Influence of different cultivated areas on the processing-grade tuber yield, specific gravity, reducing sugar content and tuber qualities of five promising varieties was studied to get the basic information for selecting potato varieties with good processing quality under the different cultivated conditions on Korea. The average total tuber yields of 5 tested varieties at the 2nd harvest time was 3,.051kg/10a in Daekwallyoung. The processing-grade tuber yield of the late-maturing variety of Gemchip was over 3.2 tons per 10a, whereas that of late-maturing variety, NS1 was 2.8 tons per 10a. The dry matter content of the tubers produced from Daekwallyoung was the highest, followed by the order of Bosung, atlantic was the highest in dry matter content, followed by the order of NS1, NS2, Superior and Gemchip. Degrees of change in glucose content as affected by change of cultivated areas were different among varieties. Atlantic and NS1 showed less change of glucose content as compared with other varieties.
This study was conducted to dentify the optimum plantlet type of 'Solara' potatoes (Solanum tuberosum L.) for growth in an aeroponics system. Plantlets of 'Solara' were transplanted on March 16, 2015 in a greenhouse, and growth and yield characteristics were investigated at 70 and 78 days after transplanting, respectively. Stem length was shorter in plantlet of 15-day-old stem cuttings and acclimatization of culture, and the stem length of plantlets of stem cuttings tended to increase with increasing stem cutting age. The fresh weight of plants was the highest in the plantlets of 40-day-old stem cuttings and the lowest in non-rooted stem cuttings and acclimatization of culture. The highest number of first stolons was obtained in 35-day-old stem cuttings. The number of second stolons was the highest in plantlets of 35-day-old stem cuttings, acclimatization of culture, and 30- day-old stem cuttings. The total number of tubers was higher in plantlets of 35-day-old stem cuttings and acclimatization of culture, and the number of tubers above 3 g was the highest in plantlets of 35-day-old stem cuttings. The weight of tubers above 3 g was the heaviest in plantlets of 35-day-old stem cuttings(1,947 g per 10 plants), followed by plantlets of 30-day-old stem cuttings. These results indicate that plantlets of 30 to 35-day-old stem cuttings could be the best for production of 'Solara' potato tubers in an aeroponics system.
Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

Althow underdrainage has been studied for long time, it is the first attempt in Korea to execute using PVC(Plastic) suction pipes in the low and wet field. First, an execution plot and a control plot were set, and the drainage method and soil temprature in the excuted plot have been examined. The growth of crops and the yeild, the improvement of soil and water quality of irrigation are to be dealt during the next experimental period. The experimental method and the results obtained through the experimentations are as follows: Method 1) Depth: 1meter. interval: 5meters Trench was performed by labor. 2) PVC(plastic) sucking pipe filters were wound with glass nylon. 3) Two. horizontal looks were set in the 5a. plot. Results 1) The soil temprature in the excuted plot went up by $1.2^{\circ}C$ in average than in the control plot during the two years(1966-67) of irrigation period, and the maximum temprature raised a day was $3^{\circ}C$ 2) The under ground water level in the executed plot went down by 45cm. 3) The yield increases were 64% in potato, 57% in barley, and 21% in rice. The yield, soil, and the quality of irrigated water will be experimented during the next experemental period.
A new cultivar of oak mushroom Lentinula edodes 'Jadam' was bred from monokaryotic strains of 'Hwadam-18' and 'GMLE36295-22'. The optimum temperature for mycelial growth of 'Jadam' on potato dextrose agar was 22-25℃. Total cultivation period of the new cultivar, from inoculation to its first harvest, was 122-124 days, shorter to that of the control cultivar 'Hwadam'. Total yield of 'Jadam' was 623.8 g per 3 kg substrate, and is higher than that of 'Hwadam' (455.2 g). In the fruiting body of the new cultivar, the stipe was shorter and thinner than those of 'Hwadam'. A farmhouse field test showed that the period of mycelial growth and browning was shorter than that of control cultivar 'L808'. The total yield for 2 flush was 543.3 g for 'Jadam'. It was similar to the 585.3 g of the control cultivar. Therefore, the new cultivar 'Jadam' could be a substitute for 'L808' in the field of farms.
