홍색 유황세균 Thiocapsa roseopersicina가 생산하는 수소생산 효소의 정제 : Ammonium sulfate 침전 및 열처리의 효과 (Purification of Hydrogenase from Thiocapsa roseopersicina : Effect of Ammonium Sulfate Precipitation and Heat-Treatment)

  • Effect of $(NH_4)_2SO_4$ precipitation and heat-treatment on hydrogenase which was extracted from the cytoplasmic fraction of the phototrophic purple sulfur bacterium Thiocapsa roseopersicina NCIB 8347 was studied. Crude enzyme extract was prepared by centrifugation($28,000{\times}g$, $400,000{\times}g$) after sonication of cells grown under photosynthetic condition for 96 hrs. Various conditions of $(NH_4)_2SO_4$ precipitation and heat-treatment were examined and the effect of protein concentration was analyzed by SDS-electrophoresis between the treatments. Optimum conditions for $(NH_4)_2SO_4$ precipitation and heat-treatment for evolution hydrogenase activity were 40-60% saturation and $60^{\circ}C$ for 20 min, respectively, which exhibited the specific hydrogenase activity of 0.78 U/mg-protein. Specific hydrogenase activity was decreased to 31.6% when the heat-treatment at $60^{\circ}C$ increased from 20 min to 5 hrs.

GIS와 PRISM을 이용한 고해상도 격자형 강수량 추정 (Estimation of High Resolution Gridded Precipitation Using GIS and PRISM)

  • In this study, in order to estimate high resolution precipitation with monthly time scales, Parameter-elevation Regressions on Independent Slopes Model (PRISM) was modified and configured for Korean precipitation based on elevation, distance, topographic facet, and coastal proximity. Applying this statistical downscaling model to Korean precipitation for 5 years from 2001 to 2005, we have compiled monthly grid data with a horizontal resolution of 5-km and evaluated the model using bias, root mean square error (RMSE), and correlation coefficient between the observed and the estimated. Results show that bias, RMSE, and correlation coefficient of the estimated value have a range from 0.2% to 1.0%, 19.6% (June) to 43.9% (January), and 0.73 to 0.84, respectively, indicating that the modified Korean PRISM (K-PRISM) is reasonably worked by weighting factors, i.e., topographic effect and rain shadow effect.

Effect of reaction temperature and time on the formation of calcite precipitation of recycled concrete aggregate (RCA) for drainage applications

  • Recycled concrete aggregate (RCA) is widely used as a construction material in road construction, concrete structures, embankments, etc. However, it has been reported that calcite (CaCO3) precipitation from RCA can be a cause of clogging when used in drainage applications. An accelerated calcite precipitation (ACP) procedure has been devised to evaluate the long-term geochemical performance of RCA in subsurface drainage systems. While the ACP procedure was useful for the French Drain application, there remained opportunities for improvement. In this study, key factors that control the formation of calcite precipitation were quantitatively evaluated, and the results were used to improve the current prototype ACP method. A laboratory parametric study was carried out by investigating the effects of reaction temperature and time on the formation of calcite precipitation of RCA, with determining an optimum reaction temperature and time which maximizes calcite precipitation. The improved ACP procedure was then applied to RCA samples that were graded for Type I Underdrain application, to compare the calcite precipitation. Two key findings are (1) that calcite precipitation can be maximized with the optimum heating temperature (75℃) and time (17 hours), and (2) the potential for calcite precipitation from RCA is not as significant as for limestone. With the improved ACP procedure, the total amount of calcite precipitation from RCAs within the life cycle of a drain system can be determined when RCAs from different sources are used as pipe backfill materials in a drain system.

