Cathepsin D (CtsD), an aspartyl peptidase, is involved in apoptosis, resulting in the release of cytochrome C from mitochondria in cells. Here, we investigated microRNA regulation of CtsD expression in 3T3-L1 cells. First, we observed the expression of CtsD in cells in response to doxorubicin (Dox). As expected, the level of CtsD mRNA increased in 3T3-L1 cells exposed to Dox in a dose-dependent manner. The cellular viability of ectopically expressed CtsD cells was decreased. Next, we used the miRanda program to search for particular microRNA targeting CtsD. MiR-145 was selected as a putative controller of CtsD because it had a high mirSVR score. In a reporter assay, the luciferase activity of cells containing the CtsD 3'-UTR region decreased in cells transfected with a miR-145 mimic compared to that of a control. The level of CtsD expression was down-regulated in preadipocytes ectopically expressing miR-145 and up-regulated by an miR-145 inhibitor. Cells also suppressed miR-145 expression when exposed to Dox. The miR-145 inhibitor reduced the cellular viability of 3T3-L1 cells. Taken together, these data suggest that miR-145 regulates CtsD-mediated cell death in adipocytes. These findings may have valuable implications concerning the molecular mechanism of CtsD-mediated cell death in obesity, suggesting that CtsD could be a useful therapeutic tool for the prevention and treatment of obesity by regulating fat cell numbers.

Progressive collapse is defined as a collapse caused by sectional destruction of a structural member which links to other surrounding structures. Currently the design guidelines for the prevention of progressive collapse is not available in Korea. So, structural engineers have a difficulty in evaluating progressive collapse. In this study, the static and dynamic analysis to evaluate the methods and procedures are conducted using commercial analysis program for RC moment resisting frames. According to the study, DCR value of RC moment resisting frame system based on code in Korea is over 2 and it shows that it can't provide alternate load paths due to the progressive collapse. And additional reinforcement should be considered for the progressive collapse resistance. As a result of vertical deflection and DCR value of linear static analysis and linear dynamic analysis, the results of dynamic analysis were underestimated more than the result of static analysis. Thus, the dynamic coefficient value of 2 provides conservative estimation.
Ballasted track on an asphalt roadbed can be beneficial for its various effects such as (i) decreasing of roadbed thickness by dispersing train load; (ii) prevention of both strength reduction and weakening in roadbed system by preventing rainwater penetration; and (iii) reducing maintenance cost by preventing roadbed mud-pumping and frostbite. With these beneficial effects, ballasted track on asphalt roadbed has been widely used in Europe and Japan, and relevant research for applying such ballasted track on asphalt roadbed systems in Korea is ongoing. In this study, full-scale static and dynamic train load tests were performed to compare the performance of ballasted track on asphalt roadbed and ballasted track. The optimum thickness levels of asphalt and reinforced roadbeds, corresponding to the design criteria for reinforced roadbed of high-speed railway, was estimated using the FEM program ABAQUS. Test results show that the earth pressure on reinforced roadbed of ballasted track on the asphalt roadbed was relatively low compared with that of simple ballasted track. The elastic and plastic displacements of simple ballasted track on the asphalt roadbed were also lower than those of ballasted track. These test results may indicate that the use of ballasted track on asphalt roadbed is an advantageous system in view of long-term maintenance.
he purpose of this study was to examine the awareness of dental hygienists about medical malpractice and dispute. The subjects in this study were 206 dental hygienists at dental clinics, on whom a survey was conducted. After SPSSWIN 14.0 program was utilized to analyze the collected data, the following findings were given: 1. As for experiences of being complained from patients by age, the dental hygienists who were at the age of 21 to 23 made up the largest age group (39.2%) who had ever meet patients who had grievances. By career, those who had worked for less than a year constituted the greatest group (30.4%) who had the same experiences. It indicated that the dental hygienists who were younger and had a less career had been more complained from patients. 2. In terms of problems posed by patients, scaling came into question the most (14.5%), followed by diagnosis (14.4%), impression (12.9%) and radiography (11.0%). 3. Regarding perception of scaling treatment, 90.8 percent, the greatest percentage, kept records of patients about scaling. As to how to do that, 34.5 percent, the largest percentage, wrote it down and then signed their name. Concerning the way of explaining how to take care of oral health, 82.5 percent gave an explanation without any prints. 4. As to knowledge on the medical law, they got a mean of 12.34 out of possible 16 points. 5. Concerning the necessity of education about the prevention of medical accidents and countermeasures, 70.4 percent felt the strong and urgent need for that.
