내염소성 원생동물인 크립토스포리디움, 지아디아 등 병원성 미생물의 제거에 대한 신뢰성을 확보하기 위해 정수처리 공정에서 막여과 시스템이 각광받고 있으며, 이러한 신뢰성을 보증하기 위해 막 완결성에 대한 연구가 중요시되고 있다. 막 완결성 시험은 크게 직접법과 간접법으로 분류되는데, 직접법 중 압력기반의 시험은 버블포인트 이론을 근간으로 병원성 미생물의 최소 크기인 $3{\mu}m$ 크기 이상을 감지할 수 있는 감도로 USEPA Guidance Manual에서 제시하고 있다. 간접법은 온라인 상태에서 연속적인 운전이 가능하다는 점에서 널리 사용되지만, 직접법에 비해 현저히 낮은 막 손상 감지 감도를 가지고 있으며, 손상 부위를 특정 지을 수 없는 한계가 있기에 이러한 감지 감도를 개선해야 할 필요가 있다. 따라서 본 연구에서는 압력 손실 시험에 의한 막 완결성 시험을 통해 막 파단 형태별, 파단 개수, 초기설정압력값에 따른 압력손실률과 LRVDIT 신뢰성 범위를 상호 비교함으로써 막 손상에 따른 감도의 결과를 정량화하여 UCL 기준을 나타내어 비교 평가하였다.
멍게 물렁증 원인규명의 일환으로 2011년 3월에 경남 통영에서 채집된 물렁증 멍게와 건강한 멍게를 대상으로 병리학적 검사를 실시한 결과 물렁증 멍게의 경우 체액량, 비만도 및 피막지수가 현저히 감소하였다. 물렁증 멍게의 피막 imprint 관찰과 배양 시 2개의 편모를 가진 원생동물이 확인되었으며, 물렁증 멍게에 대한 이들 편모충의 감염률은 97.5%인 반면 건강한 멍게에서는 검출되지 않아 본 편모충은 물렁증 멍게에서 특이적으로 검출되는 것으로 조사되었다. 조직학적 검사결과 물렁증 멍게에서 피막섬 유질다발의 붕괴 현상이 관찰되었으며, 편모충으로 추정되는 원생동물들이 다수 관찰되었다. 통영산 물렁증 멍게에서 확인된 편모충은 일본에서 검출된 그것과 매우 유사한 형태적 특징을 갖고 있으며, 향후 공격실험 등 물렁증 유발과 편모충과의 연관성에 대한 추가 연구가 요망된다.
지난 1984년의 제1차 보고에 이어 1984년부터 1992년까지 9년간 서울 백병원 외래 및 입원환자 중 기생충검사를 의뢰한 52.552명에 대하여 포르말린-에테르 집란법 및 직접 도말법으로 대변검사를 시행하였던 바 그 결과는 다음과 같았다. 총 윤충류(13종) 충란 및 원충류(5종) 포낭 양성률은 각각 평균 6.5% 및 2.5%이었다. 윤충류 종별 양성률(양성자수)은 간흡충 3.2% (1.667). 편충 2.1%(1 089) 요꼬가와흡충 1 2%(613), 회충 0.2%(100) 동양모양선충 0.06%(34). 유무구조충 0.05%(28). 왜소조충 0.03%(18). 구충 0.03%(17). 극구흡충류 0.03%(15), 폐흠충 0.02%(12). 요충 0.02%(10) 분선충 0.01%(6), 광절열두조충 0.004% (2)이었다. 원충 별로는 대장아메바 1.1%(588). 왜소아메바 0.8%(402), 람블편모충 0.3% (173), 이질아메바 0.3%(164). 장트리코모나스 0.004%(2)로 나타났다. 조사년도별로 볼 때 회 충 및 편충 등 토양매개성 윤충은 뚜렷한 감소를 보이는 반면 간흡충을 비롯한 패류 매개성 윤충과 장내 원충류는 양성률의 감소를 인정할 수 없었다.
