소나무 우량 임분의 구과 생산량에 따른 종자 및 발아 특성 (Seed Characteristics and Germination Properties according to Change of Cone Production in Pinus densiflora Stands)

  • 종자 공급 방안 개선에 필요한 기초 자료 획득을 위해 소나무 우량 임분 3개소를 대상으로 2003년부터 2006년까지 구과 생산량에 따른 종자 및 발아 특성을 분석한 결과 다음과 같은 결과를 얻었다. 구과 생산량은 2년을 주기로 하여 풍 흉 상태를 보였다. 주요 종자 특성 중 종자 생산 능력과 충실 종자율은 대체적으로 구과 생산량과 유사한 경향을 보인 반면 성숙 종자율과 종자 효율은 반대 경향을 나타내었다. 종자 특성과 구과 생산량과의 회귀분석 결과에서도 종자 생산 능력 ($R^2=0.682$, p<0.01)과 충실 종자율($R^2=0.l61$, non-significant)은 정(+)의 경향을, 성숙 종자율($R^2=0.418$, p<0.05)과 종자 효율($R^2=0.308$, non-significant)은 부(-)의 경향을 나타내었다. 발아 특성에 있어서 발아율은 대체적으로 구과 생산량과 유사한 경향으로 나타났으나, 평균 발아 일수는 반대의 경향을 보였다. 회귀분석결과에서도 발아율은 구과 생산량과 정(+)의 관계($R^2=0.576$, p<0.01)를 보인 반면, 평균 발아 일수는 부(-)의 관계($R^2=0.400$, p<0.05)를 보여 구과 생산량이 많은 시기에 이를 채취하여 사용할 때 발아율도 높고 발아에 소요되는 기간도 짧아져 즉, 발아 효율이 높아짐을 알 수 있었다.

Characterization of L-(+)-Lactic Acid Producing Weizmannia coagulans Strains from Tree Barks and Probiogenomic Evaluation of BKMTCR2-2

  • This study aimed to isolate and identify L-(+)-lactic acid-producing bacteria from tree barks collected in Thailand and evaluate the potential strain as probiotics. Twelve strains were isolated and characterized phenotypically and genotypically. The strains exhibited a rod-shaped morphology, high-temperature tolerance, and the ability to ferment different sugars into lactic acid. Based on 16S rRNA gene analysis, all strains were identified as belonging to Weizmannia coagulans. Among the isolated strains, BKMTCR2-2 demonstrated exceptional lactic acid production, with 96.41% optical purity, 2.33 g/l of lactic acid production, 1.44 g/g of lactic acid yield (per gram of glucose consumption), and 0.0049 g/l/h of lactic acid productivity. This strain also displayed a wide range of pH tolerance, suggesting suitability for the human gastrointestinal tract and potential probiotic applications. The whole-genome sequence of BKMTCR2-2 was assembled using a hybridization approach that combined long and short reads. The genomic analysis confirmed its identification as W. coagulans and safety assessments revealed its non-pathogenic attribute compared to type strains and commercial probiotic strains. Furthermore, this strain exhibited resilience to acidic and bile conditions, along with the presence of potential probiotic-related genes and metabolic capabilities. These findings suggest that BKMTCR2-2 holds promise as a safe and effective probiotic strain with significant lactic acid production capabilities.

  • Crop-animal systems which form the backbone of agriculture in the tropics are discussed with reference to their characteristics, economic importance of animals, genesis and types of crop-animal systems, relevance and potential importance, and priorities for research and development. These production systems are found across all agroecological zones: rain-fed temperate and highland systems, semi-arid and arid tropics, and sub-humid and humid tropics: the last four are priority areas in Asia. The potential importance of these systems in Asia is reflected in their advantages, synergism and complimentarity, economic benefits and contribution to sustainability. Illustrative case studies are cited which are appropriate to the two broad types of mixed farming systems: systems combining animals and annual cropping, and systems combining animals with perennial cropping with reference to Indonesia, Philippines, Malaysia, Sri Lanka, China and Nigeria. Priorities for research and development should address more complete use of the animal genetic resources, intensive utilisation of the feed resources, development-oriented utilisation of research results, minimizing animal diseases, and implementation of appropriate institutional and policy issues.

  • The traditional methods for determining the quality of milk, cheese and meat are tedious and expensive, with a significant wastage of chemicals which pollute the environment. To overcome these disadvantages, the potential of near infrared spectrophotometry (NIR) for monitoring the quality of milk and meat has been evaluated by a number of researchers. While most studies indicate that NIR can be used to predict chemical composition of milk and meat, and to monitor the cutting-point during cheese manufacturing, one study demonstrated the potential of NIR to predict sensory characteristics (e.g. hardness and tenderness) of beef. These calibrations were developed on a small number of samples, limiting their value for adoption by the industries. Now that the sophisticated computer software is available, more robust calibrations need to be developed to monitor both chemical and physical characteristics of meat and meat products simultaneously.

