EFFECTS OF DIETARY FULL-FAT SOYBEAN LEVELS ON GROWTH PERFORMANCE AND FEED UTILIZATION AND PHOSPHORUS EXCRETION OF CARP (Cyprinus carpio)

  • In order to estimate the nutritive value of roasted full-fat soybean (FFS) in carp diet, growth performance and excretion of protein and phosphorus were examined using carps having mean body weight of 111 g. Growth trial was conducted for 4 weeks using the fishes fed 5 diets (Control, $F_{24}S_{13}$, $F_{16}S_{27}$, $F_8S_{40}$ and $F_0S_{56}$) containing 32%, 24%, 16%, 8% and 0% of fish meal (F) and 0%, 13%, 27%, 40% and 56% of full-fat soybean, respectively. A total of 800 fishes were allotted randomly by groups of 40 to 5 treatments with 4 replicates per treatment. Fishes were fed to station eight times daily. Feeding trial was conducted for 4 weeks. As dietary FFS increased from 0% to 56%, weight gain of fish decreased from 91 g to 39 g and feed conversion ratio increased from 1.06 to 1.95. Protein efficiency ratio (PER) was highest (2.35) in control group which had the highest protein intake, while PER significantly decreased with decrease in protein intake as dietary FFS level increased. Although dietary protein and energy levels were maintained constant, protein excretion per kg weight gain varied from 273 g to 579 g for the groups control and $F_0S_{56}$, respectively. However, the value for control group was not significantly different to those for the groups $F_{24}S_{13}$ and $F_{16}S_{27}$. On the other hand, the highest protein retention efficiency was found in group fed the diet $F_{24}S_{13}$. Fish fed the diet $F_8S_{40}$ excreted the lowest phosphorus (P) based on kg weight gain showing the highest P retention efficiency of 62%. P excretion per kg feed intake was in the range of 5 g to 10 g for the groups $F_8S_{40}$ and $F_{24}S_{13}$, respectively. The present results indicated that as dietary FFS level increased, growth performance and feed utilization decreased while excretion of protein and phosphorus increased. Therefore, it was concluded that more than 25% substitution by FFS for fish meal could exert negative effects on growth and feed utilization of carp.

Development of Lactose-hydrolyzed Milk with Low Sweetness Using Nanofiltration

  • A lactose-hydrolyzed milk with low sweetness was developed using nanofiltration. Raw milk was treated with 0.03% ${\beta}$-galactosidase at $4^{\circ}C$ for 24 h to hydrolyze lactose partially. The resultant lactose-hydrolyzed milk containing 0.43% lactose was then concentrated using a nanofiltration membrane to reach concentration factor of 2.13. The concentration factors and coefficients of retention of milk components in nanofiltration were determined. The concentration factor of milk fat was 2.20 which was the highest of the milk components. The coefficient of retention of calcium and riboflavin was 0.96 and 0.76, respectively. However, the coefficient of retention of glucose, galactose, and sodium was 0.21, 0.15, and 0.22, respectively. Raw milk was treated with 0.1% ${\beta}$-galactosidase at $4^{\circ}C$ for 40 h to hydrolyze lactose fully and then concentrated to reach a concentration factor of 1.6 by using nanofiltration. The concentrated milk was reconstituted with water. The lactose-hydrolyzed milk had sweetness similar to milk. The compositional ratios of crude protein, calcium, sodium, and riboflavin of lactose-hydrolyzed nanofiltrated milk to those of raw milk were 99%, 97%, 77%, and 80%, respectively. This study showed that nanofiltration of lactose-hydrolyzed milk to remove galactose and glucose did not cause significant loss of calcium. The lactose-hydrolyzed nanofiltrated milk contained 0.06% lactose and had sweetness similar to milk.

Immobilized Metal Ion Affinity Chromatography of Genetically Engineered Hirudin Variants

  • Immobilized metal ion affinity chromatography (IMAC) was used to separate various types of recombinant hirudins from the culture broth. The wild type hirudin exhibited a retention in Cu(II)-chelated affinity chromatgoraphy since it contained a single exposed histidine at position 51. To obtain a stronger retention on an IDA-Cu(II) column, the hirudin variants were genetically engineered to contain one or two histidine (s) more than the wild type. While the affinity of the variants for IDA-Cu(II) ligand increased in comparison to that of the wild type, the antithrombin activities reduced to a certain degree. Cu(II), Ni(II) and Zn(II) ions were applied separately to the metal chelate column to investigate ligand specificity with respect to protein retention. As a result, the Cu(II) chelated chromatography gave the best resolution for all the hirudins tested and appeared to be the only IMAC that could be used generally for the purification of hirudins with a decreasing pH gradient.