융설효과와 한계침투량을 고려한 한강유역의 지하수위와 강우이동평균간의 상관관계 (The Correlation between Groundwater Level and the Moving Average of Precipitation considering Snowmelt Effect and Critical Infiltration in Han River Watershed)

  • 한강 유역의 강우량과 지하수위 관측 자료를 이용하여 강우이동평균을 이용하여 융설 효과와 한계침투량을 고려한 지하수위와의 상관관계를 분석하였다. 한강 유역에 위치한 14개 연구 지점을 분석해본 결과 관측소마다 다른 상관계수 및 지하수위의 변동을 볼 수 있었다. 지하수위의 계절적, 연도별 변동 추이 분석 결과 상관계수가 높게 나오는 지역일수록 계절적 특성을 따르는 지하수위의 변동을 보이는 것으로 나타났다. 상관관계 분석 결과 대체로 높은 상관계수를 얻을 수 있었으며 동일한 관측소의 암반층과 충적층의 상관계수도 큰 차이는 없었지만 대체로 암반층에서 조금 더 높은 값을 보여주었다. 유역평균 일최대침투량인 한계침투량을 고려하여 지하수위와의 상관관계를 분석한 결과 이동평균만을 이용한 상관관계 분석보다 더 큰 상관계수를 가지는 것으로 나왔다. 또한, 다른 유역에 비해, 적설량이 많은 한강 유역이므로 융설 효과를 고려할 때 더 높은 상관계수를 가지는 것으로 분석되었다.

주목 식물세포(Taxus chinensis) 배양 유래 Paclitaxel 정제를 위한 분별침전에서 제타전위 영향 (Effect of Zeta Potential on Fractional Precipitation for the Purification of Paclitaxel from Plant Cell Cultures of Taxus chinensis)

  • 본 연구에서는 주목 식물세포(Taxus chinensis) 배양 유래항암물질 paclitaxel을 정제하기 위하여, 반응액 부피당 표면적이 증가된 분별침전에서 표면적증가물질 실리카-알루미나의 제타전위가 분별침전 양상(순도, 수율, 침전시간, 침전물형태와 크기)에 미치는 영향을 조사하였다. 실리카-알루미나의 제타전위가 양(+)의 값으로 증가할수록 paclitaxel 수율은 증가하였으며 침전 시간은 감소하였다. 표면적증가물질로 제타전위가 가장 큰 알루미나(제타전위: +35.41 mV)를 이용한 경우 표면적증가물질을 이용하지 않은 경우에 비해 침전에 소요되는 시간을 12시간 정도 단축시킬 수 있었다. 반면 표면적증가물질 실리카-알루미나의 전타전위 변화에도 paclitaxel 순도는 거의 변화가 없었다. 또한 분별침전 과정에서 제타전위 절대값이 증가할수록 paclitaxel 침전물의 크기는 감소하여 제타전위 절대값에 반비례함을 알 수 있었다. 이러한 개선된 분별침전 공정은 바이오매스 유래 paclitaxel 정제 공정에 효과적으로 기여할 것으로 판단된다.

이상기후 (엘니뇨, 라니냐) 기간의 유역별 건조지수, 강수효율, 유출량의 영향성 평가 (Evaluation the Climatic Influence during El Nino and La Nina Periods of Aridity Index, Precipitation Effectiveness and Runoff in Basins)

  • The comparison between the spatial and temporal variability of aridity index, precipitation effectiveness and runoff during El-Nino and La-Nina periods and that of the normal period was conducted to evaluate the regional impacts of El-Nino, La-Nina in hydrologic variables. Aridity index and precipitation effectiveness were estimated using 59 nationwide weather stations data and runoff data of WAMIS were used. The ratio of the difference between El-Nino, La-Nina year value and that of normal year was analyzed. Temporal variation demonstrated that aridity index, precipitation effectiveness and run-off discharge increase in March, April, August, November, December and decrease in February, June, September, October according to El-Nino effect. Aridity index, precipitation effectiveness and run-off discharge increase in March, May, September and decrease in June, August, November, December according to La-Nina effect. The spatial variation of those variables analyzed for different basins showed that impacts in the Han river basin relatively higher than that of other basins.