  • Synthetic biology approaches offer potential for large-scale and sustainable production of natural products with bioactive potency, including ginsenosides, providing a means to produce novel compounds with enhanced therapeutic properties. Ginseng, known for its non-toxic and potent qualities in traditional medicine, has been used for various medical needs. Ginseng has shown promise for its antioxidant and neuroprotective properties, and it has been used as a potential agent to boost immunity against various infections when used together with other drugs and vaccines. Given the increasing demand for ginsenosides and the challenges associated with traditional extraction methods, synthetic biology holds promise in the development of therapeutics. In this review, we discuss recent developments in microorganism producer engineering and ginsenoside production in microorganisms using synthetic biology approaches.

  • 2l 발효조에서 pH6.9, 온도 $32^{\circ}C$일 때 당밀배지를 이용하여 Corynebacterium glutamicum의 영양요구성 유사체 내성변이주에 의한 라이신 발효시 산화환원 전위 (ORP)가 라이신 발효속도의 특성에 미치는 영향을 조사하였다. 희석률이 0.1$h ^1$일때 탄소원이 제한되건 로이신이 제한되건 산소가 제한되지 않는 한 최대의 대당수율 24를 보였으며, 이 때의 산화환원 전윈 값은 -60mV와 -100mV 범위에 해당하였다. 산화화원 전위 값이 -130mV의 매우 낮은 용존산소 조건하에서는 대당수율 밀 $q_s, q_p$ 등의 발효 반응속도 상수값들이 크게 감소하였으며 glvcine, alanine, valine을 포함하는 발효 부산물의 축적량이 매우 높아졌다.

Optimal Conditions for the Production of Sphimin, a Sphingomyelinase Inhibitor from Steptomyces sp. F50970

  • We isolated a sphingonyelinase (SMase) inhibitor, which would be a potential reagent to regulate cell proliferation, oncogenesis, and inflammation, from a strain of Streptomyces sp.. In this paper, we report the optimal conditions for the production of SMase inhibitor, designed as sphinin, from Streptomyces sp. F50970. The optimal carbon and nitrogen source were 1% soluble starch and 0.05%-0.15% trypton. Most of monosaccharides and high concentration of soluble starch above 1.0% caused falling of pH and sphinin production. Zn2+, Cu2+, Fe2+, Mn2+, and Co2+inhibited cell growth and the production of sphinin. Inorganic phosphate promoted the sphinin production. Optimal initial pH for the production of sphinin was 7.5-8.0. Addition of CaCO3 to the medium resulted in an increase of inhibitor production. Based on these results, we designed a fermentation medium for the production of a SMase inhibitor, sphinin, from Streptomyces sp. F50970.

시설농업부산물의 잠재메탄발생량 평가 및 사일로 저장에 따른 메탄 발생 변화 (Biochemical Methane Potential of Agricultural Residues and Influence of Ensiling on Methane Production)

  • 본 연구에서는 BMP (Biochemical Methane Potential) test를 통해 얼갈이배추, 딸기, 토마토, 오이, 참외 시설농업부산물의 잠재메탄발생량을 조사하였다. 또한, 시설농업부산물을 사일로에 저장하고 저장 전후의 잠재메탄발생량을 비교하여 사일리지 저장기술이 메탄 생산에 미치는 영향을 분석하였다. 대상 시료의 잠재메탄발생량과 생분해도는 각각 149~286 mL-$CH_4/g$-VS, 27~48%(by vol.)의 범위를 나타내었으며 메탄발생량은 얼갈이배추 > 참외 > 딸기 ${\approx}$ 오이 > 토마토 순으로 조사되었다. 사일로 저장 후, 원시료와 비교하였을 때 VS 기준 메탄발생량이 -11~36%(by vol.) 증감하여 시료 별로 상이한 결과를 보였다. 저장기간 중 유기산 증가, 섬유소 분해로 메탄발생량이 증가하고 화학 성분의 변화, 암모니아 저해로 메탄발생량이 감소한 것으로 판단된다.

Implementation of Remote Monitoring Scenario using CDMA Short Message Service for Protected Crop Production Environment

  • Protected vegetable production area is greater than 26% of the total vegetable production area in Korea, and portion of protected production area is increasing for flowers and fruits. To secure stable productivity and profitability, continuous and intensive monitoring and control of protected crop production environment is critical, which is labor- and time-consuming. Failure to maintain proper environmental conditions (e.g., light, temperature, humidity) leads to significant damage to crop growth and quality, therefore farmers should visit or be present close to the production area. To overcome these problems, application of remote monitoring and control of crop production environment has been increasing. Wireless monitoring and control systems have used CDMA, internet, and smart phone communications. Levels of technology adoption are different for farmers' needs for their cropping systems. In this paper, potential of wireless remote monitoring of protected agricultural environment using CDMA SMS text messages was reported. Monitoring variables were outside weather (precipitation, wind direction and velocity, temperature, and humidity), inside ambient condition (temperature, humidity, $CO_2$ level, and light intensity), irrigation status (irrigation flow rate and pressure), and soil condition (volumetric water content and matric potential). Scenarios and data formats for environment monitoring were devised, tested, and compared. Results of this study would provide useful information for adoption of wireless remote monitoring techniques by farmers.
