Genetic characterization of alloherpesvirus (cyprinid herpesvirus-2 and koi herpesvirus) and poxvirus (carp edema virus) identified from domestic and imported cyprinids in Korea

  • Cyprinids are popular species for aquaculture worldwide, with Asia being a significant contributor to their production. In Korea, common carp (Cyprinus carpio), koi carp (Cyprinus rubrofuscus), and goldfish (Carassius auratus) are cultivated domestically and imported for ornamental or human consumption purposes. Among the viruses that infect cyprinids, cyprinid herpesvirus-2 (CyHV-2), koi herpesvirus (KHV, also known as cyprinid herpesvirus-3), and carp edema virus (CEV) are of particular concern as they cause substantial economic losses to the aquaculture industry. In this study, we investigated these viruses in both of domestic and imported cyprinids. Our results revealed that CyHV-2 was only detected in imported goldfish from Thailand. To further investigate the genetic characteristics of them, the marker A region was analyzed. Despite belonging to the same cluster with isolates from China, France, Poland, and Israel, CyHV-2 detected in this study showed distinct differences in their repetitive sequence sizes. Furthermore, two different forms of KHV/CEV coinfection were identified from domestic koi carp, both of which exhibited typical symptoms. Phylogenetic analysis showed that one KHV isolate (ScKc-2105-K) was of the Asian type and closely related to isolates from Japan, Indonesia, Belgium, Taiwan, and China. Two CEV isolates (ScKc-2105-CE and GhKc-2207-CE) be- longed to the IIa type and showed high similarity with isolates from the USA, France, and Korea. Notably, koi carp injected with cultured KHV (ScKc-2105-K) showed 78.0% cumulative mortality within 14 days post-injection (dpi). Our findings support the importance of regular surveillance of viral diseases in cyprinids.

  • Growing stringent global regulations in Korea poses a threat to corporate sustainability. Companies must respond strategically to navigate these regulations and avoid greenwashing. Objective of this research was to analyze how Korean companies are responding to the global trend of reducing plastic use and propose improved management strategies. Seven indicators were developed to assess companies' post-plastic strategies and applied to analyze the sustainability reports of Amore Pacific and LG Household & HealthCare. These indicators included, 1) disclosure of plastic raw materials used by weight or volume, 2) disclosure of recycled plastic raw materials used by weight or volume, 3) disclosure of waste recycling, reuse amounts, and disposal using waste treatment method 4) strategies to reduce environmental impact of plastics, 5) plastic packaging, reduce, recycle, reuse, and composting (in the real environment), 6) plastic management roadmap for the circular economy, and 7) education for sustainable plastic management. Based on the review of considered companies, we propose in-listed sustainable plastics management strategies: disclosing the ratio of plastic raw materials and recycled raw materials for all products, considering recycling rate throughout the product value chain, and not only for the production phase, reviewing carbon dioxide emissions based on life cycle assessment rather than reducing plastic consumption, studying the biodegradability of biodegradable plastics in natural environment such as soil, considering the consumer's perspective.

Effect of supplement of SCM in culture medium for in vitro development of bovine in vitro fertilized oocytes

