Proteomic Analysis of Bovine Pregnancy-specific Serum Proteins by 2D Fluorescence Difference Gel Electrophoresis

  • Two dimensional-fluorescence difference gel electrophoresis (2D DIGE) is an emerging technique for comparative proteomics, which improves the reproducibility and reliability of differential protein expression analysis between samples. The purpose of this study was to investigate bovine pregnancy-specific proteins in the proteome between bovine pregnant and non-pregnant serum using DIGE technique. Serums of 2 pregnant Holstein dairy cattle at day 21 after artificial insemination and those of 2 non-pregnant were used in this study. The pre-electrophoretic labeling of pregnant and non-pregnant serum proteins were mixed with Cy3 and Cy5 fluorescent dyes, respectively, and an internal standard was labeled with Cy2. Labeled proteins with Cy2, Cy3, and Cy5 were separated together in a single gel, and then were detected by fluorescence image analyzer. The 2D DIGE method using fluorescence CyDye DIGE flour had higher sensitivity than conventional 2D gel electrophoresis, and showed reproducible results. Approximately 1,500 protein spots were detected by 2D DIGE. Several proteins showed a more than 1.5-fold up and down regulation between non-pregnant and pregnant serum proteins. The differentially expressed proteins were identified by MALDI-TOF mass spectrometer. A total 16 protein spots were detected to regulate differentially in the pregnant serum, among which 7 spots were up-regulated proteins such as conglutinin precursor, modified bovine fibrinogen and IgG1, and 6 spots were down-regulated proteins such as hemoglobin, complement component 3, bovine fibrinogen and IgG2a three spots were not identified. The identified proteins demonstrate that early pregnant bovine serum may have several pregnancy-specific proteins, and these could be a valuable information for the development of pregnancy-diagnostic markers in early pregnancy bovine serum.

  • The early diagnosis of bovine pregnancy is an essential component of successful reproductive planning on farms, because lack of bovine pregnancy over the long term results in reproductive failure and low milk yield-the latter of which is a special concern on dairy farms. This study was designed to identify early pregnancy-specific whey proteins in bovine, by comparing milk samples collected from cattle during pregnancy (Days 30 and 50) and from non-pregnant cattle. In this study, differentially expressed proteins in five pregnant and five non-pregnant Holstein dairy cattle were investigated and compared, using proteomics analysis. The first dimension was applied to a pH $3.0{\sim}10.0$ strip, by loading a 2-mg milk protein sample. After the second-dimension separation was performed, the gels were stained with colloidal Coomassie brilliant blue. The stained gels were scanned and the images were analyzed, to detect variations in protein spots between non-pregnant and pregnant cattle milk protein spots, using ImageMaster, this was followed by analysis with MALDI TOF-MS. Analysis of the 2-DE gel image resulted in a total of approximately $500{\sim}600$ protein spots, of which 12 spots were differentially expressed, six spots were up-regulated, and four spots were down-regulated; two spots were identified as pregnancy-specific proteins. These proteins were identified as lactoferrin, NA-DH dehydrogenase subunit 2, albumin, serum albumin precursor and transferrin. Our results via 2-D PAGE analysis revealed composite profiles of several milk proteins related to early bovine pregnancy, implying the possible use of these milk proteins in the early detection of bovine pregnancy.

Production of Retinol-binding Protein by Caprine Conceptus during the Time Period of Maternal Recognition of Pregnancy

  • The purpose of the study were to characterize the proteins secreted by elongating caprine conceptus, to identify a group of low molecular weight proteins as retinol-binding protein (RBP), to identify RBP cell-specific localization in conceptus tissue, and to demonstrate that the conceptuses secreted continuously RBP during the time period maternal recognition of pregnancy. Caprine conceptuses were removed from the uterus between days 16 and 22 of pregnancy, the time period maternal recognition of pregnancy. Isolated conceptuses were cultured in a modified minimum essential medium in the presence of radiolabeled amino acids. Proteins synthesized and secreted into medium were analyzed by fluorography of two-dimensional polyacrylamide gel electrophoresis and fluorography. At least five proteins showed consistently a grouping of spots with characteristic location on two-dimensional gels. A major low molecular weight protein consisted of two major isoforms (pI 5.3-6.0) of similar molecular mass (21 kDa) was identified as RBP by using antiserum against RBP. Presence of RBP in conceptus culture medium and uterine flushings between days 16 and 22 of pregnancy were determined by immunoprecipitation and Western blotting using anti-RBP serum. In immunocytochemical study, strong immunostaining for RBP was localized in trophectoderm and endoderm of conceptus. These results clearly demonstrated that the caprine conceptus was active in protein synthesis as early as day 16 of pregnancy. Secretion of RBP by caprine conceptuses (days 16-22) coincident with the rapid transformation of the conceptus from a spherical blastocyst to a filamentous structure. Production of RBP by the elongating conceptuses may be indicative of an important role for conceptus RBP in the transport, availability and metabolism of retinol during maternal recognition of pregnancy.

