Historical Record of Mushroom Research and Industry in Korea

  • Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.

Interspecific Relationships within the Fungal Genus Pleurotus by Isozyme Analysis (동위효소 분석에 의한 느타리속의 종간 유연관계)

  • Thirty six strains of Pleurotus spp., from world-wide nations, were examined for interspecific isozyme variation. A comparison of isozymes in mycelial extracts of the fungal genus Pleurotus was made by polyacrylamide gel isoelectric focusing. A total of one hundred and sixty six bands was resolved from six isozymes. A cluster analysis was done based on the zymograms for esterase, glucosephosphate isomerase, leucine aminopeptidase, malate dehydrogenase, peroxidase, and phosphoglucomutase. From the isozyme analysis, esterase showed higher degree of variability, while it was observed less variability for the enzymes such as glucosephosphate isomerase, malate dehydrogenase, and phosphoglucomutase. The species P. ostreatus, whose taxon is controversial, was discriminated from P. pulmonarius, while P. florida was classified as a distinct taxon. The clustering of P. sapidus and P. spodoleucus strains appeared to be more difficult. It was found that some strains were included to another cluster based on electrophoretic banding patterns. These results show that this lack of congruence among data sets may help explain the taxonomic difficulty within the genus Pleurotus. A dendrogram of genetic similarities was presented, and applications of isozyme data to the systematics of these commercially important fungi was discussed.

  • Nutritional characteristics and cultural conditions for trehalose synthesis and mycelial yield of Pleurotus sajor-caju and Pleurotus ostreatus 201 in submerged culture were investigated. The results were as follows: Among the carbon sources, glucose was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of glucose was 1%. Among the nitrogen sources, peptone was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of peptone was 0.05%. The optimal concentration of $KH_2PO_4$ and $MgSO_47H_2O$ for trehalose synthesis and mycelial yield was 0.1%,0.04% and 0.2%,0.04-0.08%, respective­ly. The optimal temperature and pH for trehalose synthesis were $25^{\circ}C$ and pH 5.5. but optimal temperature and pH for mycelial yield were $30^{\circ}C$ and pH 5.5. The maximum yield of trehalose was obtanined after 10 day cultivation.

Studies on Mycelial Growth and Artificial Cultivation of Pleurotus eryngii (큰느타리버섯(Pleurotus eryngii)의 균사배양 및 인공재배에 관한 연구)

  • This study was carried out to investigate the optimal mycelial growth and an artificial cultivation conditions of Pleurotus eryngii. The optimal medium for the mycelial growth and density was MYPA medium. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 6.0, respectively. The modified optimal medium composition were obtained to be soluble starch 3%(w/v), malt extract 0.25%(w/v), yeast extract 0.25%(w/v) and $CaCl_2{\cdot}2H_2O$ 0.05% (w/v). From the results of experiment on the nutritional requirements, the modified optimal medium was higher than MYP medium in mycelial production and growth yield (Yx/s) of Pleurotus eryngii. The optimal sawdust species of solid culture for the mycelial growth and density was Quercus spp. The optimal concentration of additives (rice bran and wheat bran) and moisture content for the mycelial growth were about 30%(v/v) and 70%(v/v), respectively. On the other hand, the optimal concentration of additives for the production of fruiting body was 20%(v/v) of rice bran.

Effect of functional material addition on the growth and physiological activity materials in Pleurotus ostreatus (기능성물질 첨가시 느타리버섯 생육 및 생리활성물질의 변화)

