Dyamics of Plant Communities under Human Impact in the Green-Belt nearby Seoul -Structure of Forest Communities and Secondary Succession- (인간간섭하의 수도권 그린벨트 내 식물군집의 동태 - 삼림군집의 구조와 이차천이 -)

  • In order to elucidate the relationship among the several forest types in the green-sbelt nearby Seoul, the vascular plant species data collected preferentially from 52 stands were subjected to DCA (detrended correspondence analysis). Eight types of forests were arranged on the I/II plane fo DCA stand ordination, Quercus mongolica, Alnus hirsuta, Populus alba $\times$ glandulosa, Robinia pseudo-acacia, Pinus densiflora, Pinus koraiensis, Larix leptolepis and pinus rigida forest. Correlation analysis between the stand scores of DCA and environmental factors revealed that the axis I of DCA stand ordination implied the gradient of altitude, organic matter, total nitrogen and depth of A horizon. The axis II implied the gradient of total nitrogen and soluble phosphorus. The results of PCA (principal components analysis) by environmental data were in good agreement with that of DCA by floristic data. The recruitments of Quercus spp. Occurred in all kinds of forest types in the green-belt and their recruitments were closely related with altitutde: Quercus dentata in the foot. Quercus serrata in the midslope and Quercus mongolica in the uperslope of the mountain.

  • Substrates used were hardwood-Suwon poplar-(Populus alba${\times}$glandulosa L.) and softwood-pitch pine-(Pinus rigida M.). And these substrates were steam exploded then treated with sodium chlorite at 75$^{\circ}C$ with occasional stirring in order to obtain samples which had different lignin contents and crystallinity. And then this resulting samples incubated with a commercial cellulase derived from Trichoderma ressei. The contents of Klason lignin were decreased as the increasing of the ratio of sodium chlorite in the two species. The effect of hardwood was more effective than that of softwood in the same ratio of sodium chlorite. The minimum contents of Klason lignin were 0.8% and 5.1% respectively. And the crystallinities of cellulose were increased very little as increasing of the ratio of sodium clorite. The hydrolysis extent of the two species were increased as the increasing of delignification. Especially, the hydrolysis extent of hardwood was more higher than that of softwood. The maximum hydrolysis extent were 89.8% and 71.1%, respectively.

  • We have cloned an LTP gene (PoLTP1) from poplar (Populus alba ${\times}$ P. tremula var. glandulosa) suspension cells and examined changes in its expression levels in response to various stresses and ABA treatment. The full-length PoLTP1 cDNA clone encodes a polypeptide of 116 amino acids with typical characteristics of LTPs, notably a conserved arrangement of cysteine residues. Southern blot analysis indicate that two or three copies of the PoLTP1 are present in the genome of the investigated hybrid poplar. In addition, northern analysis of samples from soil-grown plants indicate that PoLTP1 is tissue-specifically expressed in the leaves and flowers. The gene is significantly up-regulated by treatment with mannitol, NaCl and ABA, but not by either cold or wounding. These results indicate that PoLTP1 is involved in osmotic stress responses in poplar plants and suspension cells.

Wood Biomass Production of Twelve Tree Species in Coppice Plantations Managed Under 1-, 2- and 3- year Rotations (12수종(樹種)에 대(対)한 단벌기(短伐期) 맹아림(萌芽林)의 Biomass 생산(生産))

