Estimation of Indicator PCBs Exposure in Infants via Breast-Feeding Period (수유기간에 따른 모유 중 Indicator PCBs 분석을 통한 영유아의 인체노출평가)

  • The levels of indicator PCBs were determined in 132 human milk samples (Colostrum, 30th days after delivery, 60th days after delivery) collected in 2006 according to World Health Organization's (WHO) protocol. In addition, concentrations of indicator polychlorinated biphenyls (PCBs) were measured by the isotope dilution method with high resolution gas chromatograph/high resolution mass detector (HRGC/HRMS), which gave accurate and precise data for estimations of international trend and comparisons. Average daily intakes (ADIs) of all analytes were calculated. The median level of total indicator PCBs was 14 ng/g lipid, which is significantly lower than the level found in individuals from other countries. PCB congeners 138, 153 and 180 accounted for about 65% of the sum of the indicator congeners analyzed. The concentration of the most abundant PCB 153 is positively correlated with the concentration of the sum indicator PCBs (p<0.001). In this study, concentrations of total indicator PCBs significantly decreased with increasing number of children for a mother, indicating that parity and breast-feeding period an important role in the accumulation and elimination of these compounds in lactating women. The ADI for the median of total indicator PCBs for an infant was evaluated at $0.077{\mu}g/kg{\cdot}bw/day$ from birth to 6 months of age. The present study shows that concentrations of PCB 118 primipara breast milk have decreased 25~35 since 1998, and that this trend is continuing.

A Study on Polybrominated Diphenyl Ethers and Organochlorine Compounds Residues Persistent in Korean Coastal Bivalves (한국 연안 이매패류에 잔류하는 PBDEs 및 OCs에 관한 연구)

  • Bivalve samples from 30 locations Korean coastal waters were analyzed to characterize the contamination with and distribution of polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). The congeners of PBDE in 30 bivalve samples were analyzed by high resolution gas chromatography with a high resolution mass detector. The levels of PBDEs in bivalve samples ranged from 0.092 to 13.5 ng/g wet weight. These values for PBDEs concentrations in bivalves from Korea were higher than or comparable to available values for bivalves from other countries. In the congener profiles, BDE-47 (mean contribution=42.2%) was predominant, followed by BDE-99 (23.1%), BDE-100 (11.9%) and BDE-154 (3.5%). Among the OCs analyzed, $\Sigma$PCBs and $\Sigma$DDTs levels were highest, followed by $\Sigma$HCHs, $\Sigma$CHLs and HCB. The wet weight based $\Sigma$PCBs and $\Sigma$DDTs ranged from 0.28 ng/g to 19.4 ng/g, and from 0.19 ng/g to 4.86 ng/g, respectively. The $\Sigma$PCBs and $\Sigma$DDT slevels in bivalves along the entire Korean coast were lower than those of the USA, and below the Food and Drug Administration (USA. FDA) limits for fish and shell fish. However, the human health criteria and wild life criteria of the USA. Environmental Protection Agency (USA. EPA) were exceeded for DDE and $\Sigma$HCHs at most of the sampling points.

The Effects of Physico-chemical Properties of soils on PCBs Analysis (토양의 이화학적 특성이 PCBs 분석에 미치는 영향)

  • The effects of physico-chemical properties of soils on PCBs analysis in Korea was studied Three kinds of extraction solvents(toluene, acetone:hexane, dichloromethane) was selected to apply three different soils. The silicagel, florisil and alumina column cleanup also performed to compare the elution patterns, and three different methods (EPA, Japan, Proposed method in this study) of 62 PCBs also compared the individual peaks recoveries. The solvent average recovery was surveyed the order of toluene, acetone:hexane, dichloromethane as 77.94%, 58.59%, 54.20% for soil A, 53.65%, 80.32%, 68.27% for soil B and 44.52%, 60.35%, 56.36% for soil C, respectively. The average recovery was depended on the soil characters. The highest recoveries of each soil were obtained the toluene for soil A, acetone:hexane for soil B and C. However, the coplanar PCBs was obtained the highest recovery with dichloromethane. Thus, to select the solvent for the analysis of PCBs in solid, the selected compounds have to consider to get good result. The silicagel, florisil, alumina I and alumina II column cleanup process were surveyed the range of 38.73%~98.26%, and the higher chlorinated compounds was obtained the lower recovery compared to the low chlorinated compounds, generally. This results are also consistent with the coplanar PCBs isomers. The compared results of three different countries were obtained the 37.15% for USA, 45.92% for Japan and 44.46% for proposed method in this study.

