Effects of Protein and Lipid Levels in Extruded Pellets on the Growth and Body Composition of the Olive Flounder Paralichthys olivaceus during the Summer and Whiter Seasons

  • Feeding trials were performed at two different water temperatures (summer and winter seasons) to identify suitable protein and lipid (energy) levels to be used in formulating extruded pellets for olive flounder. Experiments were conducted to determine the effects of protein and lipid levels in extruded pellets on the growth and body composition of the flounder in both feeding trials. Six experimental diets were prepared containing three protein levels (46%, 51%, and 56%) and two lipid levels (10% and 17%). In the first experiment, during the summer season ($22{\pm}2.2^{\circ}C$), a triplicate group of fish (initial weight, 114 g) were fed to satiation one of the six diets for 11 weeks. The highest weight gain was observed in fish fed the 56/17 (% protein/% lipid) diet, but this weight gain was not significantly different from that of fish fed the other diets, except for those fed the 46/10 diet. The feed efficiency and protein efficiency ratio of fish fed the 17% lipid diets were higher than those of fish fed the 10% lipid diets at each protein level. In the second experiment, during the winter season ($13{\pm}1.5^{\circ}C$), a triplicate group of fish (initial weight, 107 g) were fed to satiation one of the six diets for 9 weeks. Weight gain was not significantly different among all groups. The feed efficiency and protein efficiency ratio tended to increase with increasing dietary lipid level at each protein level. The whole-body crude lipid content of the of fish fed the 17% lipid diets was significantly higher than that of fish fed the 10% lipid diets at each protein level in both feeding trials. Based on the data obtained in this study, the inclusion of dietary protein at a level of 46% appears to be sufficient to support optimal growth, and increasing the dietary lipid level from 10% to 17% had no beneficial effects on the growth and feed utilization of olive flounder (110-300 g), except for fish fed a 56% protein diet in the summer season.

  • Protein-energy malnutrition, PEM, and increased hs-CRP level are considered to be associated with increased risk of cardiovascular disease (CVD) in hemodialysis (HD) patients. This is commonly referred to as the vicious circle of malnutrition-inflammation-atherosclerosis cardiovascular disease (MIA syndrome) in chronic kidney disease (CKD). Low protein intake can decrease the serum level of albumin and increase inflammational markers; further, both low serum albumin and high hs-CRP are independent risk factors for all-cause mortality in HD patients. The aim of this study is comparing the serum levels of albumin and hs-CRP in HD patients according to the protein intake levels. The total number of subjects was 60 hemodialysis patients; they were grouped by dietary protein intake: low protein intake group (LPI, protein intake < 1.0 g/kg IBW, 11 men and 19 women) and adequate protein intake group (API, protein intake ${\geq}$ 1.0g/kg IBW, 12 men and 18 women). Blood biochemical parameters, nutrient intake, and dietary behaviors were compared between the LPI and API groups. The LPI group showed a significantly lower serum level of albumin and higher serum level of hs-CRP than the API group (p < 0.05). The LPI group showed a significantly lower intake of most nutrients than the API group (p < 0.05). Index of Nutritional Quality of most nutrients of the LPI and API groups were lower than 1.0. Dietary protein intake was positively correlated with the serum level of albumin (r = 0.306, p < 0.05) and negatively correlated with the serum level of hs-CRP (r = -0.435, p < 0.01). The serum level of hs-CRP was negatively correlated with that of albumin (r = -0.393, p < 0.01). According to these result, serum albumin and hs-CRP in HD patients were influenced by the protein intake levels. To prevent MIA syndrome, it is necessary to improve nutritional status, especially in protein and energy.

  • In two experiments 640 starcross replacement pullets between 25 and 154 days of age were fed ad libitum on either of 16 diets formed by the combination of $4CP{\times}4ME$ levels to study the interaction of CP and ME on growth performances. In both experiments, feed intake decreased, but protein intake, energy intake, live weight gain and feed conversion efficiency increased and sexual maturity hastened with the increase of dietary protein and/or energy level. The protein conversion efficiency decreased with the increase of dietary protein level. The energy conversion efficiency, however, did not show any relationship with dietary energy level. There was a greater improvement of growth performance due to simultaneous increase of dietary protein and energy level than that of increasing protein or energy alone.

