핵자기공명 영상법과 핵자기공명 분광법을 위한 뒤집음-탐침의 개발에 대한 연구 (Development of inside-out probes for both Nuclear Magnetic Resonance Imaging and Nuclear Magnetic Resonance Spectroscopy)

  • 핵자기공명(Nuclear Magnetic Resonance)에서 신호 획득 시에 가장 중요한 부분중의 하나인 rf-탐침(radio-frequency-probe)은 실험대상이 되는 시료의 기하학적 모양과 획득하고자 하는 정보 등에 따라서 적절히 설계 및 제작되어야 최적의 신호를 얻을 수 있다. 일반적으로, rf-탐침은 시료를 둘러싸는 형태와 시료 표면에 위치하는 형태 등 두 가지로 제작된다. 그러나, 시료나 인체 부위가 관모양을 하고 있으며 관 모양 주위로부터의 신호를 관측하는 경우, 앞에서 기술한 두가지 형태의 탐침을 이용해서는 신호대 잡음비의 저하로 인해 만족할 만한 신호 획득이 어려워진다. 이때, 탐침을 관 모양의 시료 내부에 위치시키고, 관찰하고자 하는 부위에 최대로 가까이하며 탐침의 품격인자를 높이는 것이 최적의 신호획득에 중요한 조건이 되게 된다. 본 논문에서는 탐침을 시료 내부에 넣고 탐침 외부로부터 신호를 얻는 뒤집음-탐침을 역-솔레노이드 saddle 및 이중표면코일 등의 세가지 형태로 제작하고 최적신호의 획득에 대한 시도를 해보았고, 컴퓨터를 이용한 계산을 통하여 기하학적인 모양의 차이에 따른 rf-자기장 발생 상태를 알아보았다. 아울러, 시험시료로부터 발생되는 영상신호를 얻고 영상 영역 내에서 신호대잡음비를 측정하여 탐침들의 성능을 비교 분석했다. 본 연구에서 제작된 뒤집음-탐침의 경우, 어떠한 상품화된 탐침과 비교할 때, 뛰어난 신호대잡음비와 탐침 주위에서 매우 높은 균일성이 유지되었다. 또한, 제작된 형태의 뒤집음-탐침을 이용한 인체 진단에의 응용성은 매우 높을 것으로 예상된다.

  • An aspheric microlens array to improve the properties of multi optical probes was designed and fabricated. To generate multi optical probes with good qualities, a microlens array with the minimum spherical aberration was designed by ray tracing. Using the reflow process, a master pattern of aspheric microlens array was made and finally with the ultraviolet-imprinting (UV-imprinting) method, the aspheric microlens array was replicated. The reflow condition was optimized to realize the master pattern of the microlens array with the designed aspheric shape. The intensity distribution of the optical probes at the focal plane showed a diffraction-limited shape.

Optimization of a Diagnostic DNA Chip for Fish Rhabdovirus

  • A DNA chip that rapidly and accurately detects the viral genes in rhabdovirus-infected fish was developed. The N, Ml, and G proteins of three rhabdovirus strains, infectious hematopoietic necrosis virus (IHNV), viral hemorrhagic septicemia virus (VHSV), and flounder rhabdovirus (HIRRV), were selected for use as probes. The sequences of the corresponding genes were obtained, and probes were prepared by PCR using specific primer sets. The specificity of the probes was confirmed by cross PCR. The prepared probes were spotted on poly-L-lysine- or aminosilane-coated glass slides and hybridized with target DNA under several different conditions in order to determine the optimal hybridization temperature, glass-slide coating, and target cDNA concentration.

  • Molecular imaging technologies have been used to provide a new pathway for therapies and diagnosis of human disease. Especially, imaging probes have been much development in the molecular imaging field. Combining imaging probes for positron emission tomography (PET) and magnetic resonance imaging (MRI) have suggested the potential of multiple methods in living body. This review discusses the cancer or lymph node-targeting probes that are suitable for PET/MRI based diagnosis.

