Laccase Production Using Pleurotus ostreatus 1804 Immobilized on PUF Cubes in Batch and Packed Bed Reactors: Influence of Culture Conditions

  • The feasibility of laccase production by immobilization of Pleurotus ostreatus 1804 on polyurethane foam (PUF) cubes with respect to media composition was studied in both batch and reactor systems. Enhanced laccase yield was evidenced due to immobilization. A relatively high maximum laccase activity of 312.6 U was observed with immobilized mycelia in shake flasks compared to the maximum laccase activity of free mycelia (272.2 U). It is evident from this study that the culture conditions studied, i.e. biomass level, pH, substrate concentration, yeast extract concentration, $Cu^{2+}$ concentration, and alcohol nature, showed significant influence on the laccase yield. Gel electrophoretic analysis showed the molecular weight of the laccase produced by immobilized P. ostreatus to be 66 kDa. The laccase yield was significantly higher and more rapid in the packed bed reactor than in the shake flask experiments. A maximum laccase yield of 392.9 U was observed within 144 h of the fermentation period with complete glucose depletion.

Pleurotus ostreatus 미토콘드리아의 7.2 kb 선상 플라스미드 염기서열 분석 (Nucleotide Sequence of 7.2 kb Mitochondrial Linear Plasmid DNA in Pleurotus ostreatus)

  • Pleurotus ostreatus에서는 10.2 kb와 7.2 kb의 미토콘드리아 선상 플라스미드 DNA가 존재함이 발견되었다. 이 플라스미드 DNA의 양쪽 5'말단에는 단백질이 공유 결합되어있다고 알려져 있다. 최근에 P.ostreatus의 10.2 kb 미토콘드리아 선상 플라스미드 DNA를 다른 곰팡이의 선상 플라스미드 DNA에 존재하는 open reading frame(ORF)와 비교 분석하여 Podospora anserina의 플라스미드 pAL2의 DNA polymerase 및 RNA polymerase 암호부위와 유사성이 있음이 확인되었다. 본 연구에서는 7.2 kb선상 DNA를 HindIII잘라서 얻은 2조각의 절편(4.7 kb, 2.3 kb)을 클로닝하고 염기 서열을 분석하였다. 클로닝된 7 kb 절편에는 3개의 ORF,즉 ORF1(2982 bp, 993 amino acids), ORF2(2703 bp, 900 amino acids), ORF3(771 bp, 256 amino acids)가 존재함을 확인하였다 또한, 이들의 ORF를 분석한 결과, DNA polymerase와 RNA polymerase 암호부위와 유사성 이 있음을 확인하였다.

Function of mORF1 Protein as a Terminal Recognition Factor for the Linear Mitochondrial Plasmid pMLP1 from Pleurotus ostreatus

  • The mitochondrial plasmid pMLP1 from a white-rot fungus, Pleurotus ostreatus, is a double-stranded DNA containing 381 bp terminal inverted repeat (TIR) whose 5'-ends are covalently bound by terminal proteins. The plasmid contains two major open reading frames (ORFs), encoding putative DNA and RNA polymerases, and a minor ORF encoding a small, highly basic protein. To identify the DNA binding activity that recognizes the TIR region of pMLP1, gel retardation assays were performed with mitochondrial extracts. A specific protein binding to a region between 123 and 248 nt within TIR was observed. We examined whether the gene product of mORF1 bindes to this region specifically. E. coli cell extract which contains an overproduced mORF1 protein formed a complex specific to the region between 123 and 248 nt. Inclusion of mORF1 protein in the specific complex formed between P. ostreatus mitochondrial extract and TIR was confirmed by a supershift assay using polyclonal antibodies against the mORF1 protein. Our result suggest that the product of mORF1 may function as a terminal region recognition factor (TRF), recognizing an internal region in TIR.

Development of a Novel Spawn (Block Spawn) of an Edible Mushroom, Pleurotus ostreatus, in Liquid Culture and its Cultivation Evaluation

  • Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.

