Effects of Phytase Supplementation of Diets with Two Tiers of Nutrient Specifications on Growth Performance and Protein Efficiency Ratios of Broiler Chickens

  • In two feeding experiments male and mixed-sex broiler chicks were offered diets based on sorghum and a wheatsorghum blend with two tiers of nutrient specifications, without and with microbial phytase (600 and 800 FTU/kg), from 7-25 and 1-42 days post-hatch, respectively. The nutrient specifications for protein, amino acids, energy density and phosphorus (P) of standard diets were reduced to formulate the modified diets on a least-cost basis. Calculated differences in nutrient specifications between standard and modified diets ranged from 14.3 to 17.1 g/kg crude protein, 0.24 to 0.40 MJ/kg apparent metabolisable energy (AME) and 1.06 to 1.20 g/kg available P. In both experiments, reduced nutrient specifications had a negative impact on growth rates and feed efficiency and phytase supplementation had a positive influence on growth performance and protein efficiency ratios (PER). Phytase addition to the less expensive, modified diets either partially or entirely compensated for reduced growth performance and, consequently, feed costs per kg of live weight gain were reduced. In Experiment 1, phytase increased (p<0.001) nitrogen-corrected AME (AMEn) from 15.39 to 15.89 MJ/kg dry matter. For nitrogen (N) retention there was an interaction (p<0.05) between diet type and phytase as the effects of phytase on N retention were more pronounced in the modified diets, with an increase from 0.512 to 0.561. These results demonstrate the positive effects of phytase on protein and energy utilisation, in addition to its established liberation of phytate-bound P and illustrate the feasibility of assigning nutrient replacement values to the feed enzyme for consideration in least-cost ration formulations. Further work is, however, required to define the most appropriate reductions in nutrient specifications in association with phytase supplementation.

Influence of Yeast Fermented Cassava Chip Protein (YEFECAP) and Roughage to Concentrate Ratio on Ruminal Fermentation and Microorganisms Using In vitro Gas Production Technique

  • The objective of this study was to determine the effects of protein sources and roughage (R) to concentrate (C) ratio on in vitro fermentation parameters using a gas production technique. The experimental design was a $2{\times}5$ factorial arrangement in a completely randomized design (CRD). Factor A was 2 levels of protein sources yeast fermented cassava chip protein (YEFECAP) and soybean meal (SBM) and factor B was 5 levels of roughage to concentrate (R:C) ratio at 80:20, 60:40, 40:60, 20:80, and 0:100, respectively. Rice straw was used as a roughage source. It was found that gas production from the insoluble fraction (b) of YEFECAP supplemented group was significantly higher (p<0.05) than those in SBM supplemented group. Moreover, the intercept value (a), gas production from the insoluble fraction (b), gas production rate constants for the insoluble fraction (c), potential extent of gas production (a+b) and cumulative gas production at 96 h were influenced (p<0.01) by R:C ratio. In addition, protein source had no effect (p>0.05) on ether in vitro digestibility of dry matter (IVDMD) and organic (IVOMD) while R:C ratio affected the IVDMD and IVOMD (p<0.01). Moreover, YEFECAP supplanted group showed a significantly increased (p<0.05) total VFA and $C_3$ while $C_2$, $C_2:C_3$ and $CH_4$ production were decreased when compared with SBM supplemented group. In addition, a decreasing R:C ratio had a significant effect (p<0.05) on increasing total VFA, $C_3$ and $NH_3$-N, but decreasing the $C_2$, $C_2:C_3$ and CH4 production (p<0.01). Furthermore, total bacteria, Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminococcus albus populations in YEFECAP supplemented group were significantly higher (p<0.05) than those in the SBM supplemented group while fungal zoospores, methanogens and protozoal population remained unchanged (p>0.05) as compared between the two sources of protein. Moreover, fungal zoospores and total bacteria population were significantly increased (p<0.01) while, F. succinogenes, R. flavefaciens, R. albus, methanogens and protozoal population were decreased (p<0.01) with decreasing R:C ratio. In conclusion, YEFECAP has a potential for use as a protein source for improving rumen fermentation efficiency in ruminants.

