The Effects of Vitamin B6 Deficiency on Stored Fuel Utilization During 3 days Fasting or 6 days underfeeding in Rats

  • The effects of vitamin B6 deficiency on energy utilization during fasting or underfeeding were studied in rats. Fifteen rats were fed a vitamin B6 deficient(-B6) diet and another 15 rats wee fed a control (+B6) diet. These rats were fed for 5 weeks with respective diet, and then subdivided into 3 groups : non-fasted group, fasted group, underfed group. Rats of the fasted group were fasted for 3 days and those of underfed group for 6 days. At the respective time (non-fast, 3 day-fast, 6 day-underfeed at 5 weeks), animals were sacrificed. Feed efficiency ratio of - B6 rats was significantly lower than that of +B6 rats. In - B6 rats, the liver and kidney weights were significantly heavier than those of +B6 rats but spleen and heart weights were not. In non-fasted group, liver protein and triglyceride level of - B6 rats were significantly higher than that of +B6 rats. After - B6 rats were fasted for 3 days, plasma free fatty acid level was significantly lower but liver glycogen level was higher than that of +B6 rats and muscle protein level of +B6 was decreased while that of - B6 was not changed. Vitamin B6 deficiency had little effect on the energy utilization with 6 days underfeeding. These results suggest that vitamin B6 deficiency may impair the utilization of stored fuel during fasting.

Vitronectin regulates osteoclastogenesis and bone remodeling in a mouse model of osteoporosis

  • Vitronectin (VN) is an extracellular matrix protein with a crucial role in regulating bone remodeling. In this study, we aimed to investigate the effect of VN deficiency in a mouse model of osteoporosis induced by ovariectomy (OVX). The findings revealed that the absence of VN led to an increase in the activity of tartrate-resistant acid phosphatase (TRAP), a marker for osteoclasts, in the plasma of OVX-operated mice. TRAP staining further demonstrated that VN deficiency resulted in a higher number of osteoclasts within the femurs of OVX-operated mice. X-ray micro-computed tomography analysis of the femurs in OVX-operated mice indicated that VN deficiency significantly suppressed the OVX-induced increase of marrow area and total volume of bone. Additionally, we assessed structural model index (SMI) and degree of anisotropy (DA) as indices of osteoporosis. The results showed that VN deficiency effectively attenuated the OVX-induced increase in SMI and DA among OVX-operated mice. In summary, our study demonstrates the vital role of VN in regulating osteoclastogenesis and bone remodeling in the mouse model of osteoporosis.

Sulphur Supply Level Effects on the Assimilation of Nitrate and Sulphate into Amino Acids and Protein in Forage Rape (Brassica napus L.)

  • Sulphur deficiency has become widespread over the past several decades in most of the agricultural area. Oilseed rape (Brassica napus L.) is a very sensitive to S limitation which is becoming reduction of quality and productivity of forage. Few studies have assessed the sulphur mobilization in the source-sink relationship, very little is known about the regulatory mechanism in interaction between sulphur and nitrogen during the short-term sulphur deficiency. In this study, therefore, amount of sulphur and nitrogen incorporated into amino acids and proteins as affected by different S-supplied level (Control: 1 mM ${SO_4}^{2-}$, S-deficiency: 0.1 mM ${SO_4}^{2-}$, and S-deprivation: 0 mM ${SO_4}^{2-}$) were examined. The amount of sulphur in sulphate (S-sulphate) was significantly decreased by 25.8% in S-deprivation condition, compare to control, but not nitrogen in nitrate (N-nitrate). The markedly increase of sulphur and nitrogen incorporated amino acids (S-amino acids and N-amino acids) was observed in both S-deficiency and S-deprivation treatments. The amount of nitrogen incorporated proteins (N-protein) was strongly decreased as sulphur availability while the amount of sulphur incorporated into proteins (S-protein) was not affected. A highly significant ($p{\leq}0.001$) relationship between S-sulphate and S-amino acid was observed whereas the increase of N-amino acids is closely associated with decrease of N-proteins. These data indicate that increase of sulphur and nitrogen incorporated into amino acids was from different nitrogen and sulphur metabolites, respectively