  • The sex steroid hormone progesterone is essential for normal development and maturation of the endometrium in preparation for the embryo implantation and the maintenance of pregnancy. Insulin-like growth factor (IGF) system that is composed of IGF-I, IGF-II, IGF binding proteins (IGFBPs) is also involved in the maintenance of pregnancy. In addition, liver, kidney, and uterus is a target tissue for IGF system. However, the effect of exogenous progesterone on IGF system was not elucidated in female rats. Therefore, we investigated the effect of progesterone on insulin-like growth factors (IGFs) and IGF-binding proteins in serum, liver, kidney, and uterus in female ovariectomized rats. IGFs concentration was measured by radioimmuoassay (RIA) and IGFBPs levels by western ligand blotting(WLB). IGF-I concentration was increased in serum, liver, and uterus, but not in kidney of progesterone-treated ovariectomized rats, compared to control (P<0.05). IGF-II concentration was decreased in liver, but not in serum, kidney, and uterus of progesterone-treated rats, compared to control (P<0.05). IGFBP-3 was increased in serum, but not in liver of progesterone-treated rats, compared to control. IGFBP-2 was decreased in kidney, but not in others tissues of progesterone-treated rats, compared to control. These results suggest that progesterone may exert diverse physiological functions via the tissue-specific regulation of IGFs/IGFBPs system in female rats.

체외수정 후 태반단백들을 이용한 초기임신의 예후 추정 (Prediction of The Course of Early Pregnancy after In Vitro Fertilization by Placental Proteins)

  • Maternal serum ${\beta}$-specific human chorionic gonadotropin(${\beta}$-hCG) and pregnancy-specific ${\beta}_1$-glycoprotein($SP_1$) levels were determined more than one per week during 11-41 days post embryo transfer(ET) in 21 consecutive pregnancies after in vitro fertilization(IVF), which included 8 normal singleton pregnancies, 3 twin pregnancies, 4 clinical abortions, 1 ectopic pregnancy, and 5 preclinical abortions. The sensitivity of serum ${\beta}$-hCG and $SP_1$ radioimmunoassays was 3mIU/ml and 0.7ng/ml relatively. At the 7th to 8th week of gestation, ultrasonographic confirmation of fetal pole and fetal heartbeat was performed. Both serm ${\beta}$-hCG and $SP_1$ levels showed logarithmic increase, but log[$SP_1$] had more steep rising curve and had wider variation than log[${\beta}$-hCG] in normal singleton pregnancies. In 3 twin pregnancies and one ectopic pregnancy, both serum ${\beta}$-hCG and $SP_1$ levels located within the 95% confidence interval of the mean levels of 8 normal singleton pregnancies(normal range). In 2 clinical abortions which had a fetal pole without heartbeat, serum ${\beta}$-hCG level showed lower limit of the normal range or just below, but all $SP_1$ levels showed within the normal range. In other 2 clinical abortions which were diagnosed as blighted ovum, both serum ${\beta}$-hCG levels from 11 days post-ET and serum $SP_1$ levels from later days compared with ${\beta}$-hCG were below the normal range. In 5 preclinical abortions, serum $SP_1$ levels were within the normal range but serum ${\beta}$-hCG levels were far below the normal range. In conclusion, both serum ${\beta}$-hCG and $SP_1$ levels increased exponentially with similar pattern in normally conceived pregancy after IVF-ET. Both serum ${\beta}$-hCG and $SP_1$ levels could predict outcome of early pregnancy to a certain degree, but log[${\beta}$-hCG] levels had more significant correlation with outcome of pregnancy compared with log[$SP_1$] levels. In addition, ultrasonographic examination of fetal poles and fetal heartbeats gives very important clinical information and prognosis.

초기 임신돈에 있어서 자궁분비단백질의 전기영동상과 면역억제력에 관한 연구 (A Study on Electrophoretic Pattern and Immunosuppressive Activity of Uterine Secretory Protein in Early Pregnant Sows)

  • The experiment was carried out to study the profile of uterine specific protein during early pregnancy in sows and to test it's immunosuppressive activity. Uterine protein samples were obtained by flushing the uterine horn on Day 3, 6, 9, 12 and 15 of the estrous cycle and the pregnancy respectively and the protein concentration of each sample was determined. The change of uterine protein was examined by sodium dodecyl sulfate(SDS)-PAGE. The immunosuppressive activity of uterine secretory protein was investigated according to the lymphocyte blastogenesis response to mitogen. The results of this experiment are summarized as follows ; 1. The uterine protein during estrous cycle and early pregnancy was relatively constant up to Day 9, but increased on Day 12. Maxium total protein values were found on Day 15. The concentration of serum proteins were about 82-95 mg during estrous cycle, but decreased to about 70-82 mg during early pregnancy. 2. The proteins components similar electrophoretic patterns(PAGE) that were no differences (band ; a, b, c, d, e, f, g, I) on Days 3, 6 and 9 of the estrous cycle and pregnancy. But there were more 2 bands specifically on Day 12 of the pregnancy and on Day 15 of estrous cycle and showed more 4 bands on Day 15 of early pregnancy. They seemed to be acidoprotein and their average molecular weight were 38,000, 22,300 and 12,600. 3. When uterine protein were added 500$\mu\textrm{g}$/ml, there was no immunosuppresive activity on Day 3 of estrous cycle and lymphocyte blastogenesis was slightly suppressed on Day 3 of pregnancy. The immunosuppressive activity on Day 9 of estrous cycle and pregnancy appeared in 500$\mu\textrm{g}$/ml and 150$\mu\textrm{g}$/ml, respectively the uterine protein on Day 12 and 15 showed immunosuppresive activity, which at the level of 150$\mu\textrm{g}$/ml during non-pregnancy and at the level of 100 to 125$\mu\textrm{g}$/ml during early pregnancy, respectively.

