Xenopus Oocyte의 $K^{+}$ Channel에 관한 연구

  • 목적: Guinea pig heart의 ATP sensitive $K^{+}$ channel xenopus oocyte에 발현시켜 연구하고져 본 실험을 행하였다. 실험방법: 기니픽 심장으로부터 ,RNA를 분리하여 50ng/$\mu$l의 농도로 50nl를 xenopusdp 주입하였다. Xenpus oocyte에서 conventional electrode를 이용 막전휘를 측정하였고, pH selective 미세전극으로 세포내 pH를 측정하였다. 막전위에 미치는 potassium channel opener, blocker, KCN의 작용을 관찰하였다. 결과: 기니픽 심장 mRNA를 주입하거나 주입하지 않은 xenopus oocyte에서 $K^{+}$channel opener인 cromakalin, RP49356등은 과분극을 일으키지 못하였다. 그러나 세포내 ATP 감소제인 KCN은 농도 의존적으로 과분극을 일으켰으나 ,glibenclamide에 의해 차단되는 않았다. mRNA를 주입한 oocyte에서 Na-H 자극제인 NH$_4$Cl은 pH 변동을 일으켜 NA-H exchange를 expression 시켰다. 결론: Xenopus oocyte는 cromakalin등에 의해 open되는 $K^{+}$channel 은 없었고, 기니픽 심근의 ATP sensitive $K^{+}$channel로 expression 되지 않았으나 Na-H exchange 는 expression 됨을 알 수 있었다. KCN으로 open 되는 $K^{+}$channel이 있었으나 glibenclamide에는 차단되지 않는 channel이였다.

신규 합성 $K_{ATP}$ 통로 개방제인 SKP-450과 대사체 SKP-818의 흰쥐 적출 심장 및 대동맥에 대한 작용 (Cardiovascular Effects of Novel Potassium Channel Opener SKP-450 and SKP-818 in Isolated Rat Heart and Aorta)

  • The effect of potassium channel openers, SKP-450, SKP-818 and lemakalim have been compared in rat heart and aorta. In rat isolated heart, SKP-450 had a greater negative inotrop ic effect than lemakalim and KR-30818 against left ventricular developed pressure (LVDP) and double product of heart rate and LVDP (DP). In addition, SKP-450 had a greater effect than lemakalim and KR-30818 in increasing coronary flow, indicating a more potent vasodilating effect in coronary artery. Negative inotropic effect and coronary vasodilating effect of SKP-450 and SEP-818 were significantly reduced by 10 min-perfusion with $10^{-6}M$ glyburide, a selective blocker of ATP-sensitive potassium channel. In rat aorta, SKP-30450 and SKP-30818 as well as lemakalim induced powerful concentration-dependent relaxations against norepinephrinc-induced tone ($EC_{50},\;{\mu}M$ : SKP-30450, $0.107{\pm}0.009$; SKP-30818, $0.476{\pm}0.022$ ; lemakalim, $0.565{\pm}0.039$ ). These relaxant effects were significantly reduced by pretreatment with glyburide. In sununary, SKP-30450 and SKP-30818 showed greater negative inotropic and vasorelaxant effect than lemakalim in rat aorta with order of potency of SKP-30450 > SKP-30818 > lemakalim. These actions are suggested to be mediated at least in part by a mechanism which involves the opening of ATP-sensitive potassium channel.

$K^+$ 통로개방제 Pinacidil이 종양이식 생쥐에서 Tl-201의 체내분포에 미치는 영향 (Effects of Pinacidil, a Potassium-Channel Opener, on Biodistribution of Thallium-201 in Tumor-Bearing Mice)

