumulation of P. fluorescens in response to soil supplemented with soil extracts, exudate and solutions was generally greater in soil extracts compared to control and other solutions, but Pseudomonas sp. and E. carotovora were not much response to supplemented extracts, exudate and solutions. Accumulation of the bacteria in capillaries containing various exudates from fungal propagules with not attracted to the exudates. For an accumulation of bacteria in rubber tubes containing soil inoculated with fungal propagules, the Pseudomonas sp. was not attracted in soil inoculated by the organisms as attractant but P. fluorescens and E. carotovora to fungi were attracted to F. solani, F. oxysporum and mixed organism Alternaria panax did not affect on bacterial movement except E. carotovora. The organic matter conten in Kangwha and Kimpo soil were low in diseased and healthy soil. The K content was especially high in Kimpo healthy soil. Bacterial population in Goesan and Kangwha were more abundant than other soil. The number actinomycetes was populated abundant in healthy soil of Goesan and diseased soil of Poonggi.
In this study, to reduce the production cost of poly(3-hydroxyalkanoates) (PHA), optimal cell growth and PHA biosynthesis conditions of the isolated strain Pseudomonas sp. EML8 were established using waste frying oil (WFO) as the cheap carbon source. Gas chromatography (GC) and GC mass spectrometry analysis of the medium-chain-length PHA (mcl-PHAWFO) obtained by Pseudomonas sp. EML8 of WFO indicated that it was composed of 7.28 mol% 3-hydrxoyhexanoate, 39.04 mol% 3-hydroxyoctanoate, 37.11 mol% 3-hydroxydecanoate, and 16.58 mol% 3-hydroxvdodecanoate monomers. When Pseudomonas sp. EML8 were culture in flask, the maximum dry cell weight (DCW) and the mcl-PHAWFO yield (g/l) were showed under WFO (20 g/l), (NH4)2SO4 (0.5 g/l), pH 7, and 25℃ culture conditions. Based on this, the highest DCW, mcl-PHAWFO content, and mcl-PHAWFO yield from 3-l-jar fermentation was obtained after 48 hr. Similar results were obtained using 20 g/l of fresh frying oil (FFO) as a control carbon source. In this case, the DCW, the mcl-PHAFFO content, and the mcl-PHAFFO yields were 2.7 g/l, 62 wt%, and 1.6 g/l, respectively. Gel permeation chromatography analysis confirmed the average molecular weight of the mcl-PHAWFO and mcl-PHAFFO to be between 165-175 kDa. Thermogravimetric analysis showed decomposition temperature values of 260℃ and 274.7℃ for mcl-PHAWFO and mcl-PHAFFO, respectively. In conclusion, Pseudomonas sp. EML8 and WFO could be suggested as a new candidate and substrate for the industrial production of PHA.
The gene responsible for dechlorination of 4-chlorobenzoate (4CBA) was cloned in E. coli XL1-Blue from Pseudomonas sp. DJ-12. The cloned cell of E. coli Cjl had the hybrid pBluescript SK(+) plasmid, into which about 9.5 kb genomic DNA fragment of PseudOmonas sp. DJ-12 was inserted. The subclone of pCJlOl was constructed by inserting the 3.4 kb EcoRI-HindIII fragment of pCJl into the vector. Those cloned cells could be simply selected by halo formation around the colonies which was the precipitate of AgCl produced by reaction of AgNO$_{3}$ and chloride ion liberated by bacterial dechlorination of 4CBA- Such a plate assay method was standardized by the procedure that the colonies grown for 2 days on the Cl$^{-}$-free plate medium containing 1 mM 4CBA were flooded with 0.1 M AgNO$_{3}$ solution.
