Differential inhibitory effects of alpha-viniferin, resveratrol trimer on inflammatory mediators

  • Alpha-viniferin was isolated from Carex humilis (Cyperaceae), and showed anti-inflammatory effects on carrageenin or histamine-induced paw edema in mice. To understand mode of the anti-inflammatory action. effects of alpha-viniferin on cyclooxygenase (COX)-2, iNOS, oxygen radicals and proinflammatory cytokines have been analyzed. Alpha-viniferin showed selective inhibitory effect with an IC50 value of 5 $\mu\textrm{m}$ on COX-2 activity but showed weak inhibitory effect on the synthesis of COX-2 transcript which was identified by RT-PCR.   • HDAC and HAT (histone acetyltransferase) are involved in co-regulation in chromatin remodeling and the functional regulation of gene transcription. Abnormal recruitment of HDAC is related to carcinogenesis. Thus, the identification of potent histone deacetylase (HDAC) inhibitor has been considered as very intriguing approach for development for cancer chemotherapy. More recently, anti-inflammatory activity of SAHA cytokines was reported via reduction of proinflammatory cytokinres in vitro and in vivo. (omitted)

  • Mast cells play an important role in inflammation by functioning as a source of histamine, tryptase, and proinflammatory cytokines. Lonicera Japonica (Caprifoliaceae. Lc) has been used to treat inflammation.   • Celastrus hindsii (family Celastraceae) is located abundantly in the United States, China, and Vietnam, where it is utilized as a traditional herbal and traditional drug for the care of cancer. However, the antioxidant and anti-inflammatory effects of Celastrus hindsii extract are unknown. In our research, the antioxidant activity of Celastrus hindsii leaf extract was investigated, and then anti-inflammatory efficacy of C. hindsii extract in lipopolysaccharide (LPS)-induced RAW264.7 cells. First, our results revealed that C. hindsii extract showed powerful antioxidant capability. Moreover, the application of C. hindsii extract significantly reduced nitric oxide (NO) production without cytotoxic effects. Furthermore, C. hindsii extract reduced the generation of pro-inflammatory cytokines, like as interleukin (IL)-6, IL-1β, and TNF-α. Our results are the first to confirm the anti-inflammatory capability of C. hindsii extract in RAW264.7 cells.

Immunomodulatory Effects of Dioscoreae Rhizome Against Inflammation through Suppressed Production of Cytokines Via Inhibition of the NF-${\kappa}B$ Pathway

  • Dioscoreae Rhizome (DR) has been used in traditional medicine to treat numerous diseases and is reported to have anti-diabetes and anti-tumor activities. To identify a bioactive traditional medicine with anti-inflammatory activity of a water extract of DR (EDR), we determined the mRNA and protein levels of proinflammatory cytokines in macrophages through RT-PCR and western blot analysis and performed a FACS analysis for measuring surface molecules. EDR dose-dependently decreased the production of NO and pro-inflammatory cytokines such as IL-$1{\beta}$, IL-6, TNF-${\alpha}$, and $PGE_2$, as well as mRNA levels of iNOS, COX-2, and pro-inflammatory cytokines, as determined by western blot and RT-PCR analysis, respectively. The expression of co-stimulatory molecules such as B7-1 and B7-2 was also reduced by EDR. Furthermore, activation of the nuclear transcription factor, NF-${\kappa}B$, but not that of IL-4 and IL-10, in macrophages was inhibited by EDR. These results show that EDR decreased pro-inflammatory cytokines via inhibition of NF-${\kappa}B$-dependent inflammatory protein level, suggesting that EDR could be a useful immunomodulatory agent for treating immunological diseases.

Expression of ${\alpha}_1$-Acid Glycoprotein and Inflammatory Cytokines during Differentiation of HL-60 Cells

  • In order to understand the role of AGP on the differentiation of promyelocytic leukemia cells, the AGP expression and its relation to cytokines were investigated during granulocytic or monocytic differentiation of HL-60 cells. When HL-60 cells were treated with all-trans-retinoic acid (ATRA) for 5 days, the cells were fully differentiated into granulocytes, and the AGP mRNA and protein levels were continuously increased up to 5 days in a dose- and time- dependent manner. However, in the case of the monocytic differentiation of HL-60 cells by tetradeanoyl phorbol acetate (TPA), the AGP gene expression was not induced. In addition, $IL-1{\alpha}$, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNAs were also enhanced during granulocytic differentiation. These cytokine transcripts showed a peak level 3 days after the ATRA treatment. It decreased gradually thereafter. However, direct addition of recombinant cytokines ($IL-1{\beta}$, IL-6 and $TNF-{\alpha}$) and dexamethasone to the HL-60 cell cultures showed no AGP induction. These findings suggest that the AGP and proinflammatory cytokines are expressed in ATRA-treated promyelocytic cells. However, these cytokines do not act as autocrine inducers on AGP expression. This fact implies that the AGP expression during granulocytic differentiation of HL-60 cells is induced through a signal pathway different from hepatocyte signaling in inflammation.

