Influence of Ph and Temperature on Polyphenol Oxidase in the Leaves of Perilla frutescens var. japonica (들깨잎 폴리페놀 산화효소의 pH 및 온도에 의한 영향)

  • Polyphenol oxidase-catalyzed oxidation of substrates (t-butylcatechol, 4-methylcatechol, chlorogenic acid, caffeic acid and pyrocatechol) were performed in the Ph range 4~8. Co ncentrations of substrate's major oxidation products were monitored by high performance liquid chromatograph. The nature and amounts of products formed were highly pH dependent. They also were ifluenced by kinds of substrates. Major oxidation product of 4-methylcatechol appeared the maxium value at pH 5, them of chlorogenic acid, caffeic acid and pyrocatechol at pH 6.0 and that of t-butylcatechol at pH 5~7. Time-dependent PPO activity was determined at $4^{\circ}C\;and\;30^{\circ}C$. PPO extracted by phosphate buffer containing triton X-114 (t-PPO) was more stable than PPO by phosphate buffer (b-PPO). The result of electrophoresis, at first PPO was showed only a band at 48 kd. After 1~3 days a partial degrade band was appeared in b-PPO and three partial degrade bands in t-PPO. No activity band was appeared in PPOs at $30^{\circ}C$ and b-PPO at $4^{\circ}C$ after 4 days. And a band (37 kDa) in t-PPO was remained finally and disappered. PPO from Perillae leaves has two activity bands at 48 and 37 kDa in previous paper. It was supposed that PPO in the leaves of Perilla frutescens was a protein having one molecular weight as 48 kDa. And 37 kDa protein, relatively proteolysis-resistant, was a proteolyzed form of a major form.

Effects of Combined Treatment with Ultrasound and Ascorbic Acid on the Storage Qualities of Fresh-cut 'Jonathan'Apples (초음파와 ascorbic acid의 병용처리가 신선절단 '홍옥' 사과의 저장 중 품질에 미치는 영향)

  • The effects of ultrasound treatment, in combination with ascorbic acid, on the quality of fresh-cut 'Jonathan' apples was investigated. Prepared apple slices were ultrasonicated in distilled water (US) or in 1%(w/v) ascorbic acid solution (UA) and the other samples were just dipped in 1%(w/v) ascorbic acid solution (AA). All samples were stored at$10^{\circ}C$ for 12 days. UA-treated samples showed high $L^{*}$ and hue values and low $a^{*}$, $b^{*}$, chroma, and ${\Delta}E$ value. Both control and US-treated samples showed considerable browning. A significant inhibition of polyphenol oxidase activity was observed after UA treatment. The level of total phenolics in UA-treated samples was higher on the day of treatment compared with other samples. Total soluble solids, pH, titratable acidity, and gas concentrations were similar in all samples. This study demonstrated that the simultaneous treatment of ultrasound and ascorbic acid was effective in preventing enzymatic browning of fresh-cut 'Jonathan' apples and maintaining total phenolics contents.

Genotype and Environment Effects on Gliadin Content and Polyphenol Oxidase Activity in Wheat

  • The environment in which a given genotype is grown may influence its grain quality characteristics. When varieties are $\times$ evaluated over numerous environments, a variety environment interaction usually is observed, but the relative magnitude of environmental(E), genetic(G), and G $\times$ E effects on quality is unclear. In order to determine relative contribution of genotype, environment, and G $\times$ E interaction to the variations observed in grain quality characteristics, 18 Korean wheat cultivars and experimental lines were evaluated in two environments in 1998 and 1999. Correlation coefficients between grain quality and agronomic characteristics were also estimated. The analysis of variance for the optical density obtained by reaction bet- ween gliadin and anti-gliadin polyclonal antibody (AGPab) indicated that gliadin content measured by Enzyme-Linked Immunosorbent Assay(ELISA) was significantly in- fluenced by environment and cultivar differences. The significant differences of year and year $\times$ location were also found. The ratio of the variances associated with environmental effects to the variances associated with genetic effect gave relatively greater influence of environmental factor on gliadin content. The different protein content from same genotype grown in different environment might be associated with degree of storage protein accumulations. Significant relationships between ELISA and protein content, yield, ten spike weight, and ten spike number were detected. Polyphenol oxidase (PPO) activity was significantly influenced by year, location, cultivar and year $\times$ location. The variance in grain PPO activities among growing years appeared larger than the variation produced by the cultivar examined. This suggested that the growing environment contributed more to variability in grain PPO concentration.