Effects of Dietary Protein and Lipid Levels on Growth, Feed Utilization and Body Composition of Adult Starry Flounder (Platichthys stellatus)

  • A 25-week feeding trial of two dietary protein (47 and $52\%$) and three dietary lipid level (7, 12 and $17\%$) factorial design with three replications were conducted to determine effects of dietary protein and lipid levels on growth, feed utilization and body composition of adult starry flounder (Platichthys stellatus), average initial weight 332 g, during the winter season. Survival of fish was not affected by either dietary protein or dietary lipid level. Weight gain, feed efficiency and protein efficiency ratio improved with dietary protein and lipid levels except for those of fish fed the $52\%$ protein diet with $17\%$ lipid. The best growth and feed utilization were observed in the $52\%$ protein diet with $12\%$ lipid, but were not significantly different from those of fish fed the $52\%$ protein diet with $17\%$ lipid or the $47\%$ protein diets with $17\%$ lipid levels. Hepatosomatic and visceral somatic indexes were significantly influenced by dietary protein level, but not by dietary lipid level. None of moisture, crude protein, crude lipid, or glycogen contents of dorsal muscle or liver in starry flounder except for crude lipid in dorsal muscle was significantly influenced by either dietary protein or dietary lipid level. Plasma cholesterol concentration was significantly influenced by both dietary protein and dietary lipid levels. The results of this study suggest that the diets containing $47\%$ protein with $17\%$ lipid or $52\%$ protein with $12-17\%$ lipid are optimal for growth and feed utilization of adult starry flounder under these experimental conditions.

Effects of Dietary Protein Sources on Growth and Body Composition in Olive Flounder (Paralichthys olivaceus)

  • A 6-week feeding trial was conducted to investigate the effects of six different dietary animal protein sources on growth and body composition of olive flounder, Paralichthys olivaceus in recirculating system. White fish meal (WFM), flounder muscle (FLM), carp muscle (CM), blood meal (BM), squid liver powder (SLP) and casein (CA) were used as the main animal protein sources in the six experimental diets. Fish averaging $2.9\pm0.03g$ $(mean\pm SD)$ were distributed to each aquarium as a group of 15 fish and were fed one of the six experimental diets to each treatment of triplicate groups. After 6-week of the feeding trial, fish fed white fish meal (WFM) and flounder muscle (FLM) diets showed a significant higher weight gain $(WG\%)$ (P<0.05) than those of fish fed the CM, BM, SLP and CA diets. Fish fed BM diet showed the lowest WG among all the dietary treatments. Feed conversion ratio (FCR) and protein efficiency ratio (PER) showed the similar trend as WG. Hematocrit and hemoglobin were not affected by the dietary treatments. Fish fed the FLM and CM diets showed significant higher survival rate than those of fish fed BM diets, and there was no significant difference in survival of fish fed WFM, FLM, CM, SLP and CA diets. These results indicated that WFM and FLM are the best dietary protein sources tested in olive flounder.

건조비지 첨가 두부의 영양적 품질평가 1. 단백질의 영양가 (Nutritional Evaluation of Tofu Containing Dried Soymilk Residue(DSR) 1. Evaluation of Protein Quality)