To understand protozoan, viral, and bacterial infections in diarrheal patients, we analyzed positivity and mixedinfection status with 3 protozoans, 4 viruses, and 10 bacteria in hospitalized diarrheal patients during 2004-2006 in the Republic of Korea. A total of 76,652 stool samples were collected from 96 hospitals across the nation. The positivity for protozoa, viruses, and bacteria was 129, 1,759, and 1,797 per 10,000 persons, respectively. Especially, Cryptosporidium parvum was highly mixed-infected with rotavirus among pediatric diarrheal patients (29.5 per 100 C. parvum positive cases), and Entamoeba histolytica was mixed-infected with Clostridium perfringens (10.3 per 100 E. histolytica positive cases) in protozoan-diarrheal patients. Those infected with rotavirus and C. perfringens constituted relatively high proportions among mixed infection cases from January to April. The positivity for rotavirus among viral infection for those aged $\leq$ 5 years was significantly higher, while C. perfringens among bacterial infection was higher for $\geq$ 50 years. The information for association of viral and bacterial infections with enteropathogenic protozoa in diarrheal patients may contribute to improvement of care for diarrhea as well as development of control strategies for diarrheal diseases in Korea.
Giardia intestinalis infections arise primarily from contaminated food or water Zoonotic transmission is possible, and at least 7 major assemblages including 2 assemblages recovered from humans have been identified. The determination of the genotype of G. intestinalis is useful not only for assessing the correlation of clinical symptoms and genotypes, but also for finding the infection route and its causative agent in epidemiological studies. In this study, methods to identify the genotypes more specifically than the known 2 genotypes recovered from humans have been developed using the intergenic spacer (IGS) region of rDNA. The IGS region contains varying sequences and is thus suitable for comparing isolates once they are classified as the same strain. Genomic DNA was extracted from cysts isolated from the feces of 5 Chinese, 2 Laotians and 2 Koreans infected with G. intestinalis and the trophozoites of WB, K1, and GS strains cultured in the laboratory, respectively. The rDNA containing the IGS region was amplified by PCR and cloned. The nucleotide sequence of the 3' end of IGS region was determined and examined by multiple alignment and phylogenetic analysis. Based on the nucleotide sequence of the IGS region, 13 G. intestinalis isolates were classified to assemblages A and B, and assemblage A was subdivided into A1 and A2. Then, the primers specific to each assemblage were designed, and PCR was peformed using those primers. It detected as little as 10 pg of DNA, and the PCR amplified products with the specific length to each assemblage (A1, 176bp; A2, 261 bp; B, 319 bp) were found. The PCR specific to 3 assemblages of G. intestinalis did not react with other bacteria or protozoans, and it did not react with G. intestinalis isolates obtained from dogs and rats. It was thus confirmed that by applying this PCR method amplifying the IGS region, the detection of G. intestinalis and its genotyping can be determined simultaneously.
Waterborne infectious disease is induced by several pathogenic microbes such as bacteria, viruses and protozoans, and the cases caused by viral infection is currently increasing. Water treatment process could reduce the number of virus in the water, but there were many difficulties to completely remove the virus particles from water. Therefore, the membrane separation technology which was reported to effectively remove pollutants from raw water has attracted increasing attention and demand. Since its efficiency has been introduced, demands for evaluation method toward the membrane filtration process are increasing. However, progression of the method development is slow due to the difficulties in cultivation of several waterborne viruses from animal models or cell culture system. To overcome the difficulties, we used adenovirus, one of the commonly isolated pathogenic waterborne viruses which can grow in cell culture system in vitro. The adenovirus used in this study was identified as human adenovirus C strain. The adenovirus was spiked in the raw water and passed through the microfiltration membrane produced by Econity, a Korean membrane company, and then the viral removal rate was evaluated by real-time PCR. In the results, the amount of virus in the filtered water was decreased approximately by 5 log scale. Because coagulant treatment has been known to reduce filtering function of the membrane by inducing fouling, we also investigated whether there was any interference of coagulant. In the results, we confirmed that coagulant treatment did not show significant interference on microfiltration membrane. In this study, we found that waterborne virus can be effectively removed by membrane filtration system. In particular, here we also suggest that real-time PCR method can rapidly, sensitively and quantitatively evaluate the removal rate of virus. These results may provide a standard method to qualifying membrane filtration processes.