  • 목적: 생체 내에서 potassium과 유사한 역학을 보이는 thallium은 종양의 영상에 널리 사용된다. $K^+$ 통로개방제는 세포 내의 potassium을 외부로 배출되게 하는 기능이 있어 Tl-201을 이용한 종양영상에도 영향을 미칠 수 있을 것이라 생각된다. 본 연구는 강력한 $K^+$ 통로개방제의 하나인 pinacidil이 Tl-201을 이용한 종양의 국소화에 어떠한 영향을 미치는 가를 알아보기 위하여, 종양을 가진 생쥐에서 pinacidil에 의한 Tl-201의 체내분포 변화를 알아보았다. 대상 및 방법: 생쥐 유방암세포주를 이식받은 Balb/c 생쥐를 3주간 사육한 후 실험에 이용하였다. Tl-201 185 KBq를 꼬리정맥을 통해 주입한 후 일정시간에 실험동물을 희생시켜 Tl-201의 체내 분포를 알아 보았으며, pinacidil $100{\mu}g$ 투여에 따른 분포 변화를 알아보았다. 또한 Tl-201 3.7 MBq를 꼬리정맥을 통해 주입하여 Tl-201의 시간에 따른 전신 잔류율을 측정하였고, pinacidil 투여에 의한 전신 잔류율 변화를 구하였다. 결과: pinacidil 투여시 대조군에 비해 혈액 내 Tl-201의 방사능치를 약간 감소시키나 신장에서는 현저한 감소를 일으켰다. 또한 간, 근육, 및 장관의 방사능은 pinacidil 투여에 의해 변하지 않았다. 종양 내 Tl-201 섭취율 및 종양조직/혈액 무게당 섭취비는 대조군에 비해 pinacidil 투여군에서 낮았으며, Tl-201의 24시간 전신 잔류율도 pinacidil 투여군에서 낮았다. 결론: $K^+$ 통로개방제는 Tl-201의 체외 배설을 촉진시키고, 신장 섭취를 감소시켜나, 종양 섭취량도 감소시켰다. 그러므로 Tl-201 종양영상 판독시 $K^+$ 통로개방제를 사용하는 경우에는 오히려 Tl-201 종양영상의 질이 향상되기 보다는 저하될 수 있다는 사실을 고려하여야 할 것으로 생각된다.

  • Neuronal hyperexcitability followed by high level of intracellular calcium and oxidative stress play critical roles in neuronal cell death in stroke and neurotrauma. Hence, KR-31378, a novel benzopyran derivative was designed as a new therapeutic strategy for neuroprotection possessing both anti-oxidant and potassium channel modulating activities. In the present study, we tested for its neuroprotective efficacy against oxidative stress-induced cell death in primary cortical cultures and further investigated its neuroprotective mechanism. Pinacidil causes depresor action, catecholamine release and vasorelaxation in the normotensive rat

  • The present study was conducted to investigate the effects of pinacidil, a potassium channel opener, on arterial blood pressure, catecholamine release and vascular contractile responses in the normotensve rats and to establish the mechanism of action. Phenylephrine (an adrenergi $_1$-receptor agonist) and high potassium (a membrane- depolarizing agent) caused greatly contractile responses in the isolated aortic strips, respectively. These phenylephrine (10$\^$-5/ M)-induced contractile responses were dose-dependently depressed in the presence of pinacidil (25 ∼ 100 ${\mu}$M). (omitted)

$K^+$ Channel 개방제인 BRL 34915의 신장작용 (Renal Action of BRL 34915, a $K^+$ Channel Opener, in Dog)

  • The effect of BRL 34915, a $K^{+}$ channe$Na^{+}$l opener, on renal function was investigated in anesthetized dog. BRL 34915, when given into the vein, elicited the decrease of urine volume accompanied with the reduction of renal plasma flow (RPF), osmolar clearance ($C_{osm}$) and amounts of sodium excreted into urine ($E_{na}$), whereas reabsorption rate of sodium in renal tubules ($R_{na}$), ratio of $K^{+}$ against $Na^{+}$ in urine ($K^{+}$ /$Na^{+}$) were elevated significantly with a partial fall of mean arterial pressure (MAP). BRL 34915 injected into a renal artery produced the diuretic action along with the increase in RPF $C_{osm}$, $E_{na}$ and amounts of potassium excreted in urine ($E_{k}$), and the decrease in $R_{na}$, reabsorption rate of potassium in renal tubules ($R_{k}$), free water clearance ($C_{H20}$) and $K^{+}/Na^{+}$ ratio in only ipsilateral kidney, however changes of the renal function were not observed in control kidney. BRL 34915 given into carotid artery exhibited the same aspect as changes of renal function induced by intravenous BRL 34915. These results suggest that BRL 34915 has dual effects, renally acting diuretic and centrally acting antidiuretic action.n.

