Bacillus thuringiensis의 내독소 단백질의 분리1 (Purtification of Parasporal Protein Crystals of Bacillus thuringiensis)

  • 해충의 미생물적 방제를 위하여 Bacillus thuringiensis 살충제 개발에 관한 기초자료를 얻고자 3종의 B. thuringiensis var. kurstaki, dendrolimus, aizawai를 이용하여 배지내에서 생장특성을 조사하고 이들의 내독소단백질의 분리를 위해 Renograffin 불연속중층법을 이용한 새로운 분리법을 시도하였다. 1. 각공시균주의 생장곡선을 조사한 결과는 3균주 모두 접종후 3시간 이내에 exponential phase에 이르렀고, 7-8시간 정도에서 stationary phase에 이르렀다. 2. 배지내 pH변화는 exdponential phase때 원래 배지에서 1.4정도 저하하였다가 sporulation의 개시에 따라 회복되기 시작하여 후기에 거의 평형상태를 나타내서 균주간에 차이가 없는 것으로 확인되었다. 3. Renograffin 불연속중층법을 이용한 새로운 B. thuringiensis 내독소단백질 분리법은 50$m\ell$ 원심분리관에 10$m\ell$의 시료를 중층, fixed angel rotor를 이용하여 27,000g에서 1시간 원심했을 경우 99.0% 순도와 5.8%의 회수율을 나타내어 지금까지 보고된 방법들보다 더욱 효율적인 것으로 확인되었다.

호밀과 유채의 혼파비율이 생육특성, 사초수량 및 영양소 수량에 미치는 영향 (Effect of Sowing Rate of Mixture on the Growth Chracteristics, Forage Yield and Quality of Rye and Rape)

  • Rye(Secale cereale L.) shows lower dry matter yield and nutrient content when it was harvested before wintering. This study was conducted to investigate how effect forage rape(Brassicu napus Subsp oleifera), which carries early maturity and can be harvested before wintering, on the increase of the forage yield and nutrient content when rye was sown mixed with the rape. Optimum sowing rate of the rye/rape mixture was also pursued in this study. The five treatment of rye single, rape single, ryel20+rape 1Okg/ha, rye84+rape 7kg/ha and rye60+rape 5kg/ha were sown on 2 September 1989. Rape single plot was cut one time(l9 November 1989) and the other plots were cuted two times(19 November 1989 and 20 May 1990). In botanical composition of the mixtures, rape recorded dominant ratio with 64~69% at the autumn cut. The rye84+rape 7kg/ha mixture plot marked the highest dry matter yield with 4.46t/ha among all the experimental plot at the autumn cut(P<0.05). In year total yield of dry matter(t/ha), rye single, rape single, rye120+rape IOkg/ha, rye84+rape 7kg/ha and rye60+rape 5kg/ha marked 13.6, 4.1, 12.7, 12.9 and 11.8, respectively. In crude protein content, the rye single plot was 17.2% at the autumn cut on the other hand the mixture plots showed increasing tendency with 18.0~19.9%. In crude fiber content, the mixture plots were remarkably lower than rye single plot. In year total yield of crude protein, rye84+rape 7kg/ha mixture plot showed the highest yield with 1.6lt/ha among all the experimental plots. Ratio of crude protein yield to crude fiber yield, rye84+rape 7kg/ha tended to be higher than the other mixture plots. It is confirmed that the rye/rape mixture can produce more forage than the rye single cropping when they are harvested in late autumn, besides the mixture forage contains higher crude protein than that rye single forage. It can be said that the rydrape mixture is more useful than the rye single cropping, and optimum sowing rate of the mixture is 84kg/ha of rye+7kg/ha of rape.

