A Parametric Study of Sheet Pile Wall Near the Laterally Loaded Pile (횡방향 재하 말뚝 주변의 널말뚝에 관한 변수연구)

  • Construction of sheet pile retaining walls in urban and coastal regions has resulted in sheet pile walls in close proximity to laterally loaded pile foundations. However, there is currently little information available in the literature to assist engineers for quantifying the response of sheet pile walls. This study provides a quantitative method for estimating sheet pile wall response due to loads imposed from a nearby laterally loaded pile. Three dimensional finite element analyses using commercial software, ABAQUS, were performed to assess the response of a sheet pile wall and nearby laterally loaded pile. The soils were modeled using Drucker-Prager constitutive model with associated flow rule, and the sheet pile wall and pile foundation were assumed to behave linear elastic. Four parameters were investigated: sheet pile wall bending stiffness, distance from the pile face to the wall, excavation depth in front of the sheet pile wall, and elastic modulus of the soil. Results from the analyses have been used to develop preliminary design charts and simple equations for estimating the maximum horizontal displacement and maximum bending moment in the sheet pile wall.

Optimum Conditions for the Separation of Lecithin from Egg Yolk by Response Surface Methodology (반응표면 분석법을 이용한 계란 난황 레시틴 분리 최적조건)

  • The optimum conditions for the extraction of lecithin from egg yolk were determined using response surface methodology (RSM). On the basis of the results of preliminary experiments, the most effective values were selected. The effects of three independent variables, dilution ratio, solvent composition, and extraction temperature on the response of crude egg lecithin (g) were then determined. The optimum conditions for egg lecithin separation obtained using ridge analysis were 6.51, 95.83%, and $40.2^{\circ}C$ for the dilution ratio, solvent composition, and extraction temperature, respectively. Using the optimum conditions, 55.04% of crude lecithin in total phospholipid can be obtained from 100 g liquid egg yolk. The experimental values (56.21% crude lecithin in total phospholipid) agreed with the predicted values.

Phase II Study of Preoperative Intra-Arterial Epirubicin, Etoposide, and Oxaliplatin Combined with Oral S-1 Chemotherapy for the Treatment of Borrmann Type 4 Gastric Cancer

  • Purpose: A phase II study was conducted to evaluate the safety and efficacy of preoperative, intra-arterial perfusion of epirubicin, etoposide, and oxaliplatin combined with oral chemotherapy S-1 (SEEOX) for the treatment of type 4 gastric cancer. Materials and Methods: A single-center, single-arm phase II trial was conducted on 36 patients with histologically proven type 4 gastric cancer without distant peritoneal or organ metastasis. Patients received 3, 21-day courses of SEEOX preoperative chemotherapy. The primary endpoint was overall survival (OS) and the secondary outcomes assessed were chemotherapeutic response, radical resection rate, pathological regression, toxicities, postoperative morbidity, and mortality. Results: All patients were at an advanced stage of cancer (stage III or IV) and completed the entire course of treatment. Based on changes in tumor volume and peritoneal metastasis, the objective response rate was 55.6% (20/36; 95% confidence interval [CI], 38.5%-72.6%) and the disease control rate was 69.4% (25/36; 95% CI, 53.6%-85.3%). The radical resection rate was 75% (27/36; 95% CI, 60.1%-89.9%) and the proportion of R0 resections was 66.7% (21/36; 95% CI, 50.5%-82.8%). The pathological response rate was 33.3%, of which 13.9% showed complete pathological regression. The median survival was 27.1 months (95% CI, 22.24-31.97 months), and the 2-year OS was 48.5% (95% CI, 30.86%-66.1%). Conclusions: Preoperative SEEOX is a safe and effective treatment for type 4 gastric cancer. Based on these preliminary data, a phase III study will be conducted to confirm the superiority of this regimen over standard treatment.

HIV-1 Vaccine Development: Need For New Directions

  • The AIDS epidemic continues unabated in many part of the world. After near two decades, no vaccine is available to combat the spread of this deadly disease. Much of the HIV -1 vaccine effort during the past decade has focused on the viral envelope glycoprotein, largely because it is the only protein that can elicit neutralizing antibodies (Nabs). Eliciting broadly cross-reactive Nabs has been a primary goal. The intrinsic genetic diversity of the viral envelope, however, has been one of the major impediments in vaccine development. We have recently completed a comprehensive study examining whether it is possible to elicit broadly acting Nabs by immunizing monkeys with mixtures of envelope proteins from multiple HIV -1 isolates. We compared the humoral immune responses elicited by vaccination with either single or multiple envelope proteins and evaluated the importance of humoral and non-humoral immune response in protection against a challenge virus with a homologous or heterologous envelope protein. Our results show that (1) Nab is the correlate of sterilizing immunity, (2) Nabs against primary HIV -1 isolates can be elicited by the live vector-prime/protein boost approach, and (3) polyvalent envelope vaccines elicit broader Nab response than monovalent vaccines. Nonetheless, our findings clearly indicate that the increased breadth of Nab response is by and large limited to strains included in the vaccine mixture and does not extend to heterologous non-vaccine strains. Our study strongly demonstrates how difficult it may be to elicit broadly reactive Nabs using envelope proteins and sadly predicts a similar fate for many of the vaccine candidates currently being evaluated in clinical trials. We have started to evaluate other vaccine candidates (e.g. genetically modified envelope proteins) that might elicit broadly reactive Nabs. We are also exploring other vaccine strategies to elicit potent cytotoxic T lymphocyte responses. Preliminary results from some of these experiments will be discussed.

