흰쥐에서 식이 단백질 수준이 수유를 통한 수동면역에 미치는 영향 (Effect of Dietary Protein Level on Immune Substances in Milk and its Transfer to Pups in Rats)

  • This study was performed to determine the effect of maternal protein intake on 1) the concentration of immune substances in milk 2) degree of passive immunity to pups via lactation, and 3) specific antibody production to a specific antigen, $\beta$-lactoglobulin(BLG). 4) the effect of passive immunity that pups received from mother during lactation on the production of antibodies when the pups were challenged to the same antigen. Part of the female rats were immunized with BLG before and during pregnancy. The pregnant rats were placed into either 25% or 10% isolated soy protein diet throughout gestation and lactation. After weaning, pups from each group continued to be fed the same diet. At 18 weeks of age, all the pups were challenged with BLG. Total IgA and IgG, lysozyme, BLG-specific IgA and IgG were measured in dam's serum, dam's milk, and pup's serum. Total IgG, and lysozyme in dam's serum and milk were higher in high protein group. Total IgA and IgG in pup's serum remained higher in high protein group from 5 to 18 weeks of age. BLG-specific antibodies were found in the milk and serum of immunized dams, and in serum of pups born to immunized dams but not in the non-immunized group. BLG-specific IgA and IgG were again higher in high protein group and declined with time. The concentration decreased faster in the low proetein group than in the high protein groups. After immunization the pups with LBG, serum BLG-specific antibodies were not differ between rats born to immunized dams and those born to non-immunized dams. Therefore passive immunity rats received via milk as a pup had no effect on the BLG-specific antibody production later in life. This study shows the importance of protein status of mother and strongly support to the endorsement of breast feeding.

Proteome characterization of hormone-induced diploid and tetraploid roots of Platycodon grandiflorum

  • Plants, including Platycodon grandiflorum have been used globally across varied cultures as a safe natural source of medicines. From time immemorial, humans have relied on plants that could meet their basic necessities such as food, shelter, fuel and health. This study was executed to profile proteins from the hormone induced diploid and tetraploid roots using high throughput proteome approach. Two dimensional gels stained with CBB, a total of 64 differential expressed proteins were identified from the diploid root using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 20 differential expressed protein spots ( ${\geq}1.5-fold$) were analyzed using LTQ-FTICR MS whereas a total of 13 protein spots were up regulated and 7 protein spots were down-regulated. However, in the case of tetraploid root, a total of 78 differential expressed proteins were identified from tetraploid root of which a total of 28 differential expressed protein spots (${\geq}1.5-fold$) were analyzed by mass spectrometry whereas a total of 16 protein spots were up regulated and a total of 12 protein spots were down-regulated. However, proteins identified using iProClass databases revealed that the identified proteins from the explants were mainly associated with the nucleic acid binding, oxidoreductase activity, transporter activity and isomers activity. The exclusive protein profile may provide insight clues for better understanding the characteristics of protein function and its metabolic activity that can help for the development of the nutritional and breeding aspects of this economically important medicinal plant.

Comparative proteome analysis of diploid and tetraploid root in Platycodon grandiflorum

  • In spite of the potential medicinal significance and a wide range of pharmacologic properties of Platycodon grandiflorum, the molecular mechanism of its roots is still unknown. The present study was conducted to profile proteins from 3, 4 and 5 months aged diploid and tetraploid roots of Platycodon grandiflorum using high throughput proteome approach. Two-dimensional gels stained with CBB, a total of 68 differential expressed proteins were identified from the diploid root out of 767 protein spots using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 29 differential expressed protein spots (${\geq}2-fold$) were analyzed using LTQ-FTICR MS whereas a total of 24 protein spots were up-regulated and 5 protein spots were down-regulated. On the contrary, in the case of tetraploid root, a total of 86 differential expressed proteins were identified from tetraploid root out of 1033 protein spots of which a total of 39 differential expressed protein spots (${\geq}2-fold$) were analyzed using LTQ-FTICR MS whereas a total of 21 protein spots were up-regulated and a total of 18 protein spots were down-regulated. It was revealed that the identified proteins from the explants were mainly associated with the nucleotide binding, oxidoreductase activity, transferase activity. Taken together, the identified proteins may be helpful to identify key candidate proteins for genetic improvement of plants.

