Identification of genes related to ER stress in bZIP28 gene transgenic potato plant

  • Potato (Solanum tubersosum L.) is susceptible to various environmental stresses such as frost, high temperature, and drought. Enhancement of potato drought tolerance can reduce yield loss under drought that has negative effect on potato tuber growth. Genetic engineering can be utilized to achieve this goal, but such approaches using endogenous potato genes have rarely been applied. Since unpredictable global weather changes cause more severe and frequent water limiting conditions, improvement of potato drought tolerance can minimize such adverse effects under drought and can impact on sustainable potato production. Genetic engineering can be utilized to improve potato drought tolerance, but such approaches using endogenous potato genes have rarely been applied. We were obtained AtbZIP28 gene transgenic potato plants. It is identified transcript levels at various stress conditions, polyethylene glycol (PEG), NaCl, abscisic ${\underline{acid}}$ (ABA). Also, For identification to regulate ER stress response genes in AtbZIP28 gene transgenic potato plant, we screened seven potato genes from RNA-seq analysis under TM treatment. Five and two genes were up- and down-regulated by TM, respectively. Their expression patterns were re-examined at stress agents known to elicit TM, DTT, DMSO and salt stress.

Changes in Growing Period and Productivity under Double Cropping of Spring Potato and Summer Cereals in Paddy Fields of Southern Korea (남부지역 논에서 봄감자와 하작물 이모작에 따른 생육기간 및 생산성 변화)

  • Changes in growing periods and productivities of crops under double cropping of potato-rice, potato-soybean and potato-maize, were investigated at the Paddy Experimental Fields in Miryang City from 2015 to 2018. Spring potatoes planted in early March showed a yield of 2.1-2.3 ton/10a and a period of 90 days. In double cropping, growing period of rice, soybean, and maize was about 130, 125 and 115 days, respectively. The potato yield obtained was as much as 616, 330 and 815 kg/10a under double cropping with rice, soybean and maize, respectively. It is beneficial to sow the spring potatoes as early as possible to increase the yield and to secure the growing period of sequential crops. The introduction of summer medium-late variety grain crops into double cropping of spring potato and rice as well as into double cropping of spring potato and soybean/maize, was possible because of no sowing in the fall and plants were able to reach the heading growth stage before the safe heading limit of rice in particular. In the case of maize, the growth period was different according to the change in temperature over the year. The introduction of upland crops such as soybeans and maize instead of rice improved soil physicochemical properties in a short period of time, contributing to the increase of spring potato yields, but there was also a risk of damage by successive cropping for more than three years. Spring potato-maize showed higher yield in terms of starch production, and spring potato-soybean was found to be advantageous for net income.

Estimating the Yield of Marketable Potato of Mulch Culture using Climatic Elements (시기별 기상값 활용 피복재배 감자 상서수량 예측)

  • The object of this study was to evaluate the effects of climatic elements on potato yield and create a model for estimating the potato yield. We used 35 yield data of Sumi variety produced in mulching cultivation from 17 regions over 11 years. According to the results, some climatic elements showed significant level of correlation coefficient with marketable yield of potato. Totally 22 items of climatic elements appeared to be significant. Especially precipitation for 20 days after planting (Prec_1 & 2), relative humidity during 11~20 days after planting (RH_2), precipitation for 20 days before harvest (Prec_9 & 10), sunshine hours during 50~41 days before harvest (SH_6) and 20 days before harvest (SH_9 & 10), and days of rain during 10 days before harvest (DR_10) were highly significant in quadratic regression analysis. 22 items of predicted yield ($Y_i=aX_i{^2}+bX_i+c$) were induced from the 22 items of climatic elements (step 1). The correlations between the predicted yields and marketable yield were stepwised using SPSS, statistical program, and we selected a model (step 2), in which 4 items of independent variables ($Y_i$) were used. Subsequently the $Y_i$ were replaced with the equation in step 1, $aX_i{^2}+bX_i+c$. Nutrient Uptake and Productivity as Affected by Nitrogen and Potassium Application Levels in Maize/Sweet Potato Intercropping System

