  • Embryonic germ (EG) cells are undifferentiated stem cells isolated from cultured primodial germ cells (PGC). These cells share many characteristics with embryonic stem cells including their morphology and pluripotency. Undifferentiated porcine EG cell lines demonstrating capacities of both in vitro and in vivo differentiation have been established. (omitted)

  • DEAE-Cellulose, Sephadex column chromatography and polyacrylamide gel electrophoresis were used to purify acid phosphatase, aryl sulfatases, ${\beta}-glucuronidase$ and cathepsin D in n-butyl alcohol extracts of porcine leukocyte Iysosomes. The degree of purification was quite high for all enzymes studied and some could be identified by histochemical reactions.

  • The purpose of this study was to survey seroprevalence of porcine reproductive and respiratory syndrome (PRRS) virus and porcine circovirus-2 (PCV-2) in Gyeongbuk province by enzyme-linked immunosorbent assay (ELISA). A total of 966 samples collected from 21 pig farms were tested. The sero-positive rate of PRRS and PCV-2 were 77.6% (750/966) and 76.4% (738/966), respectively.

  • The persistence of porcine epidemic diarrhea virus(PEDV) infection was demonstrated in 7 swine farms employing continuous pig flow management even after seasonal outbreaks. Clinically, sporadic postweaning diarrhea was a major concern in those farms. Subsequently circulatory antibody detection using serum neutralizing test made useful for confirmation of PEDV persistent infections. The persistence of PEDV in the premise might have induced recurrence over the period of time.

  • In this paper enantioselective production of levofloxacin by porcine liver esterase was investigated, To enhance the produc0-tivity various factors which affect the enzyme activity and the enantioselectivity were optimized, In terms of temperature and pH 45$^{\circ}C$ and 4.8 were found to be the best conditions for enzyme reaction. Addition of ofloxacin butyl ester the substrate at the concentration of 5 g/L was desirable to avoid the product inhibition and the activity of porcine liver esterase was maintained up to 72 hours.In addition to enhance the availability of substrate effect of solvent was also examined. It was found that the application of 5% (v/v) of acetone acetonitrile and dimethylsulfoxide did not increase the conversion of substrate and the presence of 5%(v/v) butanol inhibited the enzyme activity significantly.

  • Porcine circoviruses(PCV) are the smallest nonenveloped DNA viruses containing a unique single-stranded circular genome. No recognized link was found between PCV infection of pig and disease. But the PCV consistently identified from postweaning multisystemic wasting syndrome(PMWS) and researches indicate that there are strong relationships between PCV and PMWS. Clinical signs were emaciation, dyspnea, high fever with normal appetite. Necropsy findings showed respiratory disease complex lesion and lymph node anomalities. An indirect-immunofluorescent antibody procedure was used to assay swine sera for the presence of PCV atibodies. Antibodies against PCV were found in an average of 20% of the samples tested. The PCV DNA was amplified from lymph nodes collected from pigs. PCV specific primers were successfully amplified PCV DNAs. Further studies are needed to determine the possible role this virus might have in disease.

  • We purified a protein carboxyl O-methyltransferase (protein methylase II) from porcine spleen to homogeneity. The molecular weight of the porcine spleen protein methylase II (ps-PM II) was estimated to be 27,500 daltons on SDS-PAGE. Amino acid sequence of N-terminal 28 residues for ps-PM II was identified. Amino-terminal three amino acid residues of ps-PM II were deleted when compared to those of other protein carboxyl methytransferase. S-Adenosyl-L-homocysteine competitively inhibits ps-PM II with a K, value of $1.63{\times}10^{-7}M$. Myelin basic protein exhibited the highest methyl-accepting capacity among the proteins tested.

Xenotransplantation of Pig Spermatogonia into Mouse Testis

  • The objective of the present study was to investigate the survival effect after transplantation of pig spermatogonia cells into mouse testis. Donor cells were collected from porcine testis and the isolated spermatogonial stem cells were labeled with a fluorescent marker before transplantation and transplanted into testes of busulfan-treated recipient mice. Testes were examined for the presence and localization of labeled donor cells immediately after transplantation or every week for 4 wk. Transplanted germ cells were present in the seminiferous epithelium at 4 weeks after the transplantation, but any differentiating porcine-derived cells were not detected in mouse testis. These results indicate that porcine-derived spermatogonial stem cells can be survived in the recipient, but suggest that porcine-derived male stem cells can not proceed to further differentiating step without helping of immunosuppressor agents.

  • This study was conducted to investigate the effects of preincubation time, concentration and exposure time of sperm on in vitro fertilization of porcine follicular oocytes rnatured in in vitro. The results obtained are as follows ; 1. Effect of preincuhation time for porcine sperm capacitation on in vitro fertilization in medium with heparin was investigated. Normal fertilization rate was highest in 15 min(26.4%). However, there were no significant differences among preincuhation times of 5~90 min, 2. Normal fertilization rates of sperm concentrations were 17.0~26.5%, and normal fertilization rate from l$\times$ l05cell /ml concentration was also higher than those of other sperm concentration. 3. Normal fertilization rates of sperm exposure time of 4, 8, 12, 16 and 20 hours were 6.1, 20.8, 27.8, 25.0 and 26.7%, respectively. Normal fertilization rate from sperm exposure time of 12 hours was also higher than that of other sperm exposure times.

  • For the inspection of the occurrence situation of porcine pleuropneumonia and serotyping of Actinobacillus pleuropneumoniae strains isolated from lung lesions of pig in Korea, a series of experimentation have been carried out by the isolation and identification of A pleuropneumoniae, serotyping by coagglutination test, observation of lung lesion and clinical signs from 360 cases of porcine pneumonia in Clinical Pathology Laboratory, Bayer Veterinary Medical Research Institute. The results could be summarized as follows. The reaction of coagglutination between the reference antigens and the specific reagents of A pleuropneumoniae was strongly agglutinatied within 30 seconds without cross reaction. The 89 strains of A pleuropneumoniae were isolated from 360 cases of porcine pleuropneumonis and the biochemical properties of the isolates were same as the reference strains. The 89 isolated strains could be serotyped 39 strains as setotype 5, 34 strains as serotype 2, 8 strains as serotype 3, 2 strains as serotype 7 by coagglutination test, respectively. The clinical signs of pleuropneumonia were weakness, fever, anorexia, dyspnea and laboured breath in the later stages. The gross lesions of lung were haemorrhages, enlargement of interlobular septa, nodular formation and adhesion of the pleura.