Evaluation of calcium, magnesium, vitamin D and some biomarker parameters levels in children with favsim disease in the Basrah Governorate-Iraq

  • Essential minerals are important for human health because they support biochemical reactions in metabolism and may play a role in the development of glucose-6-phosphate dehydrogenase deficiency (G6PD). We investigated the relationship between calcium, magnesium, urea, creatinine, total protein, glucose and vitamin D levels in G6PD deficiency in this study. The control group consisted of 40 people (23 females and 17 males) and the patient group consisted of 50 people (20 females and 30 males), all of whom were between the ages of (1-12 years). The findings revealed that the calcium level in patients, depending on sex factor, has a highly significant increase (p <0.0001) when compared to the control group, especially in children who are females rather than males who are affected by G6PD deficiency. In addition, the level of magnesium was found to be significantly different (p <0.0001) in children male patients when compared to the control group. On the other side, the level of total protein was found to be significantly high in children patients (p <0.01) when comparing with control group, and the levels of urea, creatinine and glucose were found to be highly significant increase (p <0.001) in patients when comparing to healthy groups, vitamin D levels were significantly lower (p <0.0001) with G6PD deficiency comparing to control group. In conclusion, the low and high significant associations between vitamin D, calcium, magnesium, urea, creatinine, and glucose indicate that more research is needed to better understand their roles in G6PD development.

Pyridoxine결핍이 뇌의 신경전달물질에 미치는 영향 - Catecholamine 대사 - (Pyridoxine Deficiency on Neurotransmitters in the Developing Rat Brain - Catecholamine Metabolism-)

  • Pregnant rats were fed a pyridoxine deficient diet during the gestation and lactation. DEF I group received the deficient diet from delivery ; DEF II group, from the 15 th day of gestation. Body and brain weights, brain protein, DNA, RNA, plasma GOT and GPT, and catecholamines were measured. Effect of MAO inhibiting drug, pargyline, was determined. Brain protein, DNA, and RNA of offsprings of deficient groups were significantly lower than the control group, but RNA/ DNA, brain weight/DNA, and protein/DNA show that cell number were more affected than cell size by the pyridoxine deficiency during the 3rd week of gestation and lactation. Plasma GOT activities were more significantly different than plasma GPT between the control and deficient group. Brain norepinephrine of offsprings of deficient group were significantly lower than the control, but brain dopamine content was not significantly different from the control. At 2nd and 3rd week, norepinephrine was significantly depressed in deficient groups. Pargyline treatment affected a 1.2 fold increase in catecholamines in 3hr while the control had a 1.5 fold increase. Thus norepinephrine and dopamine synthesis was depressed in the deficient groups. Dopaminergic neurons may be less dependent on pyridoxine level than neurons from norepinephrine. Pyridoxine deficiency in maternal diet is not so critical to brain catecholamines of offspring except to the neonatal rats.

  • A successful outcome of pregnancy requires an efficient uteroplacental vascular system. Since this system may be compromised by disorders of haemostasis associated with a prothrombotic state, maternal thrombophilia might be a risk factor for fetal loss. Hereditary deficiencies of the naturally occuring anticoagulants are well recognized conditions predisposing to recurrent venous thromboembolism. Since thrombotic phenomena have been implied as a cause of abortion and stillbirth, these deficiencies might increase the risk of fetal demise. We have experienced a case of antiphospholipid syndrome associated with protein C deficiency in patient with recurrent spontaneous abortion. So we report this case with a brief review of literatures.