  • 두부 제조공정 중의 부산물인 비지의 효율적인 이용을 위하여, 원료 대두 중량의 10%에 해당하는 건조비지를 첨가시킨 두부를 제조하여 이의 영양적 품질을 검토하였다. 일반성분은 수분의 경우 건조비지 첨가 두부가 80.6%, 일반 두부가 77.5%로써 비지 첨가 두부가 다소 높았으며 단백질의 함량은 건조비지 첨가 두부와 일반 두부 각각 38.5%, 45.8%였으며, 지질의 함량은 각각 34.3%, 26.9%로써, 단백질과 지질 함량이 낮은 건조비지 첨가로 인해 일반 두부에 비해 건조비지 첨가두부가 낮은 함량을 보였다. Trypsin inhibitor (TI) 함량은 비지의 경우 잔존량이 6.9 (mg/g시료)정도였고, 건조비지의 경우에도 거의 비슷하였다. 건조비지 첨가두부에는 원료 대두에 있었던 TI의 12% 정도만 남아있었다. 단백질의 효소소화율 (enzyme digestibility)은 비지, 건조비지 각각 87%, 86% 정도로 높았고 건조비지 첨가 두부도 일반 두부와 거의 동일한 91% 정도였다. 효소소화율은 TI함량이 저하함에 따라 역상관관계를 가지면서 상승하였다. 구성 아미노산 조성은 일반 두부와 건조비지 첨가 두부는 거의 차이가 없었으며, 총아미노산에 대한 필수 아미노산의 비율은 일반 두부가 40.4%, 비지첨가 두부가 40.6%였으며, lysine 함량이 높은 건조비지로 인하여 건조비지 첨가 두부는 lysine의 보충 효과가 있었다. 단백품질을 Computed Protein Efficiency Ratio (C-PER)로 계산할 때 일반 두부는 1.95, 비지 첨가 두부는 1.4였으나 Discriminant Computed Protein Efficiency Ratio (DC-PER)로 계산할 때는 오히려 비지 첨가 두부경우가 높았다. 여러 연구자들의 대두가공품에 대한 생체실험 결과와 비교할 때 일반 두부를 제외하고는 두유, 비지 및 비지첨가두부의 영양평가는 DC-PER 방법이 유리한 것으로 생각되었다.

조피볼락 육석용 배합사료와 시판사료의 사육효과 비교 (Evaluation of Experimental Formulated Diets and Commercial Diets for Growing Korean Rockfish (Sebastes schlegeli))

  • 실용적인 사료의 효과를 검토하기 위해 단백질함량이 $41\~45\%$인 4종류의 실험사료를 dry pellet 형태로 제조하였다. 단백질원으로 어분을 $58\%$ 첨가한 사료를 대조구(사료 1)로 하고, 대조구의 어분을 육분, 콘글루텐 밀, 대두박 및 혈분으로 각각 다른 비율로 대체하여 어분 함량을 $40\%$ 전후로 감소시킨 실험사료들(사료 $2\~4$)을 설계하여 시판되고 있는 해산어용 배합사료 A, B, C (단백질 함량 : $51\~53\%$) 및 양어가들이 선호하는 RMP 사료(넙치용 분말사료와 냉동 메가리를 1 : 1의 비율로 혼합한 moist pellet, 단백질 함량 : $58\%$)와 그 효과를 사육 실험을 통해 비교하였다. 평균체중 125 g의 조피볼락을 선별하여 실험사료마다 2반복으로 15주간 사육실험한 결과, 최종 평균체중, 증체율, 사료효율, 일일사료섭취율, 일일단백질섭취율, 단백질효율 및 간 중량지수가 RMP 구를 제외한 실험사료와 상품사료를 급여한 실험구들은 대조구와 통계적인 차이는 없었다(P>0.05). RMP 섭취구의 증체율이 대조구와는 유의한 차이가 없었지만, 실험사료(사료 $2\~4$)와 상품사료(사료 $A\~C$)보다 유의적으로 높은 값을 보였다(P<0.05). 또한, 단백질효율은 RMP 구가 가장 높았고, 상품사료 B가 가장 낮은 값을 보였다. 대조사료에서 1 kg의 어체를 증체시키는데 소요되는 사료비용을 100으로 환산하였을 때, 사료 3 급여구는 92, 사료 4 급여구는 79로 나타나, 대조구보다 $10\~20\%$ 정도의 사료비가 절감되었고, RMP 사료 급여구는 대조구보다 $27\%$의 사료비가 더 소요되는 것으로 나타났다. 실험 종료시 간과 근육의 수분, 단백질, 지질 및 회분 함량은 모든 실험구간에 유의차는 없었다(P>0.05).

