The effect of alteration of light-darkness cycle on the protein metabolism was studied in the rat. The light-darkness cycle was altered either every 3 or 9 days, and animals consumed diets containing 8 or 25% casein. The results were summarized as follows : 1) Food consumptions and body weight gains of the 25% casein groups were higher than those of the 8% casein groups, and, among the animals consumed 25% casein diet, the light-darkness cycle altered group had lower food consumption and body weight gain than the unaltered group. 2) Weights of liver and adrenal gland per l00g body weight were not different with the dietary protein levels, but, at the end of experimental period, the 8% casein diet group of which light-darkness cycle altered every 9 days had the smallest liver weight and the largest adrenal gland weight. 3) Liver nitrogen and plasma protein concentrations of the 25% casein groups were slightly higher than those of the 8% casein groups. 4) Percentages of nitrogen retention of the 25% casein groups at period III were slightly lower in the light-darknerr cycle altered animals than that of the unaltered group.
Immunohistochemical stains for RB and p53 tumor suppressor gene products were performed on 72 cases of resected primary lung cancer tissues to study the correlation between their expressions and the histologic types, the clinical stage, and the survival rate. The results were as follows. 1. The RB protein was altered or absent in 38 cases (52.8%), and the mutant p53 protein was detected in 35 cases (48.6%). 2. The incidences of RB and p53 protein expression were significantly different among the histologic types (p<0.05) but were not correlated with the clinical stages of lung cancer (p>0.05). 3. The two year survival rate of patients with alteration of both RB and p53 genes (RB-/p53+) was 22. 4%, and that with no alteration of both genes (RB+/p53-) was 63.1%. This difference was statistically significant (p=0.01). 4. It was shown that alteration of RB protein greatly affects the prognosis of lung carcinoma by multivariate analysis of prognostic factors. The presence or absence of RB and mutant p53 protein in tumor cells is closely related to the survival of primary lung cancer patients, and it is suggested that RB gene expression is an independent prognostic factor of primary lung cancer.
Obesity is a major nutritional problem in the developed countries. The prevalence of obesity may range from 10 to 50 per rent or mort of adult population and it may be increasing tendency. Many efforts have been made to understand the pathogenesis of obesity, but except a few metabolic obesities in the most of obese patients, the mechanisms are not understood. The treatment modalities of obesity, ranging from dietary and pubilc health intervention through the pharmacological and surgical therapy, have been developed and tested. In the obese patients mortalities and mobilities are significantly increased than non obese subjects due to hypertension, diabetics, and other problems. There are four possible mechanisms by which energy balance might be altered to enhance metabolic efficiency. futile metabolic pathway, alteration of protein rum over, alteration in sodium-potassium ATPase and alteration in uncoupled oxidation in brown adipose tissue are considered as possible mechanisms. Low calory and very low calory diets are recommended as a dietary program. Several pharmacological agent such as benzphetamine, fenfluramine, mazindol and fluoxetin are currently popular drugs for the treatment of obesity.

Changes in biochemical constituents and physiological alteration were studied in various intensities (1-5%, 6-15%, 16-30%, 31-50% and > 50%) of leaf spot (Myrothecium roridum) on mulberry leaves and compared with healthy leaves. Chlorophyll, total soluble sugar and total protein were decreased (P < 0.01), but total phenol increased due to pathogen infection. Changes in biochemical constituents showed significant correlation with intensity of disease. Chlorophyll ($r^2$= 0.92), and protein (($r^2$= 0.83) possessed negative while phenol (($r^2$= 0.61) possessed positive correlation. Photosynthetic rate, transpiration rate, stomatal conductance, moisture content (%) and physiological water use efficiency (pWUE) were decreased, but stomatal resistance increased in the infected leaves. Physiological parameters also possessed significant (P < 0.01) correlation with disease intensity. Photosynthetic rate (($r^2$= 0.96), transpiration rate ($r^2$=0.88), stomatal conductance (($r^2$= = 0.65), physiological water use efficiency (($r^2$= 0.88) and moisture content (r = 0.85) were negatively but stomatal resistance (($r^2$= 0.75) was positively correlated to disease intensities.
Purpose: Multidrug resistance-associated protein (MRP) 2 is a glutathione conjugate in the canalicular membrane of hepatocytes. Early graft damage after liver transplantation (LT) can result in alteration of MRP2 expression. The purpose of this study was to evaluate the relationship between the pattern of MRP2 alteration and graft outcome. Methods: Forty-one paraffin-embedded liver graft tissues obtained by protocol biopsy within 2 months after LT; these were stained using monoclonal antibodies of MRP2. We selected 15 live donor biopsy samples as a control, that showed homogenous canalicular staining for MRP2. The pattern of canalicular MRP2 staining of graft was classified into 3 types: homogenous (type C0), focal (type C1), and no (type C2,) staining of the canaliculi. Results: In total, 17.1% graft tissues were type C0, 36.6% were type C1, and 46.3% were type C2. The median operation time was longer in patients with type C2 (562.6 minutes) than in patients with type C0 (393.8 minutes) (P = 0.038). The rates of posttransplant complications were higher in patients with type C2 (100%) than in patients with type C0 (42.9%) and C1 (73.3%) (P < 0.001). Conclusion: MRP2 expression pattern was altered in 82.9% after LT. The pattern of MRP2 alteration was associated with longer operation time and higher rates of post-LT complications.