Juice were extracted from fresh leaves of 70 species of plants growing in Korea by mincing and pressing the resulting pulp through a cotton cloth. Leaf protein concentrates could be prepared from many species of land and water plants that are at present economically unimportant. The choice of plants is of considerable important. Total-N, protein-N and pH determinations were made on the extracts, and total-N remaining in the fibre were calculated. Leaf protein concentrates were precipitated from the extracts at $75{\sim}80^{\circ}C$, and analysed total-N as protein-N of products. The present paper deals with the calculated yields of leaf protein concentrates from various plants, relations between yield of leaf protein concentrates and total-N of leaves, or pH of extracts, and the amino acid compositions of leaf protein concentrates. Results are summarized as follows. 1. Spinach and radish were the best sources of easily extractable, but good results were also obtained with indian mustard, kail, chenopod, red bean, cucumber, squash, houndberry, white flowered gourd, potato, Humulus japonicus, arrowroot and soybean as a good resources for the production of leaf protein concentrates. 2. In general, the greater the protein content of leaves the greater the yield of leaf protein concentrates. However, there are some plants difficult to make a adequate protein extraction by a simple mechanical process. 3. It was to be expected that leaf protein concentrates would be more extractable with the higher pH of extracts. There were a poor yield of the leaf protein concentrate in the pH values lower than 5.50 of the first extracts. 4. Protein content of the leaf protein concentrate shows marked differences, depending on species and season. It ranged between 29 to 80% of protein contents. However, the majority of plants yielded products containing more than 50% of protein. Products containing more than 75% of protein were obtained from two species of radish and indian mustard. Cabbage and Digitaria sanguinalis cilialis (summer) made products containing 29 to 32% of protein. 5. The amino acid composition of leaf protein concentrates was not greatly altered by species of plants. On an amino acid compositional basis, the leaf protein concentrate has a favorable balance of essential and non-essential amino acids, the only exception being methionine, which was usually low in all cases.

This experiment was conducted to solve the failure of fruiting body production in the bottle cultivation of the oyster mushroom cultivar 'Heuktari'. The effects of incubation temperature on primordium formation and fruiting body yield of the oyster mushroom cultivar 'Heuktari' were investigated. The proper temperature for mycelium growth of 'Heuktari' on potato dextrose agar (PDA) medium is 23-26℃. The mycelial growth of 'Heuktari' was faster than that of Chunchu 2ho. During mycelial culture in sawdust medium, the temperature of the medium in the bottle initially increased, reached the highest point in the middle of the culture, and then decreased. The higher the set temperature, the shorter the incubation period. When the incubation temperatures were 20℃ and 24℃, respectively, the undeveloped primordium formation rates were low (1.8% and 4.2%, respectively). However, the rate of undeveloped primordium formation increased, and the yield decreased at incubation temperatures of 16℃ and 28℃. Mushroom farms that set incubation temperatures to 18℃ and maintained the medium temperature at less than 28℃ showed undeveloped primordium formation rates ranging between 0.3-0.8%. The rate of undeveloped primordium formation increased and the yield decreased in the farms with high incubation temperatures (above 28℃). We found that in order to reduce undeveloped primordium formation, the air inside the incubation room should be circulated continuously so that the temperature of the medium does not rise above 28℃, and dense incubation conditions should be avoided.
