Recent industrial society has human widely exposed to PAHs (polynuclear aromatic hydrocarbons) that are comming from the incomplete combustion of organic material as wider spread environmental contaminants. Biological activities of PAHs are not known although PAHs are considered as carcinogens. Our laboratory have been studied the effect of PAHs in the mouse liver hepa 1 cells. In this study, we examined the mouse liver hepa-l cells as a new bioassay system to evaluate bioactivity of PAHs. We have selected 13 PAHs to examine bioassay using cyp1a1-luciferase reporter gene expression system where cyp1a1 1.6 Kb 5flanking region DNA was cloned in front of luciferase reporter gene and this plasmid was transfected into hepa 1 cells transiently. This cells then used for the study to observe the effect of PAHs. We demonstrated that PAHs induced the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase (EROD) activities in a concentration-dependant manner. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD. Acenaphthene, anthracene, fluorine, naphthalene, pyrene, phenanthrene, carbazole were weak responders to cyp1a1 promoter activity stimulation and EROD induction in hepa 1 cells and these chemicals seemed to respond less to EROD than cyp1a1 promoter activity. Benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, chrysene, and dibenzo(a,h)anthracene showed strong response to cyp1a1 promoter activity stimulation and also EROD induction in hepa 1cells. Results of dose response study suggested that four strong responding PAHs, such as benzo(a)anthracene benzo(k)fluoranthene, chrysene, and dibenzo(a, h)anthracene might be mediated through arylhydrocarbon receptor system in hepa1 cells.
Due to their slow degradation properties, hydrophobic organic contaminants in estuarine sediment have been a concern for risks to human health and aquatic organisms. Studies of fate and transport of these contaminants in estuaries are further complicated by the fact that hydrodynamics and sediment transport processes in these regions are complex, involving processes with various temporal and spatial scales. In order to simulate and quantify long-term attenuation of Polycyclic Aromatic Hydrocarbons (PAH) in the Elizabeth River, VA, we develop a modeling approach, which employs the U.S. Environmental Protection Agency's water quality model, WASP, and encompasses key physical and chemical processes that govern long-term fate and transport of PAHs in the river. In this box-model configuration, freshwater inflows mix with ocean saline water and tidally averaged dispersion coefficients are obtained by calibration using measured salinity data. Sediment core field data is used to estimate the net deposition/erosion rate, treating only either the gross resuspension or deposition rate as the calibration parameter. Once calibrated, the model simulates fate and transport PAHs following the loading input to the river in 1967, nearly 4 decades ago. Sediment PAH concentrations are simulated over 1967-2022 and model results for Year 2002 are compared with field data measured at various locations of the river during that year. Sediment concentrations for Year 2012 and 2022 are also projected for various remedial actions. Since all the model parameters are based on empirical field data, model predictions should reflect responses based on the assumptions that have been governing the fate and sediment transport for the past decades.
Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in the environment. They are highly toxigenic and carcinogenic. Probiotic bacteria isolated from fermented foods were tested to check their ability to degrade and/or detoxify PAHs. Five probiotic bacteria with distinct morphologies were isolated from a mixture of 26 fermented foods co-cultured with benzo(a)pyrene (BaP) containing Bushnell Haas minimal broth. Among them, B. velezensis (PMC10) significantly reduced the abundance of BaP in the broth. PMC10 completely degraded BaP presented at a lower concentration in broth culture. B. velezensis also showed a clear zone of degradation on a BaP-coated Bushnell Haas agar plate. Gene expression profiling showed significant increases of PAH ring-hydroxylating dioxygenases and 4-hydroxybenzoate 3-monooxygenase genes in B. velezensis in response to BaP treatment. In addtion, both live and heat-killed B. velezensis removed BaP and naphthalene (Nap) from phosphate buffer solution. Live B. velezensis did not show any cytotoxicity to macrophage or human dermal fibroblast cells. Live-cell and cell-free supernatant of B. velezensis showed potential anti-inflammatory effects. Cell-free supernatant and extract of B. velezensis also showed free radical scavenging effects. These results highlight the prospective ability of B. velezensis to biodegrade and remove toxic PAHs from the human body and suggest that the biodegradation of BaP might be regulated by ring-hydroxylating dioxygenase-initiated metabolic pathway.