Effects of Dietary Protein and Lipid Levels on Growth and Body Composition of Sub-adult Flounder Paralichthys olivaceus During the Summer Season

  • A $3{\times}2$ factorial experiment was conducted to investigate the proper dietary protein and lipid levels for the growth of sub-adult flounder Paralichthys olivaceus reared during the summer season. Six experimental diets were formulated to contain three levels of protein (45%, 50% and 55%) and two levels of lipid (9% and 14%). Duplicate groups of fish (initial body weight of 298 g) were hand-fed to apparent satiation during the summer season ($21.8{\pm}1.7^{\circ}C$) for 60 days. Survival of each group was over 83% and there was not significant difference among all groups. Weight gain of fish fed the 45% protein diet with 14% lipid was not significantly different from that of fish fed the 50% and 55% protein diets with 9% and 14% lipids, but weight gain of fish fed the 45% protein diet with 9% lipid was significantly lower than that of fish fed the 55% protein diets with 14% lipid. Feed efficiency tended to increase with increasing dietary lipid level at each protein levels, although no significant differences were observed at 50% and 55% protein levels. Protein efficiency ratio, daily feed intake, condition factor and hepatosomatic index were not significantly affected by dietary protein and lipid levels. Crude lipid content of the liver tended to increase with increasing dietary lipid level at the same protein levels, but the opposite appearance was found for moisture content. The contents of moisture, crude protein and crude lipid of the dorsal muscle were not significantly affected by dietary protein and lipid levels. Based on data obtained form this study, inclusion of dietary protein at level of 45% appears sufficient to support optimal growth, and an increase of dietary lipid level from 9% to 14% has beneficial effects on feed utilization of sub-adult flounder during the summer season.

사육 수온에 따른 쏘가리(Siniperca scherzeri) 치어의 성장 및 체조성 변화 (Effect of Water Temperature on Growth and Body Composition of Juvenile Mandarin Fish Siniperca scherzeri)

  • This study was conducted to investigate the effects of different water temperatures on growth, survival, biochemical composition, and blood physiological parameters of juvenile mandarin fish Siniperca scherzeri. Feed-trained juvenile fish were subjected to four water temperatures (20, 23, 26 and 29℃) with two replicate groups. The fish were fed to apparent satiation twice daily using a formulated diet containing 55% crude protein and 6% crude lipid. After the 8-week feeding trial, survival was >96% in all groups. Weight gain and feed efficiency of fish reared at 26 and 29℃ were higher than those reared at 20℃. The protein efficiency ratio, daily feed intake, and whole body proximate composition were not affected by water temperature. These results indicate that a suitable water temperature range for optimal growth and feed efficiency of juvenile mandarin fish is 26-29℃ under these experimental conditions.

Studies on Heated Protein Quality Using Homoarginine Method

  • To determine the quality of heated protein, in vitro method, invluding lysine, lysionalanine, and fructose-lysine as well as homoarginine by guanidination of lysine, was assessed using heated casein with of without glucose. In vivo methods such as PER, digestibility and BV were also tried on homoarginine, lysinoalanine, fructoselysine, and lysine. The nonreactive lysine for huanidination was hardly digestive, while the non heat damaged lysine side chanis in the protein were accessible for guanidination as well as for the digestion. A linear correlation(${\gamma}$=0.80) was obstained between PER and digestibility of the analysed lysine. Digestibility of homoarginine was higher that of true protein. However, in the guanidinated heated casein with glucose, digestibility of homoarginine was significantly reduced. It is suggested that the homoarginine method may mislead to over- or underestimation of the damaged protein quality.

Dietary Protein Requirement for Young Far Eastern Catfish Silurus asotus

  • A feeding trial was conducted to determine the optimum dietary protein requirement of young far eastern catfish Silurus asotus. Five isocaloric diets were formulated to contain graded levels of protein (35, 40, 45, 50, and 55%). Triplicate groups of fish (initial body weight of 44 g) were hand-fed to apparent satiation for 9-weeks. Weight gain and specific growth rate of fish fed 55% protein diet were significantly higher than those of fish fed 35 and 40% protein diets, but not significantly different from those of fish fed 45 and 50% protein diets. The feed efficiency of fish fed 50 and 55% protein diets was significantly higher than that of fish fed 35, 40 and 45% protein diets. The protein efficiency ratio of fish fed 40% protein diet was significantly higher than that of fish fed 45, 50 and 55% protein diets, but not significantly different from that of fish fed 35% protein diet. The dietary protein level significantly affected whole body lipid and moisture contents. The results of this study suggest that the 45% protein in the diet is optimal for maximum growth of young far eastern catfish weighing in the range of 44 to 227 g.