  • In recent years, many researchers have attempted to make use of 2D nanoparticles as molecular imaging probes since extensive investigations proved that 2D nanoparticles in the body tends to accumulate certain lesions by enhanced permeability and retention (EPR) effect. For example, graphene and carbon nitride which have high surface area and modifiable properties showed good biocompatibility and targetability when it used as imaging probes. However, poor dispersibility in physiological mediums and its uncontrolled size limited its usage in bio-application. Therefore, oxidation process and mechanical exfoliation have been developed for overcoming these problems. In this paper, we highlight the several major methods to synthesize biocompatible 2D nanomaterials like graphene and carbon nitride especially for molecular imaging study including positron emission tomography (PET).

DNA Microarray Probe Preparation by Gel Isolation Nested PCR

  • To develop a simplified method that can rapidly prepare DNA microarray probes in a massive scale, a lambda phage genomic DNA-fragments library was constructed for the microarray-probes collection. Four methods of DNA band recovery from the first PCR products were tested and compared. The DNA microarray probes were collected by a novel method of nested PCR that was mediated by gel isolation of the first PCR products. This method was named GIN-PCR. The probes that were prepared by this GIN-PCR technique were used as subjects to fabricate a DNA microarray. The results showed that a wooden toothpick was superior to the other 3 methods, since this technique can steadily transfer the DNA bands as the template of the second PCR after the first PCR. A group of probes were successfully collected and DNA microarrays were constructed using these probes. Hybridization results demonstrated that this technique of DNA recovery and probe preparation was rapid, efficient, and effective. We developed a cost-effective and less labor-intensive method for DNA microarray probe preparation by nested PCR that is mediated by wooden toothpick transfer of the DNA bands in the gel after electrophoresis.

접지임피던스 측정시 보조전극의 배치 기준 연구 (Placement Standard Research of Auxiliary Probes when Measuring Ground Impedance)

  • Among ground impedance measurement methods, the fall-of-potential method is the most thorough and reliable method. In the fall-of-potential method, ground electrode and auxiliary probes are placed in a straight line, and then, auxiliary potential probe is moved away from the ground electrode. The point at which plotted resistance curve flattens out is taken as right position of auxiliary potential probe. However, in some cases, it is hard to place ground electrode and auxiliary probes in a straight line. Therefore, we provided alternative placement method in this research. The method can be easily applicable to placing auxiliary probes. Also, this paper analyzed and compared ground impedance measurement standards of large grounding systems. Based on the analysis, practical measurement method using an earth tester was proposed. The proposed methods presented in this paper will be useful when determining locations of auxiliary probes in alternative positions, and the methods can be applied practically and easily.

  • The advent of microarray technologies gives an opportunity to moni tor the expression of ten thousands of genes, simultaneously. Such microarray data can be deteriorated by experimental errors and image artifacts, which generate non-negligible outliers that are estimated by 15% of typical microarray data. Thus, it is an important issue to detect and correct the se faulty probes prior to high-level data analysis such as classification or clustering. In this paper, we propose a systematic procedure for the detection of faulty probes and its proper correction in Genechip array based on multivariate statistical approaches. Principal component analysis (PCA), one of the most widely used multivariate statistical approaches, has been applied to construct a statistical correlation model with 20 pairs of probes for each gene. And, the faulty probes are identified by inspecting the squared prediction error (SPE) of each probe from the PCA model. Then, the outlying probes are reconstructed by the iterative optimization approach minimizing SPE. We used the public data presented from the gene chip project of human fibroblast cell. Through the application study, the proposed approach showed good performance for probe correction without removing faulty probes, which may be desirable in the viewpoint of the maximum use of data information.

원자 현미경용 콜로이드 탐침 수직 스프링 상수 측정 (Measurement of Normal Spring Constant of Colloidal Probes for Atomic Force Microscope)

  • A modified thermal noise method was proposed to measure the normal spring constants of the colloidal probes for an atomic force microscope. We used commercial tipless cantilevers (length 150, width 30, nominal k 7.4 N/m) and borosilicate spheres with a diameter of 20 to fabricate colloidal probes. The inverse optical lever sensitivity of both the tipless cantilever and colloidal probes were used to measure the normal spring constant of the colloidal probes. We confirmed the accuracy and usefulness of our method by comparing the measurement results with those obtained using the nanoforce calibrator (NFC), which reportedly has an uncertainty of 1.00%. The modified thermal method showed a good agreement (~10% difference) with the NFC, allowing us to conclude that the modified thermal method could be employed for the effective measurement of the normal spring constants of colloidal probes.
