Turbidity Reduction for Construction Runoff Using Polyacrylamide (고분자응집제를 이용한 건설현장 탁수처리)

  • 많은 건설사업장의 경우 토사 유출에 따른 고농도 탁수가 발생하며, 그 탁도를 낮추기 위해 침전지 혹은 토사탈수백 등의 Best Management Practices(BMPs)를 이용한다. 본 연구에서는 유입되는 탁수에 고분자응집제인 polyacrylamide(PAM)를 액상으로 주입하고, 침전지와 토사탈수백을 통해 탁도를 저감하는 field-scale 실험을 수행하였다. 일반적으로 침전지 중간에는 유입되는 물의 힘을 분산시키기 위해 배플이라는 구조물이 설치되는데, 본 연구결과에 따르면 PAM을 통한 화학적 처리 없이 배플 자체로는 탁도를 낮추는 데 그 효과가 낮았으나, PAM주입을 할 경우 배플 설치 여부에 상관없이 90-98%까지 방류수 탁도를 낮출 수 있는 것으로 나타났다. 침사지 이후에 설치한 토사탈수 백은 그 효과가 낮았으나, 침사지 없이 토사탈수백만 사용할 경우 PAM을 이용한 화학적 처리 없이는 만족할 만한 방류수 탁도를 기대하기 어려웠다. 본 연구결과는 향후 미국에서 면적 10에이커 이상의 공사현장에 적용되는 탁도 규제에 대한 대응 방안으로 PAM을 이용한 탁수 처리의 효용성을 보여준다.

Efficient Treatment of Sewage Sludge by Effective Microorganisms (유효미생물에 의한 하수슬러지의 효율적 처리)

  • This study was conducted to evaluate the effects of microbial inoculation on sewage sludge composting. The number and species of microorganisms in sewage sludge sampled on February were higher than those sampled on August. The composting of sewage sludge is inhibited by the polyacrylamide cation, which was used as a coagulant and known to repress the growth of microorganisms. The growth of all microorganisms was inhibited by the addition of the polyacrylamide cation at a concentration of more than 0.8%. The species and viable counts of microorganisms were observed to increase during composting sewage sludge by inoculation of the effective microorganisms and addition of the pine tree sawdust as a bulking agent, compared with those without inoculation. A variety of organisms in compost(sewage sludge plus sawdust) were observed after composting for 30 days, such as Fragilaria sp., Proales sp., Vorticella sp., Schizothrix sp., Anabaena sp., Zoothaminium sp., Epstylis sp., Arcella sp., Balantidium sp., Actinophrys sp., Synedra sp., Euglypha sp., Ulothrix sp., Anacystis sp., and Clostium sp.

Purification of Isocitrate lyase Produced from Microbacterium laevaniformans (Microbacterium laevaniformans가 생성하는 Isocitrate lyase의 정제)

  • Purification of the isocitrate lyase extracted from Microbacterium laevaniformans was investigated. The isocitrate lyase was purified 43.6 folds by the following continuous treatment with ammonium sulfate fraction, DEAE-cellulose, DEAE-sephacel and Sephadex G-200 chromatography. The purified isocitrate lyase was showed to be a single protein band by polyacrylamide gel electrophoresis. The molecular weight of the purified isocitrate lyase was estimated 54,000 Da by the SDS-polyacrylamide gel electrophoresis. The Km and Vmax values for isocitrate were estimated to be 0.83mM and 0.33units/ml, respectively. Activity of isocitrate lyase was inhibited by cystein-HCl and glutathione.

  • The neurotoxin of Clostridium botulinum type B was purified from a liquid culture. The purification steps consist of ammonium sulfate precipitation of whole culture, treatment of Polymin P(0.15%, v/v), gel filtration on Sephadex G-100 at pH5.6 and DEAE-Sephadex charomatography at pH8.0. The procedure recovered 17% of the toxin assayed in the starting culture. The toxin was homogeneous by sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis and had a molecular weight of 163,000. Subunits of 106,000 and 56,000 molecular weight were found when purified toxin was treated with a disulfide-reducing agent and electro phoresed on SDS-polyacrylamide gels.

