Efficient Fertile Plant Regeneration from Protoplasts of Javanica Rice and Their Ploidy Determination by Flow Cytometry (Javanica 벼 원형질체로 부터 효율적인 식물체 재분화와 flow cytometry에 의한 ploidy 검정)

  • The Southeast Asian javanica rice variety Tinawen was investigated for efficient protoplast culture and plant regeneration from cell suspension-derived protoplasts using a feeder cell culture method. Feeder cells of both Lolium multiflorum and Oryza ridleyi, either alone, or in combination, were employed and plants were regenerated from protoplast-derived colonies on several plant regeneration media. Dehydration of protoplast-derived colonies was also investigated as a means of enhancing plant regeneration. In the presence of L. multiflorum or O. ridleyi feeder cells, the protoplast plating efficiency ranged from 0.09% to 1.48%, depending on the feeder cell type and the age of the cell suspension. L. multiflorum feeder cells induced approximately 6-fold higher plating efficiency compared with those of O. ridleyi. The plant regeneration frequencies were 19.3-31.7% with L. multiflorum, 13.0-18.0% with O. ridleyi and 18.0-22.0% with a mixture of both in various plant regeneration media when protoplast-derived colonies were dehydrated, while for the non-dehydrated colonies, the values were 2.0-7.0%, 3.0-5.0% and 0-4.0%, respectively. Flow cytometric analysis of 34 protoplast-derived plants showed that the majority of plants were diploids and only 2 plants were tetraploids. The plants which were transferred to glasshouse were fertile.

Breeding of Garden Chrysanthemum Cultivar 'Nuri Ball' (Dendranthema grandiflorum Ramat.) with White Color Petals and Semi-Decorative Type Characteristics (흰색 반겹꽃의 화단국화 '누리볼' 육성)

  • The new garden chrysanthemum (Dendranthema grandiflorum Ramat.) cultivar 'Nuri Ball' was developed at Yesan Chrysanthemum Experiment Station of Chungcheongnam-do Agricultural Research and Extension Services in 2011. 'Nuri Ball' was bred through a cross between the '02-145-01' line as the female parent with yellow flower color and '02-04-32' as the male plant with white flower color in 2004. Three years of adaptation trials were conducted from 2006 to 2009 under natural conditions. This study compared the external shape type with that of 'White Miri' and conducted ploidy and RAPD (Random amplified polymorphic DNA) marker analyses. These tests showed that 'Nuri Ball' cultivar has its own characteristics compared with the control 'White Miri'. 'Nuri Ball' was a shrub type variety with semi-double flowers of 4.0 cm in width with white petals. It could produce 1025.2 flowers per plant in autumn. Compared with the control 'White Miri', 'Nuri Ball' was similar in terms of shape and color of flowers, but was different in flower size and number. The natural flowering time of 'Nuri Ball' was late September. It had very vigorous growth and an early budding plant. 'Nuri Ball' was demonstrated to be a new cultivar based on ploidy test and RAPD analysis. 'Nuri Ball' is intended for use as a bed chrysanthemum and expected to contribute to farm incomes in landscaping.

Histopathological and DNA Content Analysis of a Dermal Sarcoma in the Soft-shelled Turtle Pelodiscus sinensis

  • A dermal sarcoma was found in a freshwater, soft-shelled turtle Pelodiscus sinensis. The neoplasm consisted of proliferating fibrous tissue and extended from the dermis. The overlying epidermis was hyperplastic and partially folded. The deeper dermis and hypodermis contained three large, discrete necrotic foci of -10 mm diameter. Numerous eosinophilic granule cells and macro phages surrounded the necrotic areas. A mixed population of cells with nuclear pleomorphism was observed between the papillary layers of vessels. This area also had regions of different histological structures: (l) regularly arranged, spindle-shaped cells with compact nuclei in a fine-fibrillar matrix; (2) haphazardly arranged cells ($\leq$ 23 11m diameter) with ovoid, highly hypertrophic, faintly stained nuclei; and (3) cells (3.6-5.8 11m diameter) with irregularly shaped nuclei and marginal condensed chromatin in a myxomatous matrix. Some mitotic figures, binucleate cells, and multinucleate giant cells of up to 50 11m in length were also found. Flow cytometry of propidium iodide-stained cells yielded different histograms for the normal skin and the skin (primarily epidermis) and fibrous dermis of the tumor, indicating DNA heterogeneity in the dermal portion of the tumor. The ploidy indices for the dermal cells were 1.91 and 0.78, as compared to normal cells.

