Growth and Pigment Formation of Genus Monascus on Medium Compositions (배지에 따른 Genus Monascus의 증식도와 색소함량)

  • Growth and pigment formation in Genus Monascus(M. pilosus and M purpureus) related 15 kinds of culture media(Lin medium, SP medium, YM medium, YE medium, GMIN medium, SMO medium, MY medium, GY medium, Nishikawa medium, sucrose medium, stock culture, Mizutani medium, modified Lin medium, Toya medium and rice medium) were investigated. Mizutani medium and Lin medium among 15 kinds of the culture media showed good growth for M. pilosus, M purpureus, fresh mycelium weight cultivated for 10 days at 30$^{\circ}C$ was each 24.5∼26.9 or 15.9∼17.2 g/100 mL. The culture media which showed higher content of pigment production in two fungi were Lin medium(OD: 1.2 ∼ 1.6 and Mizutani medium(OD: 0.8 ∼ 1.0) that showed higher in M. pilosus.

Effects of Relative humidity Conditions on the Compressive Strength Changes of Corrugating Mediums (상대습도조건에 따른 골심지의 압축강도 변화에 관한 연구)

  • Changes in the compressive strength of four typical corrugating mediums (K$_2$A, AS and S) as affected by relative humidity conditions were compared and their relative cost effectiveness was analysed. All mediums lost their compressive strength as relative humidity increased. at the relative humidity of 93% , As medium lost 58% of its initial strength while S medium did about 40%. Calculations of compress factor and compress factor by price indicated that $K_2$ medium was the most cost effective and maintained the highest compressive strength among the mediums tested. It was recommended that $K_2$ medium could be effectively used to make corrugated fiberboard especially for fresh agricultural product packaging .

The Effect of Self-Leadership on the Lntention of Start-up in Women: The Mediation Effect of Self-Efficacy (여성의 셀프리더쉽이 창업의도에 미치는 영향 : 자기효능감의 매개효과)

  • This study analyzed the impact of women's self-preservation on their intention to start a business by using self-efficacy as a mediation, reflecting the importance of women's self-preservation to adapt to the new environment of start-up. In particular, in addition to the overall self-preservation, the sub-factors of behavioral delay, compensatory natural strategy, and constructive thinking strategy were also validated. As a research method, 198 people who responded to the online survey were analyzed using Model 4 of the PROCESS macro proposed by Hayes(2018), and indirect effects were analyzed by designating 5,000 bootstrapping samples. The findings were as follows. First, self-efficacy has been shown to have a mediating effect in which women's self-preservation influences their intention to start a business. At this time, the medium effect was shown as a partial medium. Second, the sense of self-efficacy was found to have a mediating effect on women's behavior-oriented strategies affecting their intention to start a business. At this time, the medium effect was shown as a partial medium. Third, self-efficacy was found to have a mediating effect in which women's natural compensatory strategies affect their intention to start a business. At this time, the medium effect was shown as a partial medium. Fourth, self-efficacy was found to have a mediating effect in which women's constructive thinking strategies affect their intention to start a business. At this time, the medium effect was shown as a complete medium. This shows that the higher a woman's self-preservation, the higher her self-efficacy and willingness to start a business. Based on the results of these studies, he made a developed proposal for women's start-ups.

A Study on Chamber Design for the Efficient Preservation of a Small Amount of Archives (소량 기록물의 효율적 보존을 위한 보존챔버 설계에 관한 연구)

  • The archives preservation chamber is an equipment that is able to store important archives with air-cleaning and isothermal-isohumidity control functions to permanently store archives in a highly safe environment. The chamber can only store small amounts of archives but it is crucial for organizations preserving archives to economically store a variety of archives, be it a medium or a small amount of documents. An arm of this study researches the current preservation status of the local record center to determine the need for such a chamber. In addition, in order to improve the air-cleaning and isothermal-isohumidity functions, the researchers of this study have conducted analyses to design the core parts. The preservation chamber that had been designed includes an archives preservation area, air-cleaning equipment, and environment measurement equipment. A preservation chamber is used to preserve small amounts of important archives safely. Thus, when a local record center is preserving various types of archives in small or medium quantities in a stack room, the preservation chamber can be used to aid their long-term preservation.

Development of hypothermic preservation solution for the human dermal fibroblast using protein hydrolysates (단백질 가수분해 물을 이용한 인간 피부 섬유아세포의 저온 보존액 개발)

  • Stable cell preservation is an essential factor in the regenerative medicine for cell therapies and transplantation of biologic materials. In this study, we studied to provide more stable hypothermic preservation by protection of cell damage during the preservation at $4^{\circ}C$. The result of searching for key components that have excellent efficacy in hypothermic preservation of cells, we have identified the fact that the hypothermic preservation adding protein hydrolysates such as yeast hydrolysate is far superior to others. All protein hydrolysates that are derived from animal, plant and microbe sources have superior efficacy, especially the peptides which have molecular weights under 10 kDa have the best efficacy among the components of protein hydrolysate. The protein hydrolysates prevented the decrease of ATP level in the cells caused by hypothermic environment and they inhibited the generation of ROS. Adding antioxidants and control agents of osmotic pressure were showed to have more superior efficacy in hypothermic preservation. Finally, KUL261 solution (DMEM/F12 1 : 1 medium, yeastolate 1%, $\alpha$-tocopherol $100{\mu}M$, dextran 2.5%), the preservation solution developed in this study, showed the best efficacy in both cell viability and cell growth more than other conventional preservation solutions. In conclusion, the improved hypothermic preservation solution that contains the protein hydrolysates as a key component provide the best preservation efficacy. It provides better efficacy than other preservation solutions and will contribute to both the development of regenerative medicine and global commercialization in this therapeutic field.