고지방식을 섭취한 흰쥐의 체내 지질 함량에 대한 대두 단백질과 칼슘의 섭취 효과 (Effects of Dietary Soy Protein and Calcium on Blood and Tissue Lipids in Rats Fed Fat-Enriched Diet)

  • This study was carried out to examine how dietary protein and calcium levels in rats fed fat-enriched diet affect the total lipid and cholesterol contents of blood and tissues. Male Sprauge-Dawley rats weighing approximately 200g were fed six purified diets which contained 18%(w/w) beef tallow, 1% (w/w) cholesterol, two source of protein, casein or isolated soy protein (ISP) and three levels of dietary calcium, 0.1%, 0.4% and 1.0%, first, for four weeks, and second, for eight weeks. The contents of the total lipid, cholesterol and triglyceride in blood, liver, heart and feces were determined. After four weeks feeding serum lipid and cholesterol concentrations significantly decreased in rats fed 1.0% (w/w) level calcium, regardless of dietrary protein sources. After eight weeks, these concentrations were significantly lower in the rats fed soy protein than in casein-fed rats. As dietary calcium level increased serum and tissue lipid and cholesterol contents were decreased and fecal lipid excretion increased. It is concluded that hypolipidemic and/or hypocholesterolemic effects of soy protein and calcium were partly due to decrease in lipid absorption.

Effect of the Dietary Protein Level on Plasma Glucose, Lipids and Hormones in Streptozotocin-Diabetic Rats

  • Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.

Identification of a Cellular Protein Interacting with Murine Retrovirus Gag Polyproteins

  • The retroviral Gag polyprotein directs the assembly of virion particles and plays an important role in some events after entry into a host cell. The Gag polyprotein of a virus mixture is responsible for inducing murine acquired immunodeficiency syndrome (MAIDS) when injected into susceptible strains of mice. In order to identify the host cellular proteins which interact with the MAIDS virus Gag proteins and possibly mediate the function of the Gag proteins, mouse T-cell leukemic cDNA expression library was screened using the yeast GAL4 two hybrid system. Of 11 individual positive clones, the clone Y1 was selected for the study of protein-protein interaction. Its DNA sequence revealed that it was an exact match to the murine SH3 domain-containing protein SH3P8. It is expressed as 2.4 kbp transcripts in testis at higher levels and in various tissues tested at lower levels. Glutathione S-transferase-Y1 fusion protein binds tightly to $Pr60^{def-gag}$ as well as $Pr65^{eco-gag}$.

산란계 사료의 에너지와 단백질 수준이 산란율과 란중에 미치는 영향 (Effects of Dietary Protein and Energy Levels on Egg Production and Egg Weight of Laying Hens)

  • 사료의 에너지와 단백질 수준이 산란계의 능력에 미치는 영향을 조사하기 위하여 3회의 시험을 실시하였다. 시험1에서는 352일령의 마니나 브라운종을, 시험 2에서는 280일령의 갈색 와렌종을, 시험 3에서는 317일령의 갈색 와렌종을 각각 이용하였다. 각 시험에서 공히 총 360수의 공시계를 18수씩 20개구로 나누어 4개처리에 5반복으로 배치하였다. 시험 설계는 2가지 수준의 에너지(2500과 2800 kcal ME/kg)와 2가지 수준의 단백질 (13% 및 16 %)을 조합한 2$\times$2 요인시험을 실시하였다. 모든 공시계는 산란계용 Cage에 cage 당 2수씩 수용하였으며 시험 1과 2에서는 4주간의 시험기간동안 시험사료와 물을 자유로이 급여하였고 시험3에서는 2주간의 시험기간동안 사료섭취량을 1일 수당 130g으로 제한하여 급여하였다. 시험 1과 2에서 고에너지 사료를 섭취한 닭이 산란율이 다소 높았으나 유의성은 없었다. 그러나 시험 1에서는 단백질 수준이 산란율에 영향을 미치지 못하였다. 사료를 제한 급여한 시험3에서는 고에너지 사료에서 산란율이 개선되었으며(P〈0.05) 단백질 수준도 산란율에 영향을 미쳤으나 유의성은 없었다. 시험 1과 2에서 란중은 에너지 수준이나 단백질 수준에 의하여 영향을 받지 않았으나 시험 3에서는 고에너지 수준에서 란중도 약간 개선되는 경향을 보였다. 3회의 시험 모두에서 사료의 단백질이나 에너지 수준이 증가할 때 egg mass가 증가하는 경향을 보였다. 사료의 에너지나 단백질 수준이 증가할 때 사료섭취량은 감소하고 사료효율이 개선되는 경향을 보였다. 또한 사료의 에너지 수준이 증가할 때 단백질 효율(egg mass/단백질섭취량)이 개선되었으며 사료의 단백질 수준이 증가할 때 에너지 효율(egg mass /ME 섭취량) 이 개선되었다.