  • Background: The successful production of superior or transgenic offspring from in vitro produced embryos in cattle relies heavily on the quality of blastocyst stage embryos. In order to enhance the developmental competency of these embryos, a novel culture method was devised. Methods: This study utilized stem cell culture medium (SCM) from hESCs as a supplement within the culture medium for bovine in vitro produced embryos. To gauge the efficacy of this approach, in vitro fertilized embryos were subjected to culture in CR1aa medium enriched with one of three supplements: 0.3% BSA, 10% FBS, or 10% SCM. Results: The blastocyst development and hatching rates of one-cell zygotes cultured in CR1aa medium supplemented with SCM (23.9% and 10.2%) surpassed those cultured in CR1aa medium supplemented with BSA (9.3% and 0.0%) or FBS (3.1% and 0.0%) (p < 0.05). Furthermore, post-zygotic gene activation, cleaved embryos cultured in CR1aa medium supplemented with SCM (57.8% and 34.5%) exhibited notably higher rates (p < 0.05) compared to those cultured with BSA (12.9% and 0.0%) or FBS (45.7% and 22.5%) supplementation. Furthermore, the microinjection of SCM into the cytoplasm or pronucleus of fertilized zygotes resulted in elevated blastocyst development and hatching rates, particularly when the microinjected embryos were subsequently cultured in CR1aa medium supplemented with SCM from the 8-cell embryo stage onwards (p < 0.05), in contrast to those cultured with FBS supplementation. Conclusions: In conclusion, this study conclusively demonstrated that the incorporation of SCM into the culture medium significantly enhances the developmental progress of preimplantation embryos.

Identification of heat shock protein70-2 and protamine-1 mRNA, proteins, and analyses of their association with fertility using frozen-thawed sperm in Madura bulls

  • Objective: This study aims to identify heat shock protein70-2 (HSP70-2) and protamine-1 (PRM1) mRNA and protein in Madura bull sperm and demonstrate their relation as bull fertility biomarkers. Methods: The Madura bull fertility rates were grouped based on the percentage of first service conception rate (%FSCR) as high fertility (HF) (79.04%; n = 4), and low fertility (LF) (65.84%; n = 4). mRNA of HSP70-2 and PRM1 with peptidylprolyl isomerase A (PPIA) as a housekeeping gene were determined by quantitative real-time polymerase chain reaction, while enzyme-linked immunoassay was used to measure protein abundance. In the post-thawed semen samples, sperm motility, viability, acrosome integrity, and sperm DNA fragmentation index were analyzed. Data analysis was performed on the measured parameters of semen quality, relative mRNA expression, and protein abundance of HSP70-2 and PRM1, among the bulls with various fertility levels (HF and LF) in a one-way analysis of variance analysis. The Pearson correlation was used to analyze the relationship between semen quality, mRNA, proteins, and fertility rate. Results: Relative mRNA expression and protein abundance of HSP70-2 and PRM1 were detected and were found to be highly expressed in bulls with HF (p<0.05) and were associated with several parameters of semen quality. Conclusion: HSP70-2 and PRM1 mRNA and protein molecules have great potential to serve as molecular markers for determining bull fertility.

Major histocompatibility complex genes exhibit a potential immunological role in mixed Eimeria-infected broiler cecum analyzed using RNA sequencing

  • Objective: This study was conducted to investigate the differential expression of the major histocompatibility complex (MHC) gene region in Eimeria-infected broiler. Methods: We profiled gene expression of Eimeria-infected and uninfected ceca of broilers sampled at 4, 7, and 21 days post-infection (dpi) using RNA sequencing. Differentially expressed genes (DEGs) between two sample groups were identified at each time point. DEGs located on chicken chromosome 16 were used for further analysis. Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis was conducted for the functional annotation of DEGs. Results: Fourteen significant (false discovery rate <0.1) DEGs were identified at 4 and 7 dpi and categorized into three groups: MHC-Y class I genes, MHC-B region genes, and non-MHC genes. In Eimeria-infected broilers, MHC-Y class I genes were upregulated at 4 dpi but downregulated at 7 dpi. This result implies that MHC-Y class I genes initially activated an immune response, which was then suppressed by Eimeria. Of the MHC-B region genes, the DMB1 gene was upregulated, and TAP-related genes significantly implemented antigen processing for MHC class I at 4 dpi, which was supported by KEGG pathway analysis. Conclusion: This study is the first to investigate MHC gene responses to coccidia infection in chickens using RNA sequencing. MHC-B and MHC-Y genes showed their immune responses in reaction to Eimeria infection. These findings are valuable for understanding chicken MHC gene function.