Distribution and Human Risk Assessment of Polycyclic Aromatic Hydrocarbons and Polychlorinated Biphenyls in Sediments and Fish from Suyoung Estuary of Busan, Korea (부산 수영강 하구역의 퇴적물과 어류에서 다환방향족탄화수소 및 폴리염화비페닐의 농도분포와 인체 위해도 평가)

  • Distributions of 15 PAHs and 82 PCBs were investigated in sediment and fish samples collected from the Suyoung Estuary of Busan, one of the most urbanized and newly contaminated areas in Busan. The concentrations of $S_{15}PAH$ in sediments ranged from 17.9 to 777 ng/g-dry and were mainly originated from combustion processes. The concentrations of $S_{82}PCB$ in sediments ranged from 0.55 to 12.5 ng/g-dry and were significantly correlated with those of 12 dioxin-like PCBs and 6 non-dioxin like PCBs. Higher concentrations of PAHs and PCBs were found in the upper river and the inner Suyoung Bay than in marina and the outer Suyoung Bay. No sites exceeded the ecotoxicological values of PAHs and PCBs in marine sediments. Benzo[a]-pyrene was not detected in fish samples and the concentrations of $S_4PAH$ (0.15~0.45 ng/g-wet) were lower than the maximum level set by EU. The levels of PCBs in fish samples were at less than 1% of the Korean maximum level and 2% of the EU maximum level. The concentrations of dioxin-like PCBs were $0.01{\sim}0.32pg-TEQ_{WHO-2005}/g-wet$, and the highest concentration was found in eels, which were at less than 5% of the EU maximum level. Dietary intakes of PAHs and PCBs through fish consumption were estimated, and their lifetime cancer risk and non-cancer risk were much lower.

Evaluation of accuracy for measurement of Dioxins (PCDDs/PCDFs) by using certified reference material (CRM) (인증표준물질(Certified reference materials, CRM)을 이용한 이옥신류(PCDDs/PCDFs) 측정의 정확도 평가)

  • In our study, the accuracy for measurement of seventeen 2,3,7,8-substituted PCDDs/PCDFs in certified reference material (CRM) which is the sample of homogeneous sediment matrix taken from an area known to have significant chemical contamination, particularly PCBs (polychlorinated biphenyls), was evaluated. Though the methodology in this study followed the official method of unintentionally produced persistent organic pollutants (UPOPs) announced by the Ministry of Environment of the Republic of Korea in 2007, there were slight changes using additional purification step by activated carbon column because the interferences of sample were not sufficiently removed when only multi-silica column and alumina column have been used for purification. The |En| number proposed by the Korea Research Institute of Standards and Science was used for a valuation basis of the accuracy. The |En| numbers of seventeen 2,3,7,8-substituted PCDDs/PCDFs have been indicated as 1 and below, they were decided "Pass" in this test, when DB-5MS column and SP-2331 column were used together. Because 1,2,3,7,8-PeCDD and #169-HxCB were not separated on DB-5MS column, the ions of 1,2,3,7,8-PeCDD were selected at M/M+2 instead of M+2/M+4 suggested by EPA 1613. It is possible to distinguish them in HRGC/HRMS analysis.