Comparison of Viability in Basidiomycetes After Low Temperature Storage According to Storage Period (저온 보존 기간에 따른 담자균류의 생존율 비교)

  • Short-preservation of basidiomycetes is generally being conducted in slant tubes containing solid medium based on agar. In this study, we investigated the vitality of 28 species and 76 strains preserved on potato dextrose agar (PDA) at $4^{\circ}C$ for 2~7 years. The survival rates of the fungi were 82%, 86%, 94%, 96%, 94%, and 94% for seven, six, five, four, three, and two years old preservation, respectively. The volume of medium in Lentinula edodes showed decrease after 2 years preserved. The pH of preserved medium was 5.42 in 2007 (two years old), but it became nearly neutral as increasing preservation term.

Character and Sustainability of Built Heritage Listed in the Eight Scenery

  • The purpose of this study is to examine the characters and sustainability of built heritage featured in the eight-scenery listings of Gyeonggi municipalities in South Korea. Eight scenery is a place marketing medium that reflects the community's appreciation of the selected resources. For a research method, we utilize framework analysis to understand the cultural resources in three categories: heritage value identification, preservation, and dissemination. Primary sources for data collection include the chosen eleven cities' eight-scenery webpages, the National Cultural Heritage Portal, and articles about the built heritage resources included in the eight scenery. The results of this study show the public's acknowledgment of diverse built heritage, from prestigious monuments to local heritage interpretations, and various preservation and public program development approaches. The conclusion discusses the research findings regarding historic preservation, sustainable development, and place marketing.

EVALUATION OF PERIODONTAL LIGAMENT CELL VIABILITY IN RAT TEETH AFTER FROZEN PRESERVATION USING IN-VIVO MTT ASSAY (급속냉동된 쥐 치아의 in vivo MTT 검색법을 이용한 치주인대세포 활성도 평가)

  • The purpose of this study was to examine the viability of PDL cells in rat molars by using in vivo MTT assay, which was used to compare fast cryopreservation group by liquid nitrogen $(-196^{\circ}C)\;with\;4^{\circ}C$ cold preservation group. A total of 74 Sprague-Dawley white female rats of 4 week-old with a body weight of 100 grams were used. The maxillary left and right, first and second molars were extracted as atraumatically as possible under ketamine anesthesia. Ten teeth of each group were divided as six experimental groups depending upon the preservation. Cryopreservation groups were Group 1 (5% DMSO 6% HES in F medium) Group 2 (10% DMSO in F medium), Group 3 (5% DMSO 6% HES in $Viaspan^(R)$). Group 4 (10% DMSO in $Viaspan^(R)$) which were cryopreserved for 1 week and cold preservation groups were Group 5 (F medium) , Group 6 ($Viaspan^(R)$) at $4^{\circ}C$ for 1 week. Immediate extraction group was used as a control. After preservation and thawing, the in vivo MTT assay was processed. Two way ANOVA and Duncan's Multiple Range Test was performed at the 95 % level of confidence, Another 2 teeth of each group were treated as the same manner and frozen sections $10{\mu}m$ thick for microscopic observation. The value of optical density obtained after in vivo MTT analysis was divided by the value of eosin staining for tissue volume standardization. Group 1, 2 had significantly higher optical density than Group 3 and 4 which had the lowest OD value. Group 6 had higher OD value than in Group 5 (P<0.05). Histological findings of periodontal ligament cell, after being stained with MTT solution were consistent with the in vivo MTT assay results. In this study, the groups which were frozen with DMSO as a cryoprotectant and the groups with F medium showed the best results.

Factors Affecting on Quality Evaluation of Citrus unshiu Produced in Cheju (제주산 온주밀감의 품질평가에 미치는 요인)

  • Soluble solids($^{\circ}$Brix), acid content and edible part ratio of Citrus unshiu collected at sorting Places in south Cheju area wee more lowered, and peel thickness was more thickened with increasing fruit size. Compared to early variety of Citurs unshiu(C. unshiu Miyakawa Mar), peel thickness of medium variety of Citurs unshiu(C. unshiu Hayashi Mar) was more thickened, and edible part ratio was more lowered with increasing fruit size. Soluble solids of medium variety of Citurs unshiu were lowered, and acid contents were increased compared to early variety. Processing properties for pressing concentrated juice was good for early variety of Citurs unshiu, especially on fruit diameter of 50-65mm. Soluble solids, acid contents and juice ratio were decreased with increasing peel thickness. The quality properties for fresh fruit with peel thickness had a good correlation on early variety of Citurs unshiu, and these data are supposed to be applied to the quality evaluation of Citurs unshiu produced in Cheju.

Change in Growth of alcohol Fermentation Yeast with Addition of Deep Seawater (해양 심층수 첨가에 따른 알콜발효 효모의 증식 변화)

  • In order to study optimum culture condition of yeast medium added deep seawater, we examed samples with 9 yeast strains. The growth rate were measured for Saccharomyces cerevisiae 10, 11, 12, 901 and RCY and Saccharomyces kluyvery DJ97, Saccharomyces cerevisiae YJK, JK99, CMY-28 etc.. The growth of S. cerevisiae 12 was found most active in the deep seawater(hardness 500). The growth rate of S. cerevisiae 901 on medium containing deep seawater(hardness 1000) was faster than that of the yeast on medium without deep seawater. The use of deep seawater on the growth of Sacch.cerevisiae kluyvery DJ97 revealed maximum growth under the condition of hardness 200 of deep seawater and 10% of sugar concentration.
