To observe the antigenic protein fractions in saline extract of Spirometra mansoni plerocercoid (sparganum), the crude extract was separated in reducing conditions of sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE). The proteins, transferred by celctrophoresis to introcillulose paper, were reacted with sera from 15 surgically confirmed sparganosis and 24 cysticercosis patients for immunoblotting. Out of 30 identified protein bands in the extract, bands of 29 and 36 kilodaltons (kDa) were the strongest and the most frequently reacting with specific antibody (IgG) in sparganosis sera. Bands of highter molecular weight also reacted with the sera but their frequency of reactions was lower. Sera of cysticercosis reacted with different protein bands in saline extract of sparganum, but the cross reactions were observed in strong antigenic bands of 29 and 36 kDa.
Ever since the expression of new tumor-specific antigens was reported during malignant transformation, studies on separation, purification and characterization of these proteins have been so activated recently. Following experiment was performed to observe tumor-specific antigens by implanting DMBA pellet into submaxillary gland of rat for inducing salivary gland tumor. After dividing 280 rats into 2 groups, in control group, sham operation was performed on right submaxillary gland and, in experimental group, DMBA pellet (5mg) was implanted into right submaxillary gland. Then proteins from excised submaxillary gland by killing 10 rats every two weeks for 28 weeks were extracted with 3M KCl, and SDS-PAGE and PAS-staining were carried out for biochemical examination. The obtained results were summarized as follows; 1) At 12th week since implantation of DMBA pellet, tumor mass formation was inspected. And dysplasia at 6th week and invasive epidermoid carcinoma at 10th week were observed by microscope. 2) In control group, the weight ratio of both submaxillary glands had no any change, however, in experimental group, the ratio was increased remarkably. And at 28th week after DMBA implantation, there was more than 15 times of differences in weight between control and experimental group. 3) There was no DMBA remnant after 22nd experimental week. 4) In the SDS-PAGE, high molecular protein bands (more than 100 kd) were appeared much, and new prominent protein bands (66, 48, 41.5, 39, 37, 37.5 kd) were appeared after 4th week since DMBA implantation. However, 38, 27, 22kd protein bands were disappeared. 5) In PAS-staining, high molecular proteins were proteins were all glycoproteins and 37.5kd protein was proved as to be glycoprotein. And 38kd glycoprotein was disappeared after 4th week since DMBA implantation.
The effect of polyamine on chlorophyll amount, chloroplast peroxidase and chloroplast thylakoid protein in rice leaf segments which were grown for 10 days(16 hrs, light : 8 hrs, dark) in a hormone-free MS medium containing polyamine was studied. Polyamine treatment increased the chlorophyll contents compared with the control in rice leaf segments. Especially spermine was most effective. Also, in rice leaf segments treated with polyamine chloroplast peroxidase activity was higher than in the control. The treatment with lmM of spermidine increased the enzyme activity by 100%. In polyamine treatment and control two major polypeptide bands corresponding to 56 and 25Kd molecular weight were clearly resolved with other minor bands by SDS-PAGE in the insoluble protein fraction. However, in these bands (56, 25Kd), the total area of protein in treating with polyamine were higher than that of the control. These results suggest that polyamine was an important factor in the chloroplast development of rice seedlings.

TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin) and related halogenated aromatic hydrocarbons elicit a diverse spectrum of biochemical and toxic responses in laboratory animals and mammalian cells in culture. Toxicity and carcinogenicity of TCDD is well established but the molecular mechanism is still poorly understood. Here, we found the noble responsive genes to TCDD using the differential display analysis. Treatment of HepG2 cells with TCDD showed a significantly different mRNA expression pattern from the untreated cells in differential display analysis. The differentially displayed bands were isolated and used as probes in dot blot and Northern blot analyses. Of thirty-five isolated differentially displayed bands, only two bands were confirmed as positive in dot blot and Northern blot analyses. The nucleotides sequences of these clones were analyzed and the search of Genebank database revealed that one clone is highly homologous with RanBP2 (Ras-related nuclear protein binding protein2; 92%) and the other is an unknown gene. RanBP2 is a nucleoporin with SUMO E3 ligase activity that functions in both nucleocytoplasmic transport and mitosis and its role as a novel tumor suppressor has been recently proposed. Thus, these results may suggest the clue elucidating the toxic mechanism of TCDD through RanBP2.
Aspergillus niger van Tieghem was cultured by the method of submerged and synchronized culture for the study of differentiation. Acid-phenol soluble cell proteins of the fungus were extracted from four stages during development. Those acid-phenol soluble proteins were separated by polyacrylamide gel electrophoresis to determine the protein patterns. A new protein band was observed from the pre-sporulation body, color density of the stained protein bands in four tubes differed according to the differentiation stages. The number of protein bands in 10% gels varied from 18 to 16, 17, and 19 according to the course of spore germination stage, conidiophore stage, phialide maturation stage and sporulation stage.