  • Wood biomass production at 1-year, 2-year, and 3-year rotations on both low and upper hills at 2m 2m spacing (25,000 trees/ha) was studied for a six-year period with following 12 species; Lespedeza cyrtobotrya Amorpha fruticosa. Robinia pseudoacacia, Acer saccharinum, Platanus orientalis Populus alba ${\times}$ P. glandulosa $F_1$, Salix alba, Pinus rigida, Alnus hirsuta var. sibirica, A. inokumai A. gultinosa, and A. incana. In One-year rotation, Lespedeza cyrtobotrya produced largest amoung of biomass (2.6 t/ha/year, fresh weight) and Populus alba ${\times}$ P. glandulosa $F_1$ the second largest (2.2 t/ha/year) on low hill. In two-year rotation, the latter produced the largest amount (4.8 t/ha/year) and Alnus hirsuta var. sibirica second largest (2.8 t/ha/year) on low hill. In three-year rotation, the largest weight (11.2 t/ha/year) was produced by Robinia pseudoacacia and the second largest (6.2 t/ha/year) by Alnus hirsuta var. sibirica on low hill Amorpha fruticosa, Acer saccharinum, Platanus orientalis and Salix alba were not suitable for biomass or fuelwood productio due to poor growth. Biomass yield on upper hill was reduced considerably for all tewlve species, with less than 4 t/year at maximum Only nitrogen fixing species (Robinia and Alnus species) are recommended on upper hill for biomass production wood sprouting ability of species was generally associated with good biomass production. Calori values of ovendry wood ranged from 4,485 cal/g for Salix alba to 5,125 cal/g for Alnus glutinosa. For maximum biomass production a three-year ratation with coppice is preferred to one-year and two-year roataions The best species appeared to be Robinia pseudoacacia and Alnus hirsuta var sibirica.

Comparative Analysis of Resistance to Antibiotics in Populus alba$\times$P. glandulosa Transformed by nptII or hpt Gene (NPTII 및 HPT 유전자가 삽입된 현사시의 항생제에 대한 저항성 분석)

  • This study was peformed to find out the optimal conditions for the selection of transformed cells using already established transgenic plants. Several transgenic poplar (Populus alba$\times$P giandulosa) lines carrying npt II or hpt gene as a selectable marker were tested against kanamycin or hygromycin. Two culture explants, leaf discs and nodes, were compared regarding their sensitivity to the antibiotics. When leaf discs of untransformed control plants were cultured on callus inducing media in the presence of varying levels of kanamycin or hygromycin, only those cultured on the media containing lower than 50 mg/L kanamycin or 2 mg/L hygromycin formed callus. However, much higher concentration of kanamycin was needed to suppress the growth of axillary buds of untransformed plants. On the other hand, hygromycin at the concentration of 5 mg/L effectively suppressed shoot growth of untransformed plants. Root induction from untransformed plants could also be suppressed at the concentration of 50 mg/L kanamycin or 5 mg/L hygromycin. The transgenic plants showed resistance to 100 mg/L kanamycin or 50 mg/L hygromycin in the growth of callus, shoots, and roots. Hygromycin appeared to be more efficient in selecting untransformed cells than kanamycin.

Nitrogen Storage Potential in Aboveground Biomass of Three-year-old Poplar Clones in a Riparian Area (하천연변에 식재된 3년생 포플러 클론의 지상부 biomass의 질소 저장능력 추정추정)

  • We estimated the biomass productivity and the storage potential of nitrogen, the major contributor of non-point source pollution, with four three-year-old four poplar clones in a riparian woody buffer established in the Anseong River in Anseong, Korea. Stem of Populus alba ${\times}$ P. glandulosa clone 72-31 and Populus deltoides ${\times}$ P. nigra clone Dorskamp showed the highest percentage of aboveground biomass components, followed by branch and leaf. Nitrogen content in aboveground biomass components of two poplar clones was the highest in leaf and the lowest in stem. Nitrogen content in leaf and branch of clone 72-31 was higher than that of clone Dorskamp, while it in stem was lower. Populus deltoides clone Ay48 showed the highest above-ground biomass productivity, which was estimated as $37.5ton\;ha^{-1}$ at age 3. However, clone 72-31 was the lowest in above-ground biomass productivity. Nitrogen storage potential in aboveground biomass of 3-year-old poplar clones was high in order of aboveground biomass. Clone Ay48 showed the highest nitrogen storage potential in aboveground biomass, which was estimated as $218.3kg\;ha^{-1}$ at age 3.