Effects of Dietary Protein and Lipid Levels on Growth and Body Composition of Sub-adult Flounder Paralichthys olivaceus During the Summer Season

  • A $3{\times}2$ factorial experiment was conducted to investigate the proper dietary protein and lipid levels for the growth of sub-adult flounder Paralichthys olivaceus reared during the summer season. Six experimental diets were formulated to contain three levels of protein (45%, 50% and 55%) and two levels of lipid (9% and 14%). Duplicate groups of fish (initial body weight of 298 g) were hand-fed to apparent satiation during the summer season ($21.8{\pm}1.7^{\circ}C$) for 60 days. Survival of each group was over 83% and there was not significant difference among all groups. Weight gain of fish fed the 45% protein diet with 14% lipid was not significantly different from that of fish fed the 50% and 55% protein diets with 9% and 14% lipids, but weight gain of fish fed the 45% protein diet with 9% lipid was significantly lower than that of fish fed the 55% protein diets with 14% lipid. Feed efficiency tended to increase with increasing dietary lipid level at each protein levels, although no significant differences were observed at 50% and 55% protein levels. Protein efficiency ratio, daily feed intake, condition factor and hepatosomatic index were not significantly affected by dietary protein and lipid levels. Crude lipid content of the liver tended to increase with increasing dietary lipid level at the same protein levels, but the opposite appearance was found for moisture content. The contents of moisture, crude protein and crude lipid of the dorsal muscle were not significantly affected by dietary protein and lipid levels. Based on data obtained form this study, inclusion of dietary protein at level of 45% appears sufficient to support optimal growth, and an increase of dietary lipid level from 9% to 14% has beneficial effects on feed utilization of sub-adult flounder during the summer season.

식이 단백질 수준이 한쪽 신장을 절제한 흰쥐에서 나이 증가에 따른 신장의 기능 및 조직 변화에 미치는 영향 (Effect of Dietary Protein Level on Renal Functions and Structure in Uninephrectomized Aging Model in Rat)

  • This study was performed to investigate the effect of dietary protein level on renal senescence. Male rats of 337.8$\pm$5.7g body weight were underlateral nephrectomy or shamoperation. The rats were divided into high protein(40% casein), normal protein(15% casein) and low protein(8% casein)diets and fed experimental diets ad libitum for 24 weeks. The results are summarized as follows. There was a hypertophy of the remnant kidney of uninephrectomized rats of 40% or 15% protein group, coming up to the comparable weights of both kidneys of sham-operated rats. However, the hypertrophic effect was not seen in uninephrectomized rats of 8% protein group. Serum albumin was lower in uninephrectomized rats. With increasing dietary protein level blood urea nitrogen was increased, whereas, urinary urea nitrogen excretion was decreased. Urinary solute excretion was higher in uninephrectomized group than in sham-operated group. However, effect of dietary protein level on urinary solute excretion varied dpending on th solutes tested. GFR and urinary protein excretion, throughout experiment, increased with feeding period and with dietary protein level. Proteinuria was most severe in uninephrectomized rats fed 40% casein diet. Maximum urine concentration ability measured after dehydration was not different among the experimental groups. Light microscopic examination showed focal segmental glomerulosclerosis and mild increas of glomerular mesangial matrix in uninephrectomized rats fed 40% and 15% protein diet, however, which was not observed in uninephrectomized rats fed 8% protein diet and in sham-operated rats fed 40% diet. Immunofluorescence studies revealed segmental deposits of albumin in the mesangium and capillary loops in high protein and uninephrectomized groups. Minimal granular deposition of IgG was noted in the mesangium of all experimental groups. In conclusion, high protein intake accelerated deterioration of renal function and it was correlated with morphological change. Low protein intake was effective in preventing these changes.