한우의 임신 초기 혈장단백질 발현 양상 분석 (Proteomic Analysis of Plasma Protein during Early Pregnancy in Korean Native Cattle (Hanwoo))

  • This study was performed to identify and analyze the specifically expressed plasma proteins during early pregnancy in both pregnant and non-pregnant Hanwoo. Blood samples were collected at 0 (the day of AI), 2, 3, 4, 7, and 11 weeks after AI from pregnant (n=3) and non-pregnant (n=4) Hanwoo, respectively. The hematological parameters were measured. After 2-dimensional electrophoresis using serum, normalized protein spots were selected for the significant expression variation deviated over two fold in its expression level between two groups. Among 17 spots selected, 15 were identified as albumin, IgG1 heavy chain constant region, haptoglobin, ferrochelatase, fibrinogen, hemopexin. 5 spots were expressed only in non-pregnant specific. The spot identification of 1105 and 6106 was decreased after 3 weeks from AI. However, 2/17 spots were still unidentified. Further studies are needed to analyze the function of the proteins associated with early pregnancy.

非姙娠 및 姙娠한 女子의 血淸蛋白質 패턴의 比較 (The Comparison of Protein Patterns of Sera in Non-Pregnant and Pregnant Women)

  • 사람의 혈액중 혈청으로써 임신의 진행에 따른 단백질의 변화 양상을 조사함에 있어서 정상인 남자, 비임신 여자, 임신한 여자 및 분만 직후 여자의 혈청을 SDS/polyacrylamide gel 전기영동과 2차원 전기영동 및 아미노산분석을 시도하였다. 각 실험 방법에 의해 분석된 결과는 다음과 같다. 1. SDS/polyacrylamide gel 전기영동에 의해 분자량 약 10,000에서 110,000 dalton 사이의 정상인 남자와 비임신 여자를 비교하였을 때의 숫적 양상은 서로 동일하였으나 bands 3 (22,000 dalton)과 6(39,000)등은 남자에 있어서 여자보다 적은 양으로 나타났다. 비임신 여자와 2주 간격으로 혈액을 채취한 임신한 여자의 혈청단백질 패턴을 비교하면 여러 band들의 양적인 증감이 관찰되었다. 임심한 여자의 단백질은 비임신에 비해 band 3(22,000)이 16주까지는 비임신의 경우와 거의 비슷한 양상을 보였으나 18주부터 임신 말기까지는 단백질의 양이 계속하여 감소되었다. 반면 bands 4(24,000), 9(69,000), 10(70,000), 12(80,000), 13(83,000), 14(86,000), 15(91,000) 및 16(94,000)은 임신이 진행됨에 따라 일반적으로 증가되나 그 이후에는 약간씩 감소하였다. 분만 직후의 혈청단백질의 패턴을 임신 말기와 비교하였을 때 bands 12(80,000), 15(91,000) 및 16(94,000)들의 양이 비교적 증가하였고 여아를 분만한 경우 남아를 분만한 경우보다 bands 4(24,000), 7(51,000) 및 10(70,000)들의 단백질 양이 많음이 관찰되었다. 2. 2차원 전기영동으로 남자와 비임신 여자의 혈청단백질 패턴을 비교하면 spot a(22,000)의 3개의 spot가 남자에 있어서는 나타나지 않았고, spotc(39,000)의 군은 남자에 있어 여자보다 농도가 매우 낮았다. 임신한 여자에 있어 albumin이 임신 10주와 12주에 매우 감소하였고 그 이후에 다시 회복되었다. 그리고 spot f(70,000)는 임신 10주에 매우 감소 되었다가 그 이후에 증가되었다. 3. 각 군들의 아미노산을 분석한 결과 임신한 여자에 있어서 전반적으로 glutamic acid, lysine, aspartic acid, leucine 및 valine이 대체로 많은 양으로 나타났고, methionine, isoleucine 및 glycine이 대체로 적은 양을 가지는 것으로 나타났다. 전체 아미노산의 양은 임신중기에 현저히 증가하였다가 말기에는 중기에 비해 감소되었다. 이상의 결과를 볼 때 임신기간 중에는 모체의 혈청을 구성하는 단백질이 특정한기에 따라 증가하므로 임신한 여자의 혈청단백질을 조사함으로써 임신의 각기에 나타나는 독특한 단백질 양상 변화를 밝혀내는 동시에, 앞으로 좀더 연구함으로써 태아의 성을 단백질 패턴으로 명확히 구별해 낼 수 있을 것으로 생각된다.