Morphometrical analysis of chicken Cryptosporidium baileyi in various stages of life cycle in the bursa of Fabricius were carried out by electron microscope to establish a differential point for identification of C baileyi. By avidin-biotin complex method, protozoans of the bursa of Fabricius were identified Cryptosporidium spp. The size and area on each developmental stages of C baileyi, as measured by Morphomat 10 attached to electron microscope were as follows. Trophozoites' size with range of $3.21{\pm}0.70{\times}2.49{\pm}0.59{\mu}m$, area with range of $118.82{\pm}41.92{\mu}m^2$; meronts' size $3.99{\pm}1.07{\times}2.96{\pm}0.52{\mu}m$, area $210.11{\pm}57.11{\mu}m^2$; merozoites' size $1.98{\pm}0.43{\times}0.60{\pm}0.18{\mu}m$, area $24.10{\pm}5.97{\mu}m^2$; microgametes' size $1.36{\pm}0.83{\times}0.50{\pm}0.23{\mu}m$, area $20.23{\pm}6.73{\mu}m^2$; macrogametes' size $4.57{\pm}0.65{\times}4.02{\pm}0.55{\mu}m$, area $258.37{\pm}51.83{\mu}m^2$; oocytes' size $4.39{\pm}0.56{\times}3.44{\pm}0.50{\mu}m$, area $187.21{\pm}62.68{\mu}m^2$. In conclusion, the size and area on each developmental stages of Cryptosporidium baileyi is different from that of other Gryptosporidia spp. It suggests, with considering tissue tropism and life cycle, morphometrical analysis can be quite a good way to identify C baileyi.
T. japonicus가 서식하고 있는 천연 tide pool의 실태를 알기 위하여, 신내천현 삼포시 유호주변에 있는 성상이 다른 tide pool 세곳(Fig.1. A, B, C)을 대상으로 $6{\sim}10$월까지 조사를 하였다. 이 tide pool 해수중의 환경요인인 염분농도는 장마철 강우기를 제외하고는 거의 $35\%0$였고, 수온은 $14{\sim}33^{\circ}C$, pH는 $7.4{\sim}8.9$, POC (현탁태유기탄소)는 $255{\sim}3980\;{\mu}g-c/l$, 전 인산염은 $1{\sim}27\;{\mu}g-at/l$의 심한 변동을 보였다. 해수중의 세균수 및 녹조유부착균수는 각각 $10^4{\sim}10^7/ml,\;10^6{\sim}10^8/g$의 범위로 시기적인 변동이 있었고, 해수중의 세균상은 Acinetobacter spp., Moraxella spp., Flabacterium spp., Pseudomonas spp.등의 세균주가 각각 $38\%,\;29\%,\;16\%$ 및 $14\%$의 비율로 나타났고, Vibrio spp. 균주나 효모의 출현비율은 낮았다. tide pool 내의 T. japonicus 최고개체수 및 포난개체수는, 8월과 9월 단세포 조류 및 원생동물의 대량발생이 관찰 되였던 B-pool이 높았고, 각각 해수 1l중 약 $2,200{\sim}7,000$ 및 $200{\sim}2,800$의 범위였다. 이상의 POC, 전 인산염 및 세균수, 세균상등의 조사결과, T. japonicus의 서식하는 천연환경이 상당히 부영양적인 것으로 추찰되었다.
가시아메바(Acanthamoeba spp.)와 파울러자유아메바(Naegleria fowleri)는 자유생활아메바로 자연계에 널리 분포하며 사람과 동물에게 치명적인 질병을 일으킨다. 본 연구에서는 가사아메바와 파울러자유아메바를 물 환경에서 조사하기 위해 기존에 보고된 네 종류의 분자생물학적 방법과 상용 real-time PCR 키트의 분석 민감도를 비교하였다. 그 결과 duplex real-time PCR 방법이 민감도가 가장 좋았으며, 동시에 두 종류의 자유생활아메바를 검출할 수 있었다. 따라서 이 방법을 사용하여 한국의 대전시에 위치한 3개 하천, 6개 지점을 대상으로 그 분포를 2회 조사하였다. 가시아메바는 12개 시료 중 10개 시료에서 검출되었으며(83.3%), 파울러자유아메바는 2개 시료에서 검출되었다(16.6%). 향후 이러한 유해 아메바로부터 먹는 물의 안전성을 확보하기 위해 지속적인 분포조사가 필요할 것이다.