Improvement of Productivity by Forest Tree Breeding Work in Korea (우리나라에서의 임목육종(林木育種)에 의(依)한 생산성(生産性) 증가(增加))

  • Improvement of productivity by forest tree breeding work in Korea was estimated for a few important tree species. Progenies of 17 plus trees of red pine (Pines densiflora) outgrew by 57 percentage compared with progenies of unselected trees at age 15. If best three families are selected among the 17, more than double in volume grow-th is expected. The hybrid Pinus rigida ${\times}$ P. taeda showed more than double volume growth compare to P. rigida at a southern plantation at age 15. However, the superiority of the hybrid decreased at northern plantations, mainly because of low coldhardiness of the hybrid. At a northern plantation, the hybrid grew less than the P. rigida on upper hill, while the hybrid grew much better than the P. rigida on flat area. Another hybrid Populus alba ${\times}$ P. glandulosa grew faster than both parents by two to two and half times according to planting sites at age 10. Introduction of Pinus rigida also showed increased volume growth. Volume increase by selection of best five provenances among 45 at age 12 was estimated as 53 percent compare to progenies of plus trees in Korea, Additional 19 percent of volume increase was expected by selection of the best families within the best provenances. Annual production of chestnuts reached about 70,000 M/T by planting resistant clones to chestnut gall wasp (Dryocosmus kuriphilus), which killed almost all susceptible trees. Although polyploid trees and mutants have been produced by colchicine treatments in over 10 tree species, none of them is economically important Remarkable improvement of productivity is expected by biotechnology in future through selection, hybridization, introduction of foreign genes at cell, cell organelle and gene level, and gene transformation. At present, mass propagation of superior planting materials by tissue culture will increase the productivity.

Transgenic poplar expressing AtNDPK2 exhibits enhanced biomass in the LMO field

  • Nucleoside diphosphate kinase 2 (NDPK2) is known to regulate the expression of antioxidant genes and auxin-responsive genes in plants. Previously, it was noted that the overexpression of Arabidopsis NDPK2 (AtNDPK2) under the control of an oxidative stress-inducible SWPA2 promoter in transgenic poplar (Populus alba ${\times}$ P. tremular var. glandulosa) plants (referred to as SN plants) enhanced tolerance to oxidative stress and improved growth (Plant Biotechnol J 9: 34-347, 2011). In this study, growth of transgenic poplar was assessed under living modified organism (LMO) field conditions in terms of biomass in the next year. The growth of transgenic poplar plants increased in comparison with non-transgenic plants. The SN3 and SN4 transgenic lines had 1.6 and 1.2 times higher dry weight in stems than non-transgenic plants at 6 months after planting, respectively. Transgenic poplar also exhibited increased transcript levels of auxin-response genes such as IAA1, IAA2, IAA5 and IAA6. These results suggest that enhanced AtNDPK2 expression increases plant biomass in transgenic poplar through the regulation of auxin-response genes.

Extractives of the Wood of Black Locust and the Bark of Poplar as Decay-Resistant Hardwood Tree Species (내후성 활엽수종인 아까시나무 목질부와 현사시나무 수피의 추출성분)

  • Black locust(Robinia pseudoacacia) and poplar(Populus alba ${\times}$ glandulosa) trees were collected, extracted with acetone-$H_2O$(7:3, v/v) after drying, fractionated with hexane, chloroform and ethylacetate, and freeze dried to get some brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluting solvents. The wood extractives of black locust contained (+)-leucorobinetinidin as flavan, robtin as flavanone and dihydrorobinetin as flavanonol, and robinetin as flavonol. The poplar bark extractives contained various kinds of phenolic compounds : (+)-catechin as flavan, naringeoin, eriodictyol, sakuranetin, aromadendrin and taxifolin as flavanonol, salireposide as salicin derivative, and minor compounds such as aesculin and p-coumaric acid. However, aesculin has not been reported as a constituent of the poplar bark in Korea yet. NMR and FAB-MS analyses were done to elucidate the structures of isolated phenolic constituents.

  • Bio-kraft pulping of Populus alba${\times}$glandulosa pretreated with white-rot fungus, Phanerochaete chrysosporium KCCM 34740, was investigated The biopulping efficiency was evalued based on fungal pretreatment time and properties of bio-kraft pulp (pulp yield, freeness, WRY, kappa number and brightness) in comparison to the controls. Pretreatment of poplar wood chips with Phanerochaete chrysosporium KCCM 34740 for 10days resulted in a some increase in screened yield (by 2%). According to increase of fungal incubation time, decrease in freeness (CSF) and increase in WRY were observed And bio-kraft pulping also led to improvement of physical properties of handsheet. As a result of bio-beating effect, we expect the saving of chemicals in kraft cooking process and energy consumption in beating process.