Relation between sludge properties and filterability in MBR: Under infinite SRT

  • A laboratory-scale submerged membrane bioreactor (MBR) was continuously operated for 100 d at an infinite sludge retention time (SRT) with the aim of identifying possible relation between the filterability of mixed liquor and sludge properties, such as extracellular polymeric substances (EPS), soluble microbial products (SMP), viscosity of mixed liquor, zeta potential of flocs and particle size distributions (PSD). Research results confirmed that MBR can operate with a complete sludge retention ensuring good treatment performances for COD and $NH_3-N$. However, the long term operation (about 40 d) of MBR with no sludge discharge had a negative influence on sludge filterability, and an increase in membrane fouling rates with the time was observed. There as a strong correlation between the sludge filterability and the fouling rate. Among the different sludge properties parameters, the concentration SMP and EPS had a more closely correlation with the sludge filterability. The concentrations of SMP, especially SMP with MW above 10 kDa, had a strong direct correlation to the filterability of mixed sludge. The protein fractions in EPS were biodegradable and available for microorganism metabolism after about 60 days, and the carbohydrates in EPS had a significantly negative effect on sludge filterability in MBR at an infinite SRT.

  • The moisture-dependent gelation characteristics of five different proteins are evaluated to understand the modification of gel strength when they are added in surimi gel. Compressive force and penetration force of protein gels gradually decreased with increase of moisture level, with showing markedly decrease at certain point of moisture level called critical moisture level. The critical moisture level for gelation of SPI-1, SPI-2, EW, WPC and LA were 79.4%, 81.6%, 91.4%, 87,8% and 84.7%, respectively. Beyond this critical level of water, protein gel matrix begins to lose its water binding and structural integrity. The mnisture that was not re tained by a protein was available to diluting the protein matrix and eventually weakened the overall gel strength. EW and MPI showed higher water retention than those of SPI, WPC and LA. The compressive force of SPI, WPC and LA-incorporated surimi gel at the varying moisture levels strongly correlated with the amount if water retained at corresponding moisture level within those protein (r=0.99).

EFFECTS OF DIETARY CELLULOSE AND PROTEIN LEVELS ON GROWTH PERFORMANCE, ENERGY AND NITROGEN UTILIZATION, LIPID CONTENTS AND DEVELOPMENT OF INTERNAL ORGANS IN GROWING CHICKS

  • In order to investigate the effects of dietary cellulose and protein levels on chick performance, four semi-purified diets were formulated so as to contain cellulose at levels of 5% (LC) and 20% (HC) in combination with 10% (LP) and 20% (HP) protein, and fed ad libitum to 1-week-old White Leghorn male chicks for 3 weeks. There were no significant differences in feed intake, body weight gain and feed efficiency between the LC-HP and HC-HP groups. All parameters were lower in the LP groups; the HC-LP group consumed very small amount of feed and lost body weight during the experiment. The retention rates of DM, ash, nitrogen and energy were higher in the HP than the LP groups. The triglyceride concentration of carcass was lower in the HC-LP group and that of liver was higher in the LC-LP group. The carcass total cholesterol level was higher in the HC-HP group. The relative weight of most digestive organs was higher in the HP group irrespective of the cellulose level. In conclusion, the chick performance was primarily influenced by dietary protein level, and when the chicks were fed inadequate levels of protein, the low cellulose level gave a better performance than the high cellulose level.

