Absorption and Fluorescence Studies of 3-Ethenylindoles

  • Synthesis, absorption and fluorescence properties of 3-methyl indole (1), N-(benzenesulfonyl)-3-(3-oxo-but-1-enyl)-indole (2) and 1H-3-(3-oxo-but-1-enyl)-indole (3) are described. Extended conjugation at C-3 of indole as in 3 causes moderate resolution of $^1L_a$ and $^1L_b$ bands. However, 2 having an electron-withdrawing group at indolic nitrogen shows only the $^1L_a$ band. While the $^1L_b$ band largely remains solvent polarity independent, the $^1L_a$ band undergoes moderate red shift in polar solvents. The fluorescence in 2 and 3 originates from the $L_b$ transition. Additionally, interaction of 2 and 3 with BSA indicates that these compounds bind to the hydrophobic site of BSA with the formation of a highly fluorescent BSA-probe complex.

Synthesis of a squaric acid-derived molecular probe for near-infrared fluorescence and photoacoustic imaging

  • Dual-modality imaging strategy using near-infrared fluorescence (FLI) and photoacoustic imaging (PAI) demands a suitable probe to enable dual-modular signal production. Herein, we demonstrate a synthetic protocol of small molecular dye for dual-modular FLI and PAI. A condensation reaction between squaric acid and carboxypentyl benzoindolium, and followed by basic hydrolysis to give the benzoindole derived squaraine (BSQ) dye in 49% yield. Next, the carboxylic acid group of BSQ was further functionalized with N-hydroxysuccinimide or azide group for an efficient conjugation with a targeting biomolecule. BSQ showed a maximum fluorescent emission at around 680 nm and the photoacoustic signal reached a maximum intensity at 680-700 nm. Based on these results, we conclude that BSQ analogs will be useful probes for dual-modular (FLI/PAI) imaging studies in animal models.

In-situ Synthesis and Investment Casting of Titanium Matrix (TiC+TiB) Hybrid Composites (Ti기 (TiC+TiB) 하이브리드 복합재료 반응생성합성 및 정밀주조)

  • The aim of the present work is to investigate the possibility of in-situ synthesis and net-shape forming of the titanium matrix (TiC+TiB) hybrid composites using a casting route. From the scanning electron microscopy, electron probe micro-analyzer, X-ray diffraction and thermodynamic calculations, the spherical TiC and needle like TiB reinforced hybrid titanium matrix composites could be obtained in-situ by the conventional melting and casting route between titanium and $B_4C$. No melt-mold reaction occurred between the titanium matrix (TiC+TiB) hybrid composites and the SKK mold, since the mold is consisted with interstitial and substitutional metal-mold reaction products. Not only the sound in-situ synthesis but also the economic net-shape forming of the titanium matrix (TiC+TiB) hybrid composites could be possible by the conventional casting route.

  • In this study, 1-3 type composite specimens were fabricated with PZT powders prepared by the molten-salt synthesis method and Eccogel polymer matrix. A virtual level-meter was fabricated with a 1-3 type composite probe and electronic unit for underwater application. There was no difference in values between a virtual and measured level on its level meter.

  • The development of metal catalysts that can polymerize or copolymerize "polar" $Ch_2=CHX$ monomers by insertion mechanisms would significantly expand the scope of metal-catalyzed polymerization and enable the synthesis of new materials with enhanced properties. We have studied the reactions of single-site olefin polymerization catalysts with vinyl chloride, acrylonitrile, and vinyl ethers, in order to probe monomer coordination trends, insertion rates and regioselectivity, and the structures and reactivity of metal alkyls that contain functional groups on the alpha and beta positions of the alkyl chain. These studies provide insights to the key issues that underlie the "polar monomer" problem. Copolymerization of olefins and selected vinyl ethers has been achieved.