Balb/C mouse의 폐장대식세포에서 유리규산 자극에 의한 Proinflammatory Cytokine과 TGF-$\beta$의 생성 및 상관관계 (The Production and Correlation of Silica Induced Proinflammatory Cytokines and TGF-$\beta$ from Monocytes of Balb/C Mice)

  • 연구배경: 유리규산(silica) 자극에 의해 폐장내 대식세포에서 형성되는 proinflammatory cytokines인 IL-1$\beta$, IL-6, TNF-$\alpha$와 fibrogenic cytokine인 TGF-$\beta$, PDGF는 섬유화 과정에 매우 중요한 역할을 한다. 그러나 이러한 cytokine 의 역할과 상호관련성에 대해서는 아직 확실히 밝혀진 바없다. 본 연구는 Balb/C mouse의 폐장단구세포를 silica로 자극시 형성되는 IL-6, TNF-$\alpha$ 폐섬유화의 발생과 유지에 중요한 역할을 할 것으로 기대되는 TGF-$\beta$ 의 생성을 관찰하고 TGF-$\beta$의 생성에 proinflammatory cytokine이 미치는 영향에 대해서 알아보고자 하였다. 방 법: Balb/C 의 폐장대식세포를 silica로 자극한 후 IL-6, TNF-$\alpha$ TGF-$\beta$의 양을 bioassay로 측정하였고 mRNA의 발현을 in situ hybridization을 통해 관찰하였으며, 대식세포의 TGF-$\beta$ 생성에 대한 IL-1$\beta$, IL-6, TNF-$\alpha$의 영향을 알기 위하여 L-1, IL-6, TNF-$\alpha$에 대한 중화항체를 이용하여 대식세포의 TGF-$\beta$의 생성 억제 유, 무를 관찰하였다. 결 과: Silica로 대식세포자극시 TNF-$\alpha$는 초기 4 시간째 최고의 생성능을 보였으며, IL-6 는 silica 0.2 ${\mu}g/ml$의 농도에서 배양 8 시간째 생성능이 가장 높았고 배양 12 시간째는 다시 감소하였다가 24시간째 증가하는 biphasic한 소견을 보여 주었다. TGF-$\beta$는 silica의 양과 관계없이 배양 24시간째 자연생성능뿐 아니라, silica로 자극시에도 증가되었다. Silica와 TNF-$\alpha$ 50ng/ml을 함께 자극시 TGF-$\beta$의 생성은 현저히 증가되었으며, TNF-$\alpha$ 50 ng/ml과 IL-6 50 ng/ml으로 동시 자극시에는 TNF-$\alpha$에 의한 TGF-$\beta$의 형성이 증가하는 경향을 보였다. TNF-$\alpha$의 중화항체를 전처치 후 silica 자극시 TGF-$\beta$의 생성능을 억제시킴을 관찰할 수 있었다. Anti-IL-$\beta$, anti-IL-6, anti-TNF-$\alpha$ antibody 모두를 통시에 투여하였을 때도 silica에 의한 TGF-$\beta$의 생성능을 억제시킴을 관찰할 수 있었다. Silica 자극 후 대식세포의 in situ hybridization에서 TNF-$\alpha$ mRNA는 2시간째부터 발현되었고 IL-6 mRNA는 4 시간째부터 발현된 후 8 시간째 소실되었고 TGF-mRNA는 12 시간째 가장 강한 발현을 보였다. 결 론: Silica 자극에 의해 폐장대식세포는 자극 초기에는 TNF-$\alpha$가 가장먼저 형성되고 후기에 TGF-$\beta$가 형성되며 TNF-$\alpha$가 TGF-$\beta$ 형성에 중요한 역할을 할 것으로 사료되었다.