  • A tetraploid cyprinid loach, Misgurnus anguillicaudatus was induced by applying heat shock. The efficiency of inductions changed with the time of initiation of treatment and the duration of treatment. The highest incidence of tetraploid was obtained at the treatment of $41^{\circ}C$ for 3 min, 28 min after fertilization. Ploidy level was estimated by kariological examination and flow cytometric analysis. The tetraploid cyprinid loach had 4n = 100 chromosomes, while the diploid had 2n = 50. The erythrocytic volume of the tetraploid was 1.65 times larger than that of the diploid. The DNA content of the tetraploid and diploid showed 6.547 pg/cell and 3.067 pg/cell, respectively.

  • 포유동물의 정자형성과정 (spermatogenesis) 은 유사분열과 감수분열이 동시에 일어나는 매우 복잡하지만, 효율적으로 생식세포를 증식, 분화시키는 시스템이다 정상적인 spermatogenesis가 일어나는 testis에서는 haploid (IN), diploid (2N), 그리고 tetraploid(4N)과 같은 핵형을 갖는 세포들이 일정한 비율로 존재한다. DNA flow cytometry(DNA FCM) 는 세포의 핵형(ploidy)을 신속·정확하게 측정하여, 1N, 2N 그리고 4N에 대한 비율을 예측할 수 있어서, 생식세포를 포함한 다양한 유형의 세포주기를 분석하는데 적용되어져 왔다. 세포주기 분석법 중 이와 같은 FCM이외에, flow cytometer와 static image cytometer를 결합시켜 새롭게 고안된 laser scanning cytometer (LSC)가 있다. 그리고, 이제까지 LSC를 사용한 spermatogenesis에 관한 연구에 대해서는 보고된 바가 없다. 본 실험은 설치류에 있어 각기 다른 발달단계에 있는 정상적인 정소세포를 분리하여 PI (propidium iodide) 로 DNA를 염색한 후, DNA함량을 LSC로 분석하였다. 이것을 FCM에 의한 정소세포의 DNA분석과 비교·검토하였으며, 이 방법을 정상적인 spermatogenesis 가 일어나지 않는 동물시스템에 적용시켰다. 생식세포를 소멸시키기 위해 항암제인 busulfan과 비타민 A를 결핍시켜 이것이 세포주기의 어떤 시점에서 어떻게 작용하여, 생식세포를 소멸시키는지 알아보았다. 위의 실험·분석결과로부터 LSC를 사용한 DNA함량과 핵형의 결정은 FCM과 동일한 수준의 정확성을 제시하였다. busulfan 또는 비타민 A의 결손은 살아있는 세포의 80% 이상이 2N의 핵형에 해당하는 G0/G1 기에 있는 것으로 나타났다. 그리고 1N:2N 및 4N:2N의 핵형비율의 변화를 가져왔다. 이러한 자극은 생식세포주기제어에 관여하며, 생식세포가 증폭하고 분화로 들어가는 단계를 차단, G0/G1 기에서 정체(arrest)되는 것으로 시사된다.

Chromosome numbers of eight taxa of Aconitum L. in Korea and their systematic significance (Ranunculaceae)

  • Various aneuploidy and polyploidy have been reported in the genus Aconitum L. (ca. 300 species worldwide, Ranunculaceae), and there is a demonstrated association between major lineage diversification and polyploidy. This study reports chromosome counts of eight Aconitum from Korea, including the first counts for A. japonicum Thunb. subsp. napiforme ($H. L{\acute{e}}v.$ & Vaniot) Kadota (2n = 32) and A. longecassidatum Nakai (2n = 16). The study also includes chromosome numbers for two taxa on the Critically Endangered species list in Korea. Among Korean native species, chromosome numbers in Aconitum subgenus Aconitum range from 2n = 16 to 2n = 64 with diverse levels of polyploidy (2x, 4x, and 8x), whereas Aconitum subg. Lycoctonum exhibits only diploids (2n = 16). Greater chromosome number diversity in subg. Aconitum than subg. Lycoctonum might explain higher species diversity within the former subgenus (more than 250 species worldwide). Investigating chromosome number diversity of Aconitum in a phylogenetic framework will be a critical step to understand species richness of the genus.

