The goal of this research was (1) to describe the conditions and parameters required for the cell cycle synchronization and the accumulation of large number of metaphase cells in maize and other cereal root tips, (2) to isolate intact metaphase chromosomes from root tips suitable for characterization by flow cytometry, and (3) to construct chromosome-specific libraries from maize. Plant metaphase chromosomes have been successfully synchronized and isolated from many cereal root-tips. DNA synthesis inhibitor (hydroxyurea) was used to synchronize cell cycle, follwed by treatement with trifluralin to accumulate metaphase chromosomes. Maize flow karyotypes show substantial variation among inbred lines. thish variation should be sueful in isolating individual chromosome types. In addition, flow cytometry is a useful method to measure DNA content of individual chromosomes in a genotyps, and to detect chromosomal variations. Individual chromosome peaks have been sorted from the maize hybrid B73/Mol7. Libraries were generated form the DOP-PCR amplification product from each peak. To date, we have analyzed clones from a library constructed from the maize chromosome 1 peak. Hybridization of labeled genomic DNA to clone inserts indicated that $24\%,\;18\%,\;and\;58\%$ of the clones were highly repetitive, medium repetitive, and low copy, respectively. Fifty percent of putative low cpoy clones showed single bands on inbred screening, blots, and the remaining $50\%$ were low copy repeats. Single copy clones showing polymorphism will be mapped using recombinant inbred mapping populations. Repetitive clones are being characterized by Southern blot analysis, and will be screened by in situ hybridization for their potential utility as chromosome specific markers.
Identification of fusion gene is of prominent importance in cancer research field because of their potential as carcinogenic drivers. RNA sequencing (RNA-Seq) data have been the most useful source for identification of fusion transcripts. Although a number of algorithms have been developed thus far, most programs produce too many false-positives, thus making experimental confirmation almost impossible. We still lack a reliable program that achieves high precision with reasonable recall rate. Here, we present FusionScan, a highly optimized tool for predicting fusion transcripts from RNA-Seq data. We specifically search for split reads composed of intact exons at the fusion boundaries. Using 269 known fusion cases as the reference, we have implemented various mapping and filtering strategies to remove false-positives without discarding genuine fusions. In the performance test using three cell line datasets with validated fusion cases (NCI-H660, K562, and MCF-7), FusionScan outperformed other existing programs by a considerable margin, achieving the precision and recall rates of 60% and 79%, respectively. Simulation test also demonstrated that FusionScan recovered most of true positives without producing an overwhelming number of false-positives regardless of sequencing depth and read length. The computation time was comparable to other leading tools. We also provide several curative means to help users investigate the details of fusion candidates easily. We believe that FusionScan would be a reliable, efficient and convenient program for detecting fusion transcripts that meet the requirements in the clinical and experimental community. The gold (LC-AuNPs) and silver (LC-AgNPs) nanoparticles were rapidly synthesized by fruit extract of Lycium chinense within 1.15 and 25 min respectively in an eco-friendly way. The synthesized nanoparticles confirmed by relevant surface plasmon resonance peaks for gold and silver nanoparticles at 536 and 480 nm, respectively. FE-TEM results revealed that LC-AuNPs were 20-50 nm and LC-AgNPs were 50-100 nm. The maximum distribution of gold, silver elements and the crystallographic nature of synthesized were confirmed using EDX, elemental mapping and XRD. LC-AgNPs showed inhibitory activity against pathogenic microorganisms such as E. coli and S. aureus, whereas LC-AuNPs did not show inhibitory activity. The LC-AgNps nanoparticles exhibited significant cytotoxicity to human breast cancer MCF7 cell line and less cytotoxicity to non-diseased RAW264.7 (murine macrophage) cells whereas LC-AuNps showed minimal toxicity to both cell lines. In-depth research on this rapid, facile and greenery nanoparticles may play a potential role in biomedical applications.

In this paper, we propose colored-hear recognition that distinguishing feature of synesthesia for human sensing by shared vision and specific sense of hearing. We perceived what potential influence of human's structured object recognition by visual analysis through the camera, So we've studied how to make blind persons can feel similar vision of real object. First of all, object boundaries are detected in the image data representing a specific scene. Then, four specific features such as object location in the image focus, feeling of average color, distance information of each object, and object area are extracted from picture. Finally, mapping these features to the audition factors. The audition factors are used to recognize vision for blind persons. Proposed colored-hear transformation for recognition can get fast and detail perception, and can be transmit information for sense at the same time. Thus, we were get a food result when applied this concepts to blind person's case of image recognition.

