  • Sep gene, which is one of the cell division genes coding for penicillin binding protein 3 was subcloned from ${\lambda}607sep^{+2}$ to plasmid pBR322. which has a strong promotor such as lac UV5(lacP). It was confirmed that the sep gene cloned to pLJ3 was in the proper orientation for expressionfrom lactose promotor. To analyze the expression efficiency of sep gene within the plasmids newly constructed, sep mRNA was assated by using ${\lambda$\mid$\;607sep^{+2}$ DNA as a probe. Sep mRNA level was increased 25 times in the cells carrying sep gene cloned to pBR322 compared to E. coli C600 which has wild type sep gene within the chromosome instead of plasmed. Furthermore, the cells carrying sep gene cloned to pLJ3 derected the synthesis of about 50 times as much sep mRNA as did cells carrying sep gene cloned to pBR 322, representing that the sep gene was successfully cloned to pLJ3.

Loss of p15INK4b Expression in Colorectal Cancer is Linked to Ethnic Origin

  • Colorectal cancers remain to be a common cause of cancer-related death. Early-onset cases as well as those of various ethnic origins have aggressive clinical features, the basis of which requires further exploration. The aim of this work was to examine the expression patterns of $p15^{INK4b}$ and SMAD4 in colorectal carcinoma of different ethnic origins. Fifty-five sporadic colorectal carcinoma of Egyptian origin, 25 of which were early onset, and 54 cancers of Finnish origin were immunohistochemically stained with antibodies against $p15^{INK4b}$ and SMAD4 proteins. Data were compared to the methylation status of the $p15^{INK4b}$ gene promotor. $p15^{INK4b}$ was totally lost or deficient (lost in ${\geq}50%$ of tumor cell) in 47/55 (85%) tumors of Egyptian origin as compared to 6/50 (12%) tumors of Finnish origin (p=7e-15). In the Egyptian cases with $p15^{INK4b}$ loss and available $p15^{INK4b}$ promotor methylation status, 89% of cases which lost $p15^{INK4b}$ expression were associated with $p15^{INK4b}$ gene promotor hypermethylation. SMAD4 was lost or deficient in 25/54 (46%) tumors of Egyptian origin and 28/48 (58%) tumors of Finnish origin. 22/54 (41%) Egyptian tumors showed combined loss/deficiency of both $p15^{INK4b}$ and SMAD4, while $p15^{INK4b}$ was selectively lost/deficient with positive SMAD4 expression in 24/54 (44%) tumors. Loss of $p15^{INK4b}$ was associated with older age at presentation (>50 years) in the Egyptian tumors (p=0.04). These data show for the first time that $p15^{INK4b}$ loss of expression marks a subset of colorectal cancers and ethnic origin may play a role in this selection. In a substantial number of cases, the loss was independent of SMAD4 but rather associated with $p15^{INK4b}$ gene promotor hypermethylation and old age which could be related to different environmental exposures.

  • 조촉매(Promotor)인 Na은 수성가스전이(Water Gas Shift, WGS) 반응 시 생성된 포름산염의 C-H결합을 쉽게 분해하는 역할을 한다. 본 연구에서는 $Pt/CeO_2$ 촉매의 성능 향상을 위해 Na의 담지량을 변화시켜 촉매적 활성을 비교하여 보았다. 제조된 담체는 침전법(Precipitation)을 사용하여 제조하였으며 $500^{\circ}C$에서 6시간 소성하였다. Pt 담지량은 1wt%로 고정하였고 Na 담지량은 1 wt%~5 wt%로 변화를 주어 동시(공)-함침법(Co-incipient wetness method)으로 담지 시켰다. 반응 실험은 공간속도(Gas Hourly Space Velocity, GHSV) $45,385h^{-1}$에서 수행하였다. WGS 반응 결과 3 wt%의 Na이 담지된 $Pt/CeO_2$ 촉매의 경우를 제외하고 나머지 Na이 담지된 촉매들은 비교적 높은 CO의 전환율을 나타내었다. 특히 2 wt%의 Na이 담지된 $Pt/CeO_2$ 촉매는 가장 높은 CO의 전환율을 나타내었다. 따라서 Na 담지량의 변화가 포름산염의 C-H결합 분해에 영향을 미친다는 것을 알 수 있다.