Hormone Use is Associated with Lymphovascular Invasion in Breast Cancer

  • Background: Risk of developing breast cancer increases with short breastfeeding and the use of hormones. The prognosis of breast cancer is better if the tumours are hormone receptor positive. Since breast feeding affects estrogen and progesterone receptors, we wanted to investigate how such reproductive factors as breastfeeding and the use of hormones interact with known prognostic markers and specific tumour characteristics in women with breast cancer. Materials and Methods: A total of 250 women treated for breast cancer from a larger cohort completed a questionnaire on breastfeeding, number and age at births and use of hormones. A logistic regression analysis was made to search for connections between known prognostic markers on the one hand (type of cancer, grade, tumor size, estrogen receptor and progesterone receptor, lymphovascular invasion and DNA-ploidy) and reproductive data, breastfeeding, and hormone use on the other. Results and Conclusions: Hormone use, but not breastfeeding, was significantly associated, also on multivariate analysis, with the prognostic variable lymphovascular invasion, connected to a worse prognosis. No other hormone use or breast feeding correlations with prognostic variables were found.

Microspore Division and Plant Regeneration from Shed Pollen Culture in Rice

  • An efficient system of rice microspore culture could contribute to the production of genetically modified rice. The microspores were isolated by mechanical or shed methods. The number of microspores per 100 anthers isolated at uninucleate stage was higher than (or similar to) those at binucleate stage in isolation method with pestle or spatular, but microspore divisions were not easily observed on both stages. On the other hand, pollen division in shed pollen culture was observed more frequently at uninuclear than at binuclear stage. Cold pretreatment at 1$0^{\circ}C$ for 10 days resulted in the best multicellular division to produce microcalli at 12.5% efficiency in shed microspores. Heat shock at 33$^{\circ}C$ for one hour before or after pollen shedding enhanced cell division and callus formation. Out of twelve green regenerants, two were haploids and ten were diploids based on the chromosome analysis of root tips. The size of stoma was 12$^{m}$ m in haploids and 15 ${\mu}{\textrm}{m}$ in diploids determined by scanning electron microscope (SEM).

  • The influence of triploidization on cell and nucleus size characteristics of the same tissues of erythrocyte, retina, kidney, hepatocyte and midgut epithelium in marine medaka, Oryzias dancena has been determined histologically. Induced triploid fish are produced by cold shock treatments. Likewise, the size of horizontal cell nucleus in inner nuclear layer of retina, ganglion cell nucleus in ganglion cell layer of retina, proximal tubule cell of kidney, hepatocytes and nuclear height of midgut epithelium all appear to be significantly larger than diploid (P<0.05). On the other hand, retina thickness is larger in diploid than induced triploid (P<0.05). Induced triploid shows low density of cell number. Results of this study suggest that same characteristics in the induced triploid exhibiting larger cells and nucleus sizes with fewer number of cells than the diploid can be useful criteria for the distinction between diploid and induced triploid, and also the ploidy level in marine medaka.

  • Meiosis is a specialized cell division, essential in most reproducing organisms to halve the number of chromosomes, thereby enabling the restoration of ploidy levels during fertilization. A key step in meiosis is homologous recombination, which promotes homologous pairing and generates crossovers (COs) to connect homologous chromosomes until their separation at anaphase I. These CO sites, seen cytologically as chiasmata, represent a reciprocal exchange of genetic information between two homologous non-sister chromatids. RAD51, the eukaryotic homolog of the bacterial RecA recombinase, plays a central role in homologous recombination (HR) in yeast and animals. Loss of RAD51 function causes lethality in the flowering plant, Arabidopsis thaliana, suggesting that RAD51 has a meiotic stage-specific function that is different from homologous pairing activity.
