Effects of Acupuncture at the Acupoint Yongok$(KI_2)$, Umgok$(KI_10)$, Sobu$(HT_8)$. Sohae$(HT_3)$ on Aquaporin-2 expression, RSNA, MAP, HR, serum ANP, renin and norepinephrine in Rats (족소음신경(足少陰腎經) 연곡(然谷).음곡(陰谷)과 수소음심경(手少陰心經) 소부(少府).소해(少海) 침자(鍼刺)가 신장(腎臟) Aquaporin-2 발현(發現)과 신교감신경활성(腎交感神經活性), 혈압(血壓) 및 혈청(血淸) ANP, renin, norepinephrine에 미치는 영향(影響))

  • Objectives : This research was performed to investigate the effect of acupuncture at the $KI_2\;KI_{10}\;HT_8\;HT_3$, on Aquaporin-2(AQP2) expression related with the renal functions in rats. Methods : Acupuncture was performed during 100-seconds, 6-times, at 150-seconds intervals under anesthesia in rats. We observed rats' mean arterial pressure(MAP), heart rate(HR), renal sympathetic nerve activity(RSNA) during acupuncture and AQP2 expression by western blot method and atrial natriuretic peptide(ANP), renin, norepinephrine of plasma after decapitation. Results : The AQP2 expression was significantly increased in $HT_8$ group, but decreased in $KI_{10}$ group. Average MAP during 6-times acupuncture was significantly increased in $HT_8$ group. Average HR was significantly increased in $HT_8$ group, Average RSNA was increased in $KI_{10}$ group, but that was marginally increased in $KI_{10}$ group. Plasma renin concentration was increased in $KI_2,\;HT_3$, groups. Plasma ANP show a tendency to decrease in $KI_10\;HT_3$ groups, increased in $KI_2,\;HT_3$ but not significant. Plasma norepinephrine concentration was significantly decreased in $KI_{10},\;HT_3$ groups. Conclusions : These results suggested that acupuncture at $HT_8$ activate renal function to reuptake, but $KI_{10}$ show a decline on effect of AQP2 expression, blood pressure, nerve activity and renin.

Effects of Aqua-Acupuncture of Jibaikjihwangtang on the Blood Pressure in Hypertensive Rats (지백지황탕(知柏地黃湯) 약침(藥鍼)이 혈압(血壓)에 미치는 영향(影響))

  • Objective : This study was performed to investigate the effects of aqua-acupuncture of Jibaikjihwangtang in two-kidney one clip Goldblatt hypertensive rats and spontaneously hypertensive rats. Methods : we injected aqua-acupuncture solution into Shin-Soo ($BL_{23}$) which corresponds to human acupuncture point in two-kidney one clip Goldblatt hypertensive rats and spontaneously hypertensive rats. Systolic blood pressure, renin activity, aldosterone and atrial natriuretic peptide (ANP) plasma levels were tested. Results : Systolic blood pressure decreased significantly after aqua-acupuncture of jibaikjihwangtang. Acupuncture group in two-kidney one clip Goldblatt hyper-tensive rats had deference with control group. In plasma levels of atrial natriuretic peptide, acupuncture group of spontaneously hypertensive rats increased meaningfully but to two-kidney one clip Goldblatt hypertensive rats it was decreased meaningfully. In Serum Aldosterone density, the acupuncture group of spontaneously hypertensive rats had significant alteration than control group, but the acupuncture group of two-kidney one clip Goldblatt hypertensive rats had decreased alteration than control group. Conclusion : According to these results, after Aqua-Acupuncture of Jibaikjihwangtang blood pressure decreased significantly and data suggest that blood pressure reduction activity connected with renin activity reduction in renal hypertensive rat.

Effect of Atrial Natriuretic Factor on the Renal Function and Renin Release in Unanesthetized Rabbit (무마취 가토 신장기능에 미치는 Atrial Natriuretic Factor의 영향)

  • Since it has been suggested that atrial receptor may be involved in the mechanism of extracellular volume regulation, it was shown that the granularity of atrial cardiocytes can be changed by water and salt depletion, and that an extract of cardiac atrial tissue, when injected intravenously into anesthetized rats, was shown to cause a large and rapid increase in renal excretion of sodium. Various natriuretic peptides were isolated and synthetized, and the effects were investigated by many workers. Most studies, however, have been carried out under anesthesia and there have teen some controversies over direct effect of the factor on the renal function. Therefore, it was attempted in this study to access the effects of an atrial extract and a synthetic natriuretic factor in unanesthetized rabbits. Intrarenal arterial infusion of atrial extract caused a rapid increase of urinary volume and excretion of sodium. Glomerular filtration rate and renal plasma flow were both increased with no change in filtration fraction. The ventricular extract produced no change in urinary excretion of electrolytes, nor in renal hemodynamics. Intrarenal infusion of synthetic atrial natriuretic factor caused increases of renal excretory rate of sodium, chloride and potassium, and $FE_{Na}$. Glomerular filtration rate, renal plasma flow increased. And free water clearance also increased. Accentuated excretory function correlated well with increased glomerular filtration rate and renal plasma flow during infusion and for 10 minutes following the cessation of the infusion. Renin secretion rate decreased during constant infusion of atrial natriuretic factor. However, no correlation was found with the changes in glomerular filtration rate, renal plasma flow, or urinary excretion of sodium. These results suggest that atrial extract or atrial natriuretic factor induces changes in renal hemodynamics, as in excretion of electrolytes either indirectly through hemodynamic changes or directly by inhibiting tubular reabsorption. At the same time, renin secretory function is affected by the factor possibly through an unknown mechanism.