완두 자엽에서 소포체 말단의 팽창에 의한 단백과립 발달 (Terminal Dilation and Transformation of the Protein-filled ER to Form Protein Bodies in Pea (Pisum sativum L. var, exzellenz) Cotyledons)

  • 완두 종자에 축적되는 저장물질은 주로 전분과 단백질로서 이러한 저장물질 때문에 고정이나 전자현미경 관찰시료를 제작하기가 쉽지 않다. 따라서 자엽을 얇게 절편을 만들고 효소를 사용하여 단일세포로 분리한 다음 고정하여 관찰하였다. 완두의 저장단백질이 축적되는 단백질 저장 액포는 종자발달의 이른 시기에 기존의 액포를 둘러싸고 발달하게 되므로서 액포는 수축되고 단백질 저장 액포는 점점 발달하여 그 가장자리에 단백질 덩어리가 축적되게 된다. 이와는 별도로 종자발달의 이른 시기에 조면소포체의 내강에 전자밀도가 높은 단백질이 축적되기 시작하여 늦은 시기에 이 소포체의 끝이 부풀어서 구형의 단백과립으로 발달하였다. 완두종자의 저장단백질은 주로 vicilin과 legumin으로서 단백과립에 대한 면역세포화학적 방법으로 확인한 결과 vicilin은 세포질에 발달된 작은 단백과립과 단백질 저장액포의 가장자리에 축적된 단백질 덩어리에 모두 반응하였으나 legumin은 세포질의 단백과립에만 반응하였다. 또한 소포체에 존재하는 단백질인 Bip은 단백질 저장액포에 축적된 단백질 덩어리의 안쪽 가장자리에만 반응하였다. 이는 단백질이 활발하게 축적되고있는 시기에 특징적으로 작용하는 Bip의 기능과 관련되는 것으로 사료된다.

단백질 이차 구조 예측을 위한 단백질 프로파일의 성능 비교 (A Performance Comparison of Protein Profiles for the Prediction of Protein Secondary Structures)

  • 단백질의 이차구조는 단백질의 진화, 구조, 기능을 연구하는데 중요한 정보이다. 단백질 서열 정보만을 이용하여 단백질의 이차 구조를 예측하는 분야에 심층 학습 방법들이 최근 들어 활발히 적용되고 있다. 이러한 방법에서 널리 사용되는 입력은 단백질 서열을 변환하여 만들어진 단백질 프로파일이다. 본 논문에서는 효과적인 단백질 프로파일을 얻기 위하여 단백질 서열 탐색 방법으로 PSI-BLAST와 더불어서 HHblits를 사용하였다. 단백질 프로파일의 구성에 사용되는 상동 단백질 서열을 결정하기 위한 유사도 문턱치와 상동 단백질 서열 정보를 반복적으로 사용하는 회수를 조절하였다. 합성곱 신경망과 순환 신경망을 사용하여 단백질 이차구조를 예측하였는데, 진화적 정보를 한번만 추가하여 만들어진 단백질 프로파일이 효과적이었다.

도메인 조합 기반 단백질-단백질 상호작용 확률 예측 틀 (A Domain Combination-based Probabilistic Framework for Protein-Protein Interaction Prediction)