Cyclodextrin glucanotransferase (CGTase) was purified from the culture broth of the Bacillus firmus var. alkalophilus, using ultrafiltration, starch adsorption/desorption, ion-exchange chromatography on DEAE-cellulose and gel filtration on Sephacryl HR-100. The molecular weight of the purified enzyme was determined as 77,000 by SDS-PAGE. The optimum pH and temperature for the CD synthesis were 6.0 and 5$0^{\circ}C$, respectively. The activity of this enzyme was stably kept at the range of pH 6.0~9.5 and up to 5$0^{\circ}C$. However, in the presence of $Ca^{2+}$, the optimum temperature for CD synthesis was shifted 55~6$0^{\circ}C$ and this enzyme was stable up to 6$0^{\circ}C$ because of the stabilizing effect of $Ca^{2+}$. The purified CGTase produced CDs with high conversion yields of 45~51% from sweet potato starch, com starch and amylopectin as substrate, especially, and the product ratio of $\beta$-CD to ${\gamma}$-CD was obtained at range of from 5.8:1 to 8.4:1 according to the kind of substrate. The purified enzyme produced mainly $\beta$-CD without accumulation of $\alpha$-CD during enzyme reaction using various starches as the substrate, indicating that the purified enzyme is the typical $\beta$-CGTase. The purified CGTase produced 25 g/l of CDs from 5.0% (w/v) liquefied com starch and the conversion yield of CDs was 50%, and the content of $\beta$-CD was 84% of total CDs after 8 hours under the optimum reaction condition.ion.
To find out the most effective method of treatments for the induction of sprouting of dormant seed potato pieces for the fall crop production of Irish cobbler, this experiment was carried out with sprout inducing bed and field performance trial. In GA treatments, about 10 days were required to sprout and resulted uniform and thin 3-4 sprouts per tuber piece, but sprouts were slender and rooting was not observed. In Ethrel treatments, 20-25 days were required, and sprouts were inferior than that of GA treatment in uniformity of sprouting, and percentage of rotten pieces and of healthy sprouted tuber pieces, but number of sprouts per tuber pieces was low, being 1-2, and sprouts were short, thick, and healthy, and showed good rooting. In GA and Ethrel mixture treatments, 1-2 more days were required to sprout than GA treatments, but sprouts were relatively healthy, and other sprouting pattern were like that of GA treatments. In Ethylene chlorohydrin and 6-Benzyl-adenine treatments, sprouting was like that of Ethrel treatments, but much more days were required than Ethrel treatments and tendency of severe rotting was observed. Optimum treating methods of promising chemicals found to be 1-2 and 2-5 ppm GA solution, 500 and 1000-2000 ppm Ethrel solution, and 1-2+250-500 and 5+250-500 ppm GA and Ethrel mixture solution for 60 min. treatment of tuber piece and whole-tuber, respectively. Induction of sprouting in dry and hot time resulted severer rotting of tuber pieces during the induction of sprouting and with the advancement of dormancy, being delayed in date of treatment, tendency of promotion of sprouting and rooting was observed. When sprouted tuber piece was transplanted at the same date, yields were in order of Ethrel, GA and Ethrel mixture, and GA treatment, indicating the correlation between yield and healthiness of sprout and rooting status of sprouted tuber piece. In all treatments, earlier transplanting resulted higher yields.

In strawberry production, among others, the high incidence of diseases by pathogenic fungi resulting in the reduction of fruit yield and quality requires the development of eco-friendly management systems rather than chemical sprays to control them. The diameter of colonies grown in media at $25^{\circ}C$ for 5 days was measured to evaluate the in vitro inhibition of mycelial growth of 5 pathogenic fungi by irradiation with ultraviolet (UV-C, 264 nm). The mycelial growth of 5 pathogenic fungi was inhibited in potato dextrose agar (PDA) by the irradiation of UV-C for 1 hour a day, and was dramatically inhibited by the irradiation of UV-C for 9-12 h a day. The irradiation of UV-C for 9-12 h a day inhibited completely the growth of the late blight pathogen, Phytophthora cactorum. The irradiation distance of 40 to 50 cm was effective for the inhibition of mycelial growth of fungi. The mycelial growth of fungi without pre-incubation was inhibited strongly by UV-C irradiation compared to fungi pre-incubated for 2 days without light. The mycelia growth of Colletotrichum gloeosprioides and Fusarium oxysporum was inhibited strongly by UV-C irradiation in vegetable 8 juice agar compared to PDA.