  • Objectives : Acupuncture has been used as treatment of disease in the korean medicine. In this study, it was investigated that effects of reduction of acupuncture techniques of five evolutive phase for appling excess in the heart, kidney on cardiovascular system as blood pressure and renin and atrial natriuretic peptide (ANP) in plasma, cardiac hypertrophy. Materials and methods : The experiments were performed on Sprague Dawley rats, 2K1C hypertension model was prepared by constricting the left renal artery with a sliver clip. Animals were then divided into seven groups, 2K1C induced and no treated group (Control), acupuncture on SP3 HT7(AC-1), LR1 KI1 (AC-2), SP3 HT7 LR1 KI1 (AC-3). The treatments were performed once a day for 10 days in rats. Results : The results are that the blood pressure was significantly decreased at 15days in AC-1 group. The cardiac hypertrophy was significantly decrease in AC-3 group. The activity of plasma ANP was increased in all groups without AC-1 group and the that of plasma Rein was decrease in AC-1, AC-2 groups than control group. Conclusions : These results suggest that acupuncture at SP3 HT7 and SP3 HT7 LR1 KI1 can be used as a therapy for controlling renal hypertension induced by 2K1C in the rats.

Effect of Sodium Intake on Responses of Blood Pressure, Renin-Aldosterone and Renal Excretions to Atrial Natriuretic Peptide in Spontaneously Hypertensive Rats (소금 섭취량을 달리한 정상 및 고혈압쥐에서 Atrial Natriuretic Peptide가 혈압, Renin-Aldosterone 및 신배설에 미치는 영향)

  • Effects of atrial natriuretic peptide (ANP) on blood pressure, plasma lenin activity, aldosterone and renal excretion were compared in conscious spontaneously hypertensive rats (SHR) and normotensive Wistar rats fed low, medium or high sodium diet (2, 10, 25 mmol NaCl/100g diet) for 6 weeks. ANP infusion (380 ng/kg/min for 20 min) produced reductions in blood pressure, plasma renin activity, and aldosterone level, but marked increases in hematocrit, urine flow, and excretions of sodium and potassium. The low sodium group showed a significantly enhanced aldosterone lowering effect of ANP than the high sodium group. However, three salt groups showed no difference in effects of ANP on blood pressure, plasma renin activity, hematocrit and diuresis. Natriuretic response to ANP was significantly greater in the high salt-than in the low sait-SHR, but was not different between the Wistar salt groups. There were strain differences in effects of ANP: SHR showed greater responses of blood pressure and natriuresis than Wistar rats. Above results indicate that aldosterone-lowering and natriuretic effects of ANP were modifed by different dietary sodium intakes. However, blood pressure- and renin-lowering, or diuretic effects of ANP were not affected by dietary sodium intakes. The mechanisms whereby dietary sodium intakes alter the effects of ANP in the pathogenesis of hypertension are not clear.

  • Acute cadmium exposure has been shown to increase sodium reabsorption in kidney through increase in aldosterone secretion in human and rodents. However, the antinatriuresis is not completely explained by hyperaldosteronism. Moreover, it is still controversial that the increase in plasma aldosterone concentration is mediated by the renin-angiotensin-aldosterone system(RAAS).(omitted)

  • To find out a convenient and reliable method of. detecting low renin status, we employed intravenous furosemide injection as a stimulatory maneuver. The results thus obtained were compared with those from the postural stimuli and basal plasma renin activity (PRA) in relation to sodium excretion. Intravenous furosemide test was performed in 66 control subjects and 44 patients with essential hypertension. The results were as follow; 1) Mean PRA in control subjects rose from $2.5{\pm}1.95$ ng/ml/hr (basal) to $4.5{\pm}2.51,\;5.2{\pm}2.49\;and\;4.2{\pm}2.44$ ng/ml/hr at 1, 2 and 3hrs after IV injection. One-hour response is more convenient in clinical practice. 2) Postural stimuli by assuming an upright posture for 3 hrs gave rise to considerable increase in PRA ($4.0{\pm}2.92\;from\;2.4{\pm}1.85$), but we found it less convenient than stimulation with furosemide. 3) The increase in PRA was much less marked in patients with essential hypertension as a whole ($2.9{\pm}2.75$). Hyporesponsiveness to furosemide stimuli was found in 34.1%. Of these hypo responders, a third had a normal basal PRA, indicating the need for this kind stimulatory procedure. 4) Younger age group showed greater renin responsiveness than older age group after furosemide stimuli. Likewise mean age of low renin patients ($52.9{\pm}5.38$ years old) was significantly higher than that of high and normal renin patients ($44.1{\pm}13.78$ years old).

Effect of Brain Angiotensin II Receptor Antagonists and Antisense Oligonucleotide on Drinking and Renal Renin in Rats

  • The physiological roles of brain angiotensin II in mediating water deprivation-induced drinking and in regulating renal renin release were assessed in male Sprague-Dawley rats. Specific $AT_1$ receptor antagonists, losartan and SK 1080, and antisense oligonucleotide (AS-ODN) directed to $AT_1$ receptor mRNA were intracerebroventricularly (i.c.v.) administered in conscious unrestrained rats. When water was given 20 min after i.c.v. injection of $AT_1$ receptor antagonists in 48-h water-deprived rats, losartan and SK 1080 produced approximatly 20% and 50% decrease in 1-h water intake, respectively. In contrast, i.c.v. treatment of the AS-ODN to $AT_1$ receptor mRNA for 24-h did not alter 1-h water intake in 24-h water-deprived rats, but prevented the increase in overnight water intake after 24-h water-deprivation. Six-day i.c.v. treatment of AS-ODN did not alter either the basal plasma renin concentration or renal cortical levels of renin and renin mRNA. The present results suggest that endogenous brain Ang II plays an important role in thirst and water intake through $AT_1$ receptors, but further studies are required to elucidate its regulatory role in renal renin synthesis.
