The Penetration Site of n-Alkanols into Synatosomal Plasma Membrane Vesicles Isolated from Bovine Cerebral Cortex (소의 대뇌피질로부터 분리된 Synaptosomal Plasma Membrane Vesicle에서의 n-Alkanols의 침투정도)

  • 세포막에서 마취제의 작용점을 규명하기 위하여, 마취제의 많은 부분을 차지하는 n-Alkanol을 이용하여, 소의 synaptosomal plasma membrane vesicles(SPMV)에서 n-Alkanol의 침투 정도를 형광 probe를 이용한 형광소광법을 통하여 검색하였다. n-Alkanols는 SPMV 외부 단층(outer monolayer)의 표면에 주로 분포하되 그 탄소수에 비례하여 소수성 부위에 분포되는 양이 증가되는 경향을 나타내었다(1-decanol은 제외). Methanol, Ethanol, 1-propanol, 1-butanol, 1-pentano, 1-hexanol, 1-heptanol, 1-octanol, 1-nonanol 및 1-decanil은 SPMV 외부 단층의 표면(친수성 부위)에 분포되는 것에 비하여 각각 949, 416.8, 214.8, 90.3, 53.7, 15.20, 6.80, 2.00, 1.03 및 2.40 배가 된다는 것을 확인하였다. 1-decanol은 $C_{10}$인데도 불구하고 $C_8$인 1-octanol에 비하여 적은 양이 소수성 부위에 침투 분포한다는 것이 확인되었다.

  • Mature spermatozoa of dark chub Zacco temmincki (Temminck and Schlegel), were examined under a scanning electron microscope (SEM) and a transmission electron microscope (TEM). The spermatozoa have a spherical, homogeneously electron-dense nucleus with an axial nuclear fossa containing two laterally oriented centrioles. The centrioles, which are arranged at about a $120^{\circ}$ angle to each other, have the 9+2 microtubule structure typical of flagella. The mature spermatozoon is of the primitive anacrosomal aquasperm type. The nuclear envelope is strongly undulated and contains nuclear vacuoles of different sizes and positions. The midpiece contains six or more mitochondria and encircles the basal body of the flagellum with an axoneme covered by the plasma membrane. Cytoplasmic vesicles lie between the axonemal doublets and the plasma membrane, and encircle the anterior part of the tail. The plasma membrane of the flagellum extends laterally and forms a pair of side fins. The species showed minor differences in number and structure of mitochondria, the angle between centrioles, and total length and occurrence of the fins. These characters, especially the side fins, appear to be apomorphic and useful for determining phylogenetic relationships at the genus or family level.

Analysis of Vasopressin-Induced $Ca^{2+}$ Increase in Rat Hepatocytes

  • To analyze vasopressin-induced $Ca^{2+}$ increase in liver cells, rat hepatocytes were isolated and attached to collagen-coated cover slips. Using fura-2, a $Ca^{2+}$-sensing dye, changes in intracellular $Ca^{2+}$ concentration by vasopressin were monitored. Results in this communication suggested that vasopressin-induced $Ca^{2+}$ increase were composed of both $Ca^{2+}$ release from internal $Ca^{2+}$ stores and influx from the plasma membrane. The $Ca^{2+}$ influx consisted of two distinguishable components. One was dependent on the presence of vasopressin and the other was not. SK&F96365 blocked vasopressin-induced $Ca^{2+}$ influx in a dose-dependent manner. Vasopressin-induced $Ca^{2+}$ release from internal stores diminished in a primary culture of hepatocytes according to the culture time. However, changes in vasopressin-induced $Ca^{2+}$ influx across the plasma membrane differed from changes in the $Ca^{2+}$ release from internal stores, suggesting two separate signalings from receptor activation to internal stores and to the plasma membrane.

Identification and Purification of a Normal Rat Liver Plasma Membrane Surface Protein which Disappears after Chemical Carcinogenesis

  • The electrophoretic patterns of plasma membrane surface proteins of normal rat liver cells and rat hepatomas were compared in 10% non-denaturing and 7-15% gradient non-denaturing gel. Chemical carcinogens, 2-Me DAB (2-methyl-4-dimethylaminoazobenzene) and DENA (diethylnitrosamine), were used to induce hepatoma in rats. One protein which disappeared in hepatoma was identified in normal rat liver by non-denaturing gel electrophoresis. Rabbit antisera were raised against this specific protein, and the protein was purified by Sephacryl S-200 column and immunoaffinity chromatography using the purified antibody. The purified protein showed two bands of molecular weights approximately 50 $kD_{\alpha}$ and 52 $kD_{\alpha}$ by SDS-polyacrylamide gel electrophoresis, which reacted specifically with the antibody. However only one band was observed in non-denaturing gel and also in isoelectric focusing with a pI value of 6.6. This study showed the existence of an unique protein on the plasma membrane surface of normal rat liver cells which disappeared in rat hepatomas induced by chemical carcinogens.

Identification and Characterization of Phytochrome-Regulated Phospholipase D in Oat Cells (Avena sativa L.)