Reductive Dechlorination of Low Concentration Polychlorinated Biphenyls as Affected by a Rhamnolipid Biosurfactant

  • We investigated whether the threshold concentration for polychlorinated biphenyl (PCB) dechlorination may be lower in biosurfactant-amended sediments compared with biosurfactant-free samples. At PCB concentrations of 40, 60, and 120 ppm, the surfactant amendment enhanced the PCB dechlorination rate at all concentrations and the rate was also faster at higher concentrations. On a congener group basis, dechlorination proceeded largely with group A (congeners with low threshold) in both surfactant-free and -amended sediments, accumulating mainly group C (residual products of dechlorination) congeners, and surfactant enhanced the dechlorination rate of group A congeners. Since the PCB threshold concentration for the inoculum in the experiment was lower than 40 ppm, we carried out another experiment using sediments with lower PCB concentrations, 10, 20, and 30 ppm. Sediments with 100 ppm were also performed to measure dechlorination at a PCB saturation concentration. Comparison between the plateaus exhibited that the extent of dechlorination below 40 ppm PCBs was much lower than that at a saturation concentration of 100 ppm. There was no significant difference in the extent of dechlorination between surfactant-free and -amended sediments. Moreover, surfactant did not change the congener specificity or broaden the congener spectrum for dechlorination at PCB concentrations below 40 ppm. Taken together, it seems that at a given PCB concentration, dechlorination characteristics of dechlorinating populations may be determined by not only the congener specificity of the microorganisms but also the affinity of dechlorinating enzyme(s) to individual PCB congeners.

Horizontal Distributions and Their Ecotoxicological Implications Relating to PCBs, DDTs, HCHs, and HCB in Sediments in the West Nakdong River (서낙동강 수계 퇴적물 내 PCBs, DDTs, HCHs 및 HCB의 수평적 분포와 잠재적 생태독성 영향 평가)

  • Horizontal distributions and their ecotoxicological implications relating to organochlorine compounds (OCs) were determined in surface sediments sampled from the West Nakdong River. The concentrations of polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethanes (DDTs), hexachlorocyclohexanes (HCHs), and hexachlorobenzene (HCB) in surface sediments were in the range of 0.57-3.68, 0.41-8.35, 0.63-1.65, and n.d.-0.56 ng/g, respectively. DDTs represent the major OC in the West Nakdong River. Concentrations of PCBs and DDTs correlate with the total organic carbon (PCBs: r=0.874, p<0.001, and DDTs: r=0.762, p<0.05, respectively), indicating that organic content in the sediments is one of the major factors for influencing horizontal distribution of the organochlorine compounds in this river. Examining the potential poisonous character of the surface sediments to benthic organisms according to the sediment quality guideline (SQG), set out by the Canadian Council of Ministers of the Environment (CCME) and the National Oceanic and Atmospheric Administration (NOAA), we found that the concentrations of total PCBs, p,p'-DDD, and ${\gamma}$-HCH were lower than the effects range-low (ER-L) and the probable effects level (PEL). As a result, it is considered that the effect of organochlorines in the region considered on benthic organisms is very low from the perspective of environmental toxicology. However, in terms of the Canadian SQC, p,p'-DDE, p,p'-DDT, and total DDTs, respectively, exceeded the interim sediment quality guidelines (ISQG) and the threshold effect level (TEL) in 30%, 17%, and 19%, respectively, of the sediment samples analyzed.