  • 완두 종자에 축적되는 저장물질은 주로 전분과 단백질로서 이러한 저장물질 때문에 고정이나 전자현미경 관찰시료를 제작하기가 쉽지 않다. 따라서 자엽을 얇게 절편을 만들고 효소를 사용하여 단일세포로 분리한 다음 고정하여 관찰하였다. 완두의 저장단백질이 축적되는 단백질 저장 액포는 종자발달의 이른 시기에 기존의 액포를 둘러싸고 발달하게 되므로서 액포는 수축되고 단백질 저장 액포는 점점 발달하여 그 가장자리에 단백질 덩어리가 축적되게 된다. 이와는 별도로 종자발달의 이른 시기에 조면소포체의 내강에 전자밀도가 높은 단백질이 축적되기 시작하여 늦은 시기에 이 소포체의 끝이 부풀어서 구형의 단백과립으로 발달하였다. 완두종자의 저장단백질은 주로 vicilin과 legumin으로서 단백과립에 대한 면역세포화학적 방법으로 확인한 결과 vicilin은 세포질에 발달된 작은 단백과립과 단백질 저장액포의 가장자리에 축적된 단백질 덩어리에 모두 반응하였으나 legumin은 세포질의 단백과립에만 반응하였다. 또한 소포체에 존재하는 단백질인 Bip은 단백질 저장액포에 축적된 단백질 덩어리의 안쪽 가장자리에만 반응하였다. 이는 단백질이 활발하게 축적되고있는 시기에 특징적으로 작용하는 Bip의 기능과 관련되는 것으로 사료된다.

Euglena의 Cytochrome C552 Methylation에 관한 연구 (Studies on the possible existence of methylarginine in cytochrome C552 isolated from Euglena gracilis)

  • Post-translational modification of protein amino acid residues is a well known metabolic phenomenon. One such side chain modification, protein methylation, occur ubiquitously in nature, in organism ranging from prokaryotic to eukaryotic and the biological significance of protein methylation has begun to emerge. The observation that cytochrome C methylation facilitates the binding of this hemoprotein to mitochondria could be placed as the one of the examples along this line. However, the detail biological meaning of cytochrome C methylation is remained to be clarified. In the aspect of such reason this research was done. The results of this experiment were; 1) pure Euglena gracilis cytochrome C552 was isolated, 2) methylarginine and methylmethionine were not found in cytochrome C552 sequence, 3) however, Unknown Peak at 20.78min of retention time was found, and 4) this Unknown Peak was found only from Euglena cytochrome C552, so far.

Genotypic and Environmental Effects on Cookie Quality of Korean Winter Wheat

  • Cookie baking properties and relationships between cookie baking properties and flour characteristics were evaluated for two years, 1997 and 1998, and at two locations, Suwon and Deokso, with Korean winter wheat cultivars and lines. Cookie baking parameters, except for cookie diameter and top grain score, were influenced by locations and years. Chokwang, Suwon 274, Suwon 275, Suwon 277 and Urimil showed larger cookie diameter and excellent top grain score compared to the other Korean winter wheat cultivars and lines. Among the flour characteristics, protein content, damaged starch content, alkaline water retention capacity and flour swelling volume showed high positive correlation coefficients with cookie baking parameters. Friabilin-present lines showed larger cookie diameter, suitable surface structure of cookie and softer snapping force than the friabilin-absent lines.

Elution Behavior of Protein and Pullulan in Asymmetrical Flow Field-flow Fractionation (AsFlFFF)

  • An AsFlFFF channel was designed and built, and then tested for analysis of pullulans and proteins. Pullulans and proteins having various nominal molecular weights were injected at various conditions of the cross-flow rate ($F_c$) and the channel-out flow rate ($F_{out}$). The retention (measured by the retention ratio R) and the zone broadening (measured by the plate height H) were measured, and then compared with theory. When the incoming flow rate, $f_{in}$ (and thus $F_{out}$) was varied with $F_c$ fixed at 2.5 mL/min, the plate height measured for the pullulan with nominal molecular weight (M) of about 100,000 showed the trend expected by the longitudinal diffusion theory (H decreases with increasing flow rate). In contrast, when $F_{out}$ was varied with the flow rate ratio, $F_c/F_{out}$, fixed constant at 5, the plate height measured for the same sample showed the trend expected from the non-equilibrium theory (H increases with increasing flow rate). Calibration plots (log D vs. log M) obtained with pullulans and proteins were not coincide, probably due to the difference in molecular conformation, suggesting the analysis of pullulans and proteins using AsFlFFF requires independent calibration. It was found that the linearity of the protein-calibration plot was improved by using a buffer solution as the carrier.