Background : Polycyclic aromatic hydrocarbons (PAH) are well known environmental pollutants. The measurement of PAH in ambient air is not commonly used, because it is quite difficult to perform and is unreliable. Using biomarkers of PAH can be an alternative approach to this problem. The PAH in ambient air is absorbed in particulate matter. Total suspended particulate(TSP) or particulate matter of less than $10{\mu}m$ in diameter (PM10) can be easily measured. Therefore, TSP or PM10 can be used as a surrogate measurements of ambient air PAH. Objectives : We investigated whether the urinary concentration of two biomarkers of PAH, 1-hydroxypyrene (1-OHP) and 2-naphthol, could reflect the total suspended particulate in the general population. Methods : In order to exclude the effects of occupational exposure and smoking, first grade middle school students were included in this study. Four middle schools within a one kilometer boundary of ambient air monitoring stations were selected. Total suspended particulate was regarded as the marker of airborne PAH. Diet and smoking data were collected by self administered questionnaires, and spot urine samples were collected. Urinary 1-OHP and 2-naphthol were analyzed by high performance liquid chromatography. Results : The correlation between urinary 1-OHP, 2-naphthol and passive smoking was not statistically significant. The correlation between urinary 1-OHP and TSP indices was not statistically significant. The correlations between urinary 2-naphthol and TSP of two lag days, one lag day, and zero lag days were statistically significant. The statistical significance of two lag days was the strongest (p=0.001), one lag day was the next (p=0.0275), and zero lag days was the weakest (p=0.0349). Conclusion : Our results imply that the urinary concentration of 2-naphthol can be applied as a PAH exposure marker for the general population with low PAH exposure.
In the soil washing process, the contaminants are usually removed by abrasion from soil particles using mechanical energy and water However, organic contaminants with low water solubility like polycyclic aromatic hydrocarbons (PAH) are remained on soil particles. Previous studies have shown that surfactant possessing amphipathic activity enhances the solubility of organic materials. For this reason solutions with surfactants have been used to improve removal of organic contaminants on soil washing process. But, in this manner, many problems were found like complete loss of surfactants and additional contamination by surfactant. The remediation method using microemulsion has been introduced to overcome these disadvantages. In this case, surfactants are recycled by phase separation of microemulsion after remediation. In microemulsion process, the surfactant will be recycled by phase separation of the microemulsion into a surfactant-rich aqueous phase and an oil phase after extraction. That is why remediation concept applying microemulsion as washing media has been Introduced. Suitable microemulsion have to be used in order to have the chance of refilling the soil after decontamination and to avoid any risk due to toxicity. The purpose of this research is to evaluate effect of microemulsion to remediation of contaminated soil. Polycyclic aromatic hydrocarbons (PAHs) in Galactic planetary nebulae (PNe) are investigated by means of the unidentified infrared (UIR) bands. Continuous near- to mid-infrared spectra of PNe are obtained with the AKARI/IRC and the Spitzer/IRS. All 19 PNe in the present study show prominent dust emissions and we investigate the variation in the intensity ratios among the UIR bands. The ionization fraction and the size distribution of PAHs in PNe are derived using the UIR band ratios. We find that the ionization fraction of PAHs in PNe is around 0.0-0.6 and that small PAHs are scarce. The present result indicates a systematic trend of the $3.4{\mu}m$ aliphatic feature to become weak as the PAH ionization fraction increases.
