Identification of pathways and genes associated with cerebral palsy

  • Cerebral palsy (CP) is a non-progressive neurological disease, of which susceptibility is linked to genetic and environmental risk factors. More and more studies have shown that CP might be caused by multiple genetic factors, similar to other neurodevelopmental disorders. Due to the high genetic heterogeneity of CP, we focused on investigating related molecular pathways. Ten children with CP were collected for whole-exome sequencing by next-generation sequencing (NGS) technology. Customized processes were used to identify potential pathogenic pathways and variants. Three pathways (axon guidance, transmission across chemical synapses, protein-protein interactions at synapses) with twenty-three genes were identified to be highly correlated with CP. This study showed that the three pathways associated with CP might be the molecular mechanism of pathogenesis. These findings could provide useful clues for developing pathway-based pharmacotherapies. Further studies are required to confirm potential roles for these pathways in the pathogenesis of CP.

  • The present study was designed to investigate the morphological developments of the stomach in the prenatal and postnatal rats. The gastric fundus of 16- to 22-day-old fetuses, neonates, 7-day-old, 14-day-old, 21-day-old, and adult rats were observed by transmission electron microscopy. 1. In the 19-day-old fetuses, the gastric pits and the parietal cells were the primitive state, but the parietal cells contained numerous microvilli and mitochondria. 2. In the 20-day-old fetuses, the intracellular canaliculus appeared in the parietal cells, and glycogen granules were localized mainly in the basal part of the epithelial cells and parietal cells. 3. In the 20-day-old fetuses, the mucous granules and zonular occludens appeared in the gastric glandular cells adjacent to the parietal cells. 4. In the 21-day-old fetuses, the chief cells were identified in the lamina propria and these cells contained granular endoplasmic reticulum and zymogen granules. 5. In the 20- to 22-day-old fetuses, the definitive parietal cells were firstly appeared in pairs or groups within the lamina propria. These cells tended to be placed near the basement membrane and blood capillary, and frequently tended to be extruded into lumen of gland. 6. The endocrine cells were appeared at the basal areas of the gastric gland after the 14-day-old.

  • Trans-placental neonatal human immunodeficiency virus (HIV) infection is common in Africa; however, it is not yet reported in the Republic of Korea. With the increasing incidence of HIV infection, especially in the reproductive age group, the risk of the vertical transmission of HIV is also increasing. We report the first case of HIV infection acquired in-utero in a newborn in Korea. The baby is growing well with normal development.

  • Pneumocystis carinii is pulmonary pathogen of immunocompromised humans or other mammals. Its infection results from activation of organism involves in latent infection or form new infection through the air. Almost all children are known to be infected within 2 to 4 years of birth, though prenatal transplacental transmission has not yet been demonstrated. In this study we observed experimental P.carinii infection in neonatal rats, thus investigating the possibility of transplacental vertical transmission by Diff-Quik staining of the lung impression smears and in-sity hybridization for lung sections. The postive rate of P.carinii infection in immunosuppressed maternal rats was 100%, but that in normal maternal rats was 0%. Cystic forms of P.carinii were observed in three of six 1-week old neonatal rats born of heavily infected mothers, but none of them was positive by in-situ hybridization. Five weeks after birth, cystic forms were detected in four neonatal rats. In the lobes of the lungs, no predilection site of P.carinii was recognized. Counts of cystic forms on smears and the reactivity of in-situ hbridization in the lungs of neonatal rats 0 were signficantly lower than in maternal rats. The present findings suggest that P.carinii is rarely transmitted through the placenta and proliferates less successfully in the lungs of neonatal rats than in mothers.

  • There needs disease-predictable signs in order to enable preventive diagnosis and therapy. Then traditional Chinese medicine applies various medical diagnostic equipments used in western medicine to diagnosing sub-healthy state. But such data are not originated from inherent oriental medicine, and not obtained easily in ordinary clinical practice. This paper is to provide synopsis of the ante-disease diagno-therapeutics partly and to show predictable data based on the facial shapes and signs, especially of gall bladder's versus bladder's body and masculine versus feminine shape. Ante-disease means not only the complete healthy state, but also the state unseen any symptoms in macrographically in the course of outbreak of disease. It contains two stages, first one is the former state of disease and second one is untransmitted state of disease. The patterns of ante-disease consist of latent disease, pre-disease, transmission type like senescent syndrome, abnormal reactive syndrome(變證), syndrome of transmission and transmutation. The classification with gall bladder and bladder type manifests the differences of shape, color and size of each organ in comparison of the universal and standard figures of the human being. On the other hand, the classification with masculine and feminine shape contrasts the innate sexual difference and the shape, characteristics originated from in itself. These two classification theories have their own pathologic types and syndrome types with each disease so that disease-predictable data can be constructed based on such a relationship. In addition, this diagnostic method by facial shapes and signs is able to be applied to whole stages from prenatal to present state of disease even if the cause and inducement are not clear. Ante-disease diagno-theraputic system by Gall Bladder's versus Bladder's Body and Masculine versus Feminine Shape is getting more important in the chronic and internal disease in comparison of the acute and traumatic disease. So this study is able to make up for the limit of diagnosis on ante-disease in the field of oriental medicine clinic.

In order to increase our physical power through raising the amount of Hb to normal level and thus enrich our national power, it is earnestly required to improve the general eating habits in the direction of taking enough animal protein (nutrition food) that is required to form Hb and adequate administrative procedures of the nursery school are to be taken at the same time. 2. The genetic amentia's development of skeleton is generally under the normal level. And so their physical condition should be inferior to the standard growth rate of the same age. But the fact is that their physical growth is almost the same as the same age with only few exceptions. Besides, considering the fact that their amount of Hb is less than that of the normal level, I think we can conclude that the majority of the amentia in the nursery school were malnutrition during their prenatal period or during their infancy. We need continuous statistical study concerning many amentia that is scattered all part of our country, to make our amentia's hereditary transmission and the expression clear. And so the Intelligence Quotient of the amentia are from 26 to 80, we consider to need nourishing meal in ordinary times to make more efficient development of intellectual faculties. 3. As to the relation between blood type and disease, there are much debates going on in many countries. To get more apodictic results, however, continuous study is desired to be made in the future.