A Mixture of Morus alba and Angelica keiskei Leaf Extracts Improves Muscle Atrophy by Activating the PI3K/Akt/mTOR Signaling Pathway and Inhibiting FoxO3a In Vitro and In Vivo

  • Muscle atrophy, which is defined as a decrease in muscle mass and strength, is caused by an imbalance between the anabolism and catabolism of muscle proteins. Thus, modulating the homeostasis between muscle protein synthesis and degradation represents an efficient treatment approach for this condition. In the present study, the protective effects against muscle atrophy of ethanol extracts of Morus alba L. (MA) and Angelica keiskei Koidz. (AK) leaves and their mixtures (MIX) were evaluated in vitro and in vivo. Our results showed that MIX increased 5-aminoimidazole-4-carboxamide ribonucleotide-induced C2C12 myotube thinning, and enhanced soleus and gastrocnemius muscle thickness compared to each extract alone in dexamethasone-induced muscle atrophy Sprague Dawley rats. In addition, although MA and AK substantially improved grip strength and histological changes for dexamethasone-induced muscle atrophy in vivo, the efficacy was superior in the MIX-treated group. Moreover, MIX further increased the expression levels of myogenic factors (MyoD and myogenin) and decreased the expression levels of E3 ubiquitin ligases (atrogin-1 and muscle-specific RING finger protein-1) in vitro and in vivo compared to the MA- and AK-alone treatment groups. Furthermore, MIX increased the levels of phosphorylated phosphoinositide 3-kinase (PI3K), protein kinase B (Akt), and mammalian target of rapamycin (mTOR) that were reduced by dexamethasone, and downregulated the expression of forkhead box O3 (FoxO3a) induced by dexamethasone. These results suggest that MIX has a protective effect against muscle atrophy by enhancing muscle protein anabolism through the activation of the PI3K/Akt/mTOR signaling pathway and attenuating catabolism through the inhibition of FoxO3a.

재래식 간장 및 된장 제조가 대두 단백질의 영양가에 미치는 영향 제2보 메주장의 숙성중에 일어나는 성분 변화 (The Effect of Korean Soysauce and Soypaste Making on Soybean Protein Quality Part II. Chemical Changes During Meju-brine Ripening)

  • 실험실에서 제조된 메주와 한국에서 보내온 재래식가정 메주와 개량메주를 사용하여 장을 담그고 8개월동안의 숙성(장담금)기간중 일어나는 화학성분변화를 검토하였고 메주 종류에 의한 차이를 비교하였다. 또한 분석된 아미노산 조성에 의거하여 과정에 일어나는 대두단백질의 영양가 변화를 추정하였다. 8개월간의 숙성기간중 일반 화학조성의 변화는 거의 일어나지 않았으나 수용성 질소율은 메주의 $13{\sim}29%$에서 $66{\sim}78%$로 증가되었다. 유리아미노태 질소율은 메주의 $4{\sim}7%$에서 8개월 숙성후 $29{\sim}35%$로 증가되었으며 암모니아태 질소율은 $1{\sim}4%$에서 $5{\sim}14%$로 증가되었다. 메주장 숙성중에 일어나는 이들 질소화합물의 성분변화는 사용된 메주의 종류에 크게 의존하였다. 숙성중 메주의 아미노산 조성도 크게 변화되었으며, 특히 methionine은 숙성 1개월동안 급격히 감소하여 원래 농도의 거의 1/2에 달하게되며 그후 숙성 7개월동안 농도의 변화가 없었다. Arginine 과 histidine 은 숙성 1개월이 지난 후부터 나머지 7개월 동안 급격히 파괴되었다. 한편 메주제조 과정까지는 거의 존재가 없던 ornithine 이 숙성과정중에 상당량이 합성되었다. 대체적으로 숙성 3개월동안은 아미노산 조성의 전반적인 큰 변화가 일어나지 않았으며, 이들 변화는 사용된 메주의 종류에 따라 큰 차이를 보였다. 3개월된 실험실 메주로 8개월 장담금을 하였을 경우 대두중의 영양가 수율은 탄수화물이 99%, 조지방이 107%, 조단백질이 93%, 총 아미노산이 74%였다. Histidine, arginine 및 methionine은 과정중 가장 심한 손상을 보였으며 회수율은 각각 25, 27, 및 49%였다. 한편 lysine회수율은 79%였다. 장 걸름 과정 즉, 간장과 된장의 분리과정에 의하여 메주장중에 함유된 약 60%의 조단백질, 거의 모든 조지방 및 80%의 탄수화물이 된장에 남아 있었다. 또한 된장은 lysine을 제외한 모든 아미노산의 농도(16g 중)가 간장보다 높았다. 대두 및 메주를 포함한 모든 시험장류의 제한 아미노산은 methionine 과 cystine 등 유황을 함유하는 아미노산이었으며 제2제한 아미노산은 원류대두에서 valine, 메주와 대부분의 메주장에서 threonine, 된장에서 lysine 그리고 대부분의 간장에서는 tryptophan 으로 나타났다. FAO 표준 아미노산 조성을 기준으로 한 원료 대두 단백질의 단백가(chemical score)는 82 였으며 이것은 대두의 삶음과정에서 77로 저하되었으며 메주발효과정에서 $71{\sim}74$로 다시 저하되었다. 재래식 가정메주의 단백가는 실험실 메주의 그것보다 낮았으며 개량메주는 실험실 메주보다 높은 단백가를 나타내었다. 메주장의 8개월 숙성후 단백가는 $51{\sim}66$으로 상당히 떨어졌으며 분리에 의하여 된장의 단백가는 $60{\sim}71$로, 간장은$45{\sim}57$의 단백가를 갖게되었다. 일반적으로 개량메주로 만든 장들이 높은 단백가를 가졌으며 가정 메주로 만든 장들은 가장 낮은 단백가를 나타내었다. 계란 단백질 조성을 기준으로한 필수 아미노산지수(EAAI)는 단백가(chemical score)와 유사한 수치를 나타내었다. 그리고 이 필수 아미노산지수는 검체단백질중의 아미노산조성의 전반적인 변화를 단백가보다 더 근사하게 나타내었다.