Reproductive management of dairy cows: an existing scenario from urban farming system in Bangladesh

  • Background: Reproductive management practices play crucial roles to maximize the reproductive performance of cows, and thus contribute to farm profitability. We aimed to assess the reproductive management of cows currently practiced in the dairy farms in an urban farming system. Methods: A total of 62 dairy farms were randomly selected considering all size of farms such as small (1-5 cattle), medium (6-20 cattle) and large farms (> 20 cattle) from selected areas of Dhaka city in Bangladesh. The reproductive management-related parameters viz. estrus detection, breeding method, pregnancy diagnosis, dry cow and parturition management, vaccination and treatment of reproductive problems etc. were obtained in a pre-defined questionnaire during the farm visit. Results: The visual observation method was only used (100.0%; 62/62) for estrus detection irrespective of size of the farms; while farmers observed cows for estrus 4-5 times a day, but only for 20-60 seconds each time. Regardless of farm size, 89.0% (55/62) farms used artificial insemination (AI) for breeding the cows. Intriguingly, all farms (100.0%) routinely checked the cows for pregnancy at 35-40 days post-breeding using rectal palpation technique by registered veterinarian. However, only 6.5% (4/62) farms practiced dry cow management. Notably, all farms (100.0%) provided nutritional supplements (Vit D, Ca and P) during late gestation. However, proper hygiene and cleanliness during parturition was not practiced in 77.4% (48/62) farms; even though 96.7% (60/62) farms treated cows by registered veterinarian for parturition-related problems. Conclusions: While farmers used AI service for breeding and timely check their cows for pregnancy; however, they need to increase observation time (30 minutes/ observation, twice in a day: early morning and early night) for estrus detection, consider dry cow management and ensure hygienic parturition for maximizing production.

Prevalence of salivary microbial load and lactic acid presence in diabetic and non-diabetic individuals with different dental caries stages

  • Objectives: This study aims to correlate caries-causing microorganism load, lactic acid estimation, and blood groups to high caries risk in diabetic and non-diabetic individuals and low caries risk in healthy individuals. Materials and Methods: This study includes 30 participants divided into 3 groups: Group A, High-risk caries diabetic individuals; Group B, High-risk caries non-diabetic individuals; and Group C, Low-risk caries individuals. The medical condition, oral hygiene, and caries risk assessment (American Dental Association classification and International Caries Detection and Assessment System scoring) were documented. Each individual's 3 mL of saliva was analyzed for microbial load and lactic acid as follows: Part I: 2 mL for microbial quantity estimation using nutrient agar and blood agar medium, biochemical investigation, and carbohydrate fermentation tests; Part II: 0.5 mL for lactic acid estimation using spectrophotometric analysis. Among the selected individuals, blood group correlation was assessed. The χ2 test, Kruskal-Wallis test, and post hoc analysis were done using Dunn's test (p < 0.05). Results: Group A had the highest microbial load and lactic acid concentration, followed by Groups B and C. The predominant bacteria were Lactobacilli (63.00 ± 15.49) and Streptococcus mutans (76.00 ± 13.90) in saliva. Blood Group B is prevalent in diabetic and non-diabetic high-risk caries patients but statistically insignificant. Conclusions: Diabetic individuals are more susceptible to dental caries due to high microbial loads and increased lactic acid production. These factors also lower the executing tendency of neutrophils, which accelerates microbial accumulation and increases the risk of caries in diabetic individuals.

고로슬래그 미분말 치환율에 따른 이산화탄소 양생 모르타르의 기초 물성 (Fundamental Characteristics of CO2-cured Mortar with Varied Rates of Blast Furnace Slag Fine Powder Substitution)

  • 본 실험에서는 고로슬래그 미분말 치환율에 따른 이산화탄소 양생 모르타르의 기초 물성을 확인하였다. 실험 결과, 이산화탄소 양생은 CaCO3의 생성을 촉진하며 공극 감소와 조기 강도 발현에 영향을 준다. Ca(OH)2가 CaCO3보다 주된 강도 발현에 영향을 주며 고로슬래그 미분말 치환율이 높은 배합일수록 Ca(OH)2 생성이 감소한다. 하지만 Ca(OH)2가 소모된 이후에도 CaCO3를 통해 강도가 유지되며 이산화탄소 양생 이후 대기 중에 노출하여도 소요의 성능을 유지할 수 있을 것으로 전망된다. 고로슬래그 미분말 치환율이 50%를 초과할 경우 이산화탄소에 의한 성능 저하를 유발하므로 치환 비율을 조정할 필요가 있다.