  • A photoactive analog of ATP, 3'(2')-O-(p-azidobenzoyl)-adenosine 5-triphosphate (AB-ATP) was synthesized by chemically coupling N-hydroxysuccinimidyl-4-azidobenzoate (NHS-AB) and ATP. The utility of AB-ATP as an effective active-site-directed photoprobe was demonstrated using catalytic subunit of protein kinase A as a model enzyme. Photoincorporation of AB-ATP was saturated with apparent dissociation constant of $30{\mu}m$ and protected completely by $100{\mu}m$ of ATP. When the enzyme was covalently modified by photolysis in the presence of saturating amounts of photoprobe, about 60% inhibition of enzyme activity was observed. These results demonstrate that AB-ATP has potential application as a probe to characterize ATP-binding proteins including protein kinases.

  • In this work, a novel strategy is provided to prepare silver nanoplates by a fibronectin (Fn) nanofibril template. First, Fn molecules were controlled to assemble into amyloid-like nanofibrils in highly concentrated ethanol aqueous solution. The resultant nanofibrils could serve as a soft template to direct the formation of silver nanoplates. It is worth noting that the silver nanoplates are excellent surface-enhanced Raman scattering (SERS) substrate with 4-aminothiophenol (4-ATP) molecule as a test probe. This high active SERS substrate can also be used to detect drug molecule, 2-thiouracil with high sensitivity.

  • This paper describes the process of nanoparticle synthesis by laser ablation of consolidated microparticles. We have generated nanoparticles by high-power pulsed laser ablation of Al, Cu and Ag microparticles using a Q-switched Nd:YAG laser (wavelength 355 nm, FWHM 5 ㎱, fluence 0.8∼2.0 J/㎠). Microparticles of mean diameter 18∼80 ㎛ are ablated in the ambient air The generated nanoparticles are collected on a glass substrate and the size distribution and morphology are examined using a scanning electron microscope and a transmission electron microscope. The effect of laser fluence and collector position on the distribution of particle size is investigated. The dynamics of ablation plume and shock wave is analyzed by monitoring the photoacoustic probe-beam deflection signal. Nanosecond time-resolved images of the ablation process are also obtained by laser flash shadowgraphy. Based on the experimental results, discussions are made on the dynamics of ablation plume.

Gram-Positive Bacteria Specific Properties of Silybin Derived from Silybum marianum

  • Silybin has a potent antibacterial activity, more potent than silymarin II, against gram-positive bacteria without hemolytic activity, whereas it has no antimicrobial activity against gram-negative bacteria or fungi. The mode of action of silybin against the gram-positive bacterial cell was examined by investigating the change in plasma membrane dynamics of bacterial cells using 1 ,6-diphenyl-1,3,5-hextriene (DPH) as a membrane probe and by assessing the inhibition of macromolecular synthesis using radiolabeled incorporation assay. The results showed that silybin inhibited RNA and protein synthesis on gram-positive bacteria.

Reliable preparation of [11C]GR205171, a selective NK1 radioligands for noninvasive imaging

  • [11C]GR205171, a Neurokinin 1 (NK1) radioligand, has been known as such a promising PET probe for quantitation of NK1 receptors in the brain by positron emission tomography (PET) imaging. First trial to synthesis of [11C]GR205171 was to use methylene chloride and tetrabutylammonium hydroxide for preactivation of precursor, but the result was not successful in radiochemical yield (0~25%) and unreliable. 7 years later, inorganic base (Cs2CO3) was tried to achieve higher radiochemical yield, and they showed higher yield (~53%). We have tried to repeat the same synthesis method, but it did not work properly, because there were the lack of the detail procedure and still reproducibility in radiochemical yield. Here we report the improved synthesis protocol to produce [11C]GR205171 in high yield via commercial automated synthesizer. The sonicator which combines water heating bath was used to activate desmethyl-GR205171, and this method showed high efficiency and reasonable yields (4.7 ± 0.6%, non-decay corrected from molecular sieve trap) with >95% radiochemical purity.