Inhibitory Effect of Rosmarinic acid Extrcted from Euonymus Alatus on Cyclooxygenase-2

  • Objectives and methods : Previous mechanistic studies suggest the cyclooxygenase-2 (COX-2) inhibitors represent the good candidates against tumor progression. MeOH extract of the stem barks of Euonymus alatus induced the strong inhibition of COX-2. A phenolic compound responsible for the anti- COX-2 known to involve in tumor adhesion and invasion has been studied through the methanol extracts. The compound, rosmarinic acid (ROS-A) was an ester of caffeic acid and 3,4-dihydroxyphenyllactic acid. ROS-A showed a strong inhibitory effect of COX-2 activity in a concentration-dependent manner. Then we have measured the IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production related the immune regulation, induction of inflammatory related genes. Results and Conclusions :Hep3B cells produce proinflammatory cytokines of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ while ROS A inhibited the cytokines production. Since IL-1${\beta}$, IL-6 and TNF-${\alpha}$ need the transcription factors such as nuclear factor- ${\kappa}$B (NF-${\kappa}$B) and activated protein-1 (AP-1), we measured the transcription factors. ROS-A inhibited the activation of p65, p50, c-Rel subunits of NF-${\kappa}$B and AP-1 transcription factors. These findings indicate that ROS A from the stem bark of E. alatus inhibits proliferation in metastatic cancer cells. It was suggested that stem barks of E. alatus could be suitable for anti-cancer drugs.

마우스 대식세포를 이용한 일당귀 물추출물의 항염효능 연구 (Anti-inflammatory Effect of Angelicae acutilobae Radix Water Extract on LPS-stimulated Mouse Macrophages)

  • Objectives : The purpose of this study was to investigate the effects of Angelicae acutilobae Radix Water Extract (AA) on the production of cytokines in RAW 264.7 cell stimulated with lipopolysaccharide (LPS). Method : RAW 264.7 cells were cotreated with AA (50 and $100{\mu}g/mL$) and lipopolysaccharide (LPS; $1{\mu}g/mL$) for 24 hours. After 24 hours treatment, using bead-based multiplex cytokine assay, concentrations of various cytokines such as interleukin(IL)-6, tumor necrosis factor-alpha(TNF-${\alpha}$) granulocyte colony-stimulating factor(G-CSF), granulocyte macrophage colony-stimulating factor(GM-CSF), and macrophage inflammatory protein(MIP)-$1{\alpha}$ were measured. Result : AA significantly inhibited LPS-induced production of IL-6 and MIP-$1{\alpha}$ from LPS-stimulated RAW 264.7 cells at the concentration of $50{\mu}g/mL$. AA significantly inhibited LPS-induced production of TNF-${\alpha}$ from LPS-stimulated RAW 264.7 cells at the concentration of $100{\mu}g/mL$. AA significantly inhibited LPS-induced production of G-CSF and GM-CSF in RAW 264.7 cells at the concentrations of 50 and $100{\mu}g/mL$. Conclusion : These results suggest that AA has anti-inflammatory effect related with its inhibition of proinflammatory cytokines such as IL-6, TNF-${\alpha}$, G-CSF, GM-CSF, and MIP-$1{\alpha}$ in LPS-induced macrophages.

Effects of in vitro immune stimulation by ginsenoside Rb1

  • Red ginseng is a classical traditional Chinese medicine. Among Chinese herbs, red ginseng has been considered as one of the tonics. Many studies indicated that red ginseng could enhance immune function of the human body. Red ginseng total saponin, ginsenoside, the most important active constituents identified in red ginseng can protect against myocardial ischaemia damage and protect endothelium against electrolysis-induced free radical injury. Macrophages play a significant role in host defense mechanisms. When activated, they inhibit the growth of a wide variety of tumor cells. The aim of this study was to determine the effects of pure ginsenoside Rb1 on immunostimulatory activity such as murine macrophage phagocytosis and proliferation of splenocytes. Furthermore, we investigated the effects of ginsenoside Rb1 on the production of nitric oxide (NO), reactive oxygen species (ROS) and proinflammatory cytokines (IL-1beta, IL-6, and TNF-alpha) in murine macrophage, RAW 264.7 cells. ROS have emerged as important signaling molecules in the regulation of various cellular processes. Ginsenoside Rb1 significantly increased production of ROS in dose dependent manner. As NO plays an important role in immune function, ginsenoside Rb1 treatment could modulate several aspects of host defense mechanisms due to stimulation. Treatment with ginsenoside Rb1 to macrophages induced the production of NO and proinflammatory cytokines and expression levels of these genes in a dose-dependent manner. Furthermore, incubation of RAW 264.7 cells with ginsenoside Rb1 showed a dose dependent increased phagocytosis activity and lymphocyte proliferation of splenocytes. Therefore, these results suggest that ginsenoside Rb1 has promising potential as a natural medicine for stimulation of the immune system.