느타리버섯 물 추출물의 항산화 및 항암 활성 효과 (Antioxidant and Anticancer Effects of Water Extract from Pleurotus ostreatus)

  • 버섯은 우리나라에서 식용으로 널리 이용되고 있는 식품으로 많은 연구들에서 다양한 생리활성 효과에 대한 연구가 활발하게 이루어지고 있다. 본 연구는 다양한 버섯 중에서 최근 많이 이용되고 있는 느타리버섯 추출물을 이용하여 항산화 활성 및 세포 독성 효과를 확인하고자 하였다. 연구결과, 느타리버섯의 총 폴리페놀 함량과 플라보노이드 함량은 각각 $30.2{\pm}0.7mg%$$20.4{\pm}0.6mg%$로 나타났다. 0.625, 1.25, 2.5, 5 mg/mL 농도에서의 DPPH radical 소거능은 0.625의 농도에서 $4.5{\pm}0.1%$, 1.25의 농도에서 $11.0{\pm}0.4%$, 2.5 mg/mL 농도에서 $23.2{\pm}0.1%$, 5.0 mg/mL 농도에서 $44.1{\pm}0.6%$의 결과를 보였다. 환원력은 5 mg/mL의 농도에서 $1.648{\pm}0.047$로 가장 높은 환원력을 보여주어 시료의 농도가 증가할수록 흡광도 값도 증가하여 높은 환원력을 나타내었다. 느타리버섯 물 추출물의 인간 정상 신장세포 HEK293을 이용한 정상세포에 대한 세포 독성을 측정한 결과에서는 느타리버섯 물 추출물 80.5%의 생존력을 나타내어 독성이 없는 것으로 보였다. 한편, 이들 버섯 추출물의 암세포 성장 억제에 미치는 영향을 검색한 간암세포 생육 저해능의 결과는 최고 5 mg/mL의 농도에서 $70.0{\pm}5.3%$의 세포 생존력을 나타내어, 암세포 성장에는 영향을 미치지 못하는 것으로 사료된다.

Pleurotus ostreatus에서 분리한 아스콜빈산 산화효소의 특성 (Properties of Ascorbate-Oxidizing Enzyme Purified from Pleurotus ostreatus)

  • Pleuratus ostreatus로부터 ascorbate oxidizing enzyme을 황산암모늄 침전, preparative polyacrylamide gel 전기영동, DEAE Sepharose CL-6B 이온교환크로마토그라피, Sephadex G-150 gel 여과크로마토그라피의 단계를 거쳐 순수분리 하였다. 이 효소의 분자량은 gel 여과크로마토그라피에 의하여 140,000 정도로, 효소의 소단위 분자량은 SDS-polyacrylamide gel 전기영동에 의하여 66,000 정도로 추정되었다. Isoelectric focusing에 의하여 이 효소는 6.0의 등전점을 갖는 것으로 밝혀졌고, 최적반응온도는 $85^{\circ}C$ 정도, 최적반응 pH는 5.2 정도인 것으로 나타났다. 본 효소는 L-ascorbic acid와 D-isoascorbic acid에 대하여 동일한 친화도를 갖는 것으로 보이며, Km값은 두가지 기질에 대해 모두 2.2µM 이였다.

느타리버섯 생장에 미치는 광 감응성 작용 스펙트럼 (An Action Spectrum for Light-induced Growth rate in Pleurotus ostreatus)

  • The action spectrum for light-induced growth of fruit body in Pleurotus ostreatus has been studied by irradiation at various wavelengths. Effective wavelengths were distributed from near ultraviolet to blue region of spectrum. The most effect of light was observed in the region between 340 to 520 nanometers. The growth weigth obtained from which has been irradiated 144 hours in the region of 340-520 nm increases 74%, on the contrary the observed growth weight has been observed in the longer wavelengths than 620 nm. The pileus size was promoted 30$\times$32% in the region of 340-500 nm, and retarded 22$\times$19% in the dark. the strip length was rearded 12% in the region of 340-500 nm, and promoted 38% in the dark. The color thickness for pileus is getting light in longer wavelengths.

느타리버섯 병재배의 톱밥 대체 배지 재료 선발 (Selection of an Substitute Sawdust Material in Pleurotus ostreatus by Bottle Cultivation)

  • 느타리버섯의 병재배를 위한 톱밥 대체재료를 선발하기 위해 팽화왕겨, 코코피트, 콘코브, 코코넛나무톱밥을 비트펄프와 면실박에 혼합하여 실험한 결과 화학적 특성은 4가지 배지재료 모두 느타리버섯 재배에 적합하였고, 균사배양 및 재배기간도 지연되지 않았으나, 코코피트 혼합배지에서 수량이 133 g/850ml으로 미송톱밥과 대등하였고 생물학적 효율이 77%로 가장 높아 코코피트를 톱밥 대체 재료로 선발하였다.

The enzyme contained FMN as a prosthetic group, which was reduced by NADH and reoxidized by ascorbyl free radical.