  • Pollutants that are persistent, bioaccurnulative, and toxic have been linked to numerous adverse effects in human and animals, PBTs include heavy metals, polychlorinated biphenyls (PCBs), dioxins, polycyclic aromatic compounds (PACs) in addition to pesticides. This study focuses on toxic effects of the PBTs except pesticides on insects. Eight PBTs were selected from subgroups: three heavy metals (Pb, Hg, and Cd), two PCB mixtures (Aroclor mixtures 1 and 2), 2,3,7,8-tetrachlorodibenzo-p-dioxin, two monophenols (4-octylphenol and 4-nonylphenol), and tetrabutyltin, Beet armyworm, Spodoptera exigua, was used as test target insect species. Three physiological markers (metamorphosis, immune reaction, and follicle patency) were assessed in each exposure to different doses of the PCBs. Heat-shock proteins as molecular markers were also analyzed in response to the PCBs. All tested PBTs were toxic to metamorphosis from larvae to pupae when they were applied with diet. Two PCB mixtures were the most toxic compounds in this assay by giving significant toxicity at 0.005 ppm, while others had from 10 to 1000 ppm. Dioxin (0.1 ppb), tetrabutyltin (0.1 ppb), Pb (10 ppb), and Hg (0,01 ppb) were potent to inhibit immune reactions analyzed by inducing phenoloxidase activity and blocked phospholipase $A_2$ enzyme, Tetrabutyltin and dioxin significantly induced follicle cell patency, but their effects were lower than that of endogenous juvenile hormone, Dioxin, Pb, Hg, and Cd could induce the expression of heat shock proteins that were detected by immunoblotting against human HSP70 monoclonal antibody. HSP78 and HSP80 were upregulated in response to the PBTs. This expression was detected from the fat body and epidermis at as fast as 4h after injection. All these results clearly suggest that PBTs give significant ecotoxicity to insects that are valuable organisms in our environment.

Distribution of Organochlorines and PCB Congeners In Korean Human Tissues

  • In order to investigate the residual amounts of organochlorines and polychlorinated biphenyls (PCBs) in Korean human tissues (blood, adipose tissue, liver, kidney cortex, and lung), the samples were collected from the autopsied cadavers of 40 men and 40 women (from teens to seventies of age). ${\alpha}-BHC,\;{\beta}-BHC,\;{\gamma}-BHC,\;{\delta}-BHC$, p,p'-DDT, p,p'-DDD, p,p'-DDE, endrin, dieldein, aldrin, and 7 marker PCBs (28, 52, 101, 118, 138, 153, and 180) were determined in human tissues. The levels of organochlorines and PCB congeners indicated that they have been widely distributed in Korean human body. Positive correlations in terms of age were observed for the following cases: p,p'-DDE, p,p'-DDT, ${\Sigma}-DDT$, PCB 118, PCB 138, PCB 153, and ${\Sigma}-PCB$ in the adipose tissue, and p,p'-DDE in the lung. Concentration of these compounds showed a significant age-related increase. Accumulation of these compounds in aged people revealed that these compounds were more slowly eliminated in our environment and risk assessment was necessary for further proper action. Significant differences in the levels of PCBs between genders were found for PCB 118 in the adipose tissue and PCB 138 in the liver. Positive correlation coefficients between tissues were detected with p,p'-DDE and ${\beta}-BHC$.

Analysis of dioxin-like PCBs in Soil samples (토양 중 dioxin-like PCBs의 분석)

  • The analytical method of 14 kinds of coplanar - PCBs was established and applied the soil sample. The three kinds of extraction solvents (toluene, acetone: n-hexane, dichloromethane) were selected to apply the soil sample. The silica gel, florisil and alumina column cleanup also performed to compare the elution recovery. The average recovery of selected solvents in soil A, B and C was surveyed the 84.25%, 56.09% and 44.69% for toluene, 52.56%, 81.42% and 58.53% for acetone : n - hexane and 55.94%, 71.33% and 61.05% for dichloromethane. The average recovery is represented 49.99% for silica gel (n - hexane 100 mL), 69.65% for florisil (6% ether/n - hexane 100 mL), and 65.23% for alumina (2% DCM : n - hexane 100 mL, 50% DCM: n-hexane 150 mL). In silica gel (n - hexane) and florisil (6% ether : n - hexane) cleanup, the 14 kinds of coplanar PCBs eluted until 40 mL. In the silica gel and florisil columns cleanup, the amounts of elution solvent can be reduced from these results, but the researcher has to confirm the elution amounts before performing the experiments. In alumina cleanup process, the result was obtained to the 100 mL of elution solvents (2% DCM: n-hexane 100 mL and 50% DCM: n-hexane 40 mL), therefore the change of elution solvent is necessary to develop the simple procedure.