  • In this study, the effects of cationic fatty acid bulky promotor on the properties of BCTMP (bleached chemithermomechnical pulp) handsheet including bulk and strength were elucidated. As results, it was observed that the bulk of BCTMP handsheet increased with the increases of the concentration of cationic fatty acid bulky agent, while mechanical properties such as tensile strength and burst strength decreased. The opacity of BCTMP handsheet also increased with the increases of the concentration of cationic fatty acid bulky agent, while brightness was almost not changed. The effectiveness of bulky agent with SwBCTMP (softwood) was higher than that with HwBCTMP (hardwood). Compared with previous research on the effect of bulky agent on BKP handsheet, the bulk increase of BCTMP handsheet was greater compared to that of BKP handsheet. However, the reduction of mechanical property in BCTMP handsheet was lower than that of BKP handsheet.

Characterization of promotor sequences for strong expression of groEx IN Escherichia coli.

  • The cloned X-bacterial gene (groEx) which is analogous to groE of E. Coli strongly expressed in E. coli when grown at the temperature 27.deg. C or higher without having to add any inducers. By S1-nuclease mapping, primer extension analysis and site directed mutagenesis, we found 4 promoters in the gene. Among them two promoters located at 5'-extended region of the gene are homologous to the promoters found in groE family of heat-shock genes ; they are , .sigma.$^{32}$ factor-dependent P1 promotor and .delta$^{70}$factor-dependet P2 promoter. The other two promoters found within the coding region of groESx were P3, 5'-TTGGCG-(18 bases)-AATACT-3' and P4, 5'-TTGGCA-(19 bases)-TAAGT which overlapped within 49 bases. These unique intragenic .delta.$^{70}$-dependent promoters are the first to be cloned and characterized in groE analogous heat-shock genes so far. These P3 and P4 promoters appeared to be responsible for the strong expression of GroElx in X-bacteria in vivo.

Immunocytochemical Localization of c-raf Protein Kinase in EC-4 Cell (EC-4 세포에 있어서 c-raf Protein Kinase의 면역세포화학적 위치)

  • c-raf protein kinase, a kind of oncogene, is a cytopiasmic serine / threonine-specific protein and is activated by mitogenic or oncogenic signals. The strncture and functions of c-raf protein kinase are considered very similar to those of protein kinase C. Using immunocytochemical approach, the time course of singal transduction of c-raf protein kinase in EC-4 cell was examined with 12-0-tetradecanoylphorbol-13-acetate (TPA) as tumor promotor and plateletderived growth factor (PDGF) as mitogenic factor. Immunoreactive c-raf was initially bound to the perinuclear membrane and then moved into the nucleus. The effect of the long-term treatment with TPA or PDGF was taken place down regulation at different time point. These results indicate that TPA and PDGF give rise to the translocation of c-raf protein kinase through the two different pathways.

Appraisal of re-irradiation for the recurrent glioblastoma in the era of MGMT promotor methylation

  • Despite recent innovation in treatment techniques and subsequently improved outcomes, the majority of glioblastoma (GBL) have relapses, especially in locoregional areas. Local re-irradiation (re-RT) has been established as a feasible option for recurrent GBL of all ages with safety, tolerability, and effectiveness both in survival and quality of life regardless of fractionation schedule. To keep adverse effects under acceptable range, cumulative dose limit in equivalent dose at 2 Gy fractions by the linear-quadratic model at α/β = 2 for normal brain tissue (EQD2) with narrow margin should be observed and single/hypofractionated re-RT should be undertaken very carefully to recurrent tumor with large volume or adjacent to the brainstem. Promising outcome of re-operation (re-Op) plus re-RT (re-Op/RT) need to be validated and result from re-RT with temozolomide/bevacizumab (TMZ/BV) or new strategy is expected. Development of new-concept prognostic scoring or risk group is required to select patients properly and make use of predictive biomarkers such as O(6)-methylguanine-DNA methyltransferase (MGMT) promotor methylation that influence outcomes of re-RT, re-Op/RT, or re-RT with TMZ/BV.