Effect of Polyphenol Oxidase Activity on Discoloration of Noodle Dough Sheet Prepared from Korean Wheats

  • Polyphenol oxidase (PPO) is implicated in discoloration of white salted noodles and other wheat based foods. PPO activity was evaluated to determine the effect on discoloration of noodle dough sheets prepared from 25 Korean wheat flours during storage and to screen experimental lines with low PPO activity in 52 Korean wheats. PPO activity was assayed with whole-seed and performed with L-dihydroxyphenylalanine (L-DOPA) as substrates. Absorbance by L-DOPA assay of 25 Korean wheats was from 0.285 to 1.368 at 475 nm. PPO activity was significantly related with grain characteristics, including 1000-kernel weight and grain colors. In flour characteristics, PPO activity positively correlated with ash and protein content (r = 0.658, P < 0.001 and r = 0.424, P < 0.05, respectively) and negatively correlated with $L^*$ value of flour (r = 0.412, P < 0.05). In the changes of color of noodle dough sheet, $L^*$ and $b^*$ values consistently decreased and $a^*$ value increased during storage. PPO activity negatively correlated with $L^*$ value of noodle dough sheet during storage (r = 0.566, P < 0.01 at 2 hr, r = 0.547, P < 0.01 at 24 hr, and r = 0.509, P < 0.01 at 48 hr). But, no significant relationship was found in between PPO activity, $a^*$ and $b^*$ values during storage. The 52 Korean wheat lines examined in this study were divided into 3 different groups, low (< 0.500), medium (0.501-0.999) and high level (> 1.000), on the basis of the level of PPO activity. Twenty two Korean wheat lines showed low level of PPO activity and Suwon 252, 277 and 280 showed lower PPO activity (< 0.200) than others.

Antibrowning Effects of Electrolyzed Oxidizing Water with/without Freezing Point Depressing Agents on Peeled Chestnut during Storage (빙점강하제 첨가 전해산화수에 의한 깐밤의 저장 중 갈변억제 효과)

  • This study was to investigate the inhibiting effect of electrolyzed oxidizing (EO) water with/without freezing point depressing agents on polyphenol oxidase (PPO) activity of peeled chestnut. 0.85% sodium chloride, 0.5% citron and 0.5% lemon juice were used to freezing point depressing agents. The content of total phenolics was 13.36 mg% at the earlier stage of storage, and then suddenly increased at around 8∼1ldays. At the 11th day, PPO activity of untreated chestnut was 1,152 units, that was higher than any ethers. EO water adding lemon and citron juice showed synergistic effects on the enzyme inhibition, and their PPD activities were 143.3 and 180.22 units after 4 weeks, respectively. Sensory analysis showed that acceptance of peeled chestnuts was dependent on color and taste, which was related to PPO activity and sweetness. The peeled chestnut treated with EO water added citron or lemon juice tended to show the highest score fur acceptance.

Inhibition of Enzymatic Browning of Crown Daisy by Maillard Reaction Products (마이얄반응생성물(Maillard reaction product)에 의한 쑥갓의 효소적갈면 억제)

  • The study was conducted to investigate the effect of Maillard reaction products (MRPs) on enzymatic browning of crown daisy (Chrysanthmum coronarium var. spatiosum). The MRPs prepared by heating various amino acid and sugar at $90^{\circ}C$ caused a strong inhibitory effect on crown daisy polyphenol oxidase (PPO, ${\sigma}$-diphenol oxygen oxidoreductase, EC 1.10.3.1). As the reaction time of the solution containing glycine and glucose increased at $90^{\circ}C$, the production of MRPs was increased, whereas the amounts of glycine and glucose were decreased. Accordingly, the inhibitory effect of crown daisy PPO activity by MRPs was increased as the amounts of synthesized MRPs were increased. The MRPs synthesized from the various amino acids and sugars significantly reduced the PPO activity, particularly MRPs prepared by glutamine and xylose. The Michealis-Menten constant value ($K_m$) of crown daisy PPO with catechol as a substrate was 22.0 mM, and MRPs were a noncompetitive inhibitor against crown daisy PPO.

