Development of a cell-laden thermosensitive chitosan bioink for 3D bioprinting

  • 3D bioprinting is a technology to produce complex tissue constructs through printing living cells with hydrogel in a layer-by-layer process. To produce more stable 3D cell-laden structures, various materials have been developed such as alginate, fibrin and gelatin. However, most of these hydrogels are chemically bound using crosslinkers which can cause some problems in cytotoxicity and cell viability. On the other hand, thermosensitive hydrogels are physically cross-linked by non-covalent interaction without crosslinker, facilitating stable cytotoxicity and cell viability. The examples of currently reported thermosensitive hydrogels are poly(ethylene glycol)/poly(propylene glycol)/poly(ethylene glycol) (PEG-PPG-PEG) and poly(ethylene glycol)/poly(lactic acid-co-glycolic acid) (PEG/PLGA). Chitosan, which have been widely used in tissue engineering due to its biocompatibility and osteoconductivity, can be used as thermosensitive hydrogels. However, despite the many advantages, chitosan hydrogel has not yet been used as a bioink. The purpose of this study was to develop a bioink by chitosan hydrogel for 3D bioprinting and to evaluate the suitability and potential ability of the developed chitosan hydrogel as a bioink. To prepare the chitosan hydrogel solution, ${\beta}-glycerolphosphate$ solution was added to the chitosan solution at the final pH ranged from 6.9 to 7.1. Gelation time decreased exponentially with increasing temperature. Scanning electron microscopy (SEM) image showed that chitosan hydrogel had irregular porous structure. From the water soluble tetrazolium salt (WST) and live and dead assay data, it was proven that there was no significant cytotoxicity and that cells were well dispersed. The chitosan hydrogel was well printed under temperature-controlled condition, and cells were well laden inside gel. The cytotoxicity of laden cells was evaluated by live and dead assay. In conclusion, chitosan bioink can be a candidate for 3D bioprinting.

  • Carbon fiber fabric-silicon carbide composites were fabricated by liquid silicon infiltration (LSI) process. The porous two-dimensional carbon fiber fabric performs were prepared by 13 plies of 2D-plain-weave fabric in a three laminating method, [0/90], [${\pm}45$], [$0/90/{\pm}45$] lay-up, respectively. Before laminating, a thin pyrolytic carbon (PyC) layer deposited on the surface of 2D-plain weave fabric sheets as interfacial layer with $C_3H_8$ and $N_2$ gas at $900^{\circ}C$. A densification of the preforms for $C_f-Si-SiC$ matrix composite was achieved according to the LSI process at $1650^{\circ}C$ for 30 min. in vacuum atmosphere. The bending strength of the each composite were measured and the microstructural consideration was performed by a FE-SEM.

  • $(Ba(Zr_{0.85}Y_{0.15})O_{3-\delta})$ oxide, showing high protonic conductivity at high temperatures and good chemical stability with $CO_2$ are referred to as hydrogen separation membrane. For high efficiency of hydrogen separation ($H_2$ flux and selectivity) and low fabrication cost, ultimate thin and dense BZY-Ni layer has to be coated on a porous substrate such as $ZrO_2$. Aerosol depostion (AD) process is a novel technique to grow ceramic film with high density and nano-crystal structure at room-temperature, and may be applicable to the fabrication process of AD integration ceramic layer effectively. XRD, SEM, X-ray mapping measurements were conducted in order to analyze the characteristics of BZY-Ni membrane fabricated by AD process. it is observed that it is homogeneous distribution for BZY-Ni. The result of $H_2$ permeation rate suggests that BZY-Ni composite is higher than BZY.