Monitoring Nutritional Status of Dairy Cows in Taiwan Using Milk Protein and Milk Urea Nitrogen

  • The climate and marketing system of raw milk in Taiwan create problems in balance feeding of protein and energy in lactating cows in Taiwan. Level of urea nitrogen both in bulk milk and serum reflects ruminal protein degradation and post-ruminal protein provision, whereas milk protein concentration responds to dietary energy intake and bacterial protein production in the rumen. Establishment of a range of reference standards in milk protein and urea nitrogen levels can be applied as a noninvasive economical feeding guide to monitor the balance of protein and energy intake. Standard reference levels of 3.0% milk protein and 11-17 mg/dL milk urea nitrogen (MUN) were established. Level of milk protein below 3.0% is regarded as indicating inadequate dietary energy whereas MUN below or above the range is regarded as a deficiency or surplus in dietary protein. Results from analysis of bulk a milk samples collected from 174 dairy herds over Taiwan showed that only one quarter (25.29%) of the herds received a balanced intake of protein and energy, 33.33% adequate protein with energy inadequate, 22.99% herds in protein surplus with energy inadequate, 10.35% herds in protein surplus with energy adequate, 4.6% protein deficiency with energy adequate, and 3.45% herds with both protein and energy inadequate. Energy inadequate herds accounted for 60% of the total dairy herds in Taiwan with 56% adequate, 38% surplus and 6% inadequate in protein. In comparing milk sampled from bulk milk on different seasons from Lee-Kang area in the southern Taiwan, the concentrations of milk fat and milk protein were significantly higher in the cool season (February) than in the warm season (August) (p<0.05), whereas the urea nitrogen in the milk was significantly lower in the cool season than in the warm season (p<0.05). This indicated that lactating cows had excess protein and/or inadequate energy intake in the warm season in this area. It appears that the major problem feeding in lactating cows is energy intake shortage, especially during the warm season in Taiwan.

난소 과자극증후군과 동반된 뇌바닥동맥 혈전증 (A Case of Basilar Arterial Thrombosis in Ovarian Hyperstimulation Syndrome)

  • 보조생식술 시행 후 혈전색전증의 발생은 매우 드물기는 하나 치명적인 합병증이다. 과배란 유도시 중증 난소 과자극 증후군은 $0.1{\sim}0.5%$에서 발생하며, 중증 난소 과자극증후군 환자의 128명 중 한 명에서 혈전색전증이 발생하는 것으로 알려져 있다. 발병기전으로는 과배란 유도에 따른 고에스트로젠 혈증에 의한 혈액응고인자의 변화, 혈관 투과성의 증가에 따른 혈액농축 및 순환혈액의 감소 등을 원인으로 추정하고 있으나 그 정확한 기전은 알려진 바가 없다. 또한 thrombophilia나, 혈전색전증의 과거력 및 가족력이 있는 경우 발병율이 증가한다. 혈전증의 발생 부위는 정맥이 67%이나, 33%는 동맥에 발병하며 주로 뇌동맥에 발생된다고 보고되었다. 본 예는 3년간의 이차성 불임을 주소로 내원하여 시험관 아기시술을 시행하였으며 hCG 투여 8일 후 난소 과자극증후군이 중등도로 발생하였다. hCG 투여 후 11일째 갑작스런 의식 소실 및 우측 상지의 운동장애가 발생하여 시행한 MRI상 뇌바닥동맥 (basilar a.) 혈전증으로 진단되었으며, 혈관내 혈전 용해술 및 풍선 확장술 시행후 재관류에 성공하였다. 시술 2일 후 의식 및 운동장애는 완전히 회복되었으며 시술 7일 후 말더듬증 역시 회복되어 후유장애 없이 건강한 상태이다. Thrombophilic study상 Protein S결핍 소견을 보였고, 또한 vWF-associated Ag.이 증가된 양상을 보였다. 지금까지 국내에서 난소 과자극증후군 이후에 발생한 뇌경색은 5예가 보고되었으며, 이중 4예에서 protein S deficiency 소견을 보였으므로 선별 검사로 유용할 것으로 사료된다. 저자 등은 3년간의 이차성 불임을 주소로 내원한 33세 환자에서 시험관 아기시술시 hCG 투여 11일째에 발생한 뇌바닥동맥 혈전증을 간단한 문헌 고찰과 함께 보고하는 바이다.