Effects of Wounding and Jasmonic Acid on Polyphenol Oxidase in Tomato Seedlings (토마토 유식물의 Polyphenol Oxidase에 미치는 상해 및 Jasmonic Acid의 영향)

  • The effects of wounding and jasmonic acid(JA) on polyphenol oxidase(PPO) in tomato(Lycopersicon esculentum Mill.) seedlings were investigated. PPO was strongly induced by wounding or JA, and the response was also shown to be systemically induced by wounding. Mechanical wounding in cotyledon or leaf produced a signal that caused the concentration of PPO to increase in the unwounded cotyledon, in the first leaves but not in the second leaves. Severity of wounding and light intensity also affected wound induced change in PPO activity, JA showed a stimulatory effect on the loss of chlorophyll and the rapid increase in PPO activity. The PPO was clearly more active in the wounded leaves than in controls. The potency and specificity of the JA indicate a close relationship between JA and wound-induced changes in PPO in tomato species. JA and abscisic acid(ABA) acted similarly on both unwounded and wounded leaves, but the amount of PPO in the wounded leaves was always more than the respective controls. The highest increase in PPO activity occurred in woundand JA-induced leaves of seedlings kept under bright lighting. Benzyladenine(BA) completely abolished JA- and ABA-induced PPO activity. The results suggest that JA-induced PPO activity is due to de novo PPO synthesis. Histochemical tests for PPO in stems of wound- and JA -treated tomato plants indicate that PPO was localized primarily, in the. outer .cortex . and xylem parenchyma. It is concluded that exogenously applied JA acts as stress agents and PPO may be a component of the inducible anti-hervivore defense response.

Changes of Enzyme Activities and Inorganic Nutrient Contents Associated with Flower Development in Tulip (Tulipa gesneriana) (튤립(Tulipa gesneriana) 꽃의 발달단계에 따른 효소 활성 및 미량요소 함량의 변화)

  • This study was carried out to investigate the changes of enzymes and micro inorganic nutrients that is associated with flower senescence during flower development in tulip cultivars, ‘Apeldoorn’ and ‘Golden Apeldoorn’. Ribonuclease, peroxidase and protease activities were gradually increased from the stage of early flowering to later Polyphenol oxidase showed the highest activity at stage 5, which the flower was in full bloom indicating that it acts at an initial stage of flower senescence. The protease activity was different in the petal extracts during flower development between the cultivars ‘Apeldoorn’ (red petal) and ‘Golden Apeldoorn’ (yellow petal). This result suggested that protease might relate to pigment biosynthesis in petal of tulip. In contrast to the decrease of inorganic nutrients K, Mn, Zn and P contents during floral development, Ca, Mg and Fe showed the gradual increasement that is similar with ribonuclease, peroxidase and protease. It suggests that they have some interaction during flower senescence.

Effects of $\beta$Carotene on the Browning of Phenolics Extracted from Potato and Polyphenol Oxidase Characteristics (감자에서 추출한 phenolics의 갈변화에 미치는 $\beta$carotene의 영향과 polyphenol oxidase 특성)

  • The effects of synthetic and carrot carotene on the browning of polyphenolics extracted from potato for eliminating the effects of other components in potato which inhibit the browning of potato juice and the potato polyphenol oxidase (PPO) activity were investigated. Total phenolics content in potato sample was 1.854mg/g D.W. and PPO activity was 100 unit. Delta 'L'value of polyphenolics extracted from potato decreased makedly, but that of potato with carrot decreased little by little. Potato with P-carotene showed a little decrease after 50 min. At the same time, polyphenolics extracted from potato were mixed with carotene extracted from carrot or with syntetic $\beta$-carotene. As the results, the delta 'L'value of the former increased but decreased similarly to the latter after 1 hour. The effects of enzyme and substrate concentration on the browning of PPO extracted from potato were investigated. Optimum enzyme concentration was 10% and optimum substrate concentration was 13.3% $\beta$-carotene concentration did not appear to influence significantly on PPO activity.

Isolation of a Novel Polyphenol from Oolong Tea and Its Effective Prevention of the Gout (우롱차로터 새로운 Polyphenol 분리 및 통풍 예방 효과)

  • Eighty percent of acetone extract were isolated from the leaves of Korean Oolong tea using Sephadex LH-20, MCI-gel, Fuji gel. The compound reacted the red and the blue with anisaldehyde and $FeCl_3$, respectively. Instrumental analysis of the derivatives of this compound showed that the chemical structure was decided to $epicatechin-(4{\beta}{\rightarrow}8)-epigallocatechin-3-O-gallate$ as a kind of dimeric proanthocyanidine, that bound with -(-)epicatechin and -(-)epigallocatechin-3-O-gallate, at the upper and the lower, respectively. Considering inhibition effect on xanthin oxidase by 62% levels at $50{\;}{\mu}mole$, this compound showed a possibility to be used as a new material for functional food.