흰쥐 배뇨근에 존재하는 potassium 통로의 특성 (Characteristics of Potassium Channel in the Isolated Rat Detrusor Muscle)

  • 흰쥐의 적출 배뇨근에 대한 수종의 potassium 통로개방제의 작용을 관찰하고, 배뇨근에 존재하는 potassium 통로의 특성을 알아보기 위하여 체중 250~350g의 흰쥐 (Sprague-Dawley)를 단두하여 희생시킨 후 방광을 적출하였다. 적출된 방광으로 부터 $1.5mm{\times}1.5cm$의 배뇨근 수평절편을 만들어 1ml의 Tyrode 영양액을 포함하는 적출근편실험조에 현수하고 등척성장력을 측정하여 polygraph에 묘기하였다. 배뇨근절편은 potassium 통로 개방제인 pinacidil, BRL 38227 및 RP 52891의 누적 농도 첨가에 의하여 그 기본장력이 농도의존적으로 감소하였는데 그 작용강도는 RP 52891, pinacidil 그리고 BRL 38227의 순이었다. 전위 의존성 potassium 통로 봉쇄제인 procaine은 배뇨근 절편의 기본장력에 영향을 미치지 못했으며, pinacidil, BRL 38227 및 RP 52891에 의한 기본장력감소작용에 대해서도 영향을 미치지 못하였다. 칼슘 의존성 potassium 통로봉쇄제인 apamin은 배뇨근의 기본장력에 유의한 변화를 가져오지 못하였고, potassium 통로 개방제들에 대하여는 상경적 길항작용을 나타내지는 않았으나 BRL 38227과 RP 52891의 최고효능을 유의하게 감소시켰다. ATP 의존성 potassium 통로봉쇄제인 glibenclamide는 배뇨근 절편의 기본장력을 증가시키고, pinacidil을 상경적으로 길항하였으며, BRL 38227과 RP 52891을 상경적으로 길항하는 동시에 그 최대효능을 감소시켰다. 췌장의 ${\beta}$-세포에서 ATP 의존성 potassium 통로를 개방시켜 인슐린의 분비를 억제하는 galanin은 흰쥐의 배뇨근을 수축시켰다. 이상의 결과를 종합하면, 흰쥐의 배뇨근에서는 새로운 potassium 통로 개방제인 RP 52891의 배뇨근 이완작용이 pinacidil보다 강한 것으로 관찰되었다. 또 흰쥐 배뇨근에서는 ATP 의존성이며, glibenclamide 반응성인 potassium 통로가 존재 한다고 생각되는데, 이는 췌장의 ${\beta}$-세포에 있는 ATP 의존성 potassium 통로와는 다른 특성을 가진 것으로 추측된다.

The Role of Mitochondrial ATP-sensitive Potassium Channel on Intestinal Pacemaking Activity

  • Interstitial cells of Cajal (ICCs) are the pacemaker cells that generate slow waves in the gastrointestinal (GI) tract. In the present study, we investigated the effect of mitochondrial ATP-sensitive potassium (mitoKATP) channel on pacemaking activity in cultured ICCs from murine small intestine by using whole-cell patch clamp techniques. Under current clamp mode, at 10μM glibenclamide, there was no change in pacemaking activity of ICCs. At $30{\mu}M$ glibenclamide, an inhibitor of the ATP sensitive $K^+$ channels, we could find two examples. If pacemaking activity of ICCs was irregulating, pacemaking activity of ICCs was changed into regulating and if in normal conditions, membrane potential amplitude was increased. At $50{\mu}M$ glibenclamide, the resting membrane potential was depolarized. At 3mM 5-HDA, an inhibitor of the mitoKATP channels, inhibited the pacemaking activity of ICCs. Both the amplitude and the frequency were decreased. At 5 mM 5-HDA, both the amplitude and the frequency were completely abolished. Diazoxide, an opener of the mitoKATP channels, was applied to examine its effect on pacemaking activity of ICCs. At $50{\mu}M$ concentration, the pacemaking activity of ICCs was inhibited. Both the amplitude and the frequency were decreased. At 1 mM concentration, both the amplitude and the frequency were completely abolished and the resting membrane potential was shaked.These results indicate that mitoKATP channel has an important role in pacemaking activity of ICCs.