Banhabaikchulcheunma-tang Down-regulates LPS-induced Production of Pro-inflammatory Cytokines

  • Banhabaikchulcheunma-tang (BBCT), a prescription composed of thirteen herbal mixtures, has been widely used in the treatment of brain disorders in Oriental Medicine. However, the mechanisms by which the formula affects on the production of pro-inflammatory cytokines in cerebral infarction (CI) patients remain unknown yet. The levels of secretory protein of pro-inflammatory cytokines, including tumor necrosis factor (TNF)-a, interlukin (IL)-1b, and IL-6, were significantly increased in lipopolysaccharide (LPS)-stimulated THP-1 differentiated macrophage-like (THP-1/M) cells and Peripheral blood mononuclear cells (PBMCs) from CI patients. However, pretreatment with BBCT significantly inhibited the secretion of pro-inflammatory cytokines, including TNF-a, IL-1b, and IL-6, in THP-1/M cells and PBMCs from CI patients with stimulus. Thus, these data indicate that BBCT may be beneficial in the cessation of inflammatory processes of cerebral infarction through suppression on the production of pro-inflammatory cytokines.

Carcass Characteristics of Goats Fed Ammoniated Neem (Azadirachta indica) Seed Kernel Cake

  • The present investigation was carried out to study the affect of feeding urea ammoniated neem kernel meal on carcass characteristics and organoleptic properties of the meat in goats. Eight local weaned kids of 3-4 months age with mean body weight of $7.85{\pm}0.42kg$ were assigned to two groups of four each in a completely randomised design and were offered diets containing isonitrogenous concentrate mixtures containing either peanut meal or urea ammoniated neem (Azadirachta indica) seed kernel meal (UANSKM) along with ad libitium oat hay or green sorghum as roughage for 13 fortnights. The animals were slaughtered at the end of the experimental period and the carcass characteristics and organoleptic evaluation of the meat was carried out. The carcass characteristics in terms of dressing percentage, meat bone ratio, percent edible and inedible did not differ between the treatments. Similarly the organoleptic characteristics and cooking loss were comparable between the treatments implying that UANSKM can be substituted for peanut meal in goat diets to alleviate the shortage and high cost of peanut meal without affecting meat quality.

Orchargrass-Red Clover 혼파이용에 관한 연구 II. Orchargrass-Red Clover 파종비율이 목초의 수량과 품질에 미치는 영향 (Study on the Use of Orchargrass-Red Clover mixture II. Effect of seed rate of orchargrass-red clover on herbage yield and quality)