급냉응고 및 열간가공된 Al-Zr계 합금의 석출거동 (Precipitation Behaviors of Rapidly Solidified and Hot Worked Al-Zr Base Alloys)

  • Rapidly Solidified (RS) Al-Zr base splats with various alloy contents were prepared by atomization-splat quenching method to understand the continuous and discontinuous precipitation in the aged alloys. And the RS alloys were consolidated by hot extrusion and swaging to analyze the effect of plastic deformation on the precipitation behavior. Discontinuous precipitation dominated at relatively low temperature in the Al-Zr alloy, whereas both V additions to Al-Zr alloys and hot metal working appeared to suppress the discontinuous precipitation. As continuous precipitation is favored in the grain interiors, the driving force for discontinuous precipitation become to disappear with a further process.

미량원소첨가강의 석출 및 재결정에 의한 제어변형 (Controlled Deformation of Microalloyed Steel by Precipitation and Recrystallization)

  • The multistage deformation and stress relaxation were carried out to investigate the strain induced precipitation by torsion tests in the range of 1000~80$0^{\circ}C$, 0.05~5/sec for V-microalloyed steel. The starting temperature and time for the initiation of precipitation were determined by stress relaxation tests. The distribution of precipitates increased, as the strain rate increased and the mean size of precipitates was found to be about 10~30nm. The precipitation starting time$(P_s)$ decreased with increasing strain rate and the amount of pre-strain. The effect of deformation conditions on the no-recrystallization temperature$(T_nr)$ was also determined in the multistage deformation. $T_nr$ Tnr decreased with increasing the strain and strain rate. In the controlled rolling simulation, grain refinement and precipitation hardening effects could be achieved by the alternative large pass strain at the latter half pass stage under the condition of low temperature and high strain rate.

지하수위 변동량과 전기전도도 변동량을 이용한 강수 효과 분석 (Analysis of Precipitation Effects Using Groundwater Level and Electrical Conductivity Fluctuations)

  • Moving average precipitation provides periodic precipitation patterns by solving precipitation irregularities. However, due to uncertain moving average periods, excessive data smoothing occurs, which limit the possibility to analyze groundwater levels in the short term. Nonetheless, groundwater level fluctuation can compensate these limitations as it can calculate appropriately for unit time and verify the effect of precipitation penetrated into groundwater in a short time period. In this study, the characteristics of groundwater level were evaluated using groundwater level fluctuation to compensate for limitations of groundwater level analysis using moving average precipitation. In addition, the groundwater quality was investigated using the electrical conductivity fluctuation. The study site was Hyogyo-ri, Yesan-si, Chungcheongnam-do. Four observation wells and an automated weather system were used. The correlation between groundwater level fluctuation and precipitation (Case 1) and the correlation between groundwater level and moving average precipitation (Case 3) were compared. In the analysis for 1 hour data, the correlation coefficient of Case 1 was higher than that of Case 3, and in the analysis for 1 day data, the correlation coefficient of Case 3 was higher than that of Case 1.

단지내 차도용 투수성 포장의 물순환 효과분석 (An Analysis on the Hydrologic Cycle Effect of Rodway Permeable Pavement in Rasidential Site)

  • In recent years, increases in impervious areas with rapid urbanization and land use changes are causing numerous hydrologic cycle and environmental problems. Impermeable pavement have a various defect such as collection rainwater, decreasing of sliding resistance, and etc. In this study, the hydrologic cycle effect of permeable pavement were analyzed by the experiment and the numerical simulation. The numerical model used was a modified SWMM especially for considering the hydrologic cycle effect of permeable pavement. The parameters of modified SWMM were revised by the experimental results. Also, the effects of runoff quantity reduction are reviewed when permeable pavement is applied to Incheon Cheongna watershed. The hydrologic cycle analysis of Incheon Cheongna watershed, continuous simulations of urban runoff were performed. The analysis results of permeable pavement setup effect on runoff are follows: the surface runoff after permeable pavement setup decreases to 74.35% of the precipitation whereas the surface runoff before permeable pavement setup amounts to 81.38% of the precipitation; the infiltration after permeable pavement setup increases to 15.13% of the precipitation whereas the infiltration before permeable pavement setup amounts to 8.32% of the precipitation.