  • This expriment was carried out on culture media which affect growth, development and physiological effect to Pleurotus ostreatus, when aqueous ferrum, red pepper powder, leaves of ginkgo biloba were added to culture media. The yield of fresh mushroom per 850ml PP bottle in chunchu-neutari was highest by 144g in 34.9g aqueous ferrum added, ferrous content don't affected to yield of mushroom, but variety of Pleurotus ostreatus affect to yield. Mineral content of fruit body and culture media in Suhan-neutari was different to mineral kind, ferrous content of furit body was similar to 0.31~0.43mg/kg when ferrous content of media was increased in 0.89mg/kg to 10.8mg/kg. The more content of red pepper powder in culture media was high, the more capsaicin content in fruit body was high. transferred capsaicin was highest by 6 mg/100g in Suhan-neutari. Total phenolics content were higher in culture media which was added aqueous ferrum and red pepper powder, but correlation don't showed total phenolics content among antioxydant activity, reducing power, chelating capacity. Macrophage activation was higher in Suhan-neutari than other Pleurotus spp.

In Vitro Antagonistic Characteristics of Bacilli Isolates against Trichoderma spp. and Three Species of Mushrooms

  • Twenty isolates of Bacillus species obtained from livestock manure composts and cotton-waste composts were tested for their antagonistic effects in vitro against three green mold pathogens of mushrooms (Trichoderma harzianum, T. koningii, and T. viridescens). However, there exists a possibility Bacillus species may have antagonistic effects against mushrooms themselves, and thus the same 20 isolates were tested in vitro against three species of mushrooms (Flammulina velutipes, Lentinus edodes, and Pleurotus ostreatus). Of the 20 Bacillus species isolates tested, two inhibited mycelial growth of T. harzianum, seven that of T. koningii, and eight that of T. viridescens. Importantly, the bacterial isolates M27 and RM29 strongly inhibited mycelial growth of all the Trichoderma spp. isolates tested. The isolate M27 was subsequently identified as the most effective in inhibiting mycelial growth of all the Trichoderma species. Interesting results of the effect Bacillus isolates had upon the mushroom species followed. It was found that most Bacillus isolates except 5T33 at least somewhat inhibited mycelial growth of the three mushroom species or some of the mushrooms. Furhermore, the antagonistic effects of the bacterial isolates against the three species of mushrooms varied depending on the mushroom species, suggesting a role for mushroom type in the mechanism of inhibition. The bacterial isolates M27 and RM29 were identified as having the most antagonistic activity, inhibiting mycelial growth of all the Trichoderma spp. as well as mycelial growth of the three species of mushrooms. These results suggest that the bacterial isolates and their antagonistic effects on green mold pathogens should be further studied for their practical use for biological control of green mold in the growing room of the mushrooms.

Nuclear DNA inheritance of intraspecific somatic hybrids by mono-mono cross in Pleurotus ostreatus based on URP-PCR analysis (URP-PCR 분석에 의한 느타리 단핵 계통간 교잡주의 핵 DNA 유전)

  • The primary objective of the present study is the characterization of the hybrids of monokaryon- monokaryon (mono-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus species strains with 85 mono-mono intraspecific hybrids of 7 combinations between six Pleurotus ostreatus strains and one Pleurotus florida strain. In this study, the results of analysis on hybridization rate, nuclear DNA patterns, and colors and yields of fruit-bodies, are presented as follows. The crossability between mono-mono crossing ranges between 50 and 93.75%. The results of the analysis on the nuclear DNA patterns of 85 hybrid strains of mono-mono crosses share the nuclei of both parents, but their genetic similarities were predominated by either parent. The hybrid strain between P. florida and P. ostreatus showed patterns more similar to P. florida, while the hybrid strain between P. ostreatus and P. ostreatus either had patterns predominated by either parent strain. The fruiting body colors of the mono-mono crosses mostly had combined colors of both parents but showed the tendency of being more similar to that of either parent. 82% of the hybrid strain indicated similar fruiting body yields compared to both parent strains, while 0% was higher and 18% were lower than both parents. The present study was able to find out and suggest superior hybrid trains by identifying the nuclear DNA patterns of hybrids between Pleurotus species as well as the characteristics of their fruiting bodies. This study expects that the advantages of the mono-mono crossing are needs to be fully utilized in mushroom breeding and it is better to develop superior strains of Pleurotus species strains together with the mono-mono crossing.