단백질과 칼슘의 급여 수준이 납중독된 흰쥐의 체내 무기질함량에 미치는 영향 (Effects of Dietary Protein and Calcium levels on Ca, Fe, Cu, Zn and Mg Level of the Tissues of the Pb-administered Rats)

  • 식이 단백질과 칼슘의 수준이 납중독된 흰쥐의 체내무기질 함량에 미치는 영향을 관찰하기 위해, 흰쥐에게 납 2,000ppm을 함유한 식수를 제한없이 주면서, 식이 단백질과 칼슘 수준을 각각 3수준, 사육 기간을 3주와 7주로한, 3$\times$3$\times$2 요인 실험으로 설계하고 대조군을 별도로 설정하여 조사한 결과는 다음과 같다. 혈청, 간 및 대퇴골의 칼슘 함량은 납 급여시 감소하였고, 혈청과 대퇴골에서는 식이 단백과 칼슘 함량이 낮을수록 감소를 나타냈으나, 간은 식이 종류에 따른 차이를 보이지 않았다. 신장의 칼슘 함량은 납 급여나 식이 종류에 따른 영향을 받지 않았다. 혈청 철 함량은 칼슘 섭취량이 많으면, 납 급여의 영향을 받지 않았지만, 간의 철 함량은 칼슘이 충분한 고단백 식이를 섭취한 경우만 영향을 받지 않았다. 혈청 구리 함량은 납 급여시 감소하였고, 식이 단백과 칼슘 함량이 적을수록 감소하였다. 간의 구리 함량은 고칼슘군은 납 급여의 영향을 받지 않았으나, 나머지군들은 감소하였다. 혈청 아연 함량은 저단백-저칼슘군에서만 납 급여의 영향을 받아 감소하였고, 간의 아연 함량은 납 급여나 식이 단백 및 칼슘의 영향을 받지 않았다. 혈청 마그네슘 함량은 식이 단백과 칼슘이 동시에 부족할 때, 납 급여의 영향을 받아 감소하였다. 간의 마그네슘 함량은 납 급여시 감소하였고, 식이 단백 함량이 낮을수록 감소하였다.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01). 시험스트레스가 대학생의 질소대사에 미치는 영향 (Effect of Examination-stress on Nitrogen Metabolism of College Students)

  • This study was performed to investigate effects of examination-stress and protein supplementation on nitrogen metabolism and blood protein levels of Korean college students. Experiment was conducted at the beginning of a academic term and during midterm examination. During midterm examination, subjects were classified into two groups randomly : protein supplemental group(male n=6, female n=10) and placebo group(male n=4, female n=9). Protein capsules(2g/day) above 10% of indispensible amino acids requirement estimates were given to supplemental group for 10 days. At the begining of the term, male students(n=12) ingested 223.15mgN/kg/d, excreted 20.7mgN/kg/d in feces, and excreted 94.31mgN/kg/d in urine. Their apparent protein protein digestibility was 90.72%, true N balance was +100.11mgN/kg/d, and the mean maintenance N requirement of mixed Korena diet calculated was 112.13mgN/kg/d. Female students(n=19) ingested 171.44mgN/kg/d, excreted 22.13mgN/kg/d in feces, and excreted 122.92mgN/kg/d in urine. Their apparent protein digestibility was 86.76%, true N blance was + 18.39mgN/kg/d, and the mean maintenance N requirement calculated was 135.31mgN/kg/d. Blood levels of serum total protein, albumin, and BUN were within normal range. During midterm examination, fecal and urinary N excretions of female subjects(n=19) were increased, especially urea N markedly, and urea N/creatinine N ratio was augumented significantly. Apparent protein digestibility of male subjects(n=10) was decreased. Examination-stress showed 8.05mgN/kg/d (7.2%) increase of mean maintenance N requirement in male and 8.55mgN/kg/d(6.3%) increase in female students in comparison with that of the beginning of the term. Serum total protein and albumin levels showed no significant change, but serum transferrin level of female were decreased significantly. During midterm examination, females supplemented with protein capsules(2g/d)had no significant increase in fecal and urinary N excretions.