Differential Proteomic Analysis of Chinese fir Clone Leaf Response to Salicylic Acid

  • Chinese fir (Latin name: Cunninghaimia lanceolata) is one of the major commercial coniferous trees. Most of Chinese fir forests are managed in successive rotation sites, which lead productivity to decline. Autotoxicity is the important reason for soil degradation of Chinese fir plantation, especially, phenolic acids are considered as the major allelopathic toxins which induce autotoxicity in Chinese fir rotation stands. We performed here proteomic approach to investigate the response of proteins in Chinese fir leaves to salicylic acid. The tube plantlets of Chinese fir clone were treated with 120 mg/L salicylic acid for 1, 3 and 5th day. 2-DE, coupled with MALDI-TOF-TOF/MS, was used to separate and identify the responsive proteins. We found 12, 7, and 12 candidate protein spots that were up- or down-regulated by at least 2.5 fold after 1, 3, and 5th day of the stress, respectively. Of these protein spots, 16 spots were identified successfully. According to the putative physiological functions, these proteins were categorized into five classes (1) the proteins involved in protein stability and folding, including 26S proteome, Grp78, Hsp70, Hsp90 and PPIase; (2) the protein involved in photosynthesis and respiration, including OEC 33 kDa subunit, GAPDH; (3) the protein related to cell endurance to acid, F-ATPase; (4) the protein related to cytoskeleton, tubulin; (5) the protein related to protein translation: prolyl-tRNA synthetase. These results give new insights into autotoxic substance stress response in Chinese fir leaves and provide preliminary footprints for further studies on the molecular signal mechanisms induced by the stress.

Design of MR dampers to prevent progressive collapse of moment frames

  • In this paper the progressive collapse resisting capacity of steel moment frames with MR dampers is evaluated, and a preliminary design procedure for the dampers to prevent progressive collapse is suggested. Parametric studies are carried out using a beam-column subassemblage with varying natural period, yield strength, and damper force. Then the progressive collapse potentials of 15-story steel moment frames installed with MR dampers are evaluated by nonlinear dynamic analysis. The analysis results of the model structures showed that the MR dampers are effective in preventing progressive collapse of framed structures subjected to sudden loss of a first story column. The effectiveness is more noticeable in the structure with larger vertical deflection subjected to larger inelastic deformation. The maximum responses of the structure installed with the MR dampers designed to meet a given target dynamic response factor generally coincided well with the target value on the conservative side.

  • This paper presents a capacitive humidity sensor using porous silicon layers formed tom the anodization of p-type silicon in HF solution. The upper electrodes consist of many aluminum strips over porous silicon, between which the porous silicon is etched away. The sensor showed a good sensitivity(20pF/%RH) and lineaity in the range of 40%RH$\sim$80%RH, a hysteresis of ${\pm}2%$ RH, and a slow transient response. These preliminary resluts show that futher improvement can still be expected.

Parametric seismic evaluation of highway overpass bridges in moderate seismic areas

  • Prior to modern seismic provisions, several bridges were not designed for seismic actions in moderate seismic areas. Precast multi-girder and slab bridges are typical highway overpass structures; they have a significant contribution to national bridge stocks. Since the seismic behavior is questionable, a preliminary parametric study is conducted to determine critical configurations and components. The results indicate that the behavior of the abutments, backfill soil, superstructure and foundation is normally satisfactory; however, the superstructure-abutment joints are critical for both single- and multi-span bridges, while the piers are also critical for longer multi-span configurations. The parametric results provide a solid basis both for detailed seismic assessment and development of design concepts of newly built structures in moderate seismic zones.

  • It is well known that the metallo-phthalocyanines (MPcs) are sensitive to toxic gaseous molecules such as NO$_2$ as well as are chemically and thermally stable. Therefore, lots of MPcs have been studied as the potential chemical sensor for NO$_2$gas using quartz crystal microbalance(QCM) or electrical conductivity. In this study, thin films of octa(2-ethylhexyloxy)copper-phthalocyanine were prepared by Langmuir-Blodgett method and characterized by using UV-VIS absorption spectroscopy and ellipsometry. Optimal transfer condition of LB films was investigated and preliminary results of current-voltage(I-V) characteristics of these films exposed to NO$_2$gas as a function of film thickness, temperature and concentration were discussed.

Histomorphometric Analysis of Two Types of Coated Implants : a Preliminary Study Using the Rabbit Tibia Model

  • Purpose : The purpose of this pilot experiment was to evaluate early bone response in two types of coated implants using the rabbit tibia model. Materials and Methods : Screw type titanium implants manufactured with a calcium metaphosphate (CMP) coating and hydroxyapatite (HA) coating were placed in the tibiae of 3 New Zealand White rabbits. The bone responses at 2 weeks after insertion were evaluated and compared by histomorphometry. Results : There was no significant difference in bone-to-implant contact between the groups (P>.05). However, some qualitative differences on histologic views were found. Conclusions : CMP-coating is suggested to be the preferred candidate for fast osseointegration over HAcoating.