  • Field experiment was conducted during 1993-94 season to determine the pattern of nutrient uptake and productivity of maize/sweet potato intercropping system. Four levels of nitrogen (0, 50, 100 and 150kg N ${ha}_{-1}$) and four levels of potassium (0, 40, 80 and 120kg $K_2$O ${ha}_{-1}$) formed treatment variables. Plants were sampled periodically to determine dry matter and tissue concentrations of N and K in the individual plant components of intercropped maize and sweet potato. Nitrogen and potassium fertilizer did not interact significantly to nutrient uptake by any plant parts of intercropped maize and sweet potato. But application of N fertilizer independently enhanced N uptake in all the plant parts of maize and sweet potato. The uptake of N in leaf, leaf sheath, stem, husk, and cob of maize increased upto 90 days after planting (DAP) but grain continued to accumulate N till its maturity. Sweet potato exhibited a wide variation in N uptake pattern. Sweet potato leaf shared the maximum uptake of N at 50 DAP which rapidly increased at 70 DAP and then declined. Declination of N uptake by petiole and stem were observed after 120 DAP whereas N uptake by tuber increased slowly upto 90 DAP and then rapidly till harvest. Rate of applied K had very little effect on the uptake patterns in different components of intercropped maize. Pattern of K uptake by leaf, petiole and stem of sweet potato showed almost similar trend to N uptake. But uptake of K by tuber increased almost linearly with the K application. Pattern of N and K uptake by grain and tuber paralleled the grain yield of maize and sweet potato respectively. Intercropped productivity of maize and sweet potato found to be better by the application of 100kg N and 120 kg $K_2$O ${ha}_{-1}$

Effect of Low Dose ${\gamma}$ Radiation on the Dormancy and Growth of in vitro Microtuvers of Potato(Solanum Tuberosum L.) (저선량 ${\gamma}$선이 기냐 생산된 감자 소괴경의 휴면과 생에 미치는 효과)

  • To observe the stimulating effect of low dose ${\gamma}$ radiation on the dormancy breaking and growth, microtubers induced in vitro of two potato (Solanum tuberosum L. cv. Dejima and cv. Superior) cultivars with different storage duration were irradiated with ${\gamma}$ radiation at the dose of $0.5{\sim}30$ Gy. Sprouting rate, growth and tuber yield of ‘Dejima’ microtuber were increased by ${\gamma}$ radiation in the range of $2{\sim}16$ Gy. In the microtuber of ‘Superior’, the sprouting rate was promoted by 2 Gy and 4 Gy irradiation, and the growth and tuber yield by 0.5 Gy and 4 Gy irradiation. There were not that much difference in chlorophyll content of potato plantlet by the low dose ${\gamma}$ irradiation. These results suggested that low dose ${\gamma}$ radiation stimulated the dormancy breaking and potato growth.

Plant growth promoting rhizobacteria influence potato tuberization through enhancing lipoxygenase activity

  • Molecular insights on the role of plant growth promoting rhizobacteria (PGPR) in potato tuberization are reported in the present study. The PGPRwere isolated from the soil collected from potato fields of Highland Agricultural Research Centre, Pyeongchang, Korea and they were identified to the genus level based on the 16S rRNA sequence analysis. These PGPR were heat-killed, filtered and the filtrates were addedindividually at a concentration of $10^7\;cfu\;mL^{-1}$ in MS (Murashige and Skoog's) medium supplemented with 7% (w/v) sucrose to study their influence on in vitro potato tuberization. Tuber initiation occurred early in untreated control, while tuber growth was pronounced in case of PGPR treatments. The control explants showed tuber formation as a result of sub-apical swelling of stolons while several sessile tubers formed directly in the axils of nodal cuttings in case of PGPR treatments, which is an indication of strong induction for tuberization. Theexplants cultured on MS medium supplemented with bacterial isolate 6 (Bacillus firmus strain 40) showed highest average tuber yield (Ca. 12.56 g per treatment) after 30 days of culture, which was 3 folds increase over the untreated control. A significant increase in lipoxygenase (LOX1) mRNA expression and activity of LOX enzyme were also detected in the tubers induced on PGPR treatments as compared to untreated control. This LOX expression level correlated with increased tuber growth and tuber yield. Further studies focused on the role of bacteria cell wall components, growth regulators and signal molecules released by PGPR are under investigation to elicit clues for PGPR-mediated signal pathway controlling potato tuberization.