  • DEAE-Cellulose, Sephadex column chromatography and polyacrylamide gel electrophoresis were used to purify acid phosphatase, aryl sulfatases, ${\beta}-glucuronidase$ and cathepsin D in n-butyl alcohol extracts of porcine leukocyte Iysosomes. The degree of purification was quite high for all enzymes studied and some could be identified by histochemical reactions.

  • L-PHA, a lectin having lymphoagglutinating activity but devoid of erythroagglutinability which is contained in Korean white kidney bean (Phaseolus vulgaris L.), was isolated and purified through several techniques such as ion exchange chromatography, hydroxyapatite column and affinity chromatography. Purified L-PHA was identified as a single band by polyacrylamide disc gel electrophoresis. The molecular weight of the L-PHA was estimated about 125, 000 daltons by polyacrylamide gel electrophoresis and it was turned out having one subunit (probably dimer of the Yachnin's model) of Mr~60, 000. Immunochemical studies also tried and these results reconfirmed the purity of this L-PHA.

  • This experiment was carried out to investigate the change in protein of pasteurized whole egg with or without added 1% of sugar during fermentation S. lactis, L. casei and S. faecalis were used in this fermentation test. Whole egg showed no phenomenen of coagulation by heating at $58^{\circ}C$ for 30 min, but the patterns in polyacrylamide gel of pasteurized whole egg without sugar added were changed by fermentation, especially in the egg fermented with L. casei. Polyacrylamide gel electrophoretic patterns of pasteurized whole egg added 1% of sugar changed more significantly by fermentation than pasteurized whole egg without sugar added and the most significant change was observed in the bands of conalbumin, preconalbumin, globulin and postalbumin by S. faecalis.

  • For the efficient recovering of collapsed sloped soil, using a soil binder that can support the soil strongly and help the growth of plants is very important. The soil binder should also have functions of recovering the soil ecologically as well as be environmental friendly materials. In this research, optimum values of the water content and permeability and direct shear strength were searched by adding the water absorbent and coagulant into the soil binder. The polyacrylamide (PAM) with various anionic strength, super absorbent polymer (SAP) and cellulose ether (CE) were used as a soil binder, water absorbent and coagulant, respectively. Effects of the soil binder on the characteristics of soil were observed by changing the mixing ratio of PAM, SAP and CE. Experimental results showed that the soil binder increased the direct shear strength tens of times and the water content around two times, whereas decreased the water permeability. Also, the addition of CE to increase the coagulation of SAP increased more of the direct shear strength and water content.

  • The purpose of this study was to improve not only wet-end performances but also paper characteristics by the modification of various factors like molecular design and ionic characteristics of polyacrylamides First of all physical characteristics were observed after modify molecular design of the cationic polyacrylamides to linear, branched and cross-linked. In addition it was found analysis method to confirm branch degree of cationic polyacrylamides to combine ionic titration characteristics and spectroscopic behavior, After application of these structure modified polyacrylamides to the multiple retention systems with inorganic microparticles, it was found adjusting of branch degree of polyacrylamides was very important to optimize wet-end improvement. Second, After polymerization of amphoteric polyacrylamide to have both of cationic and anionic functional group in the polymer, we observed not only physical characteristics but also wet-end improvement to apply recycled pulp and found that the improvement of solution stability to prevent hydrolysis and increase of ash retention dramatically to compare traditional cationic polyacrylamide retention aid, Finally, After polymerization of anionic polyacrylamide, we observed not only wet-end improvement but also paper characteristics to apply preflocculation of PCC and it was found the improvements of flocculation efficiency, retention, ash retention, optical properties of the paper and bursting strength to compare traditional preflocculant of cationic polyacrylamide.

  • To investigate on the biochemical characteristics of myofibrillar proteins between cold(pollack, salmon) and warm current fish (yellow corbina, shark), myofibrils and actomyosin were prepared, and their biological activities, effect of temperature on the myofibrillar ATPase activities and SDS-polyacrylamide gel electrophoretic patterns of myofibrils were compared. SDS-polyacrylamide gel electrophoretic analysis showed that electrophoretic patterns of myofibril vary from fish to fish. Difference in KCl concentration dependency of myofibrillar ATPase activities and ATPase activity- pH curve were found among fish species. Myofibrillar proteins from cold current fish showed higher specific activity at low temperature $(5^{\circ}C-10^{\circ}C)$ than those from warm current fish.