Nutritional Constraints and Possibilities for Pig Production on Smallholders Farms in Central Vietnam

  • This study aimed to evaluate the nutritional situation of pigs kept in three ecological zones of central Vietnam: Upland, Lowland and Coastal Area. An interview-based questionnaire was made and surveys were conducted in 27 villages and data were collected from 1,200 participating households. The current study showed that amounts of feed and crude protein content in the diets for fattening pigs and sows are deficient for all three regions. Amounts of feed as DM (kg/d) fed to growing pigs of 20-50 kg BW was deficient by 0.54 kg (29%) in Lowland, 0.53 kg (28.6%) in the Coastal area and 0.42 kg (22.4%) in Upland. The deficiency in CP in the diets of growing pigs in this period (20-50 kg) was largest at 20.7 g/d (62.1%) in Lowland, following by 22.1 g/d (66.4%) in Coastal and 23.2 g/d (69.7%) in Upland. Amount of feed as DM (kg/d) fed to growing pigs of 50-90 kg BW had a deficiency of 1.26 (48.9%), 1.25 (51.2%) and 1.14 (51.5%) kg/d in Lowland, Coastal and Upland, respectively. The deficiencies in crude protein in the growing diet during this period in Lowland, Coastal and Upland regions were 27 g/d (68.3%), 29 g/d (71.9%) and 30 g/d (74.6%), respectively. The deficiency in DM intake (kg/d) of pregnant sows in the Lowland area was 0.3 kg (15%), 0.33 kg (16%) in the Coastal area and 0.47 kg (23.5%) in the Upland area. Crude protein content in the diet of pregnant sows raised in Lowland was 8 g/d (32.0%) deficient, in the Coastal region the deficiency was 11 g/d (42.7%) and in Upland this deficiency was 15 g/d (61.2%). The deficiency in DM intake (kg/d) of lactating sows raised in Lowland was 1.47 kg (31.1%), in the Coastal area this was 1.69 kg (39.2%) and in Upland it was most deficient at 2.46 kg (57.1%). The lack of crude protein content in the diets of sows raised in Lowland was 45 g/d (63.4%), in the Coastal region it was 46 g/d (65%), and in Upland it was 55 g/d (78.9%). The low input of feed in these areas is especially due to low quality and to the insufficient intake of nutrients by the pig. As a result, production and income of farmers are low.

Interaction of Vitamin D and Smoking on Inflammatory Markers in the Urban Elderly

  • Objectives: Epidemiological studies have reported that vitamin D deficiency is associated with inflammatory disease. Smoking is a well-known risk factor for inflammation. However, few studies have investigated the interactive effect of vitamin D deficiency and smoking on inflammation. This study aims to investigate the interaction of vitamin D and smoking with inflammatory markers in the urban elderly. Methods: We used data from the Korean Elderly Environmental Panel Study, which began in August 2008 and ended in August 2010, and included 560 Koreans ${\geq}60$ years old living in Seoul. Data was collected via questionnaires that included items about smoking status at the first visit. Vitamin D levels, high-sensitivity C-reactive protein (hs-CRP), and white blood cell (WBC) counts were repeatedly measured up to three times. Results: The association of vitamin D and hs-CRP was significant after adjusting for known confounders (${\beta}=-0.080$, p=0.041). After separate analysis by smoking status, the association of vitamin D deficiency and hs-CRP in smokers was stronger than that in nonsmokers (smokers: ${\beta}=-0.375$, p=0.013; non-smokers: ${\beta}=-0.060$, p=0.150). Smoking status was an effect modifier that changed the association between vitamin D deficiency and hs-CRP (interaction estimate: ${\beta}=-0.254$, p=0.032). Vitamin D was not significantly associated with WBC count (${\beta}=0.003$, p=0.805). Conclusions: Vitamin D deficiency was associated with hs-CRP in the urban elderly. Smoking status was an effect modifier of this association. Vitamin D deficiency was not significantly associated with WBC count.