INFLUENCE OF PINACIDIL ON CATECHOLAMINE SECRETION EVOKED BY CHOLINERGIC STIMULATION AND MEMBRANE DEPOLARIZATION FROM THE RAT ADRENAL GLAND

  • It has been known that potassium channel openers are a new class of molecules that have attracted general interest because of their potent antihypertensive activity in vivo and vasorelaxant activity in vitro (Hamilton and Weston, 1989). In the present study, it was attempted to examine the effect of the potassium channel opener on catecholamine (CA) secretion evoked by cholinergic stimulation, membrane depolarization and calcium mobilization from the isolated perfused rat adrenal gland. The perfusion of pinacidil (30-300 uM) into an adrenal vein for 20 min produced relatively dose-dependent inhibition in CA secretion evoked by ACh (5.32 mM), high $K^{+}$ (56 mM), DMPP (100 uM for 2 min), McN-A-343 (100 uM for 2 min), cyclopiazonic acid (10 uM for 4 min) and Bay-K-8644 (10 uM for 4 min). Also, under the presence of minoxidil (100 uM), which is also known to be a potassium channel activator, CA secretory responses evoked by ACh, high potassium, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were also significantly depressed. However, in adrenal glands preloaded with pinacidil (100 uM) under the presence of glibenclamide (1 uM), an antidiabetic sulfonylurea that has been shown to be a specific blocker of ATP-regulated potassium channels (for 20 min), CA secretory responses evoked by ACh, high potassium, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were considerably recovered to a considerable extent of the normal release as compared to that of pinacidil only. These results, taken together, suggest that pinacidil cause the marked inhibition of CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as by membrane depolarization, indicating strongly that this effect may be mediated by inhibiting influx of extracellular calcium and release in intracellular calcium in the rat adrenomedullary chromaffin cells. Furthermore, these findings suggest strongly that these potassium channel openers-sensitive membrane potassium channels also play an important role in regulating CA secretion.

Effects of Novel Potassium Channel Opener KR-30450 and its Metabolite KR-30818 on the Smooth, Muscle of the Guinea Pig

  • The effect of potassium channel openers, KR-30450, KR-30818 and lemakalim have been compared against several spasmogens in guinea pig bronchi. In guinea pig bronchi, KR-30450 had a greater relaxant effect than lemakalim and KR-30818 against tone induced by histamine $10^{-5}M$ ($EC_{50}$ $\mu$M: KR-30450, 0.108$\pm$0.077; KR-30818, 0.403$\pm$0.023; lemakalim, 0.968$\pm$0.036) and prostaglandin $F_{2\alpha} 3\times10^{-6} M$ ($EC_{50}$ $\mu$M: KR-30450, 0.018$\pm$0.001; KR-30818, 0.028$\pm$0.003; lemakalim, 0.138$\pm$0.019). Relaxant effect of KR-30450 and KR-30818 were significantly reduced by 20 min pretreatment of tissues with $10^[-6}$ M glibenclamide, a selective blocker of ATP-sensitive potassium channel. Against acetylcholine-induced tone in guinea pig bronchi, however, these compounds had little effect. In summary, KR-30450 and KR-30818 showed greater relaxant effect than lemakalim in guinea pig bronchi (KR-30450>KR-30818>lemakalim). These relaxant actions are suggested to be mediated at least in part by a mechanism which involves the opening of ATP-sensitive potassium channel.