We are creating all-sky diffuse maps from the AKARI mid-infrared survey data with the two photometric bands centered at wavelengths of 9 and $18{\mu}m$. The AKARI mid-infrared diffuse maps achieve higher spatial resolution and higher sensitivity than the IRAS maps. In particular, the $9{\mu}m$ data are unique resources as an all-sky tracer of the emission of polycyclic aromatic hydrocarbons (PAHs). However, the original data suffer many artifacts. Thus, we have been developing correction methods. Among them, we have recently improved correction methods for the non-linearity and the reset anomaly of the detector response. These corrections successfully reduce the artifact level down to $0.1MJy\;sr^{-1}$ on average, which is essential for discussion on faint extended emission (e.g., the Galactic PAH emission). We have also made progress in the subtraction of the scattered light caused in the camera optics. We plan to release the improved diffuse maps to the public within a year.
Soil samples were obtained from 5 sites contaminated with polycyclic aromatic hydrocarbons (PAHs). These soil samples were cultured in using phenanthrene as a sole carbon and energy source, and 36 strains of phenanthrene-degrading bacteria were isolated from 3 sites. Most of them degraded 500 ppm of phenanthrene within 8 to 10 days, and these isolates could degrade a few other PAHs other than phenanthrene. Their genotypes were determined by restriction digests of the l6S rRNA genes [amplified ribosomal DNA restriction analysis (ARDRA)]. It was found that all the phenanthrene degrading isolates were included in 4 ARDRA types, and they showed a strict site endemism. l6S rDNAs of 12 strains selected from different sites were sequenced, and they were all confirmed as Sphingomonas strains. Their l6S rDNA sequences were compared for phylogenetic analysis; their sequence showed a similar result to ARDRA typing, thus indicating that these heterotrophic soil bacteria are not regionally mixed. In addition, it was found that the microbial diversity among sampling sites could be monitored by l6S rDNA PCR-RFLP pattern alone, which is simpler and easier to perform, without l6S rDNA sequence analysis.
Bioremediation of hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), is a major environmental concern due to their toxic and carcinogenic properties. Due to their hydrophobicity, the hydrophobic organic compounds are mainly associated with the soil organic matter or nonaqueous-phase liquids. A major question concerns the relationships between biodegradation and sorption. This work develops and utilizes a non- steady state model for evaluating the interactions between sorption and biodegradation of phenanthrene, a 3-ring PAH compound, in soil-slurry systems. The model includes sorption/desorption of a target compound, its utilization by microorganisms as a primary substrate existing in the dissolved phase and/or the sorbed phase in biomass and soil, oxygen transfer, and oxygen utilization as an electron acceptor. Biodegradation tests with phenanthrene were conducted in liquid and soil-slurry systems. The soil-slurry tests were performed with very different mass transfer rate: fast mass transfer in a flask test at 150 rpm, and slow mass transfer in a roller-bottle test at 2 rpm. In the slurry tests, phenanthrene was degraded more rapidly than in liquid tests, but with a similar rate in both slurry systems. Modeling analyses with several hypotheses indicate that a model without biodegradation of compound sorbed to the soil was not able to account for the rapid degradation of phenanthrene, particularly in the roller bottle slurry test. Reduced mass-transfer resistance to bacteria attached to the soil is the most likely phenomenon accounting for rapid sorbed-phase biodegradation.
The soot formation characteristics of various alkane-based single fuel droplets were studied in this work. Also, This study was performed to provide the database of the soot behavior and formation of alkane-based single fuel droplet. The experimental conditions were set to 1.0 atm of ambient pressure ($P_{amb}$), 21% of oxygen concentration ($O_2$) and 79% of nitrogen concentration ($N_2$). Combustion and soot formation of single fuel droplet was visualized by visualization system with high speed camera. At the same time, ambient pressure, oxygen concentration and nitrogen concentration were maintained by ambient condition control system. Soot formation characteristics was analyzed and compared on the basis of intensity ratio ($I/I_0$) of background image. The results of toluene fuel droplet showed the largest soot generation. Soot volume fraction ($f_v$) was almost the same under the identical fuel types regardless of various initial droplet diameter ($d_0$) since thermophoretic flux was not much changed under the same ambient conditions.