  • This experiment was conducted over a 3-year period (1990-1992) to evaluate the production and quality of orchardgrass (Potomac)-red clover (Kenland) mixture when orchardgrass + red clover mixture was sown at seed rates of 20+0. 18+2, 16+4, 14+6 and 12+8 kg/ha. Total herbage dry matter, crude protein and digestible organic matter production and organic matter intake were markedly affected by increasing rcd clover seed rate, those increased with increasing red clovcr seed rate due to the cnhancernent of red clover percentage of total herbagc in all 3-year. but significant differences occured between the orchardgrass alone and orchardgrass-red clover mixture with a seed rate of 4,6 and 8 kg/ha red clover (P<0.01). But on the other hand increasing red clover seed rate increased red clover percentage in total herbage, the mean red clover percentage of total herbage at the last cutting ranged from 54 to 65% in first year. 58 to 60% in second year, and 62 to 69% in third year, particularly it was observed that red clover was sown at seed rates of 6 and 8 kg/ha for red clover sown dominated strongly as the cutting times were progressed. Considering the maintenance of good botanical composition balance and the enhancement of production and quality of herbage, the optimized seed rate for orchardgrass-red clover mixtures, 16 kg/ha orchardgrass and 4 kg/ha red clover seems to be considered adequate.

지방질에 대한 초음파 비선형 파라메타 B/A의 의존성 (Dependence of Ultrasonic Nonlinear Parameter B/A on Fat)

  • 광대역 고분자 초음파 변환기를 이용한 초음파 비선형 파라메타 B/A 측정장치를 이용하여 생체조직에서 지방질의 농도에 대한 B/A의 의존성을 알아보기 위하여 계란의 흰자와 노른자의 혼합에 대하여 조사하였다. 이들 두 성분에 대한 음속과 밀도가 서로 1% 정도임에도 불구하고 yoshizuni등에 의해 제시된 선형성보다 이차적 의존성이 더 있음을 확인하였다. 이것은 생체조직을 fat과 nonfat의 두 부분으로 나누어 지방질의 농도에 대한 초음파 비선형 파라메타 B/A와 음속의 크기에 대하여 본 연구에서 제시한 이론식과 잘 일치하였다. 또한 지방질이 없는 skim milk의 다양한 용해도 성분에 대한 B/A와 음속을 측정한 결과 용해도의 농도가 클수록 B/A와 음속의 값이 커짐을 알 수 있었으며, 이들에 영향을 주는 것은 단백질 등으로 예상된다.

효소처리 누에 분말의 생리 활성에 관한 연구 (Functional properties of hydrolysate from silkworms fed Cudrania tricuspidata)

  • In this study, silkworms fed Cudrania tricuspidata leaves were powdered, and hydrolytic enzymes, including viscozyme, papain, and flavourzyme, were added to verify the functionality of the different mixtures. In the general component analysis, the C. tricuspidata silkworm (CS) group exhibited higher crude protein and ash content than did the other groups, and the enzyme-treated groups exhibited higher carbohydrate content than the CS group. The DPPH and ABTS radical scavenging activities were significantly higher in the CS treated with viscozyme (CSV) and the CS treated with viscozyme/flavourzyme (CSVF) than in the other groups, with the CSV group showing the highest reducing power. ACE inhibitory activity was significantly higher in the CS treated with visocozyme/papain (CSVP) than in the CS group. In conclusion, rather than using powdered silkworms fed C. tricuspidata leaves, it would be more effective to use hydrolysates from C. tricuspidata silkworms as raw materials for functional foods.

Anti-inflammatory Action of Herbal Medicine Comprised of Scutellaria baicalensis and Chrysanthemum morifolium

  • Various mixtures were prepared depending on the mixing ratio of Scutellaria baicalensis hot water extract (SB-HW) and Chrysanthemum morifolium ethanol extract (CM-E) and their anti-inflammatory activity were compared. Among them, SB-HW (80 ㎍/mL)/CM-E (120 ㎍/mL) or SB-HW (40 ㎍/mL)/CM-E (160 ㎍/mL) significantly inhibited LPS-stimulated NO and IL-6 levels in RAW 264.7 cells. The SB-HW (80 ㎍/mL)/CM-E (120 ㎍/mL) mixture, which was determined as active mixture, significantly reduced MUC5AC secretion in PMA and LPS-induced NCI-H292 cells. The active mixture also reduced the production of PGE2 and IL-8 in PMA-induced A549 cells. LC-MS/MS analysis showed that the active mixture was composed of high contents of flavone glycosides, such as baicalin and cynaroside. Western blot analysis indicated that the active mixture suppressed phosphorylation of ERK, JNK, and p38, associating with the inhibition of MAPK signaling. Taken together, our results suggest that the active mixture could be applied as a new anti-inflammatory herbal medicine