The purpose of this study was to examine the effects of the toothpick method, one of professional toothbrushing methods, on the prevention of periodontal diseases and the satisfaction level of patients with that. The subjects in this study were 33 patients who included 16 men and 17 women. After the toothpick method was applied to the selected patients from January 9 to February 28, 2008, the collected data were analyzed with SPSS (Statistical Package for the Social Science) 12.0 program. The findings of the study were as follows: First, the patients investigated showed a 0.71 reduction in PI than in the past. The index of bleeding upon probing stood at 5.12, which dropped from 8.09 in the beginning. Second, the patients gave a mean of 4.20 to the professional toothbrushing out of possible five points, which showed that they expressed satisfaction with that. Third, as for the relationship between general characteristics and satisfaction level, the women were more satisfied with that ($4.25{\pm}0.33$). By age group, those who were in their 60s and up found that more satisfactory ($4.31{\pm}0.30$). Fourth, as to changes in oral symptoms by gender, the largest number of the men and women considered their gums to become healthier after the professional toothbrushing was applied. The above-mentioned findings suggested that dental institutions should be equipped with well-educated oral health care personnels who are responsible for the periodontal health of patients, and the development of oral health promotion programs is urgently required as well.
The purpose of this study was to examine the relationship between vitamin D and periodontal diseases based on the raw data of the 5th National Health & Nutrition Examination Survey of 2010. The subjects in this study were 1,327 people, and those whose data on major variables were missing and who suffered from diabetes and/or osteoporosis were excluded. As for data analysis, R2.15.1 program and PASW Statistics 18.0 were utilized. The findings of the study were as follows: 1. As for all the respondents aged 50 and up, there was no significant relationship between vitamin D and periodontal diseases. 2. As for the post-menopausal women including the women who underwent bilateral ovariectomy, the vitamin D-deficit group 1 (<10) were 6.66-fold more likely to suffer from periodontal diseases than the vitamin D-sufficient group (${\geq}30$) (OR, 6.66; 95% CI, 1.004~44.19). The above-mentioned findings ascertained that vitamin D had a significant negative correlation to periodontal diseases among the post-menopausal women including the women who underwent bilateral ovariectomy. This finding should be taken into account in terms of the prevention and management of periodontal diseases.
This study conducted questionnaire survey with 268 workers in Ulsan city to examine influencing factors about Oral behavior and Missing teeth of some workers. The collected data were analyzed using SPSS 12.0 program. The analysis results are as follows. 1. Oral health awareness was highest in thirties (p<0.01). This paper examines the lava tubes designated as a natural monument in Jeju Island, and then makes the several suggestions for conserving and managing them. The cave entrances should be scientifically and strictly managed to prevent the deterioration, and to preserve the interior ecosystem, of the lava tubes. The land use and vegetation of the ground surface over the lava tubes should be also systematically managed; the destroyed sand dunes must be recovered for the development of speleothem in the lava tubes, and the plants with deep roots must be removed for the prevention of ceiling collapse. The provincial authorities should manage the ground surface over the lava tubes as a public land to remove the factors threatening the stability of the lava tubes. Since the roads crossing the ground surface are likely to cause the collapse of the lava tubes, new roads bypassing the caves need to be constructed, and warning signs should be also installed at the traverse points. A monitoring system on the cultural properties protection area over the lava tubes could be effectively established by encouraging residents to participate the conservation program of lava tubes. Since the falling blocks from ceilings have been usually observed in the lava tubes, it is urgent to examine their stability. In particular, the dangerous sections must be continuously monitored for taking a quick measure against the collapse. The air environments of lava tubes need to be scientifically analyzed based upon the long-term observations. The lighting equipments must be improved to minimize the green pollution in the lava tubes open to the public. The numbers of visitors should be also controlled to prevent the destruction and contamination of lava tubes caused by a large number of visitors.
The types of Alpine ski injuries have changed through the years in relationship to the development of skiing equipment. Modern skis, boots and bindings are better at protecting the tibia, which previously was almost as commonly injured as the knee. Since the 1980s, severe knee sprains, most of them involving the anterior cruciate ligament have tripled while injuries of the lower extremity below the knee diminished significantly. However, recent studies show no further improvements in either lower leg fractures or increase in the rates of ACL sprains has occurred. The use of carving skis presents an increased risk for sustaining isolated ACL injuries in more skilled skiers and less skilled skiers are more likely to sustain an ankle fracture than skiers using conventional skis. To restore the trend of diminishing lower leg injury rates, efforts will be needed to motivate skiers to have their equipment serviced by ski shop professionals following ASTM (American Society for Testing and Material) standard procedures. As of now, there are no boots, bindings or skis on the market designed to protect skiers from the ACL injury. The only method proven to reduce ACL injury Is a training program based on recognizing the circumstances that lead to ACL injury in skiing and to avoid these events.