In order to observe the protein composltlOns of soluble extracts of P. westermani, and their changes during early developmental stages, the crude saline extracts of 4, 6, 8, 10 and 12 week-old worms which were harvested from experimentally infected dogs were analysed by disc-PAGE. The results were as follows: 1. A total of 15 bands were identified from electrophoregrams of respective developmental stages. Of them, 5 bands were recognized throughout the developmental stages. 2. The number and protein amount of identified bands changed according to the worm development from 4 weeks to 12 weeks. However, tLe bar::ding patterns of 4 and 6 week-old worms and 8 and 10 week-old worms were similar each other. 3. Of 15 identified bands, Band 1 was recognized only in 12 week-old worms whereas Bands 3, 4, 8, 9, 10, 11 and 15 gradually lowered their amount according to dcvelor:ment to disappear in 12 week-old. In addition, Band 5 became a major band in 12 week-old while Band 6 turned to a minor band at the same age. The possible relations of changing patterns of protein bands with worm development were discussed.
The electrophoretic variations in protein and some enzymes during stratification of Panax ginsseng seeds were checked in relation to the differentiation and development of immature embryo, Seed-protein on PAGE showed eight major bands during the period of investigation, Four bands of Rf : 0.12, 0.24, 0.66 and 0,72 remained in dark stained condition, whereas the remainders (Rf : 0.40, 0.48, 0.56 and 0,76) became lighter in the staining condition as the treatment period was prolonged, In embryo, which was separately sampled from 75 days after the treatement, the only band of Rf 0.76 was not observed, In peroxidase, the number of bands was gradually increased and came to four bands at 45 days after treatment, and their staining color became gradually darker until the time of final samling. Also, the embryo had the same patterns as those in the intact seed. Esterase showed four bands during the period of investigation. As the treatment period was prolonged, the degree of staining condition came to darker in the band of Rf 0.36, but lighter in the band of Rf 0.56. The band of Rf 0.20 was not detected in the intact seed at 90 days after the treatment, but clearly observed in embryo. Glutamate dehydrogenase had only two bands: one (Rf : 0.16) was detected only at 60 days after the treatment and the other (Rf: 0.14) was always observed throughout the period, and showing simple pattern in the electrophoretic variation. Glutamate-oxaloacetate transaminase showed the only band of Rf 0.36 during the period of investigation and even in embryo, and showing the simplest pattern, indeed.
Dehulled pumpkin seeds produced in Korea were defatted and their composition of fatty acid was analyzed by gas chromatography. Soluble protein and insoluble protein were isolated from the dehulled and defatted pumpkin seeds. The protein component was identified by disc-electrophoresis and the amino acid composition of the protein was analyzed. (1) The pumpkin seed contained 46.5% of fat and 25.5% of protein respectively. (2) The fatty acid of the pumpkin seed was compassed chiefly of linoleic acid (51.74%), oleic acid (22.2%), palmitic acid (18.1%) and stearic acid (7.84%) (3) The amino acid composition of the soluble and insoluble seed protein contained most of all the amino acids except for S-contained amino acids. (4) It has been identified by disc-electrophoresis that the soluble protein had 13 bands and the insoluble protein had 4 bands.
The plasma protein patterns of non-pregnant women, pregnant women, and normal male individuals were analyzed by SDS/polyacrylamide gel electrophresis. When the protein patterns of plasma of normal male individuals ranging from 10, 000 to 110, 000 daltons in molecular weights are compared to non-pregnant women, their protein patterns were the same. In this study, when the plasma of non-pregnant women are compared to pregnant women, no bands were occurred newly, but the quantity of some protein bands were increased or decreased during the pregnant periods. According to the results of measuring the molecular weights of the characteristic protein patterns, which are increasing or decreasing during the pregnancy as compared to the non-pregnant women, it was observed that the proteins over 76, 000 daltons in molecular weights were concerned in the facts mentioned above. That is, the protein of 86, 000 dalton in molecular weight was not increased in quantity until the second trimester of pregnancy, but was increased in the third trimester of pregnancy. The proteins of 91, 000-105, 000 daltons in molecular weights were gradually increased in accordance with the periods of pregnancy. On the contrary, the protein of 94, 000 dalton was rather decreased by the second trimester of pregnancy, but increased in the third trimester of pregancy. And the band of 99, 000 dalton was not changed in quantity significantly until the first trimester of pregnancy, but increased continuously from the second trimester of pregnancy to the third trimester of pregnancy. We tentatively suggest that the stages (the first, the second, and the third trimester) of pregnancy can be identified by the study on the protein patterns of the specific bands in the blood plasma of pregnant women.
To extract insoluble proteins of sesame meal residue by using microorganism, the sesame meal residue was treated with crude enzyme solution from Bacillus sp. CW-1121. It was found that the solubility reached to maximum at pH 7.5, $45^{\circ}C$. Under optimum condition, the nitrogen solubility with the enzyme solution from Bacillus sp. CW-1121 reached to 60% in 2 hours. Nitrogen solubility of protein from sesame meal showed minimum value at pH 4.5 and significantly increased above pH 6.0. When the protein from sesame meal extracted with Bacillus sp. CW-1121 was subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis, water soluble protein was showed 4 bands and salt soluble protein was showed 2 bands. The amino acid composition of water soluble protein, salt soluble protein and free amino acid indicated relatively high contents of serine (17.24 mg/g), glutamic acid (10.77 mg/g) and glutamic acid (6.55 mg/g). Specially, the contents of essential amino acids were high.