Effect of Root Media Formulation and Fertilizer Application on Potato Plug Seedling Growth and Field Performance

  • Eight vermiculite-based root media prepared with addition of complete fertilizer (2 g/L; N-$P_2$$O_5$-$K_2$O, 10-10-14) for potatoes (Solanum tuberosum L.) and a commercial root medium were evaluated in 2000 to develop the root media suitable for potato plug seedling production. The eight media consisted of various ratios of vermiculite, perlite, peatmoss, and compost. In addition, four rates (0, 1, 2, or 4 g/L) of the complex fertilizer for potato were added to a root medium (70% vermiculite, 10% perlite, 10% peat moss, and 10% compost by volume) to determine the optimum addition rate of the complex fertilizer for plug seedlings. Compost addition to the media increased plant height, the number of leaves per plant, and top and root fresh weight of 15-day old plug seedlings. The seedlings raised in root media containing compost produced significantly higher total tuber yield. Addition of the complex fertilizer to root media enhanced seedling growth and increased the number of tubers per plant and tuber yields. The results suggest that root media containing 50% vermiculite, 0 to 20% peat moss, 10% perlite, 20 to 40% compost, and 2 g/L complex fertilizer for potato appear suitable for potato plug seedling production.

Quality Characteristics of Bread with Various Concentrations of Purple Sweet Potato (자색고구마 첨가 식빵의 품질특성)

  • The purpose of this study was to investigate the physicochemical and sensory properties of various breads prepared with purple-fleshed sweet potatoes and their powders. As the amount of purple-fleshed sweet potato and its powder increased, pH of the dough and bread, volume, dough yield, baking loss rate, and fermentation power of the dough decreased, whereas acidity of the bread increased. The lightness and yellowness of the breads decreased significantly, but redness increased with increasing content of purple-fleshed sweet potato and its powder. The textural properties, such as hardness, chewiness, springiness, and brittleness were significantly higher in groups with added purple-fleshed sweet potato and its powder compared to those in the control. The anthocyanin content and DPPH radical scavenging of dough and bread were higher in groups with purple-fleshed sweet potato and its powder than those in the control group. The groups with 5% and 10% purple-fleshed sweet potatoes and their powders had significantly higher scores for overall acceptability as compared to the control group.

Gelatinization Properties of Heat-Moisture Treated Potato and Sweetpotato Starches (수분 열처리한 감자 및 고구마전분의 호화 특성)

  • Gelatinization properties of heat-mositure treated potato and sweet potato starches were investigated. Water-binding capacity of starch was increased by heat-mositure treatment, which was more pronounced in sweet potato starch. Blue value was not affected by the treatment. Amylograph viscosities were decreased by heat-mositure treatment, which was more pronounced in potato starch. Critical concentration of NaOH for gelatinization of starch increased as moisture level increased. Gel volume of starch upon KSCN gelatinization was higher in potato starch. Gelatinized starches showed Binghamapseudoplastic behavior. Consistency index and yield stress were drastically decreased upon heat-moisture treatment.

Influence of Polyethylene Film Mulch and Seedling Types on Growth and tuber Yield of Fall-Grown Potato (폴리에틸렌필름 멀칭 및 종묘종류가 가을감자의 생육과 수량에 미치는 영향)

  • Normal seed tubers and plug seedlings raised from stem cutting and mini-tubers ($10\pm3$ g) were planted or transplanted on August 20,2001 in bare soil or under transparent and black polyethylene film mulches to evaluate the effects of mulch material and seedling type on the growth and tuber yield of fall-grown potato (Solanum tuberosum L. cv. Dejima) in Jeju region. Total tuber number per plant was 4.6 regardless of mulch treatment, and direct planting seed potatoes resulted in the greatest tubers (5.14)/plant and followed by transplanting seedlings raised from cutting and mini-tubers (4.15 and 5.15 tubers/plant). Transparent and black polyethylene film mulches increased total tuber yield by 16 and 8%, respectively, and average tuber weight by 14 and 12%, respectively, compared with no mulch (21.16 t/ha and 72.2 g/tuber). Transplanting plug seedlings raised from cutting resulted in the greatest tuber yield, followed by transplanting plug seedlings raised from mini-tubers and direct planting seed potatoes.