Effects of magnetic ion exchange resin with PACI coagulation on removal of natural organic matter and MF fouling (자성체 이온교환 수지와 PACI 응집에 의한 국내 주요 수계 내 자연유기물 제거 특성 및 막오염 저감 효과)

  • The application of magnetic ion exchange resin($MIEX^{(R)}$) is effective for natural organic matter(NOM) removal and for control of the formation of disinfection byproducts(DBPs). NOM removal is also enhanced by adding $MIEX^{(R)}$ with coagulant such as polyaluminium chloride(PACl) in conventional drinking water treatment systems. In the application of $MIEX^{(R)}$, it is important to understand changes of NOM characteristics such as hydrophobicity and molecular weight distributions with $MIEX^{(R)}$ or $MIEX^{(R)}$+coagulant treatment.To observe characteristics of NOM by treatment with $MIEX^{(R)}$ or $MIEX^{(R)}$+coagulant, four major drinking water sources were employed. Results showed that the addition of $MIEX^{(R)}$ to coagulation significantly reduced the amount of coagulant required for the optimum removal of dissolved organic matter(DOC) and turbidity in the all four waters. The DOC removal was also increased approximately 20%, compared to coagulant treatment alone. The process with $MIEX^{(R)}$ and coagulant showed that complementary removal of hydrophobic and hydrophilic fraction of DOC. The combined processes preferentially removed the fractions of intermediate (3,000-10,000 Da) and low (< 500 Da) molecular weight. The microfiltration test showed that membrane cake resistance was decreased for waters with flocs from $MIEX^{(R)}$+coagulant. A porous layer was formed to $MIEX^{(R)}$ on the membrane surface and the layer consequently inhibited settling of coagulant flocs, which could act on a foulant.

Effects of Mixing Condition and Filtration Velocity on Turbidity Removal in a Contact Roughing Filter (접촉여과방식 거친여과지에서 혼화조건과 여과속도가 고탁도 제거에 미치는 영향)

  • Slow sand filtrations have been widely used for water treatment in small communities, however their capacity is often limited by high turbidity in the raw water. For this reason, several pre-treatment facilities were required for a slow sand filter. Turbidity removal from the highly turbid raw water was investigated in roughing filters as a pre-treatment process. The roughing filters followed by rapid mixing tank were operated in the form of a contact filtration. In several jar tests, the predetermined optimum aluminium sulfate (alum) doses for turbid water of 30 and 120NTU were 30 and 50mg/L, respectively. At the optimum alum dose, physically optimum parameters including G value of $220sec^{-1}$ and rapid mixing time of 3 minutes were applied to the contact filtration system. Without addition of alum, the filtrate turbidity from the roughing filters, packed respectively with different media such as sand, porous diatomite ball and gravel, was in the range of 5~30NTU at filtration velocities of 30 and 50m/day. However, the application of a contact filtration to roughing filters showed stably lower filtrate turbidity below 1.0NTU at filtration velocity of 30 m/day. Although the filtration velocity increased to 50m/day, filtrate turbidity was still below 1.0NTU in both single and double layer roughing filters. At influent turbidity of 120NTU, the filtrate turbidity was over 5 NTU in the triple layer roughing filter, which shortened the filter run time. The flocs larger than $10{\mu}m$, formed in the rapid mixing tank, were almost captured through the roughing filter bed, while the almost flocs smaller than $10{\mu}m$ remained in filtrate.

Analysis for Species and Lacquer Ware of Wooden Objects Excavated from Sinchang-Dong, Gwangju (광주 신창동 유적 출토 목제품의 수종 및 칠 분석)

  • Wooden objects excavated from Gwangju Shinchang-dong, we analyzed total 40 pieces consisting of 37 pieces for analysis of species of trees, 3 pieces for analysis of lacquer. and especially it was found that in case of lacquerware and living goods like dish, diffuse-porous woods were used. As a result of this, total 7 species of trees used for wooden objects identified are Quercus spp., Cearsus spp., Prunophora spp., Acer spp., Salix spp., Populus spp. were identified. As a result of analyzing lacquered film, though there is a difference of times of lacquering between holed lacquerware and cylider type lacquerware body part 5, it was confirmed that after undercoating, black pigments and lacquer were painted on upper layer in mixture, and foundation platter for with 5 parts lacquer ware was undercoated with mixture of powdered soil, black pigments and lacquer, and then its upper layer was coated with lacquer mixed with black pigments.