사출 성형 공정에서의 변수 최적화 방법론 (Methodology for Variable Optimization in Injection Molding Process)

  • Purpose: The injection molding process, crucial for plastic shaping, encounters difficulties in sustaining product quality when replacing injection machines. Variations in machine types and outputs between different production lines or factories increase the risk of quality deterioration. In response, the study aims to develop a system that optimally adjusts conditions during the replacement of injection machines linked to molds. Methods: Utilizing a dataset of 12 injection process variables and 52 corresponding sensor variables, a predictive model is crafted using Decision Tree, Random Forest, and XGBoost. Model evaluation is conducted using an 80% training data and a 20% test data split. The dependent variable, classified into five characteristics based on temperature and pressure, guides the prediction model. Bayesian optimization, integrated into the selected model, determines optimal values for process variables during the replacement of injection machines. The iterative convergence of sensor prediction values to the optimum range is visually confirmed, aligning them with the target range. Experimental results validate the proposed approach. Results: Post-experiment analysis indicates the superiority of the XGBoost model across all five characteristics, achieving a combined high performance of 0.81 and a Mean Absolute Error (MAE) of 0.77. The study introduces a method for optimizing initial conditions in the injection process during machine replacement, utilizing Bayesian optimization. This streamlined approach reduces both time and costs, thereby enhancing process efficiency. Conclusion: This research contributes practical insights to the optimization literature, offering valuable guidance for industries seeking streamlined and cost-effective methods for machine replacement in injection molding.

Enhancement of cryopreserved rooster semen and fertility potential after oral administration of Thai ginger (Kaempferia parviflora) extract in Thai native chickens

  • Objective: Semen cryopreservation is an effective method of preserving genetic material, particularly in native chicken breeds facing a substantial decline. In this study, we evaluated the quality of frozen/thawed rooster semen treated with different concentrations of oral administrations of black ginger (Kaempferia parviflora: KP) extract and determined its fertility. Methods: Thirty-two Thai native roosters (Pradu Hang Dum, 42 weeks old) were used in this study. The treatments were classified into four groups according to the concentration of KP extract administered to the roosters: 0, 100, 150, and 200 mg/kg body weight. The quality of fresh semen was analyzed before cryopreservation. Post-thaw sperm quality and fertility potential were determined. Also, lipid peroxidation was determined. Results: The results showed that sperm concentration and movement increased in roosters treated with 200 mg/kg of KP extract (p<0.05). The malondialdehyde (MDA) in the roosters receiving 200 mg/kg KP extract was lower than that in the other but had an insignificant difference within the KP treatment groups (p>0.05). The highest MDA levels were observed in the control group (p<0.05). The percentage of motile sperm (total motility and progressive motility) after semen thawing was higher in roosters that received 150 and 200 mg/kg KP extract than in those that received 100 mg/kg KP extract and the control (p<0.05). MDA levels decreased significantly in roosters that received 150 and 200 mg/kg KP extract than in those that received 100 mg/kg KP extract and the control (p<0.05). Fertility and hatchability were greater in the KP150 and KP200 groups than in the KP100 and control groups (p<0.05). Conclusion: The optimal amount of KP extract influencing initial sperm quality was determined to be 200 mg/kg. However, 150 mg/kg was the optimal low dosage of KP extract administration that maintained sperm quality and fertility following semen cryopreservation.