  • The activation of phospholipase D (PLD) catalyzes hydrolysis of phosphatidylcholine (PC) to phosphatidic acid (PA) and choline in plants as well as animals. To determine the presence of PLD in oat cells, we prepared inside-out plasma membrane and cytosolic fractions from oat tissues. PLD activities in both cytosol and plasma membrane were detected by ion chromatography method. The activity of PLD in plasma membrane was dependent upon $Ca^{2+}$ concentration and was heat stable. To investigate whether G-protein couples to PLD, the effects of $GTP{\gamma}S$ and $GDP{\beta}S$ on the PLD activity were measured. PLD activity was dramatically increased 300~400% in the presence of 50 ${\mu}M$ $GTP{\gamma}S$ but not in the presence of 50 ${\mu}M$ $GDP{\beta}S$. These results indicate that G-protein may be involved in regulation of PLD activity. To identify whether PLD is regulated by red light receptor, phytochrome, we irradiated red, far-red, or red/far-red/red light on oat protoplasts. PLD activity has increased 5-fold and 3-fold by treatment with red light and red/far-red/red light, respectively. In contrast, irradiation with far-red light had little or no effect on PLD activity. These results suggest that phytochrome regulates PLD activity through activation of G-protein in oat cells.

  • Mutated forms of ras are found in many human tumors and the rate of incidence is significantly higher in colon and pancreatic cancers. The protein product from the ras oncogene is a small G-protein, $p21^{ras}{\;}(Ras)$ that is known to playa key role in the signal transduction cascade and cell differentiation and proliferation. Mutated Ras is unable to regulate itself and remains constantly activated, leading to uncontrolled cell growth. The function of Ras in signal transduction requires its location near the growth factor receptor at the cell membrane. However, Ras does not have a transmembrane domain. Ras requires farnesylation to increase its hydrophobicity and subsequent plasma membrane association for its transforming activity. This key post-translational modification is catalyzed by the enzyme Ras farnesyltransferase (FTase), which transfers a farnesyl group from farnesylpyrophosphate to the C-terminal cysteine of the Ras protein. The requirement has focused attention on FTase as a target for therapeutic intervention. Selective inhibition of FTase will prevent Ras protein from association with the plasma membrane, leading to a disruption of oncogenic Ras function.

  • Recently, it is necessary fir surface materials to be low cost and to keep well endurable and wear in machinery field. Since most good materials with endurance and wear are expensive, they are being studied hard to modify to surface materials with endurance and wear, which overlay with the surface membrane completely. One of them is PTA(Plasma Transferred Arc Overlaying Process) method, which gets into the spotlight. It is thought to be an optimum method, since this method of overlays properly materials with about 3∼5cm surface membrane. It is necessity for the modified materials with endurance and wear to improve within the agricultural environment. Therefore, this research was used the PTA method to endure and wear a material. This method proved to be a reliable method for overlaying the membrane on the materials of an economical agricultural machinery.

$Ca^{2+}$ Effect on Conversion of Exogenous 1-Aminocyclopropane-1-Carboxylic Acid to Ethylene in Vigna radiata Protoplasts

  • The possibility that 1-aminocyclopropane-1-carboxylic acid (ACC)-uptake may be dependent on the H+-gradient established across the plsma membrane was tested in protoplasts isolated from 2.5 day old mungbean hypocotyls. The ACC-induced ethylene production was inhibited when the H+-gradient was collapsed by the treatment with carbonycyamide-p-trifluro-methoxy-phenylhydrazone (FCCP). Moreover, the treatment with o-vanadate, a specific inhibitor of plasma membrane H+-ATPase, caused the inhibition of ethylene production. The ACC-induced ethylene production was inhibited by the treatemnt with verapamil (Ca2+-channel blocker), or ethylene glycol-bis($\beta$-aminoethyl ether) N, N, N', N'-tetraacetic acid (EGTA) (Ca2+-chelator). In contrast, the ehtylene production was stimulated by the application of A23187 (Ca2+ ionophore). The inhibitory effect of EGTA in the ethylene producton was magnified in the presence of A23187. From these results, we suggest that the external Ca2+ influx to the cytosol resulted in the stimulatin of ACC oxidase activity after ACC-uptake resulting from a H+-gradient across the plasma membrane.

환경적 스트레스에 의한 Helicobacter pylori의 형태 변화

  • Bacillary is the most common form of H. pylori observed during human infection. However, it is known that the morphology change of H. pylori from bacillary to coccoid can be occurred with a response to the environmental stresses such as the nutrient depletion, accumulation of toxic metabolites, pH alteration, and exposure to antimicrobial agents. The coccoid form of H. pylori, which is viable but non-culturable in vitro, seems to be the major cause of antibiotic resistancy and high reinfectability of H. pylori. In this regard, we studied the environmental factors that can induce the morphological change in vitro of H. pylori, and the change of fatty acid composition of plasma membrane. The morphological change from bacillary to coccoid could be observed with the depletion of nutrients, pH variation and reactive oxygen species added in the culture media. This morphologic conversion was paralleled by a dramatic decrease in unsaturated fatty acids and an increase in saturated fattv acids of plasma membrane. The change in composition of membrane fatty acid seems to be a kind of protection mechanism of H. pylori against these environmental stresses.

  • The effects of n-alkanols on the rotational relaxation time of 1, 6-dipheny-1, 3, 5-hexatriene (DPH) in synaptosomal plasma membrane vesicles isolate from fresh bovine cerbral contex were investigated. n-Alknols decreased the rotational relaxation time of 1, 6-diphenyl-1, 3, 5-hexatriene in the native membranes and the potencies of n-alkanols up to 1-nonanol increased by 1 order of magnitude as the carbon chain length increases by two carbon atoms, The cut-off phenomenon was reached at 1-decanol, where further increase in hydocabon length resulted in an increase in the rotational relaxation time of DPH in the native membranes.