Scale Effects of Stability Parameters in the Hydraulic Model Tests of Rubble Mound Coastal Structures (사석구조물(捨石構造物)의 안정성(安定性)에 관한 수리모형(水理模型)의 축척효과(縮尺効果))

  • Scale effects of stability, run-up, run-down and reflection of layered coastal structures are investigated through the experiments with 7 kinds of hydraulic scale models. The occurrence mechanism and the control method of scale effects are also discussed. As a result, it is found that the similarity of permeability of inner layers plays an important role in the occurrence of scale effects, which has been neglected in the most of conventional model tests. To assure the best scale effects for permeable coastal structures, control of Reynolds numbers of the porous media flow in each layer is recommended. It is also found that Reynolds numbers in revetment, filter, and core layer must be greater than $2{\times}10^4$, $3{\times}10^3$, and $1{\times}10^3$, respectively.

  • In recent tissue engineering field, it is being reported that the fabrication of 3D scaffolds having high porous and controlled internal/external architectures can give potential contributions in cell adhesion, proliferation and differentiation. To fabricate these scaffolds, various solid free-form fabrication technologies are being applied. The solid free-form fabrication technology has made it possible to fabricate solid free-form 3D microstructures in layer-by-layer manner. In this research, we developed a multi-head deposition system (MHDS) and used design of experiment (DOE) to fabricate 3D scaffold having an optimized internal/external shape, Through the organization of experimental approach using DOE, the fabrication process of scaffold, which is composed of blended poly-caprolactone (PCL), poly-lactic-co-glycolic acid (PLGA) and tricalcium phosphate (TCP), is established to get uniform line width, line height and porosity efficiently Moreover, the feasibility of application to the tissue engineering of MHDS is demonstrated by human bone marrow stromal cells (hBMSCs) proliferation test.

Porous Electrode manufacture by catalyst powdering method for PAFC (촉매분말법에 의한 PAFC용 다공성 전극제작)

  • Gas diffusion passes are introduced to catalyst layer so as to enlarge reaction region in cathode and anode and then improve electrode performances. The catalyst layer was manufactured with PTFE/carbon (none catalyst loaded) for gas diffusion and Pt/carbon (10 w/o Pt catalyst loaded) by varing the mixing ratio of (PTFE/carbon) to (Pt/carbon) by catalyst powdering method. The electrodes made by mixing Pt(10 w/o)/carbon powders and PTFE/carbon powders containing 60 w/o PTFE at the ratio of 7 : 3 showed the best electrode performances. It was known that by comparing the porosities to electrode performances the electrode performances were increased as both macro pore for gas diffusion and micro pore for electrolyte intrusion were formed much more. The platinum catalyst content in electrode was 0.2 mg/$\textrm{cm}^2$ and the PTFE content was 42 w/o. The electrode performance in unit cell was 220 ㎃/$\textrm{cm}^2$/0.7 V at operating temperature of 150$^{\circ}C$.

Effect of PO43-, CO32- and F- anions on the electrochemical properties of the air-formed oxide covered AZ31 Mg alloy

  • This research was conducted to investigate in detail the effect of $PO_4{^{3-}}$, $CO_3{^{2-}}$ and $F^-$ anions on the electrochemical properties of the thin air-formed oxide film-covered AZ31 Mg alloy. In this work, native air-formed oxide films on AZ31 Mg alloy samples were prepared by knife-abrading method and the changes in the electrochemical properties of the air-formed oxide film were investigated in electrolytes containing 0.01 M, 0.05 M and 0.1 M of $PO_4{^{3-}}$, $CO_3{^{2-}}$ and $F^-$ anions. It was observed that the trend of open circuit potential (OCP) transients changed only in the solution containing $PO_4{^{3-}}$ ions. The Nyquist plots obtained from electrochemical impedance spectroscopy (EIS) showed that the resistance of the new surface films formed in fluoride ion containing bath increased with the increase in concentration of fluoride ions but the resistance of surface films formed in carbonate ion containing bath decreased with the increase in concentration of carbonate ions. The potentiodynamic polarization curves illustrated that under anodic polarization, there was growth of porous passive layer only in fluoride ion containing solution while the surface layer formed in phosphate and carbonate ion containing solutions lost its passivity at high anodic potential of $2.5V_{Ag/AgCl}$.
