Simplified HPLC Method for the Determination of Pseudoephedrine Hydrochloride from Allegra D Tablet

  • A sensitive, simple and highly selective liquid chromatography method of determination for extraction of pseudoephedrine hydrochloride from Allegra D tablet was developed. The chief benefit of the present method is the minimal sample preparation, as the procedure is only filtering through pore syringe filter. Two drugs (pseudoephedrine hydrochloride, fexofenadine) were separated on a C$_{18}$ column and analyzed by high performance liquid chromatography (HPLC). The method had a chromatographic run time of 8.0 min. 1 ml of pseudoephedrine hydrochloride solution (1 mg/ml) was filtered through 0.22 um pore syringe filter. 50 ul of filtering solution was injected to HPLC pump and we knew the retention time (1.85 min) of separating of pseudoephedrine hydrochloride using UV detector at 280 nm. We used C$_{18}$ column (4.6 mm${\times}$250 mm), mobile phase solution (<0.05 mol/L NaH$_2$PO$_4$, 2 ml/L H$_3$PO$_4$>/CH$_3$CN / sodium dodesyl sulfate = 60 ml / 40 ml / 1 g). We separated psedoephedrine hydrochloride at run time of 1.85 min from Allegra D tablet solution (1 mg/ml) filtered through 0.22 um pore syringe filter using UV detector at 280 nm. Flow rate was set at 1.0 ml/min and the column temperature was set at 40$^{\circ}C$. Psedoephedrine hydrochloride solution (1 mg/ml) separated from Allegra D tablet was filtered through 0.22 um pore syringe filter and injected 50 ul. We confirmed the peak of psedoephedrine hydrochloride at same retention time and the separating solution was freeze-dried. In conclusion, A simple isocratic reverse-phase HPLC method has been developed that provides excellent separation of pseudoephedrine from Allegra D tablet.

  • A gas chromatographic procedure is described for the quantitative analysis of ephedrine hydrochloride, pseudoephediiiie hydrochloride and norephedrine hydrochloride in pharmaceutical preparations. The procedure was based upon the determination of thioureas formed from amines and phenylisothiocyanate. The recoveries of ephedrine, pseudoephedrine and norephedrine from pharmaceutical preparations were 100.7(S.E. 0.9), 99.5(0.8), 99.1(0.8) and 99.1%(O.9), respectively. The high accuracy and precision of this assay were inherent in the flame photometric detectors normally used in gas chromatography.

Hydroxypropylmethylcellulose로부터 염산슈도에페드린의 방출조절에 관한 연구 (A Study on the Control of Pseudoephedrine Hydrochloride Release from Hydroxypropylmethylcellulose Matrices)

  • Hydroxypropylmethylcelluloses (HPMC)는 친수성 cellulose 계통의 중합체로서 독성이 적고 가격이 저렴하여 경구용 서방출성 제제에 널리 이용되고 있다. 본 연구에서는 매트릭스로서 HPMC를 이용하여 새로운 서방출성제제인 HPMC/염산슈도에페드린계를 설계하고, 직타법에 의하여 정제를 제조하였으며, 이 팽윤될 수 있는 친수성 약물전달계를 이용하여 약물의 방출에 영향을 주는 여러 인자들이 약물의 방출속도에 미치는 영향에 대하여 조사하였다. 결과로서, HPMC/염산슈도에페드린계에서 매트릭스인 HPMC의 분자량이 클수록, HPMC의 함량이 많을수록 약물의 방출속도가 더 느려짐을 알 수 있었으며, 정제의 경도나 용출시 pH 변화에는 무관한 것으로 나타났다. 특히, 음이온 계면활성제인 sodium laurylsulfate는 HPMC로부터 염산슈도에페드린의 방출속도를 지연시키는데 큰 영향을 미치는 것으로 나타났으며, 이러한 현상은 음이온 계면활성제인 sodium laurylsulfate가 반대의 이온성을 갖는 약물인 염산슈도에페드린과 착물을 형성함으로써 약물의 용해성이 떨어져서 약물방출이 지연되는 것으로 생각된다.

건강보험심사평가원의 전체환자데이터셋(HIRA-NPS)자료를 이용한 정제의 분할 처방 현황 분석 (Comparative Analysis of Prescription for Splitted Tablet using the HIRA-NPS (Health Insurance Review & Assessment Service-National Patient Sample))

  • Objective: The aim of this study was to analyze the status of split tablet prescription in South Korea. Methods: We conducted this analysis using 2016 National Patient Sample data from the Health Insurance Review and Assessment Service. We computed split tablet prescription rates by sex and age and determined which medicine and medical specialties had the highest split tablet prescribing rates. Results: The proportion of prescriptions that included split tablets was 15.6% (n=6,687,35). The proportion of prescriptions that included split tablets was higher for females (56.7%) than for males (43.3%), while that of prescriptions including split tablets versus total prescriptions for each sex was higher for males (16.4%) than for females (14.9%) (p<0.001). In the age group under 19 years, the proportion of prescriptions including split tablets (53.7%) was more than half of the total. The highest tablet splitting rate was found to be 89.9% for formoterol fumarate (40 ㎍), and pseudoephedrine hydrochloride (60 mg) had the highest number of prescriptions. Pediatrics (65.6%) was the medical field with the highest rate of split tablet prescription. Conclusion: Split tablets were most prescribed to pediatric patients. To minimize the use of split tablets, it is necessary to develop lower dose tablets and establish a policy that promotes prescription of these lower-dose tablets.