  • 최근 단백질 및 도메인과 관련된 방대한 양의 데이타들이 인터넷상에 공표되고 축적됨에 따라, 단백질간의 상호작용에 대한 예측 시스템의 필요성이 제기되고 있다. 본 논문에서는 이러한 데이타를 이용하여 계산적으로 도메인 조합 쌍에 기반하여 단백질의 상호작용 확률을 예측하는 새로운 단백질 상호작용 예측 시스템을 제안한다. 제안된 예측 시스템에서는 기존의 도메인 쌍(domain pair)의 제약성을 극복하기 위하여 도메인 조합(domain combination)과 도메인 조합 쌍(domain combination pair)의 개념이 새롭게 도입하였다. 그리고 도메인 조합 쌍(domain combination pair 또는 dc-pair)을 단백질 상호작용의 기본 단위로 간주하고 예측을 시도한다. 예측 시스템은 크게 예측 준비 과정과 서비스 과정으로 구성되어 있다. 예측 준비 과정에서는 상호작용이 있는 것으로 알려진 단백질 쌍 집합과 상호작용이 없는 것으로 추정되는 단백질 도메인 쌍 집합으로부터 각각 도메인 조합 정보와 그 출현 빈도를 추출한다. 추출된 정보들은 출현 확률 배열(Appearance Probability Matrix 또는 AP matrix)로 불리는 배열 구조에 저장된다. 논문에서는 출현 확률 배열에 기반을 두어, 단백질-단백질 상호작용을 예측하는 확률식 PIP(Primary Interaction Probability)를 고안하고, 고안된 확률식을 이용하여, 상호작용이 있는 것으로 알려진 단백질 쌍 집합과 상호작용이 없는 것으로 추정되는 단백질 도메인 쌍 집합의 확률 값 분포를 생성시킨다. 예측서비스 과정에서는 예측 준비 과정에서 얻어진 분포와 확률식을 이용하여 임의의 단백질 쌍의 상호작용 확률을 계산한다. 예측 모델의 유효성은 효모(yeast)에서 상호작용이 있는 것으로 보고된 단백질 쌍 집합과 상호작용이 없는 것으로 추정되는 단백질 쌍 집합을 이용하여 검증하였다. DIP(Database of Inter-acting Proteins)의 상호작용이 있는 것으로 알려진 효모 단백질 쌍 집합의 80%를 학습 집단으로 사용했을 때, 86%의 sensitivity와 56%의 specificity를 나타내어, 도메인을 기반으로 한 기존의 예측 시스템에 비해서 우월한 예측 정확도를 보여주었다. 이와 같은 예측 정확도의 개선은 본 예측 시스템이 상호작용의 기본 단위로 dc-pair를 채택한 점과 분류를 위하여 새롭게 고안하여 사용한 PIP식이 유효했던 것으로 판단된다.

한국인 여성 노인의 단백질 섭취 수준과 근력의 상관성 연구: 국민건강영양조사 제 7기(2016-2018년) 자료를 이용하여 (Association of Low Hand Grip Strength with Protein Intake in Korean Female Elderly: based on the Seventh Korea National Health and Nutrition Examination Survey (KNHANES VII), 2016-2018)

  • Objectives: Decreasing muscle strength in old age has become a significant health problem because it increases the risk of falls or fractures and transfers to other diseases. The precise role of dietary protein intake in preventing or reducing muscle weakness is unclear. This study examined the relationship between handgrip strength and protein intake in Korean female elderly. Methods: This was a cross-sectional study that used data from the Seventh Korean National Health and Nutrition Examination Surveys (KNHANES) on female subjects aged 65 years and older. Low handgrip strength (LHGS) was defined as a handgrip strength below than 18 kg. Dietary intake data were obtained using the 1-day 24-hour recall method. Multiple regression was performed to test whether there is an independent relationship between the grip strength and protein intake, and the association between protein intake and LHGS was confirmed through multiple logistic regression. Results: The mean age of the 2,083 elderly females was 73.3 ± 0.1 years, and the prevalence of LHGS was 35% (n=734). Elderly women with an LHGS consumed less energy, total protein, and animal-based protein than those in the normal group. A multiple regression analysis after adjusting for covariate revealed a significant positive association between the handgrip strength and energy, protein, and animal-based protein intake. Multiple logistic regression analysis showed that the odds ratio (OR) of LHGS in female elderly with the highest quartiles of consumption of energy [OR, 0.65; 95% confidence interval (CI), 0.43-0.82; P for trend=0.004], and animal-based protein [OR, 0.59; CI, 0.40-0.87; P for trend=0.037] were significantly lower than those in the lowest quartiles. Conclusions: The energy intake and animal-based protein intake were negatively associated with the LHGS. These results suggest that adequate energy intake and protein intake, particularly those from animal-based sources, for elderly women in Korea are beneficial in lowering the risk of LHGS.