The self-splicing group I intron from Tetrahymena thermophila has been demonstrated to perform splicing reaction with its substrate RNA in the trans configuration. In this study, we explored the potential use of the trans-splicing group I ribozymes to replace a specific RNA with a new RNA that exerts any new function we want to introduce. We have chosen thymidine phosphorylase (TP) RNA as a target RNA that is known as a valid cancer prognostic factor. Cancer-specific expression of TP RNA was first evaluated with RT-PCR analysis of RNA from patients with gastric cancer. We determined next which regions of the TP RNA are accessible to ribozymes by employing an RNA mapping strategy, and found that the leader sequences upstream of the AUG start codon appeared to be particularly accessible. A specific ribozyme recognizing the most accessible sequence in the TP RNA with firefly luciferase transcript as a 3' exon was then developed. The specific trans-splicing ribozyme transferred an intended 3' exon tag sequence onto the targeted TP transcripts, resulting in a more than two fold induction of the reporter activity in the presence of TP RNA in mammalian cells, compared to the absence of the target RNA. These results suggest that the Tetrahymena ribozyme can be a potent anti-cancer agent to modify TP RNAs in tumors with a new RNA harboring anti-cancer activity.
Genome-wide association studies (GWASs) have greatly contributed to the identification of common variants responsible for numerous complex traits. There are, however, unavoidable limitations in detecting causal and/or rare variants for traits in this approach, which depends on an LD-based tagging SNP microarray chip. In an effort to detect potential casual and/or rare variants for complex traits, such as type 2 diabetes (T2D) and triglycerides (TGs), we conducted a targeted resequencing of loci identified by the Korea Association REsource (KARE) GWAS. The target regions for resequencing comprised whole exons, exon-intron boundaries, and regulatory regions of genes that appeared within 1 Mb of the GWA signal boundary. From 124 individuals selected in population-based cohorts, a total of 0.7 Mb target regions were captured by the NimbleGen sequence capture 385K array. Subsequent sequencing, carried out by the Roche 454 Genome Sequencer FLX, generated about 110,000 sequence reads per individual. Mapping of sequence reads to the human reference genome was performed using the SSAHA2 program. An average of 62.2% of total reads was mapped to targets with an average 22X-fold coverage. A total of 5,983 SNPs (average 846 SNPs per individual) were called and annotated by GATK software, with 96.5% accuracy that was estimated by comparison with Affymetrix 5.0 genotyped data in identical individuals. About 51% of total SNPs were singletons that can be considered possible rare variants in the population. Among SNPs that appeared in exons, which occupies about 20% of total SNPs, 304 nonsynonymous singletons were tested with Polyphen to predict the protein damage caused by mutation. In total, we were able to detect 9 and 6 potentially functional rare SNPs for T2D and triglycerides, respectively, evoking a further step of replication genotyping in independent populations to prove their bona fide relevance to traits.
Satellite and aerial remote sensing (RS) techniques have been provided to collect spatial data globally over the last few decades. However in developing countries such as China, there is still an urgent need for low cost and high resolution RS data. As an emerging RS platform, commercial Unmanned Aerial Vehicle (UAV) integrated with state-of-the-art sensors and information technologies has the potential to become a low cost tool to meet application demands. In this paper, the architecture of UAV RS operation system is mentioned. Moreover, key technologies in UAV RS system are analyzed and current work is reported.
Evapotranspiration is a key element in designing and operating agricultural hydraulic structures. The profound effect of climate change to local agro-hydrological systems makes it inevitable to study the potential variability in evapotranspiration rate in order to develop policies on future agricultural water management as well as to evaluate changes in agricultural environment. The APEX-Paddy model was used to simulate local evapotranspiration responses to climate change scenarios. Nine Global Climate Models(GCMs) downscaled using a non-parametric quantile mapping method and a Multi?Model Ensemble method(MME) were used for an uncertainty analysis in the climate scenarios. Results indicate that APEX-Paddy and the downscaled 9 GCMs reproduce evapotranspiration accurately for historical period(1976~2005). For future periods, simulated evapotranspiration rate under the RCP 4.5 scenario showed increasing trends by -1.31%, 2.21% and 4.32% for 2025s(2011~2040), 2055s(2041~2070) and 2085s(2071~2100), respectively, compared with historical(441.6 mm). Similar trends were found under the RCP 8.5 scenario with the rates of increase by 0.00%, 4.67%, and 7.41% for the near?term, mid?term, and long?term periods. Monthly evapotranspiration was predicted to be the highest in August, July was the month having a strong upward trend while. September and October were the months showing downward trends in evapotranspiration are mainly resulted from the shortening of the growth period of paddy rice due to temperature increase and stomatal closer as ambient $CO_2$ concentration increases in the future.
For preventing the accidents generated from the chemical materials, thus far, MSDS (Material Safety Data Sheet) data have been made to notify how to use and manage the hazardous and chemical materials in safety. However, it is difficult for users who handle these materials to understand the MSDS data because they are only listed based on the alphabetical order, not based on the specific factors such as similarity of characteristics. It is limited in representing the types of chemical materials with respect to their characteristics. Thus, in this study, a lots of MSDS data are visualized based on relationships of the characteristics among the chemical materials for supporting safety managers. For this, we used the textmining algorithm which extracts text keywords contained in documents and the Self-Organizing Map (SOM) algorithm which visually addresses textual data information. In the case of Occupational Safety and Health Administration (OSHA) in the United States, the guide texts contained in MSDS documents, which include use information such as reactivity and potential risks of materials, are gathered as the target data. First, using the textmining algorithm, the information of chemicals is extracted from these guide texts. Next, the MSDS map is developed using SOM in terms of similarity of text information of chemical materials. The MSDS map is helpful for effectively classifying chemical materials by mapping prohibited and hazardous substances on the developed the SOM map. As a result, using the MSDS map, it is easy for safety managers to detect prohibited and hazardous substances with respect to the Industrial Safety and Health Act standards.

This paper introduces the system for KAU-SPUAV, which is designed and developed by Korea Aerospace University, and verifies its performance through flight test. Specification of two versions of KAU-SPUAV, avionics system, and Ground Control System (GCS) are introduced. Three missions are performed with suggested UAVs: LTE signal mapping, circumnavigation of Jeju island seashore, and long endurance flight. Each mission consists of long distance and long endurance flight which takes advantage of KAU-SPUAV. Research team of KAU-SPUAV confirmed its versatility through suggested flight data. Also based on flight results, the team found the potential of performance improvement of KAU-SPUAV.