Development and Application of Weonhyeong Strain-specific SCAR Marker in Pleurotus ostreatus (느타리 버섯에서 원형 품종 특이 SCAR marker 개발)

  • Weonhyeong is one of important commercial strains. It has good characteristics of bundle formation, grey colored pilei and high productivity. We previously reported grouping of 70 strains of Pleurotus ostreatus in which one group contained 35 strains including Weonhyeong. Four strains in that group showed same profiles implicating no variety distinction for mushroom cultivation. Now we developed a specific marker for identification of Weonhyeong. Sequence Characterized Amplified Region (SCAR) marker was developed from the RAPD amplicon. SCAR marker 'S-OPO5' produced only one band specific to 2183, 2240, 2595 and 2725 strains showing similar banding patterns to Weonhyeong in RAPD-PCR results. The sequence of 'S-OPO5' marker was unknown when compared with the data in the Genbank using BLASTN. BLASTX results indicated that the marker showed significant alignment with the protein sequences in Tricholoma bakamatsutake reverse transcriptase. The results indicate that this new SCAR marker ('S-OPO5') will be valuable to distinguish the Weonhyeong similar strains from Pleurotus spp.

Characterization of Mitochondrial Plasmids from Pleurotus spp. (Pleurotus속 균주들의 미토콘드리아 플라스미드 특성)

  • Plasmid DNAs were detected from the mitochondrial fraction of four strains of whiterot fungus, Pleurotus ostreatus. The size of the plasmids were 10.2 and 7.2 kb in strain NFFA 2, 10.2 kb in NFFA 4001, 11.2 kb in NFFA 4501, and 10.2 and 11.2 kb in KFCC 11635. The two strains,NFFA 2ml and NFFA 2m2, which are mutant derivatives of NFFA 2, did not contain any plasmids. The cleavage by proteinase K indicated that these plasmids have DNA ends associated with proteins. In digestion with proteinase K all the plasm ids remained resistant to lambda exonuclease which hydrolyzes DNA from 5' ends and were sensitive to exonuclease III which hydrolyzes DNA from 3' ends. This suggests that the plasmids are linear double-stranded DNA and the terminal proteins are covalently linked to 5' ends of plasm ids. In order to find relationship between these plasmids, hybridization of plasm ids by each separate plasmid DNA was done. The result indicated that the plasmids can be classified into at least 3 groups. Plasmids of group I were present in all the P ostreatus. More mitochondrial plasmids were detected in P cornucopiae. P ,florida, P pulmonarius, P sajor-caju, and P spodoleucus. The size of plasmids ranged between 7.2 kb and 14 kb. All the species except P cornucopiae contained plasmids of approximately 10 kb which hybridized with the 10.2 kb plasmid (group I) of P ostreatus NFFA 2.

Genetic Analysis of Cultivars in Pleurotus spp. of Korea by URP-PCR Polymorphism (URP-PCR 다형성에 의한 국내 느타리버섯 품종의 유전적 특성 분석)

  • Twelve URP primers were used to assess genetic characteristics of oyster mushroom including 59 Pleurotsu ostreatus cultivars, two of P. florida cultivars, one P. sajor-caju cultivars, one P. abalonus cultivar and two P. eryngii cultivars registered in Korea. Six URP primers produced PCR polymorphic bands within and between the Pleurotus species. Primer URP2F produced distinct cultivar specific PCR polymorphic bands that profiled to 15 cultivar types. PCR polymorphic bands amplified by URP2F, URP6R, URP4R and URP2R were used for UPGMA cluster analysis. Fifty nine cultivars of Pleurotus ostreatus are genetically clustered into 5 groups, showing genetic similarity over 70% among them and P. abalonus. P. eryngii and P. sajor-caju, were involved in outside groups.