As the observed result of alteration of Mast cell group treated by Rose essential oil and Lavender essential oil for 3 weeks Luna's stain(${\times}20$), (${\times}100$), it is observed that the damaged group treated by Lavender essential oil is numerous in alteration of mast cell's number, and Mast cell's size is larger than the damaged group.
Objective: Neuropsychiatric manifestations like depression and cognitive dysfunction commonly occur in inflammatory bowel disease (IBD). In the context of the brain-gut axis model, colitis can lead to alteration of brain function in a bottom-up manner. Here, the changes in the response of the hypothalamic-pituitary-adrenal axis and inflammation-related markers in the brain in colitis were studied. Methods: Dextran sodium sulfate (DSS) was used to generate a mouse model of colitis. Mice were treated with DSS for 3 or 7 days and sacrificed. We analyzed the gene expression of brain-derived neurotrophic factor (BDNF), cyclooxygenase 2 (COX-2), and glial fibrillary acidic protein (GFAP), and the expression of GFAP, in the hippocampus, hypothalamus, and amygdala. Additionally, the levels of C-reactive protein (CRP) and serum cortisol/corticosterone were measured. Results: Alteration of inflammatory-related markers varied depending on the brain region and exposure time. In the hippocampus, COX-2 mRNA, GFAP mRNA, and GFAP expression were upregulated during exposure to DSS. However, in the hypothalamus, COX-2 mRNA was upregulated only 3 days after treatment. In the amygdala, BDNF and COX-2 mRNAs were downregulated. CRP and corticosterone expression increased with DSS treatment at day 7. Conclusion: IBD could lead to neuroinflammation in a bottom-up manner, and this effect varied according to brain region. Stress-related hormones and serum inflammatory markers, such as CRP, were upregulated from the third day of DSS treatment. Therefore, early and active intervention is required to prevent psychological and behavioral changes caused by IBD, and region-specific studies can help understand the precise mechanisms by which IBD affects the brain.
This study was undertaken to determine whether the anabolic steroid nandrolone phenylpropionate(NPP) can inhibit the muscle atrophy and reduction in muscle protein synthesis caused by glucocorticoids in female rates. Daily injections of 50mg/kg of corticosterone for eight days induced significant reductions in body weight gain and protein without affecting food intake. The mass, protein and RNA content, ratio of RNA to protein, and fractional rate of protein synthesis, measured in vivo, of gastrocnemius muscle were all significantly reduced by corticosterone treatement. Simultaneous administration of NPP at a dose of 10mg/kg with corticosteorne (50mg/kg) fully inhibited the reductions in the mass, protein and RNA content of gastrocnemius muscle, and body weight gain and protein with no alteration in food intake but the reduction in fractional rate of muscle protein syntheis was only partially prevented. The results indicate that the anabolic steroid nandrolone phenylpropionate is capable of preventing muscle atrophy in female rats treated with excess corticosterion.
Although alteration in protein phosphorylation by specific protein kinases is of importance in transducing cellular signals in a variety of neural/endocrine systems, little is known about protein phosphorylation in the hvpothalamus. The present study aims to explore whether activation of the second messenger-dependent protein kinases affects phosphorylation of specific proteins using a cell free phosphorylation system followed by SDS-polvacrylamide gel electrophoresis. Cytoplasmic fractions derived from hvpothalami of immature rats were used as substrates and several activators and/or inhibitors of CAMP-, phosphatidylinositol- and Ca2+-calmodulin-dependent protein kinases were assessed. Many endogenous proteins were extensively phosphorylated and depending on the signal transduction pathways, phosphorvlation profiles were markedly different. The present data indicate that extracellular signals may affect cellular events through protein phosphorylation by second messengers-protein kinases in the rat hypothalamus.

Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNARE) proteins, composed of two presynaptic membrane proteins [synaptosomal-associated protein of 25 kDa (SNAP-25) and syntaxin] and a presynaptic vesicular protein [vesicle-associated membrane protein (VAMP)], serve as a core of exocytotic fusion machinery, which can be affected by ischemia. Synaptic protein in core region, striatum and cortex has been shown to alter after focal ischemia, however, little is known in hippocampus. Hippocampus is remote from ischemic core, but it is one of the most vulnerable regions. Using immunohistochemistry, the present study was undertaken to investigate the alteration of expression of SNAP-25, syntaxin, and VAMP in the hippocampus of rats which were subjected to middle cerebral artery occlusion (MCAO) for 2h and allowed to reperfuse. At 2 weeks of reperfusion, the SNAP-25 and syntaxin immunoreactivity was increased in the stratum oriens of the CA1 and the stratum lucidum of the CA3 in the ipsilateral hippocampus. However, VAMP immunoreactivity didn't show significant change. These results demonstrate that the level of the presynatpic plasma membrane proteins (SNAP-25 and syntaxin) in the rat hippocampus is more sensitively affected by focal ischemia than that of the synaptic vesicle protein (VAMP).
